Application of Iranian Medicinal Plants to the Treatment of Liver Injury.

  • Matricaria Chammomillal L., Foemiculum Vulgare mill, and Plantago Psylium L. have been screened for their hepato protective activities against liver damge induced by $CCl_4$ intoxication in mice. Hydroalcoholic extractions (2:8) of herbal drugs were concentrated in vacuo and concentrated crude extracts of Matrica Chammomilla L. and Foeniculum Vulgare mill were orally administered at doses of 100 mg/kg, 200 mg/kg, 400 mg/kg, and 800 mg/kg. Plantago Psyllium was given at doses of 50 mg/kg, 100 mg/kg, 200 mg/kg, and 400 mg/kg. Liver protective activities of these herbs were determined after administration of $CCl_4$ Liver size, serum enzyme activities, sleeping time, and histopatology of the liver were examined one hour after administration of $CCl_4$. ALT and AST activities, liver weight and sleeping time decreased in groups that received 400 mg/kg of Matricaria Chammomilla L. or Foeniculum Vulgare. Histological investigation showed significant increase in hepatic cell regeneration and reduction in liver injury. The group that received 100 mg/kg Plantago Psylium showed liver protection but protection was not significant in other doses.

  • Brazilin was tested for its ability to inhibit alloxan induced lipidperoxidation. Lipid peroxide contents in liver, kidney and serum were measured by the TBA method. ICR mice receiving alloxan at a dose of 43mg/kg via the tail vein after a 24 hrs starvation showed significantly increased lipid peroxide contents as compared to untreated control. Lipid peroxide contents in liver, kidney and serum of alloxan-induced diabetic mice were dosedependently decreased by the treatment of brazilin at a dose of 10mg/kg, 50mg/kg, 100 mg/kg for 5 days.

  • The purpose of this study was to investigate the effect of nifedipine (10 mg/kg) on the pharmacokinetic parameters and the bioavailability of paclitaxel (50 mg/kg) orally coadministered and pretreated in rats. The plasma concentration of paclitaxel in combination with nifedipine was significantly (p<0.05 at 10 mg/kg coadmin., p<0.01 at pretreat.) increased compared to that of control, from 2 hr to 24 hr. Area under the plasma concentration-time curve (AUC) of paclitaxel with nifedipine was significantly (p<0.05 at 10 mg/kg coadmin., p<0.01 at pretreat.) higher than that of control (omitted)

  • The acute oral, intraperitoneal and subacute toxicity of monocrotophos, an insecticide, was studied in albino rats. The acute oral $LD_{50}$ values for female and male rats were 5.0mg/kg and 8.7mg/kg, respectively. Conversely the intraperitoneal $LD_{50}$ values for female and male rats were 4.9mg/kg and 6.0mg/kg, respectively. Plasma cholinesterase (ChE) activity in rats treated with a single dose was the most depressed 4 hours after administration, but returned to normal within 72 hours of administration. Significant depression of body weight gain and food consumption was observed in rats receiving the 3.5mg/kg/day dose level for 28 days as compared with the controls. Additionally plasma ChE activity depression was also observed.

  • This study was carried out to investigate the Pollution of heavy metals in Soil, collected from the 50 sampling sites in 1986. The results were as follows; 1. In the average contents of Cd, the highest value was 0.33mg/kg in the Industrial area, the lowest value was 0.06 mg/kg in the Greenbelt and 0.10-0.22mg/kg in the other areas. 2. In the average contents of Cu, the highest value was 33.46mg/kg in the Industrial area, the lawest value was 3.79mg/kg in the Greenbelt and 4.69-12.21mg/kg in the other areas. 3. In the average contents of As, the highest value was 1.80mg kg in the Residential area, the lowest value was 0.43mg/kg in the Greenbelt and 0.64-1.65mg/kg in the other areas. 4. In the average contents of Zn, the highest value was 29.84mg/kg in the out wall of Seoul, the lowest value was 17.32mg/kg in the Greenbelt and 19.62-26.71mg/kg in the other areas. 5. In the average contents of Pb, the highest value was 17.09mg/kg in the Industrial area, the lowest value was 5.21mg/kg in the Reclamated waste and 5.67 -14.10mg/kg in the other areas. 6. Distribution of Heavy metals was Zn>Cu>Pb>As>Cd and the relations of Zn-Cd, Pb-Cd, Pb-Cu, Pb-Zn were shown the correlation significantly (P< 0.01).

Investigation on the Pollution in the Soils of Seoul Area (서울시일원의 토양 오염도 조사)

