Acid Hydrolysis Characteristics of Yellow Poplar for High Concentration of Monosaccharides Production (백합나무를 이용한 고농도 단당류 생산을 위한 산 가수분해 특성 연구)

  • We investigated acid hydrolysis characteristics of yellow poplar woodmeal with concentrated sulfuric acid for high concentration of monosaccharides production. Woodmeal to 72% sulfuric acid ratio (w/w), $2^{nd}$ hydrolysis temperature and time were main variables for finding optimum reaction condition. Optimum woodmeal to 72% sulfuric acid ratio was 1 : 2.61 (w/w) and $2^{nd}$ hydrolysis temperature and time was $105^{\circ}C$ and 40 min as 44.8 g/L of glucose and 25.2 g/L of xylose in hydrolysis solution. In this acid hydrolysis solution, furfural, 5-HMF, low molecular weight phenolic compounds were identified. Furfural and 5-HMF concentration were increased as increasing $2^{nd}$ hydrolysis time. More than 40 min of $2^{nd}$ hydrolysis at $110^{\circ}C$, xylose concentration was decreased but glucose concentration was leveled out because xylose to furfural reaction was faster than xylan to xylose, but cellulose to glucose reaction was similar rate with glucose to 5-HMF at that $2^{nd}$ hydrolysis reaction condition.

  • Effects of pH, amino acids, hydrolyzed protein and potassium phosphate on caramelization were investigated for improvement of its reaction rate. The caramelization was performed with starch syrup at $110^{\circ}C$ and the different color functions-metric saturation(Suv), 5-hydroxymethylfurfural (HMF) contents and absorbance at 420 nm were measured. As the pH was raised from 4 to 10, the reaction rate (Suv/hr) was increased by 31.9% along with significant increase in HMF content and absorbances at 420 nm. Among the several amino acids, arginine and glycine were very effective for improvement of caramelization, which may be due to Maillard reaction. When $K_2HPO_4$ were added in different ratio with arginine, glycine, HVP or HAP, the effects of arginine and HAP on thee rate were markedly enhanced while the effects of glycine and HVP were rather reduced.

  • In order to investigate the thermal degradation of glucose and maltose solutions after high temperature and high pressure (HTHP) treatment, the samples were treated at temperatures of 110, 120, 130, 140, and $150^{\circ}C$ for 1, 2, 3, 4, and 5 h in an apparatus for HTHP treatment. Glucose and maltose solutions (20% w/w) were prepared by weighing glucose and maltose and adding distilled water in the desired proportion. Chromaticity, pH, organic acids, 5-hydroxymethylfurfural (HMF), free sugar contents, electron donating ability (EDA), and ascorbic acid equivalent antioxidant capacity (AEAC) were evaluated. With increasing heating temperatures and times, the L-, a-, and b-values decreased. The pH and free sugar contents decreased, and organic acids and HMF contents increased with greater temperatures and times. EDA (%) and the AEAC of the heating sugars increased with the increases in temperatures and times.

Dehydration Reaction of Fructose to 5-Hydroxymethylfurfural over Various Keggin-type Heteropolyacids (Keggin형 헤테로폴리산에 의한 과당의 5-하이드록시메틸퍼퓨랄로의 전환을 위한 탈수반응)

  • Four Keggin-type heteropolyacids, $H_nXM_{12}O_{40}$(X = P and Si, M = W and Mo) that were substituted with heteroatom and polyatom were applied to the dehydration reaction of fructose to 5-hydroxymethylfurfural (HMF). The results showed that the acid became stronger when the heteroatom and polyatom were substituted with P and W than the cases of Si and Mo, respectively. However, the amount of acidic sites increased with the decrease in the acid strength, resulting in the change of the catalytic activity of heteropolyacids in the dehydration reaction. The experimental results revealed that four different heteropolyacids produced similar amounts of HMF via the dehydration reaction of fructose due to the counterbalancing effect between the amount of active sites, which is related to the catalytic activity of heteropolyacids, and the softness of polyanion. In addition, it was observed that the prepared heteropolyacids showed good structural stability after heat treatment at $200^{\circ}C$.

