현재 식중독 미생물의 검사에 전통적인 배지법이 다수를 차지하고 있으나 PCR이나 면역 분석법과 같은 신속 검출 법들의 사용이 증가하는 추세이다. 그러나 대개 speciesspecific 프라이머를 이용한 PCR에 의한 DNA 증폭산물을 전기영동을 통해 확인하는 방법을 사용하고 있는데 이는 각종 균주에 대해 각기 다른 프라이머를 사용하여야하는 단점이 있다. 이러한 단점을 보완하기 위하여 본 연구에서 는 여러 가지 균주를 동시에 신속하게 검지할 수 있도록 PCR-DGGE 기술을 연구하게 되었다. 그 결과 6종(S. typhimurium, P. fluorescens, B. cereus, S. aureus, E. coli, L. monoytogenes)의 유해 미생물을 선정하였고 DGGE 상에서 확실한 분리능을 보여 유해미생물의 인공마커로 사용할 수 있음을 보였다. 또한 실제 PCR-DGGE 기법을 사용하기 위하여 채소에서의 유해미생물 검출 감도를 확인한 결과 B. cereus를 제외한 5종(S. typhimurium, P. fluorescens, S. aureus, E. coli, L. monoytogenes)의 균들은 모두 $10^1$ CFU/g까지 검출할 수 있었고 B. cereus는 $10^3$ CFU/g이상 채소에 존재할 때 검출할 수 있었다. 또한 균질화 방법에 따라서 식물 DNA와 유해 미생물 간의 경쟁적 증폭현상이 일어남도 확인할 수 있었다. 이로써 본 연구에서는 유해 미생물의 검지, 인공마커의 제조, 식물 DNA와 유해미생물간의 경쟁적 증폭 현상 방지책과 유해미생물의 검출 감도 확인 및 염기서열을 통한 동정을 통해 PCR-DGGE가 식품에서 유해 미생물의 신속 검지 방법으로써 활용할 수 있을 것으로 사료된다. 향후 국내 식품소비 패턴이 유기농 채소의 수요와 공급이 날로 증대 되고 있기 때문에 PCR-DGGE 기법이 유기농 채소들의 식중독 균에 대한 database를 구축하여 안전성을 확보하는데 기여할 수 있다고 생각된다.

To understand the situation of water contamination with Cryptosporidium spp. and Giardia spp. in the northern region of Portugal, we have established a long-term program aimed at pinpointing the sources of surface water and environmental contamination, working with the water-supply industry. Here, we describe the results obtained with raw water samples collected in rivers of the 5 hydrographical basins. A total of 283 samples were analyzed using the Method 1623 EPA, USA. Genetic characterization was performed by PCR and sequencing of genes 18S rRNA of Cryptosporidium spp. and $\beta$-giardin of Giardia spp. Infectious stages of the protozoa were detected in 72.8% (206 of 283) of the water samples, with 15.2% (43 of 283) positive for Giardia duodenalis cysts, 9.5% (27 of 283) positive for Cryptosporidium spp. oocysts, and 48.1% (136 of 283) samples positive for both parasites. The most common zoonotic species found were G. duodenalis assemblages A-I, A-II, B, and E genotypes, and Cryptosporidium parvum, Cryptosporidium andersoni, Cryptosporidium hominis, and Cryptosporidium muris. These results suggest that cryptosporidiosis and giardiasis are important public health issues in northern Portugal. To the authors' knowledge, this is the first report evaluating the concentration of environmental stages of Cryptosporidium and Giardia in raw water samples in the northern region of Portugal.
