Effects of GooSunSan(九仙散) Extract on the Contraction of Isolated guinea pig's various kinds smooth muscles (구선산(九仙散)이 GUINEA PIG의 각종(各種) 평활근(平滑筋)에 미치는 영향(影響))

  • In order to study the effects of Goosunsan known clinically for their effects of treatment for cough and asthmas the study was carried out to investigate the effect of Goosunsan extract on the contractile force of the isolated guinea pig's vaviovs kinds smooth muscles and elucidate its mechanism The result were obtained as follows: 1. The isolated trachea & ileum smooth muscles of guinea pig was suspended in the organ bath with oxygenated kreb's Henselsite bicarbonate buffer solution at $37^{\circ}C$, and the developed tension by the drug was recorded with Isometric transducer (nacro F-60). The resting tension was approximately 0.5g. 2. The isolated trachea & ileum smooth muscles of guinea pig was remakably relaxed by the administration of Goosunsan. 3. The contractile response of the trachea smooth muscle of the isolated guinea pig to histamine 10-4 M was remakably by Goosunsan extract. 4. The contractile response of the ileum smooth muscle of the isolated guinea pig to histamine 10-4 M was remakably by Goosunsan extract. 5. The contractile response of the trachea smooth muscle of the isolated guinea pig to acetylcholine 10-4 M was remakably by Goosunsan extract. 6. The contractile response of the ileum smooth muscle of the isolated guinea pig to acetylcholine 10-4 M was remakably by Goosunsan. 7. The contractile response of the trachea smooth muscle of the isolated guinea pig to 5-hydroxytryptamine 10-4 M was remakably by Goosunsan. 8. The contractile response of the ileum smooth muscle of the isolated guinea pig to 5-hydroxytryptamine 10-4 M was remakably by Goosunsan.

Vasodilation Effect of the Water Extract of Rheum palmatum L. in Rat Thoracic Aorta.

  • Rheum palmatum L. has been used for treatment of hypertension, lipemia, and paramenia in the oriental gerbal medicines for a long time. We have examined the relaxational response to the water extract of Rheum palmaum L. in isolated thoracic aorta from sprague dawley (SD) rat in the presence and absence of endothelium. Rat thoracic aorta was investigated in vessel segments suspended for isometric tension recording by polygraph. Responses to Rhizoma Rhei were investigated in vessels precontracted with 5-hydroxytryptamine. We found that the ghoracic aorta segments responded to the water extract of Rheum palmatum L. (ERP) with a dose-dependent vasorelaxation. We found that 1.The thoracic aorta sehments responded to ERP with a dose-dependent vasodiliation. 2.The 5-HT induced contraction at 10$\^$-4/M were inhibited by 85.8% after addition of the 0.1 g/mL water extract of ERP. 3. The 5-HT induced contraction at 10$\^$-4/ M with and without endothelium were inhibited by 86.4% and 85.8% after addition of the 0.1g/mL ERP. 4. After pre-treatment of the thoracic aorta with 10$\^$-4/M N$\^$G/-monomethyl-L-arginine(L-NMMA), inducible niric oxide synthase inhibitor, the vessels has not response to the contraction. In conclusion, ERP induced reaxation in the isolated rat thoracic aorta were composed of dose-dependent relaxation. and it has potent vasodilation.

Anti-stress and Anti-nociceptive Effects of the Aqueous Extracts of Chungsimyeonja-eum against Immobilization Stress and Incision Pain in Rats (청심포자음(淸心蒲子飮)의 구속 스트레스 및 절개 통증에 대한 항스트레스 작용과 진통효과)

  • The aqueous extract of Chungsimyeonja-eum has been used to treat palpitation, anemia, chronic fatigue, hypertension, and stroke. It also possesses various pharmacological effects including hypotensive, blood circulating, sedative, anti-inflammatory, diuretic, and anti-stress activities. In the present results, the latency time of plantar test was decreased in the immobilization stress and incusion pain-induction group. However the paw withdrawal latency values were increased in the immobilization stress and incision pain-induction groups after treatment with the aqueous extract of Chungsimyeonja-eum. The c-Fos expression in the paraventricular nucleus (PVN), and 5-hydroxytryptamine (5-HT) and tryptophan hydroxylase (TPH) in the dorsal raphe were enhanced in the immobilization stress and incision pain-induction groups. However, the treatment with the aqueous extract of Chungsimyeonja-eum suppressed the immobilization stress and incision pain-induced increase of c-Fos, 5-HT, and TPH expressions. Here in this study, we have demonstrated the protective effects of Chungsimyeonja-eum on immobilization and incision pain-induced stress. The present study revealed that Chungsimyeonja-eum treatment diminishes immobilization and pain stress.

