Effects of Aqueous Extract of Schizandrae Fructus on Lead-Induced Change of Monoamine Neurotransmitters in Hippocampus

  • The effects of aqueous extract of Schizandrae Fructus (AESC) on lead (Pb)-induced changes of monoamine neurotransmitters in the hippocampus (HIP) of adult rats were investigated. Male Sprague-Dawley rats were received intraperitoneal (i.p.) administration of Pb acetate (5 mg/kg/d) for 28 days and sacrificed 7 days after the last administration. Concentrations of norepinephrine (NE), dopamine (DA), serotonin (5-HT), 5-hydroxyindole acetic acid (5-HIAA) in HIP were measured by HPLC. There were significant decreases of NE, DA, 5-HT and 5-HIAA in Pb treated rats (P < 0.05), while pretreatment with AESC (100 mg/kg/d or 300 mg/kg/d, p.o., 2 h before Pb) greatly inhibited the decrease of monoamine transmitters, respectively (P < 0.05). Also, AESC (300 mg/kg/d) significantly increased the reduction of glutathione contents and superoxide dismutase activities in HIP induced by chronic Pb. These results suggest that AESC ameliorates Pb-induced depletion of monoamine neurotransmitters in HIP through its antioxidant activity.

A Study on Immunomodulation and Serotonin Metabolism Changes by Immobilization Stress in Mice Fed Tryptophan Supplemented Diet. (고 트립토판 식이를 섭취한 마우스에서 Immobilization 스트레스로 인한 면역변조와 Serotonin 대사의 변화에 대한 연구)

  • We fed high trypotophan diet(3.5% tryptophan/diet(w/w) to mice for 7 days and treated then with 3 hour immobilization(IMMB) stress to investigate tryptophan metabolism and immunomodulation. The levels of serum tryptophan, brain tryptophan, serotonin(5HT) and 5-hydroxyindoleacetic acid(5HIAA) in the tryptophan diet fed animals were higher than those of the normal diet fed animals. Feeding tryptophan supplemented diet to stressed animal significantly decreased the levels of serum and brain tryptophan and 5HT levels. However, the amount of 5HIAA which is the metabolite of serotonin was increased in brain. Plasma corticosterone level was increased by the stress in both groups but the degree of this increase was smaller in high tryptophan fed animals. The relative numbers of CD8+ T cells, CD4+ T cells and B cells in spleen were decreased in high tryptophan diet fed and stressed animals compared to control diet fed and no stressed animals. CD8+ T cells decreased more than CD4+ T cells. The decrease of CD8+ T cells in high tryptophan fed and stressed animals was similar to that in high tryptophan fed animals or normal diet fed and stressed animals. Stress and tryptophan supplement acted synergistically to decrease the number of B cells. This study suggests that stress and tryptophan supplement could modify the number of lymphocyte cells, and indicates that the interaction of stress and tryptophan supplement on immune fuction depends on the types of immune cells.

후 시상하부의 심혈관계 조절기전(I): 흰쥐 후 시상하부에서 세포외액의 모노아민과 그 대사체들에 대한 생체내 미세투석법에 의한 연구

  • 심혈관계의 중추 조절작용과 고혈압 치료제의 작용기전을 규명하기 위한 연구의 한 단계로서 생체내 미세투석법(in vivo microdialysis)을 이용하여 후 시상하부의 국소부위에서 catecholamine들과 serotonin 및 그 대사체들의 세포와 농도의 변화를 정상혈압 횐쥐 (WKY)에서 관찰하고, 자연발생성 고혈압 횐쥐(SHR)에서 이들의 기준치를 비교하였다. 뇌정위 고정장치에 의해서 미세투석관을 후 시상하부에 위치시킨 후 링거액으로 관류하였다. 모노아민들과 그 대사체들은 고속액체 크로마토그라피와 전기화학 검출기률 이용하여 정량하였다. WKY에서 imipramine(50$\mu$M)의 관류에 의하여 norepinephrine의 농도가 유의하게 증가되었고, 또한 Pargyline (75mg/kg, i.p.)에 의하여 norepinephrine의 농도는 유의하게 증가된 반면에 DOPAC, HVA 및 5-HIAA는 감소하였다. WKY와 SHR에서 비교하였을때 norepinephrine, epinephrine, dopamine, serotonin의 농도는 차이가 없었으나 DOPAC, HVA, 5-HIAA의 농도는 SHR에서 유의하게 증가된 결과를 얻었다. 상의 결과들은 모노아민중에서 norepinephrine이 정상혈압 흰쥐의 후 시상하부에서 유리되는 주된 신경전달물질임을 시사한다. 또한 자연 발생성 고혈압 흰쥐의 후 시상하부에서 dopamine과 serotonin의 교체속도가 증가되었을 가능성이 큰 것으로 사료된다.

