p53 Mutations in Ewing's Sarcoma (유잉육종의 p53 돌연변이)

  • Purpose : The p53 tumor suppressor gene is one of the most frequently altered genes in human malignancies. We try to explore the implication of p53 alteration in Ewing's sarcoma. Materials and Methods : We analyzed 35 paraffin blocks to explore the deletion and sequence alterations of p53. Results : Quantitative PCR analysis showed that 2 tumors showed a homozygous deletion of the gene. Mutational analysis of exons 4 to 9 of p53 by PCR-SSCP revealed that 3 tumors carry sequence alterations in exons 5 or 8, and DNA sequencing analysis identified missense point mutations. Conclusion : Taken together, our data demonstrate that p53 is genetically altered in a small fraction of Ewing's sarcoma.

Analysis of Plasmid pJP4 Horizontal Transfer and Its Impact on Bacterial Community Structure in Natural Soil

  • Alcaligenes sp. JMP228 carrying 2,4­dichlorophenoxyacetic acid (2,4-D) degradative plasmid pJP4 was inoculated into natural soil, and transfer of the plasmid pJP4 to indigenous soil bacteria was investigated with and without 2,4-D amendment. Plasmid pJP4 transfer was enhanced in the soils treated with 2,4-D, compared to the soils not amended with 2,4-D. Several different transconjugants were isolated from the soils treated with 2,4-D, while no indigenous transconjugants were obtained from the unamended soils. Inoculation of the soils with both the donor Alcaligenes sp. JMP228/pJP4 and a recipient Burkholderia cepacia DBO 1 produced less diverse transconjugants than the soils inoculated with the donor alone. Repetitive extragenic palindromic-polymerase chain reaction (REP-PCR) analysis of the transconjugants exhibited seven distinct genomic DNA fingerprints. Analysis of 16S rDNA sequences indicated that the transconjugants were related to members of the genera Burkholderia and Pandoraea. Denaturing gradient gel electrophoresis (DGGE) analysis of PCR-amplified 16S rRNA genes revealed that inoculation of the donor caused clear changes in the bacterial community structure of the 2,4-D­amended soils. The new 16S rRNA gene bands in the DGGE profile corresponded with the 16S rRNA genes of 2,4-D­degrading transconjugants isolated from the soil. The results indicate that introduction of the 2,4-D degradative plasmid as Alcaligenes sp. JMP228/pJP4 has a substantial impact on the bacterial community structure in the 2,4-D-amended soil.

Synthesis, Spectral Characterization, Electron Microscopic Study and Influence on the Thermal Stability of Phosphorus-containing Dendrimer with a 4,4'-Sulphonyldiphenol at the Core

  • The divergent synthesis of novel phosphorus-containing dendrimer with 4,4'-sulphonyldiphenol at the core has been accomplished involving simple condensation reactions using $P(O)Cl_3$, $P(S)Cl_3$, 3-amino-phenol, 3-hydroxy-benzaldehyde, and 2-butyn 1, 4-diol. The final compound was a Schiff's base macromolecule possessing 4 imine bonds, 8 acetylenic bonds and 8 OH groups at the periphery. The structures of intermediate compounds were confirmed by IR, NMR ($^1H$, $^{13}C$ and $^{31}P$), LC-Mass and C, H, N analysis. The structure of the final dendrimer (5) was confirmed by IR, NMR ($^1H$, $^{13}C$ and $^{31}P$), MALDI-TOF-MS, and C, H, N analysis. The surface morphological characteristics of the final dendrimer were understood by Scanning Electronic Microscopic study (SEM). The thermal stability of the final dendrimer was studied by TGA/DTA analysis.

Analysis of the factors influencing customer satisfaction of delivery food (배달음식 이용고객의 만족도에 영향을 미치는 요인 분석)

  • Purpose: This study was performed to evaluate the importance and satisfaction of the selective attributes of delivery food and to analyze the factors affecting customer satisfaction. Methods: A total of 574 responses were collected from customers who had ordered delivery food for data analysis. Statistical analyses were conducted using the SPSS program (ver. 25.0) for frequency analysis, χ2 tests, t-test, factor analysis, Pearson correlation, multiple regression analysis, and Importance-Performance Analysis (IPA). Results: The importance of delivery food selection attributes was higher in the order of 'hygiene control level (4.72)', 'taste of food (4.64)', and 'delivery accuracy (4.40)'. Satisfaction assessment was higher in the order of 'taste of food (4.32)', 'delivery accuracy (4.26)', and 'convenience of using the delivery app (4.21)'. According to the results of IPA, items that were priorities for improvement were charges for delivery, discount offers, sufficient description of the menu, and rapid handling of customer complaints. On an average, overall customer satisfaction score of delivery food was 4.01 out of 5 points. Additionally, five satisfaction factors were extracted by exploratory factor analysis. According to the results of multiple regression analysis, quality of delivery platform (p < 0.001), quality of delivery service (p < 0.001), convenience and diversity (p < 0.001), quality of delivery food (p < 0.001), and health and safety (p < 0.001) had significant positive effects on overall customer satisfaction. Conclusion: To increase customer satisfaction among delivery food customers, restaurant or delivery platform managers should consistently improve not only the quality of the delivery platform but also the quality of the delivery food and service.