  • Four plausible precursors (21, 22, 24, and 25), just prior to formation of the oxathiazepine ring in eudistomin, were synthesized by the Pictet-Spengler condensation of N-hydroxytryptamine (15) or N-hydroxytryptophan ester (19) with cysteinal derivatives (5 and 10). In the case of the parent compound (21), one of these four precursors, treatment with dihalomethane in the presence of a phase transfer catalyst gave an eudistomin analogue (26) having the oxathiazepine ring in 35-50% yield.

  • Mahwangsan is used in the treatment of asthma due to wind and cold(風寒喘). This study was carried out to investigate the effect of Mahwangsan extract and each drug composing it on the contractile force of the isolated guinea pig trachea muscle and elucidate its mechanism. The results were obtained as follows; 1. The isolated trachea smooth muscle of guinea pig was signifi-cantly relaxed by the administration of Mahwangsan extract. 2. The contractile response of the isolated trachea smooth muscle of guinea pig to histamine was significantly inhibited by the administration of Mahwangsan extract. 3. The contractile response of the isolated trachea smooth muscle of guinea pig to histamine was significantly inhibited by the administraction of Herba Ephedrae, Cortex Cinnamoni, Flos Farfarae, Fructus Chebulae and Semen Armeniacae extract. 4. The contractile response of the isolated trachea smooth muscle of guinea pig to acetylcholine was significantly inhibited by the administration of Mahwangsan extract. 5. The contractile response of the isolated trachea smooth muscle of guinea pig to acetylcholine was significantly inhibited by the administration of Herba Ephedrae, Flos Farfarae, Fructus Chebulae and Semen Armeniacae extract. 6. The contractile response of the isolated trachea smooth muscle of guinea pig to 5-hydroxytryptamine was significantly inhibited by the administration of Mahwangsan extract. 7. The contractile response of the isolated trachea smooth muscle of guinea pig to 5-hydroxytryptamine was significantly inhibited by the administration of Herba Ephedrae, Flos Farfarae, Fructus Chebulae and Semen Armeniacae extract.

An Immunohistochemical study of the gastro-entero-pancreatic endocrine cells in the alimentary tract and the pancreas of the fresh water turtle, Geoclemys reevesii (남생이 위장관 및 췌장 내분비세포에 관한 면역조직화학적 연구)

  • The regional distribution and relative frequency of gastrointestinal endocrine cells were studied immunohistochemically in the gastrointestinal mucosa and pancreas of the fresh water turtle. Ten kinds of endocrine cells were identified in the gastrointestinal tract. Cholecystokinin-8-, bovine pancreatic polypetide-and glucagon-immunoreactive cells were seen throughout the gastrointestinal tract, also among them cholecystokinin-8-immunoreactive cells were most predominant in segment III. Although gastrin- and gastrin/cholecystokinin-immunoreactive cells were found from segment III to VI and X, respectively, they were numerous in segment III. Somatostatin-immunoreactive cells were observed from segment I to VII. 5-hydroxytryptamine- immunoreactive cells were detected in segment I, III, VIII, IX and X. Human pancreatic polypeptide-immunoreactive cells were demonstrated in segment V, VI, VIII, IX and X. Insulin-immunoreactive cells were found from segment III to X except for segment VIII, but rare in segment VII. Neurotensin-immunoreactive cells were found to be restricted to segment VIII, IX and X. No porcine chromogranin-, substance P- and bombesin-immunoreactive cells were detected throughout the gastrointestinal tract of the fresh water turtle. Although typical mammalian pancreatic islets encapsulated by connective tissue were not present in this species, five kinds of endocrine cells-glucagon, insulin, somatostatin, bovine pancreatic polypeptide and 5-hydroxytryptamine-were found in forming small or large groups and scattered in the exocrine gland region. However porcine chromogranin- and motilin-immunoreactive cells could not be demonstrated in the pancreas.