Simultaneous Determination of Catecholamines, Serotonin and Their Metabolites in the Biological Sample Using HPLC/ECD (생체 시료 중 카테콜 아민류, 세로토닌 및 대사물질들의 HPLC/ECD 동시 정량분석)

  • Simultaneous monitoring of catecholamines and serotonin with their appropriate extraction from the biological samples is required in order to understand thoroughly the regulation of the central and peripheral nervous system. In the present research the segmented gradient elution with the solid phase extraction using a C18 cartridge rather than the previous isocratic elution with alumina extraction is successfully employed to determine norepinephrine (NE), epinephrine (E), dopamine (DA), serotonin (5HT), 3,4-dihydroxyphenylacetic acid (DOPAC) and 5-hydroxyindoleacetic acid (5HIAA) simultaneously within 20 minutes using 3,4-dihydroxyhydrocinnamic aicd as the internal standard (IS). Linearities were obtained in the concentration range between $5{\times}10^{-6}M$ and $1{\times}10^{-4}M$ for all 7 compounds with detection limits of 0.6~1.9 ${\mu}M$. The present HPLC/ECD method yielded reasonable accuracy (relative error; -1.4~1.1%) and precision (relative standard deviation; 0.4~1.9%) for 9 measurements of the standard solution consisting of NE, E, DA, 5HT, DOPAC and 5HIAA compounds. Recoveries of catecholamines, serotonin and their metabolites from human serum were in the range of 57%~86%. While the concentrations of NE and 5HT in the serum of normal Sprague-Dawley rat were found as $1.4{\times}10^{-6}M$ and $2.6{\times}10^{-6}M$, respectively, the contents of NE and 5HT in the serum of the stressed rat were increased 5.6 times and 1.4 times more, respectively.

  • Aloe(Aloe arborescens M$_{ILL}$) has been used as a home medicine for the past several thousand in the world, and has been studied on anti-bacterial and anti-fungal activities, hypotension, atherosclerosis, myocardiac infartion, apoplexy, diabetes as a chronic digenerative disease, tumors, gastrointestinal tract, liver and pancreas' diseases, and genitourinary tract etc. SAMP8 as a learing and memory impairment animal model were fed basic and/or experimental diets with 1.0% freezing dried(FD)-aloe for 8 months. The passive avoidance tests such as acqusition trial and retention test were significantly higher in aloe group than in control group. Grading score of senescence resulted in a marked decreases in aloe group compared with control group. Acetylcholinesterase(AChE) activity was remarkably increased in aloe group compared with control group. Neurotransmitters such as dopamine(DA) and serotonin(5-HT) almost did not change by the feeding of aloe-added diet, but their metabolites such as homovanillic acid(HVA) and 5-hydroxy-indole acetic acid(5-HIAA) in aloe group were significantly increased compared with control group. Therefore, the ratios of HVA/DA and 5-HIAA/5-HT as a ratio of metabolite on neurotransmitter were significantly increased by the feeding of aloe-added diet. These results suggest that aloe vara may be activated acetylcholinesterase, the metabolite of neurotransmitter, and ratios of metabolite on neurotransmitter, resulting ina greater prevention of learning and memory impairments such as Alzheimertype dementia.

Male rats were received intraperitoneal (i.p.) administration of CdCl2 (0.6 mg/kg/d) for 21 days and sacrificed 7 days after the last administration. Concentrations of norepinephrine (NE), dopamine (DA) in striatum and serotonin (5-HT), 5-hydroxyindole acetic acid (5-HIAA) in cortex were measured by HPLC. There were significant decreases of NE, DA, 5-HT and 5-HIAA in Cd intoxicated rats (P < 0.05), while pretreatment with AESC (20 mg/kg/d or 60 mg/kg/d, p.o., 30 min before $CdCl_2$) greatly inhibited the decrease of monoamine transmitters, respectively (P < 0.05). Also, AESC significantly increased the reduction of glutathione contents and superoxide dismutase activities in cortex induced by $CdCl_2$. These results suggest that AESC ameliorates Cd-induced depletion of monoamine neurotransmitters in brain through its antioxidant activity.

