The Roles of CCR7 for the Homing of Memory CD8+ T Cells into Their Survival Niches

  • Memory CD8+ T cells in the immune system are responsible for the removal of external Ags for a long period of time to protect against re-infection. Naïve to memory CD8+ T cell differentiation and memory CD8+ T cell maintenance require many different factors including local environmental factors. Thus, it has been suggested that the migration of memory CD8+ T cells into specific microenvironments alters their longevity and functions. In this review, we have summarized the subsets of memory CD8+ T cells based on their migratory capacities and described the niche hypothesis for their survival. In addition, the basic roles of CCR7 in conjunction with the migration of memory CD8+ T cells and recent understandings of their survival niches have been introduced. Finally, the applications of altering CCR7 signaling have been discussed.

Momordica cochinchinensis Aril Extract Induced Apoptosis in Human MCF-7 Breast Cancer Cells

  • Momordica cochinchinensis Spreng (MC) has been used in traditional medicine due to its high carotenoid content. The objective of this study was to investigate mechanisms underlying apoptotic effects of MC on human MCF-7 breast cancer cells. A lycopene-enriched aril extract of MC (AE) showed cytotoxicity and antiestrogenicity to MCF-7 cells. On DAPI staining, AE induced cell shrinkage and chromatin condensation were evident. With flow cytometric analysis, AE increased the percentage of cells in an early apoptosis stage when compared with the control group. RT-PCR analysis showed AE to significantly increase the expression of the proapoptotic bax gene without effect on expression of the anti-apoptotic bcl-2 gene. Moreover, AE enhanced caspase 6, 8 and 9 activity. Taken together, we conclude that AE of MC fruit has anticancer effects on human MCF-7 breast cancer cells by induction of cell apoptosis via both intrinsic and extrinsic pathways of signaling.

MCF-7 인체 유방암 세포에서 백화사설초(白花蛇舌草), 산자고(山慈姑), 절패모(浙貝母)의 항암 효과 (Anti-cancer Effects of Oldenlandia diffusa, Cremastra appendiculata and Fritillaria thunbergii on MCF-7 Cells)

  • Oldenlandia diffusa, Cremastra appendiculata and Fritillaria thunbergii are widely distributed in the Korea, China and Japan, and has been used in traditional medicine for various diseases, such as pharyngolaryngitis, tonsillitis, goiter and stomach ulcer. However, the anti-cancer activities in human breast cancer have not been clearly elucidated yet. In this study, it was compared the in vitro cytotoxic effects of single and complex treatment of O. diffusa, C. appendiculata and F. thunbergii. We treat human breast cancer MCF-7 cells with O. diffusa, C. appendiculata and F. thunbergii. And we evaluated viability, growth inhibition, morphological changes, apoptotic bodies formation, measurement of the cell cycle and formation of DNA fragmentation of these cells. It was found that single treatment of O. diffusa could inhibit the cell proliferation in human breast cancer MCF-7 cells. However, complex treatment of O. diffusa, C. appendiculata and F. thunbergii is weakly or not affect the cell proliferation of MCF-7 cells. And anti-proliferative effects of O. diffusa in MCF-7 cells was associated with G1 arrest of cell cycle. These findings suggest that O. diffusa may be a potential chemotherapeutic agent for the control of human breast cancer cells and further studies will be needed to identify the molecular mechanisms.

간흡충에 감염된 흰쥐 담관 상피세포의 증식 양상 (Turnover of biliaiy epithelial cells in Clonorchis sinensis infected rats)

  • 간흡충에 감염된 담관은 확장되면서 상피세포의 수가 크게 증식하여 점막층이 두꺼워지고 과식증에 의해 점막의 단면이 유두상으로 관찰된다. Bromodeoxyuridine (BrdU)이 세포가 핵산을 복제할 때에 끼어 들어가는 성질을 이용하여, 이 연구에서는 담관 상피세포 중에서 합성기(Sphase)에 있는 세포의 분율을 파악하고자 하였다. 흰쥐에 간흡충의 피낭유충을 100 개씩 경구 감염시키고, 15주까지 간의 각 엽에 있는 담관의 조직절편을 만들어서 다세포군항체와 ABC complex를 이용한 염색을 실시하였다. 그 결과 충제가 들어 있는 담관에서는 감염 1주에 상피 세포의 2.9-10.2%가 염색에 양성이었고, 2주에 7.3-12.8%가, 5주에는 7.3-13.4%가, 15주에 8.4-14.8%가 BrdU를 함유하였다. 대조군 흰쥐나 실험군 동물의 감염되지 않은 담관의 경우에는 0-2.7%가 염색에 양성으로 나타났다. 염색된 상피세포는 주로 점막 상피세포층의 기저부에서 관찰되었다. 이 결과에 의하면 간흡충과 직접 접촉하는 담관의 상피세포가 뚜렷하게 증식하고 있어 충체의 어떠한 물리적인 또는 화학적인 자극이 있더라도 가까운 부위에서 주로 작용하고 있다고 판단하였다.

