수성이상계에서 K.fragilis의 에탄올 추출발효 특성에 관한 연구 (Extractive Ethanol Fermentation Characteristics of K.fragilis in an Aqueous Two Phase System)

  • 여러 가지 조성의 PEG 20000jDx로 구성한 수성 이상계를 사용한 에탄올 추출발효실험을 통하여 K. fragilis에 의한 돼지감자 주스의 발효 특성을 초기 당농도 및 생성 알코올 농도의 함수로 규명하여 다음의 결과를 얻었다. 균체의 비증식속도 빛 에탄올 비생성속도는 PEG 및 Dx 농도가 낮을수록 증가하였다. 비증식속도는 조성에 무관하게 초기 당농도 $80g/\ell$에서 최대치를 보였고, 에탄올 비생성속도는 $120g/\ell$최대치를 보였다. 생성 알코올 농도에 의한 저해 영향은 비증식속도에 대하여 지수함수형 을, 에탄올 비생성속도에 대하여 쌍곡선함수형을 나타냈다. Dx 농도가 2.5~5% 범위에셔 PEG 농도 가 낮을수록 에탄올 저해 영향은 감소하였다. 총괄 에탄올 생산성은 초기 당농도가 증가할수록 높았으 며 생산성이 최대치에 도랄하는 소요시간도 당농도가 증가함에 따라 증가하였다. 시간별 에탄올 생산 속도는 초기 당농도 $160g/\ell$ 에서 가장 높은 값을 보 였다. 총괄 에탄올 생산성은 대조실험에 비하여 수 성이상계 추출발효를 통하여 1.2배 이상 증가하였고 초기 당농도가 높을수록 이상계 추출발효가 더욱 유리한 것으로 나타났다.

ELISA를 이용한 IL-5 분비조절 한약물 Screening (Interleukin-5 Inhibition Assay of the Oriental Materia Medica Treatment by Sandwich ELISA on Mouse Splenocytes)

  • Background and Objective : Allergy is defined as an altered reactivity to an antigen, which can result in pathologic reactions upon subsequent exposure to that particular antigen. This study was to evaluate the effect of oriental materia medica on IL-5 secretion in the mouse spleen cell. Methods : We used the splenocytes of mouse 8 weeks after its birth, and then cultivated those into the 2 experimental groups and a control group for 48 hours. The culture media of the experimental groups were made of $1{\mu}g/ml$, $10{\mu}g/ml$ oriental materia medica, representative. And the culture media of control group was given no oriental materia medica. Then, we assayed the quantity of cytokine-expression by the sandwich ELISA. The quantities of cytokine-expression of the experimental groups were compared with that of the control group, which was standardized. These methods were used for all of the oriental materia medica treated. Results : Some oriental materia medica inhibit the secretion of IL-5 in both $1{\mu}g/ml$ and $10{\mu}g/ml$ culture media. These were Acori Rhizoma, Luffae Fasciculus Vascularis, Amomi Rotundi Fructus, Schisandrae Fructus, Biotae Semen, Clematis armandii, Dioscoreae Sativa Rhizoma, Coicis Semen, Sophorae Flos, Oroxyli Semen, Aurantii Semen, Pini Nodi Lignum. Conclusion : This study indicates that some oriental materia medica inhibit the secretion of IL-5 and are beneficial for allergic disease.

Effects of Promoter Methylation on the Expression Levels of Plakoglobin Gene in Both the ARO Thyroid Cancer Cell Line and Cancer Tissues

  • Plakoglobin (PKG) is a protein linking cadherin adhesion receptors to the actin cytoskeleton and its overexpression has been known to suppress cell proliferation and tumorigenesis in thyroid cancer. We investigated the effect of 5-aza-2'-deoxycytidine (5-Aza-CdR), a DNA methyltransferase inhibitor, on the methylation status of the promoter and the expression of the plakoglobin gene in a thyroid carcinoma cell line (ARO) and papillary thyroid carceinoma. In cultures of ARO cell line incubated without 5-Aza-2'-deoxycytidine (5-Aza-CdR), five of the fifteen CpG sites in the promoter spanning -225 and -54 were methylated at 4.2 - 12.5%. When the cells were treated with 5-Aza-CdR, all the methylated CpG sites were induced to be demethylated except one. In addition, a new methylation at one CpG site, CpG4, was identified at level of 12.0%. The expression level of PKG decreased approximately 10-fold in the 5-Aza-CdR treated cells compared to untreated cells. Different pattern of promoter methylation and expression of PKG was also observed in the tissue samples. CpG10 and CpG12 sites were methylated at 9.0-27.0% in normal tissues. However, in cancer tissues, CpG5 and CpG10 sites were methylated at 10.0-22.0%. Three of ten normal thyroid tissue samples and one of thirteen papillary carcinoma tumor samples showed increased PKG mRNA expression level. PKG protein expression analyzed by the immunohistochemical staining showed higher expression in the tumor compared with normal.