Lipase-producing bacterial strains were isolated from Antarctic soil samples using the tricaprylin agar plate method. Seven strains with relatively strong lipase activities were selected. All of them turned out to be Bacillus pumilus strains by the 16S rRNA gene sequence analysis. Their corresponding lipase genes were cloned, sequenced, and compared. Finally, three different Bacillus pumilus lipases (BPL1, BPL2, and BPL3) were chosen. Their amino acid sequence identities were in the range of 92-98% with the previous Bacillus pumilus lipases. Their optimum temperatures and pHs were measured to be $40^{\circ}C$ and pH 9. Lipase BPL1 and lipase BPL2 were stable up to $30^{\circ}C$, whereas lipase BPL3 was stable up to $20^{\circ}C$. Lipase BPL2 was stable within a pH range of 6-10, whereas lipase BPL1 and lipase BPL3 were stable within a pH range of 5-11, showing strong alkaline tolerance. All these lipases exhibited high hydrolytic activity toward p-nitrophenyl caprylate ($C_8$). In addition, lipase BPL1 showed high hydrolytic activity toward tributyrin, whereas lipase BPL2 and lipase BPL3 hydrolyzed tricaprylin and castor oil preferentially. These results demonstrated that the three Antarctic Bacillus lipases were alkaliphilic and had a substrate preference toward short- and medium-chain triglycerides. These Antarctic Bacillus lipases might be used in detergent and food industries.
Indigo is utilized in various industries including textile dyeing, cosmetics, printing and medicinal products and its reduced form, leuco-indigo, is mainly used in these process. Chemical reducing agent (sodium dithionite, sodium sulfide, etc.) is preferred to use for the formation of leucoindigo in industry. In traditional indigo fermentation process, microorganisms can participate in the reduction of indigo and thus it has been known to reduce environmental pollution and noxious byproducts. However, in fermentation method using microorganisms it is difficult to standardize large scale production process due to low yield and reproducibility. In this study, we attempted to develop the indigo reduction process using microbial flora which was isolated from naturally fermented indigo vat or deduced by metagenomic approach. From the results of library analyses of PCR-amplified 16S rRNA genes from the traditional indigo fermentation vat sample (metagenome), it was confirmed that Alkalibacteriums (71%) was distinctly dominant in population. Some strains were identified after confirming that they become pure culture in nutrient media modified slightly. Four strains were separated in this process and each strain showed obvious reducing ability toward indigo in dyeing test. It is expected that the analyzed results will provide important data for standardizing the natural fermentation of indigo and investigating the mechanism of indigo reduction.

Otitis externa (OE) is a frequent disease in the ear canals of dogs. To identify the pathogens causing OE in dogs and to determine their antimicrobial resistances, specimens were collected from animal hospitals in Daejeon. The isolates were examined by morphological and biochemical tests, 16S rRNA analysis and antimicrobial susceptibility tests. We analyzed correlation between the isolated pathogens and external factors of dogs such as breed, age, gender, ear mite, hair in ears and experience with antibiotic therapy. Thirty three strains of bacteria were isolated from 26 of the 68 heads of dogs with OE. The most isolated bacteria were Enterococcus faecalis (E. faecalis) followed by Staphylococcus aureus (Sta. aureus), Sta. pseudointermedius, E. faecium, E. avium and Streptococcus canis (Strep. canis) in order of frequency of occurrence. Isolation frequency of Enterococcus spp. and Staphylococcus spp. were 51.5% and 45.5%, respectively. E. faecalis and E. faecium isolates showed VanB phenotype, which is resistant to vancomycin but sensitive to teicoplanin were 58% and 25%, respectively. Nine isolates among total twelve isolates of E. faecalis were isolated from the dogs treated with antibiotics. There was no methicillin-resistant Sta. aureus (MRSA), but were MR-Sta. pseudointermedius (MRSP) (57.1%) and vancomycin-resistant (VR)-Sta. pseudointermedius (14.3%) (VRSP) showing VanB phenotype. However, vanA, vanB and vanC genes were not detected in VR isolates from the dogs. Taken together, VR-Enterococcus spp. (VRE) is one of the major pathogens in domestic animals, as well as community-and hospital-acquired infection.