Antiestrogen, Trans-Tamoxifen Modulation of Human Breast Cancer Cell Growth

  • To gain further insight into how antiestrogens modulate cell function, the effects of antiestrogen on cell proliferation were studied in human breast cancer cells. We examined the effects of trans-tamoxifen on the proliferation of three human breast cancer cell lines that differed in their estrogen receptor contents. Trans-tamoxifen $(1{\mu}M)$ markedly inhibited the estrogen stimulated proliferation of MCF-7 human breast cancer cells that contained high levels of estrogen receptor $(1.15{\pm}0.03 pmole/mg protein)$ over that of control. In T47D cells that contained low levels of estrogen receptor $(0.23{\pm}0.05 pmole/mg protein)$, trans-tamoxifen $(1{\mu}M)$ showed minimal inhibition of estrogen stimulated cell proliferation over that of control. MDA-MB-231 cells, that contained no detectable levels of estrogen receptors, had their growth unaffected by trans-tamoxifen treatment. These results showed their sensitivity to growth inhibition by antiestrogen conrrelated well with their estrogen receptor content. Also we examined the effect of antiestrogen on cellular progestrone receptor level as well as plasminogen activator activity in MCF-7 cells. Trans-tamoxifen $(1{\mu}M)$ showed maximal inhibition of estrogen stimulated progestrone receptor level as well as plasminogen activator activity in MCF-7 cells that were stimulated by estrogen. It is not clear whether these inhibitions of progestrone receptor and plasminogen activator activity by estrogen are related to the antiestrogen inhibition of cell proliferation of MCF-7 cells. From the results of this study, it is clearly demonstrated that trans-tamoxifen is an antiestrogen in MCF-7 human breast cancer cells. Our data suggest that the biological effectiveness of trans-tamoxifen appear to result from its affinity of interaction with the estrogen receptor.

HT-29 장관세포에 있는 디펩티드수송체에 의한 디펩티드의 흡수 (Uptake of a Dipeptide by the Dipeptide Transporter in the HT-29 Intestinal Cells)

  • The peptide transporter can be utilized for improving the bioavailability of compounds that are poorly absorbed. Characterization of the dipeptide uptake into the human intestinal epithelial cells, HT-29 was investigated. The uptake of tritiated glycylsarcosine $([^3H]-Gly-Sar,\;0.1\;{\mu}Ci/ml)$ was measured in confluent or subconfluent HT-29, Caco-2, and Cos-7 cells. Uptake medium was the Dulbecco's Modified Eagle's Media (DMEM) adjusted to pH 6.0. Both HT-29 and Caco-2 cells expressed the dipeptide transporter significantly (p<0.005) but Cos-7 did not. Certain portions of passive uptake were observed in all three cell lines. Uptake of Gly-Sar was largest at 7 days after plating HT-29 cells with significant inhibition with 25 mM cold Gly-Sar (p<0.05). but expression ratio of the dipeptide transporter was 0.7, suggesting lower expression. The effect of pH on Gly-Sar uptake was not significant in the range of pH 6 to 8. Gly-Sar uptake was also inhibited with 50 mM carnosine, 25 mM Gly-Sar, and 35 mM cephalexin significantly (p<0.05). From above results the dipeptide transporter was expressed well in HT-29 cells and was similar to that in the small intestine, suggesting that large amounts of mRNA of the transporter from the cells can be obtained.

소 분할배와 호르몬, 난관상피세포, 난구세포와의 공배양이 체외발생율에 미치는 영향에 관한 연구 (Effects of Hormones, Oviduct Epithelial Cells, Cumulus Cells during the In Vitro Culture in Medium on In Vitro Developmental Rates of Bisected Bovine Embryos)

  • The study was conducted to investigate on in vitro developmental rates of bisected bovine embryos co-culture in 10% FCS+TCM-199 media containing hormones, oviductal epithelial cells and cumulus cells 0 to 7 days after bisection. In vitro developmental rates was defined as development rates on in vitro culture or FDA-test. The results are summarized as follows : 1. In vitro developmental rates of bisected bovine embryos co-cultured in 10% FCS+TCM-199 media containing PMSG+hCG, PMSG+$\beta$-estradiol, hCG+$\beta$-estradiol, PMSG, hCG 0 to 3 days and 4 to 7 days were 16.7~30.0% and 11.1~25.0%, respectively. In vitro developmental rates of bisected embryos co-cultured in 10% FCS+TCM-199 media containing hormones significantly higher than that of non co-culture. 2. In vitro developmental rates of bisected bovine embryos co-cultured 10% FCS+TCM-199 media containing oviductal epithelial cells 0 to 3 days and 4 to 7 days were 25.0% and 22.2%, respectively.