대형 어레이 안테나 기반 초광역 무선 백홀망 시스템 (Ultra Wide Area Wireless Backhaul Network System Based on Large Scale Array Antenna)

  • 5세대 이동통신 시스템은 4세대 LTE 시스템의 용량 대비 1000배의 용량 증대를 목표로 한다. 이를 달성하기 위해 많은 수의 소형셀을 활용하는 heterogeneous network 기술이 차세대 이동통신 기술로 각광받고 있다. 5세대 이동통신 시스템에서는 고정형 기지국과 이동형 기지국이 기하급수적으로 늘어날 것으로 예상되는데, 이러한 기지국을 모두 유선으로 연결하는 것은 비용 측면에서 합리적이지 않고, 이동형 기지국을 지원하기 위해서는 유선으로 백홀 링크를 지원하는 것보다 효율적인 무선 백홀망 시스템의 구축이 필요하다. 본 논문에서는 이러한 배경을 바탕으로 고정 및 이동 액세스 포인트와 무선 백홀 스위치를 기반으로 한 5세대 이동통신 초광역 무선 백홀망 시스템을 제안, 초광역 무선 백홀망의 link budget을 분석하고 대형 어레이 안테나를 활용한 빔 기반 초광역 무선 백홀망 시스템의 성능을 시뮬레이터 분석을 통해 검증을 진행하였다. 초광역 무선 백홀망 link budget 분석을 통해 1 nJ/Gbps의 에너지 효율의 달성 가능성을 확인하고, 전송 시뮬레이터를 통해 무선 백홀 스위치를 중심으로 $10km^2$의 면적에 이동 액세스 포인트가 여러 위치에 존재할 때 사이드로브를 제어함으로써 빔 간 간섭을 줄일 경우 빔 당 1 W의 전송 전력을 통해 평균 주파수 효율을 5 bps/Hz 이상 달성 가능함을 확인하였다.

  • In this study, We evaluated the Performance of PFC and the system performance of large size air-conditioner applying to outdoor condenser. PFC can meet the same cooling capacity in 40.42% of volume to fin-tube condenser. Although the fin-tube condenser requires 3600g of refrigerant charging, PFC requires 1700g, 1800g, 1900g, 2000g refrigerant charging for each 2.0mm, 2.5mm, 3.0mm and 3.5mm fin pitches. Difference of condensing and evaporation pressure is the biggest point 2.0mm fin pitch and the smallest point 2.5mm fin pitch.

  • The detection of many synthetic chemicals used in industry that may pose a genetic hazard in our environment is of great concern at present. In this respect, administrative authorities has great concern to regulate and to evaluate the chemical hazard to environment and human health. The clastogenicity of 28 synthetic chemicals was evaluated in Chinese hamster lung fibroblast cells in vitro. Glycidylacrylate which is one of the most cytotoxic chemical among 28 chemicals tested revealed clastogenicity in the range of 0.31-1.25 $\mu\textrm{g}$/$m\ell$ both in the presence and absence of metabolic activation system. Neopentyl glycol (340-1360 $\mu\textrm{g}$/$m\ell$) also revealed weak positive result both in the presence and absence of metabolic activation system. Cyanoguanidine (/$420.5-841 $\mu\textrm{g}$m\ell$) and N-butylchloride ($231.5-926 $\mu\textrm{g}$/m\ell$) revealed weak positive result only in the absence of S-9 metabolic activation system. Nevertheless total aberration percentages of N-butylchloride in the presence of metabolic activation system, and 3,4'-dichlorobenztrifluoride in the absence of S-9 metabolic activation revealed above 5% aberration, there is no statistical significance. From the results of chromosomal aberration assay with 28 synthetic chemicals in Chinese hamster lung cells, glycidylacrylate (CAS No. 106-90-0), neopentyl glycol (CAS No. 126-30-7), N-butyl chloride (CAS No. 109-69-3) and cyanoguanidine (CAS No. 461-58-5) revealed positive clastogenic results in this study.