The present work aims to use a biofilter technology(aerated submerged filters) for the aerobic transformation at laboratory-scale of olive washing water(OWW) generated in the first steps of olive oil processing, as well as the genetic profiling and identification to the species level of the bacteria involved in the formation of the biofilm, by means of TGGE. Chemical parameters, such as biological oxygen demand at five days($BOD_5$) and chemical oxygen demand(COD), decreased markedly(up to 90 and 85%, respectively) by the biological treatment, and the efficiency of the process was significantly affected by aeration and inlet flow rates. The total polyphenol content of inlet OWW was only moderately reduced(around 50% decrease of the inlet content) after the biofilter treatment, under the conditions tested. Partial 16S rRNA genes were amplified using total DNA extracted from the biofilm and separated by TGGE. Sequences of isolated bands were mostly affiliated to the $\alpha-subclass$ of Proteobacteria, and often branched in the periphery of bacteria] genera commonly present in soil(Rhizobium, Reichenowia, Agrobacterium, and Sphingomonas). The data obtained by the experimentation at laboratory scale provided results that support the suitability of the submerged filter technology for the treatment of olive washing waters with the purpose of its reutilization.
Theileria (T.) buffeli (formerly T. sergenti/T. orientalis) is the major hemo-protozoan distributed in the Far East Asian countries such as Korea, China and Japan. It is responsible for the clinical symptoms of anorexia, ateliosis, anemia, fever and icterus. It also causes abortion and sudden death under severe cases, resulting in economic losses for many livestock farms. The objective of this study was to analyze the genetic diversity of the major surface protein (Msp) gene in T. buffeli in Holstein in Korea, and we characterized the association of the diversification of the Msp gene and its relationship with the pathogenicity of Theileria. For this, complete blood counts and Theileria PCR sequence analysis were performed from 57 Holstein in Jeju Island. A total of 26 PCR positive Holstein (16 anemic and 10 non-anemic) were then randomly selected based on 18s rRNA sequence typing of the Theileria Msp gene. The DNA sequence of the T. buffeli Msp gene in Holstein showed 99.0%, 99.2%, 99.9%, 99.5%, 98.7%, 98.4% and 98.4% homology with T. sergenti, Theileria spp., T. sergenti, Theileria spp., Theileria spp., Theileria spp. and Theileria spp., respectively. The result showed a genetic variation of 57.7% (type I), 3.8% (type II), 15.4% (type III), 7.7% (type IV), 13.5% (type V) and 1.9% (type VI). Type I is the most frequent type in both anemic and non-anemic Holstein while type II was found in only non-anemic Holstein. This results of our study help confirm the diversity of Msp gene types and demonstrate that the gene type distribution of Msp genes varies among Theileria-infected Holstein in Jeju Island.
Nucleolus Organizer Regions(NORs)는 핵인을 형성하는 염색체의 특정 부위로서 rRNA 합성에 관여하는 리보좀 유전자를 함유하고 있으며 이의 활성화가 일어나는 곳이다. 본 연구에서는 한우의 NORs의 양상을 제시하기 위하여 AgNOR 염색법을 이용하여 한우의 NORs 수와 NORs 염색체 및 이의 분포 위치 등을 구명하고 한편으로 소의 품종별, 성별 및 근원이 다른 세포간의 NORs 양상을 비교 분석하였다. 본 분석에 이용된 공시축은 한우 및 홀스타인 암수 44두로서 이들의 혈액배양으로부터 염색체를 분리하였다. 조직간 세포의 NORs 비교를 위하여 귀 조직으로부터 배양된 섬유아세포의 염색체와 백혈구 배양으로부터 분리된 염색체를 분석하였다. 분리된 중기상에 AgNOR 염색 후 G-banding을 한 결과 한우의 NORs는 2번, 3번, 4번, 11번 및 28번 염색체에 존재하고, 이들의 분포 위치는 각 염색체의 말단부에 위치한다. 한우 NORs 수는 세포에 따라 최소 2개에서부터 최대 10개까지 나타나며 평균 5.6개였다. 이러한 다형적 양상은 개체 간뿐만 아니라 동일 개체 내 세포 간에서도 달리 나타나며 발현의 크기 또한 차이가 있다. 품종 간 NORs의 비교 분석에서 한우의 NORs 수가 Holstein (5.4개)에 비해 유의적으로 높게 나타났으며, 근원이 다른 세포간 비교에서는 fibroblasts (5.9개)에서의 NORs 수가 lymphocytes (5.5개)에 비해 높은 빈도를 보였고, 성 간에는 수컷 (5.7개)이 암컷 (5.4개)에 비하여 많은 NORs 수를 나타내었다. 그러나 이들의 염색체상 분포 양상에서는 공히 동일한 염색체에 출현되었으며 염색체 상 출현 빈도에서도 세포들 간에 거의 차이가 없었다.