  • The anthelmintic substance of the roasted fruits of Prunas mume against Clonorchis sinensis was isolated and its structure was identified by chemical and physical methods. The results obtained from the experiments are as follows: 1) The methanol extract of the roasted fruits of P. mume was fractionated into four parts: petroleum ether, ethyl ether, ethyl acetate and water soluble part. Among these, etherial fraction was found to have strong wormicidal effect on liberated metacercaria of Clonorchis sinensis. 2) From the etherial fraction, the wormicidal substance was isolated by silica gel, polyamide and sephadex column chromatography and identified to be 5-hydroxymethylfurfural (5-HMF) by chemical and spectral data. 3) 5-HMF was synthesized and administered to the rabbits infected with C. sinensis. On the 2nd day after administration, the EPG (eggs per gram in feces) reached to the maximal value. Among the adult worms isolated from the bile duct of the treated animal, 84% of worms were damaged morphologically. 4) The content of 5-HMF in the fruits of P. mume which were roasted in an oven at $90-110^{\circ}C$ for 52 hours and that in the fresh fruits was evaluated by HPLC. The content of 5-HMF was 0.8% in the roasted fruits and 0.02% in the fresh ones.

Enhanced 2,5-Furandicarboxylic Acid (FDCA) Production in Raoultella ornithinolytica BF60 by Manipulation of the Key Genes in FDCA Biosynthesis Pathway

  • The compound 2,5-furandicarboxylic acid (FDCA), an important bio-based monomer for the production of various polymers, can be obtained from 5-hydroxymethylfurfural (HMF). However, efficient production of FDCA from HMF via biocatalysis has not been well studied. In this study, we report the identification of key genes that are involved in FDCA synthesis and then the engineering of Raoultella ornithinolytica BF60 for biocatalytic oxidation of HMF to FDCA using its resting cells. Specifically, previously unknown candidate genes, adhP3 and alkR, which were responsible for the reduction of HMF to the undesired product 2,5-bis(hydroxymethyl)furan (HMF alcohol), were identified by transcriptomic analysis. Combinatorial deletion of these two genes resulted in 85.7% reduction in HMF alcohol formation and 23.7% improvement in FDCA production (242.0 mM). Subsequently, an aldehyde dehydrogenase, AldH, which was responsible for the oxidation of the intermediate 5-formyl-2-furoic acid (FFA) to FDCA, was identified and characterized. Finally, FDCA production was further improved by overexpressing AldH, resulting in a 96.2% yield of 264.7 mM FDCA. Importantly, the identification of these key genes not only contributes to our understanding of the FDCA synthesis pathway in R. ornithinolytica BF60 but also allows for improved FDCA production efficiency. Moreover, this work is likely to provide a valuable reference for producing other furanic chemicals.

Conversion of Glucose and Xylose to 5-Hydroxymethyl furfural, Furfural, and Levulinic Acid Using Ethanol Organosolv Pretreatment under Various Conditions

  • The objective of this study was to understand the conversion characteristics of glucose and xylose using the major monosaccharide standards for lignocellulosic biomass. The acid-catalyzed organosolv pretreatment conducted using ethanol was significantly different from the acid-catalyzed process conducted in an aqueous medium. 5-hydroxymethylfurfural (5-HMF), levulinic acid and furfural were produced from glucose conversion. The maximum yield of 5-HMF was 5.5%, at 200℃, when 0.5% sulfuric acid was used. The maximum yield of levulinic acid was 21.5%, at 220℃, when 1.0% sulfuric acid was used. Furfural was produced from xylose conversion and under 0.5% sulfuric acid, furfural reached the maximum yield 48.5% at 210℃. Ethyl levulinate and methyl levulinate were also formed from the glucose standard following the esterification reaction conducted under conditions of the combined conversion method, which proceeded under both ethanol-rich and water-rich conditions.