Antidepressant effect of Licium chinense Mill. and its influence on indoleamine and its metabolite of depression model rats (구기자의 항우울효과 및 indoleamine에 미치는 영향)

  • Depression is very common mental disorder, so many people suffer from it, which makes the treatment of depression important. Many drugs to treat depression were developed and being prescripted. But they have a lot of side effects, so it needs to develop drugs without side effects or with less side effects. Herbal medicines have been used to treat not only physical disorder but also mental disorder and it has been reported that they have less side effects. Therefore, there is the need to discover and use herbal medicine with antidepressant effect. The purpose of this study was to reseach Antidepressant effect of Licium chinense Mill. and its influence on serotonin and its metabolite of depression model rats. We used 'forced swimming test(FST)' to know antidepressant effect of Licium chinense Mill. and HPLC to check the influence on serotonin and its metabolite(5-HIAA) of Licium chinense Mill. after rats' brains were divided into cerebral cortex, striatum, hypothalamus and hippocampus. The results were obtained as follows : In the study of antidepressant effect by 'forced swimming test(FST)' method, Licium chinense Mill. had a significant antidepressant effect. In the study of influence on serotonin and 5-HIAA by HPLC, Licium chinense Mill. mainly increased serotonin and 5-HlAA of cerebral cortex and striatum signigficantly among 4 parts of rat's brain above-mentioned. These results suggest that Licium chinense Mill. has antidepressant effect that may be related with the increase of serotonin and its metabolite as its mechanism, but more precise experiments will be need to prove their relation.

  • Depression is a sort of mental disorder which is very common. To treat depression, many drugs such as TCA, MAOI are developed and used. But they have a lot of side effects, so it needs to develop drugs without side effects or with less side effects. Herbal medicines have been used to treat diseases not only physical but also mental and have less side effects. therefore, it has been thoght the need to develop herbal medicine with antidepressant effect. The purpose of this study was to reseach antidepressant effect and influence on monoamines of chunwangboshimdan thought to have antidepressant according to ancient medical book- donguibogam- and recent reports. We used 'forced swimming test(FST)' to know antidepressant effect of chunwangboshimdan and HPLC to check the influence on monoamines and their metabolites(norepinephrine, dopamine, DOPAC, HVA, serotonin, 5-HIAA) of chunwangboshimdan after divided into cerebral cortex, striatum, hypothalamus and hippocampus. The results were obtained as follows: In the study of antidepressant effect by 'forced swimming test(FST)'method, chunwang boshimdan had a significant antidepressant effect. In the study of influence on monoamines by HPLC, chunwangboshimdan mainly increased dopamine among monoamines and their metabolites(norepinephrine, dopamine, DOPAC, HVA, serotonin, 5-HIAA) significantly in 4 parts of rat's brain above-mentioned. Calculated by turnover ratio formulae of monoamine, chunwangboshimdan has more results than Imipramine. These results suggest that chunwangboshimdan has antidepressant effect that is related with the increase of monoamines by suppressing their metabolism as its mechanism.

  • A role for brain serotonin(5-HT) in regulation of the HPA axis has been suggested but remains contoversial and poorly defined. The present experiments were designed to check kinetic parameters of 5-HT turnover in rat hypothalamus and remainder brain areas before and after stress and to test whether using various different pharmacologic approaches to stimulate or eliminate the control serotonergic system have any consistent effect on the stress-induced activation of HPA system. Steady state brain serotonin and 5-HIAA concentrations during 1 min ether stress were significantly elevated without significant rise in the levels of plasma corticosterone, which highly increased 2 minutes after stress. This suggests that the increase in serotonergic neuron activity precede that in HPA activity. Furthermore, during 1 ruin-ether stress or 30 min immobilization stress there is a marked increase in hypothalamic and remainder brain serotonin (5-HT) turnover or synthesis rates assessed by both the pargline/5-HT method and pargyline/5-HIAA method. The stress-induced corticosterone levels were increased by serotonin precursors and serotonin agonist in a dose-related fashion. The stress- induced corticosterone levels were highly elevated by L-tryptophan (100 mg/kg) and Potentiated by monoamine oxidase inhibitor, pargyline or serotonin agonist, 5-MeoDMT. The stress-induced elevation of corticosterone and 5-HT levels in rat brain were not significantly decreased by the administration of 5-HT synthesis inhibitor, PCPA and 5-HT neurotoxin, 5,7-DHT. However, the stress-induced elevation of corticosterone and 5-HT levels were decreased by the destruction of midline raphe nuclei. There was a strong positive correlation between plasma corticosterone and 5-HT concentrations changed by drugs which mainly manipulating 5-HT system in the hyhothalamus and in the remainder of the brain. In conclusion, our present data stongly suggest that 5-HT is an important key neurotransmitter involved in the stress-induced activation of the HPA system.