Aquaporins(AQPs)유전자는 다양한 조직의 상피세포와 내피세포에 존재하며 다량의 물 이동을 조절하는 막성 단백질로서, 세포간 또는 세포막 사이의 물 이동에 중요한 역할을 하는 것으로 알려져 있다. 발정기의 생쥐 자궁에서는 자궁내막세포의 증식과 함께 수화되는 특징을 보이며, 자궁내강으로 물이 이동되어 자궁내액의 점성이 낮아지는 현상이 나타난다. 따라서, 본 연구에서는 생쥐의 발정주기 동안 자궁에서의 형태학적인 변화와 관련하여 AQP유전자가 물 이동의 매개체로서 중요한 역할을 할 것으로 추측하여, 생쥐 발정주기 동안에 AQP유전자의 발현 양상을 역전사 중합효소 연쇄반응을 통하여 관찰하였다. 또한, laser capture microdissection(LCf을 이용하여 자궁내 세포의 종류에 따른 AQP유전자의 발현 양상을 조사하였다. AQP-4 전령체는 발정주기의 proestrus와 estrus 시기에 발현량이 유의하게 증가하는 반면, AQP-8 전령체는 동일한 시기에 유의하게 감소하는 것으로 관찰되었다. 또한, LCM 기법을 통해서 AQP-4와 -8 전령체가 자궁기질세포보다 자궁내막세포에서 강하게 발현이 유도됨을 관찰하였다. 결과적으로, 생쥐 자궁에서 AQP-4와 -8유전자가 발정주기에 따라 발현량이 변화하는 것으로 보아 난소내 호르몬인 estrogen과 progesterone에 의해 조절될 가능성이 있으며, proestrus와 estrus 시기에 자궁내강으로 자궁내액을 수송하는데 중요한 역할을 할 것으로 사료된다.
제주지역의 감자 줄기검은병징으로부터 분리한 12균주의 유전적 특성을 분석하기 위해 Eca-specific PCR, PCR-RFLP, ERIC-PCR을 실시하여 그 결과를 E. carotovora대조균들과 비교하였다. Eca-specific PCR 결과 Eca 대조균들은 특이적 밴드를 형성한 반면, 줄기검은병균은 특이적 밴드를 형성하지 않았다. 또한, pel유전자의 RFLP분석 결과 줄기검은병균은 pattern 2를 나타내었으나, Eca 균주는 pattern 3을 나타내어 Eca와는 다른 특성을 보여주었다. 16S rDNA의 RFLP분석결과 이번 실험에 이용된 대부분의 균주가 pattern 1을 나타냈지만, 12개의 줄기검은병균 중 11개의 균이 pattern 2을 나타내어 Ecc와도 다른 특성을 보여주었다. 제주지역의 무름병징과 줄기검은병징을 나타내는 균주들의 유전적 관계를 분석하기 위해 ERIC-PCR을 실시한 결과 줄기검은병균들은 특이적 밴드를 형성하였으며, 서로 높은 유연관계를 보여주었다. 따라서 제주지역의 줄기검은병징으로부터 분리한 균주들은 Eca, Ecc균들과는 다른 특성을 가지고 있음을 알 수 있었다.