  • More people prefer using rail traffic for travel or for commuting owing to its convenience and flexibility. The railway scenario has become an important communication scenario in the fifth generation era. The communication system should be designed to support high-data-rate demands with seamless connectivity at a high mobility. In this paper, the channel characteristics are studied and modeled for the railway tunnel scenario with straight and curved route shapes. On the basis of measurements using the "Mobile Hotspot Network" system, a three-dimensional ray tracer (RT) is calibrated and validated for the target scenarios. More channel characteristics are explored via RT simulations at 25.25 GHz with a 500-MHz bandwidth. The key channel parameters are extracted, provided, and incorporated into a 3rd-Generation-Partnership-Project-like stochastic channel generator. The necessary channel information can be practically realized, which can support the link-level and system-level design of the communication system in similar scenarios.

세개의 Gauss 동을 중첩한 간섭계형 결상계의 진폭임펄스와 초분해능 (Amplitude impulse and superresolution of interferometric imaging system obtained by superposing three Gauss pupils)

  • 역변환 문제(Inverse Problem)로 접근하여 상면에서의 최초 진폭임펄스 S$_{0}$(x)를 Gauss 함수 g$_{1}$(x), -1/2g$_{2-}$ (x+.DELTA.x$_{1}$), -1/2g$_{2+}$(x-.DELTA.x$_{1}$)를 중첩하여 설정하였다. 이를 Inverse Fourier Transform으로 동함수 A(.omega.)를 구하고, 유한구경 (-.omega.$_{0}$~+.omega.$_{0}$)에서 A(.omega)를 Fourier Transform하여 회절상의 진폭임펄스(Amplitude Impulse) S(x)를 구하였다. .lambda.=193nm, NA=0.5인 광학계에서 S(x)의 반치폭, 즉 1/2(FWHM)을 수치계산하여 49nm를 얻었다. 이는 Rayleigh 한계분해능 .epsilon.$_{R}$의 반, 1/2.epsilon.$_{R}$=96.5nm 보다 작으므로 초분해능 광학계임을 알 수 있다. OTF를 구하여 광학계의 성능을 평가한 결과 고주파영역에서 성능이 우수함을 알 수 있었다. 광학계는 Twyman-Green 간섭계를 포함하는 간섭결상계가 되고, Gauss 진폭변조판은 Polysiloxane Glass Resin을 사용하여 만들 수 있음을 제안하였다.제안하였다.

  • 호접란의 화경액아배양을 호르몬의 첨가없이 대량번식에 이용할 수 있도록 PLB 형성과 증식, 유식물분화를 위한 배지조성을 구명한 결과는 다음과 같다. PLB 형성을 위해서는 1.2X VW 배지에 사과추출물과 감자추출물, sucrose 1%, PVP 1.5g/l 또는 활성 탄소 2.5g/l, gellan gum 4g/l을 첨가하는 것이 가장 양호하였다. PLB 증식을 위해서는 sucrose 2%와 사과추출물과 감자추출물을 첨가한 VW에 cotton plate를 지지물로 사용한 것이 가장 양호하였다. 유식물의 재분화를 위해서는 Hyponex 3g/l에 사과와 감자추출물을 각각 3%, 바나나추출물은 4%를 혼용첨가한 배지가 가장 효과적이었다.

참전복, Haliotis discus hannai 유생발생 및 채묘에 미치는 아연독성 (Effects of Zinc Toxicity on Larval Development and Seed Collection of Abalone, haliotis discus hannai)

  • in the present study, the zinc toxicity to larval development and seed attachment of the abalone, Haliotis discus hannai was obtained under continuous flow through system. The zinc concentration melted from zinc coating pipe for 7 months ranged from $89.00\pm2.55 \mu\textrm{g}/\ell to 15.23\pm2.58\mu\textrm{g}/\ell(Y=0.85M^2-19.71+109.96)$. Treatments were carried out with zinc concentration $0~160 \mu\textrm{g}/\ell$. The maximum and minimum of fertilization rate were $87.7\pm5.3%$ in control, $83.7\pm7.6%$ in zinc concentration $160\mu\textrm{g}/\ell$, respectively. The maximum and minimum of hatching rate were $87.5\pm4.5%$ in zinc concentration $10\mu\textrm{g}/\ell$, $79.3\pm5.6%$ in zinc concentration $160\mu\textrm{g}/\ell$, respectively. Both of the results were not significantly different (P>0.05). But the normality rate, setting rate and survival rate of abalone larvae at over zinc concentration TEX>$20\mu\textrm{g}/\ell$ decreased rapidly and showed significantly different from those of the other group(P<0.05).