Composition, the Changes of Diastase Activity and Hydroxymethylfurfural Content During Storage of the Various Honey Samples (벌꿀의 조성(組成)과 저장중(貯藏中)의 Diastase 및 Hydroxymethylfurfural함량변화(含量變化))

  • The sugers and free amino acids of honey(Western and Domestic honey) and the changes of diastase activity during storage at various temperatures and HMF were studied. The average content of sugers in the western honey was 40.20% of fructose, 32.43% of glucose and 1.0% of sucrose. The average ratio of fructose to glucose was 1.24. In the case of the domestic honey, average content of sugers was 37.37% of fructose, 31.92% of glucose and 0.74% of sucrose and the average ratio of fructose to glucose was 1.19. Of the 16 amino acids detected, proline was the most predominant free amino acid in all of the honey studied, representing 39.59 to 46.86% of total free amino acids. Diastase activity of honey during the storage at $50^{\circ}C$ was rapidly decreased, but it was notably changed during storage at low temperature ($4^{\circ}C$) and room temperature ($20^{\circ}C$). The accumlated content of HMF were 0.12 to 0.47mg% during storage at 80 days at $20^{\circ}C$ and 5.35 to 8.71mg% after 3 weeks storage at $50^{\circ}C$.

Acid-Base Bifunctional Metal-Organic Frameworks: Green Synthesis and Application in One-Pot Glucose to 5-HMF Conversion

  • The direct synthesis of metal-organic frameworks (MOFs) with acidic and basic active sites is challenging due to the introduction of functional groups by post-functionalization method often jeopardize the framework integrity. Herein, we report the direct synthesis of acid-base bifunctional MOFs with tuning acid-base strength. Employing modulated hydrothermal (MHT) approach, microporous MOFs named $UiO-66-NH_2$ was prepared. Through the ring-opening reaction of 1,3-propanesultone with amino group, $UiO-66-NH_2-SO_3H-type$ catalysts can be obtained. The synthesized catalysts were well characterized and their catalytic performances were evaluated in one-pot glucose to 5-HMF conversion. Results revealed the acid-base bi-functional catalyst possessed high activity and excellent stability. This work provides a general and economically viable approach for the large-scale synthesis of acid-base bi-functional MOFs for their potential use in catalysis field.

Effects of the Aging Conditions on the Quality Characteristics of Garlic (숙성조건이 마늘의 품질특성에 미치는 영향)

  • This study was performed to investigate the quality characteristics such as pH, total acidity, browning index, 5-hydroxymethylfurfural (5-HMF), fructose, S-allyl-L-cysteine (SAC), total polyphenol, and DPPH radical scavenging activity of aged garlic at different aging temperatures and periods. Aging temperature and period had a significant (p<0.05) effect on the quality characteristics of garlic. The pH in aged garlic significantly (p<0.05) decreased with increasing aging temperatures and periods, while the total acidity, browning index, and 5-HMF levels increased. The 5-HMF levels in garlic aged at 60 and $70^{\circ}C$ increased from 0.08~3.30 and from 0.05~106.07 mg/100 g, respectively. The fructose content in garlic aged at $60^{\circ}C$ gradually increased from 0.77 to 14.57%, while that of garlic aged at $70^{\circ}C$ increased from 1.07 to 19.75% until day 30, after which it decreased. The SAC level in raw garlic was 47.09 mg/100 g. The SAC contents of aged garlic differed significantly according to aging temperature and period. The SAC levels in garlic aged at $60^{\circ}C$ and $70^{\circ}C$ were in the range of 15.28~100.5 and 12.41~85.55 mg/100 g, respectively. The total polyphenol contents of garlic aged at $60^{\circ}C$ and $70^{\circ}C$ were 6.01 and 6.67 fold higher, respectively, than those in raw garlic. The DPPH radical scavenging activity of aged garlic also showed a tendency to increase during aging.