향신료 분말의 Esdcherichia coli 와 Staphylococcus aureus 에 대한 항균작용 (Antibacterial Activity of Powdered Spice against Escherichia coli and Staphylococcus aureus)

  • Antibacterial activities of powdered spices(garlic , ginger, cinnamon and clove) against pathogenic Escherichia coli )157:H7 and Staphyloccus auresus were investigated. Spice powder was added in was exponetial phase of each bacterial culture . Growth inhibition was determined by the absorbance at 660nm and morphological changes of the cells were observed by transmission electron microscope (TEM). Ginger powder has the highest antibacterial activity, following cinnamon , clove and garlic has the least activity.Growth of Escherichia coli O157:H7 and Staphyloccus aureus were completely inhibited within 5 hours after addition of 1 % of garlic , 0.3% of ginger or cinnamon , 0.5% of clove powder on the exponential phase of the cells. Spice untreated cells of E. coli and S. aureus, the cytoplasm was entirely surrounded by rigid cell wall and cell walls formed a smooth layer well attached to the plasma membrane. In the cells of E. coli and S. aureus treated with spice powder, cell wall and plasma membrane were lysed and severely damaged. E.coli cells growth in the presence of spice powder showed plammolysis, the loss of electron dense material, the formation of extra cellular blebs and cytoplasm burst out from the cell. S .sureus cells grown in the presence of spice powder showed swell of cell wall, the loss of electron dense material , coagulation of cell cytoplasm and formation of extra cellular blebs. Severely damaged cells of S. aureus lost whole cytoplasm and left as ghost of the cell. Spice powder stimulated autolyssi and induced cell death.

  • 본 연구는 대식세포에서 LPS를 이용하여 염증 유사 세포 모델을 만들고 염증 유사 대식세포 모델에서 성체줄기세포의 면역 조절 능력을 평가하였다. LPS 자극에 의해 증가된 IL-1β, TNF-α 및 IL-10의 생성은 성체줄기세포를 공배양한 실험군 뿐만 아니라 성체줄기세포를 배양한 상층 배양액을 처리한 실험군에서도 동일한 효과를 나타내었으며, 또한 성체줄기세포 유래 엑소좀을 염증 유사 대식세포 모델에 처리하여 유사한 결과를 관찰하였다. 이 결과는 성체줄기세포 자체의 염증 억제 기능보다는 성체줄기세포 유래 엑소좀을 포함하여 성체줄기세포가 분비하는 bioactive molecules에 의해 세포 간 신호 전달이 이루어지고 있음을 의미하며, 이러한 엑소좀은 염증 관련 질환 분야에 치료적 적용이 가능하고 또한 새로운 염증 치료제 개발의 툴로 사용될 수 있음을 시사한다.

바이칼 호 Sponge에 서식하는 세균 군집 구조의 분석 (Community Analysis of the Bacteria in Sponges of Lake Baikal by FISH Method)

  • 바이칼호에 서식하는 있는2속(genus Baikalospongia와 Lubomirskia)(5.13)의 해면 내에 서식하고 있는 세균군집 구조를 조사하기 위하여 fluorescent in situ hybridization (FISH) 방법을 적용하였다. Baikalospongia와 Lubomirskia 속에 속하는 해면에 서식하는 총세균수는 각각 $7.2{\times}10^{7}-4.2{\times}10^{8}\;cells/ml$$1.2{\times}10^{8}-1.6{\times}10^{8}\;cells/ml$였다. 인근 호숫물의 총세균수는 $2.3{\times}7.7{\times}10^{5}\;cells/ml$ 범위로 해면에 서식하는 총세균수가 약 $10^{3}-10^{4}$배 더 높았다. 총세균수에 대한 세균군집 구조의 비율은 ${\alpha}-,\;{\beta}-,\;{\gamma}-Proteobacteria\;group$과 Cytophaga-Flavobacterium group외 다른 세균군이 genus Baikalospongia는 42.0-60.3%, Lubomirskia에서는 40.7-51.9%로, 호수물에서는 22.6-46.3%와 같이 나타났다. 이는 일반 담수에서 특정 군집이 우점하는 경향과는 다른 것으로써, 바이칼의 호숫물과 해면에 있는 세균들은 독특한 군집구조를 이루고 있음을 확인하였다.