  • This work was carried out to investigate the pollution of heavy metals and pH in soil, collected from the 156 sampling sites in 1987. 1. In the average contents of Cd, the highest value was 0.40 mg/kg in the Outskirt of Seoul area, the lowest value was 0.11 mg/kg in the Greenbelt area, and 0.21∼0.39 mg/kg in the other areas. 2. In the average contents of Cu, the highest value was 32.50 mg/kg in the Motorway area, the lowest value was 2.96 mg/kg in the Greenbelt area, and 9.53∼24.41 mg/kg in the other areas. 3. In the average contents of As, the highest value was 1.32 mg/kg in the Reclamated waste area, the lowest value was 0.31 mg/kg in the Greenbelt area, and 0.93∼1.32 mg/kg in the other areas. 4. In the average contents of Zn, the highest value was 27.33 mg/kg in the Center of city area, the lowest value was 9.97 mg/kg in the Greenbelt area, and 12.47∼25.23 mg/kg in the other areas. 5. In the average contents of Pb, the highest value was 125.25 mg/kg in the Motorway area, the lowest value was 12.99 mg/kg in the Farming area, and 17.01∼77.71 mg/kg in the other areas. 6. In the average contents of pH, the highest value was pH 7.08 in the Motorway area, the lowest value was pH 4.14 in the Greenbelt area, and pH 5.67~7.04 in the other areas. 7. The order of the distribution of heavy metals was Pb>Zn>Cu>As>Cd. There was significanthy correlation between Cd-Zn, Pb-Zn, Cu-Cd, Pb-Cu and Pb-Cd (p<0.01).

A Toxicological Study of Young Fronds of Bracken Fern (Pteridium aquilinum var latiusculum) Collected in Kwang Ju Area (한국산 고사리의 독성조사에 관한 연구)

  • The results of toxicity test using mice and rats for ethanol extract of Bracken Fern collected in Kwang Ju area were shown as follows ; Up to the dose of 10g per kg of mice administered intraperitoneally there was no lethal toxicity so that it was impossible to calculate the median lethal dose $(LD_{50})$. For the first 7 days experiment all rats administered frond extract grouping in 40mg, 400mg, and 1200mg per kg of rat as the daily oral doses did not show any characterized sign in the weight gain rate, anatomical findings, and biochemical studies. For 3 weeks following the first week the weight gain rates of all test group were reduced to $4.2{\sim}7%$ below the weight gain rate of control. In this period serum GPT, GOT, and Alkaline phosphatase value were increased significantly indicating the symptoms of Bracken Fern poisonings. The pathological findings of all test groups for 28 days showed acute and chronic intestinal lesion and liver damage with steatosis especially in 1200g/kg rat groups. In this experiment the Bracken poisonings appeared slowly in rats of 400mg/kg and 1200mg/kg for two weeks and in rats of 40mg/kg for 3 weeks, showing the symptoms of lowering of weight gain rate, subacute hepatitis, hepatic steatosis and enteritis in 28 days experiment.

  • The effects of red ginseng component (water extracts, alcohol extracts, lipophilic extracts, total saponins, panaxadiol and panaxatriol) administration on glutathione (GSH) and lipid peroxidation levels in mice were investigated. 20~25 g ICR mice which were pretreated with water extracts (50 mg/kg), alcohol extracts (50 mg/kg), lipophilic extracts (50 mg/kg), total saponins (50 mg/kg), panaxadiol (50 mg/kg) and panaxatriol (50 mg/kg) for 15 days. The ability of red ginseng component to protect against oxidative damage to the mouse liver was examined by determining the level of lipid peroxidation (MDA), glutathione, and the activities of glutathione peroxidase (GPX). The GSH level was raised by all the ginseng component, but the GSSG level was lowered ]argely by all the ginseng component. The ratio of GSSG/total GSH was decreased because the level of GSSG was decreased more than that of GSH. Finally, the lipid peroxidation (MDA) level was the lowest in lipophilic extracts and panaxadiol nest. In conclusion, the order of effectiveness of anti-oxidants was to be lipophilic extracts>panaxadiol>total saponins.

  • The polysaccharide was predominantly composed of glucose (79.60%), galactose (9.68%), mannose(8.67%), fructose (1.96%;) and xylose (0.09%). Major amino acid in protein was consisted of cystein, aspartic acid, valine and glutamic acid. The tumor growth inhibition ratio of the polysaccharide against Sarcoma 180 was at highest level of 64.96%, when administerd at the concentration of 50 mg/kg and life prologation ratio showed 28.91% at the concentration of 50 mg/kg. The direct cytotoxic effect of polysaccharide was not observed in the tumor bearing mice with Sarcoma 180 in vitro.

Residual Pattern of Procymidone and Chlorothalonil in Grape During the Period of Cultivation and Storage (포도의 재배 및 저장기간 중의 Procymidone 및 Chlorothalonil의 잔류량변화)

  • In order to know the residual pattern of pesticides and predict to the degradation period until below MRL we experimented procymidone and chlorothalonil for grape which were the most detected pesticide in grape by NAQS(National Agricultural product Quality management Service) survey. In this experiment we sprayed those pesticides 10 days before harvest and analyzed 0, 1, 2, 3, 5, 7, 10 day sample to establish logical equation and to calculate $DT_{50}$. Also the same day samples stored at $4^{\circ}C$ and $20^{\circ}C$, which were compared their degradation patterns. During the cultivating period, the residue amount of procymidone was changed from 1.85 mg/kg (0 day) to 0.33 mg/kg (10 day), $DT_{50}$ was 3.5 days, and chlorothalonil was changed from 5.5 mg/kg (0 day) to 3.49 mg/kg (10 day), $DT_{50}$ was 4.4 days. During the storage period, $DT_{50}$ of procymidone and chlorothalonil at $4^{\circ}C$ were 10.5 and 7.6 days, and 6.3 and 6.1 days at $20^{\circ}C$, respectively.