Effect of Tryptophan on Serotonin and Opiate Receptor Binding in Stressed Rats (트립토판이 스트레스 받은 쥐 뇌의 세로토닌과 오피에이트 수용체 결합에 미치는 영향)

  • Brain serotonin and its utilization was investigated on stressed rats after feeding high tryptophan diet for a month. High tryptophan fed rats displayed significantly higher level of serum tryptophan, brain tryptophan, serotonin (5-hydroxytryptamine; 5-HT) and 5-hydroxyindoleacetic acid (5-HIAA) than the control diet fed rats. When rats were treated with 3 hour immobilization (IMMB) stress, serotonin turnover was slightly increased, but not statistically significant, in control diet group rats. However in high tryptophan diet rats, 3 hr IMMB stress resulted in statistically significantly (p<0.05) decreased the serum tryptophan, brain tryptophan and 5-HT level. The concentration of 5-HIAA was significantly increased indicating accelerated utilization of the brain 5-HT of the high trp. fed rat. The utilization pattern of the serotonin was found to be similar among young and adult rats. Rats on a tryptophan enriched diet displayed higher coping ability to the stress as they exhibited smaller increment of corticosterone level. A possble involvement of opioid system was suggested in serotonin utilization by measuring total $^{3}$[H]-naloxone binding in brain.

NMDA Receptor Antagonists Enhance 5-HT Receptor-mediated Behavior, Head-Twitch Response, in Mice

  • The purpose of this study was to determine the behavioral interaction between glutamatergic and serotonergic receptors. In the present study, both the competitive (AP-5 and D-CPP) and the noncompetitive (MK-801, ketamine, dextrorphan and dextromethorphan) N-methyl-D-aspartate (NMDA) receptor antagonists markedly enhanced 5-HT(5-hydroxytryptamine)-induced selective serotonergic behavior, head-twitch response (HTR), in mice. These results suggest that the glutamatergic neurotransmission may modulate serotonergic function at the 5-HT receptor. The precise relationship between glutamatergic and serotonergic system is as yet undefined. However, these are the first data available regarding glutamatergic modulation of serotonergic function at the 5-HT receptor in intact mice, and the present results support the notion that the NMDA receptors may play important roles in the glutamatergic modulation of serotonergic function at the 5-HT receptor.

  • There is no doubt that dopamine plays a critical role in the etiopathogenesis of schizophrenia. However, there appeared some limitations in explaining the complex phenomena of schizophrenia. Recent research data suggest that dysfunction in serotonergic system may be involved. Before the dopamine hypothesis of schizophrenia became established, the interest in serotonin(5-hydroxytryptamine, 5-HT) as an etiological substrate of this illness occurred. Recently the importance and extent of 5-HT's involvement in the pathophysiology and mechanism of action of antipsychotic drug is actively investigated. In recent years, therapeutic success of clozapine and risperidones has increased attention on the interaction between the 5-HT and dopamine systems in schizophrenia. This led to the concept of serotonin-dopamine antagonist for antipsychotics. The authors review the evidence for the role of 5- HT in schizophrenia and serotonin-dopamine interaction.

Effect of serotonin on the cell viability of the bovine mammary alveolar cell-T (MAC-T) cell line

  • 5-Hydroxytryptamine (5-HT), a monoamine, as a local regulator in the mammary gland is a chemical signal produced by the mammary epithelium cell. In cows, studies have shown that 5-HT is associated with epithelial cell apoptosis during the degenerative phase of the mammary gland. However, studies in other tissues have shown that 5-HT can effectively promote cell viability. Whether 5-HT could have an effect on mammary cell viability in dairy cows is still unknown. The purpose of this study was to determine: (1) effect of 5-HT on the viability of bovine mammary epithelial cells and its related signaling pathways, (2) interaction between prolactin (PRL) and 5-HT on the cell viability. The bovine mammary alveolar cell-T (MAC-T) were cultured with different concentrations of 5-HT for 12, 24, 48 or 72 hours, and then were assayed using cell counting kit-8, polymerase chain reaction (PCR) and immunobloting. The results suggested that 20 μM 5-HT treatment for 12 or 24 h promote cell viability, which was mainly induced by the activation of 5-HT receptor (5-HTR) 1B and 4, because the increase caused by 5-HT vanished when 5-HTR 1B and 4 was blocked by SB224289 and SB204070. And protein expression of mammalian target of rapamycin (mTOR), eukaryotic translation elongation factor 2 (eEF2), janus kinase 2 (JAK2) and signal transducer and activator of transcription 5 (STAT5) were decreased after blocking 5-HT 1B and 4 receptors. When MAC-T cells were treated with 5-HT and PRL simultaneously for 24 h, both the cell viability and the level of mTOR protein were significantly higher than that cultured with 5-HT or PRL alone. In conclusion, our study suggested that 5-HT promotes the viability of MAC-T cells by 5-HTR 1B and/or 4. Furthermore, there is a reciprocal relationship between PRL and 5-HT.