A Study on the Visual System of the Digital Periapical Images (디지털 치근단방사선영상에 관한 시각 특성 연구)

  • Objectives: The purpose of this study was to evaluate the optimal distance and angle of observers by modulation transfer functions(MTFs) and receiver operating characteristics(ROCs), Material and Methods: Digital periapical radiograms were taken from 43 patients who have dental diseases(19 patients : dental caries. 12 patients : periapical lesions, 12 patients : periodontal diseases). Segmental images(4×4cm) were evaluated by 4 MTFs and ROC analysis. Results: The optimal distance(magnification) using MTF by Mannos & Sakrison was 12.97. and those by Nill. Ngan and Rao were 8.39, 4.78. 5.84 respectively. The optimal distance obtained from 4 radiologists by ROC analysis was 32cm(Az value: 0.89). and it was 40cm(Az value: 0.78) from 4 non-radiologists. There were significant differences of Az values between 4 radiologists and 4 non-radiologists at 24. 32 and 40cm (P<0.05). No significant differences of optimal distances were observed using 4 MTFs among +20, +10, 0, -10, -20 degrees(P>0.05). The optimal angle obtained from 4 radiologists by ROC analysis was +20 degree(Az value: 0.91). and no significant differences of Az values were observed among +20, +10, 0, -10 and -20 degrees(P>0.05). The optimal angle obtained from 4 non-radiologists by ROC analysis was 0 degree(Az value : 0.81). and no significant differences of Az values were observed among +20, +10, 0, -10 and -20 degrees(P>0.05). And there was significant difference of Az value between 4 radiologists and 4 non-radiologists at +20 degree(P<0.05). but no significant differences of Az values were observed among +10, 0, -10 and -20 degrees(P>0.05).

Complete Nucleotide Sequence of Small Multidrug Resistance Plasmid pKH4 (Small Multidrug Resistance(smr) 플라스미드 pKH4의 염기서열 결정)

  • The complete nucleotide sequence of pKH4, a small multidrug resistance (smr) plasmid isolated from multidrug resistant Staphylococcus aureus SA5, was determined. Sequence analysis has revealed that pKH4 has two open reading frames for Rep and Smr proteins. The comparison of the amino acid sequence of Smr protein of pKH4 with those of other Smr proteins of various Staphylococcus showed that Smr protein of pKH4 is a new member of the SMR family.

P2P Network Simulation System for Performance Evaluation in Convergence Networks

  • P2P(peer to Peer) network is a distributed network architecture composed of participants that make a portion of their resources directly available to other network participants, without the need for a central server. Currently, convergence network industry using wired and mobile are grows rapidly. So P2P protocols will be used between mobile and wired network. But current P2P protocols are focused on the wired networks only and there are no simulators for performance analysis of mobile P2P. In this paper, we design a P2P simulation system for performance analysis of P2P protocols in mobile, wired and convergence networks. It is constructed by a well-known mobile network simulator and wired based P2P protocol simulator. Finally we have implemented a smart TV test-bed using our P2P test-bed for convergence networks.

Identification of a Domain in Yeast Chitin Synthase 3 Interacting with Chitin Synthase 4 by Two-Hybrid Analysis

  • It has been proposed that chitin synthase 3 (CHS3)-nediated chitin synthesis during the vegetative cell cycle is regulated by chitin synthase 4 (CHS4) of Saccharomyces cerevisiae. To investigate direct protein-protein interaction between the coding products of these two genes, a domain of Chs3p that is responsible for interaction with Chs4p was identified, using the yeast two-hybrid system. This domain of 54 amino acids, termed MIRC3-4 (Maximum Interacting Region of Chs3p with Chs4p), is well conserved among CHS3 homologs of various fungi. Some mutations in MIRC3-4 resulted in a decrease in the enzymatic activity and chitin contents. Chs3p carrying those mutations exhibited weak interactions with Chs4p, when assayed by the yeast two-hybrid system. Surprisingly, all the mutants were sensitive to Calcofluor regardless of changes in enzymatic activities or chitin contents. This report deals with a core region in MIRC3-4 that affects the interaction with Chs4p.

Proposed dynamic p-y curves on a single pile considering shear wave velocity of soil

  • The dynamic behavior of a single pile was investigated by using analytical and numerical studies. The focus of this study was to develop the dynamic p-y curve of a pile for pseudo-static analysis considering the shear wave velocity of the soil by using three-dimensional numerical analyses. Numerical analyses were conducted for a single pile in dry sand under changing conditions such as the shear wave velocity of the soil and the acceleration amplitudes. The proposed dynamic p-y curve is a shape of hyperbolic function that was developed to take into account the influence of the shear wave velocity of soil. The applicability of pseudo-static analysis using the proposed dynamic p-y curve shows good agreement with the general trends observed by dynamic analysis. Therefore, the proposed dynamic p-y curve represents practical improvements for the seismic design of piles.

Successful Factor Analysis of 540° Dwihuryeochagi to Apply Free Style Poomsae of Taekwondo (태권도 자유 품새에 적용하기 위한 540° 뒤후려차기의 성공요인 분석)

  • The purpose of this study was to provide fundamental information for success factors of techniques through kinematic analysis including coordination of lower extremities and landing stability according to the success and failure of $540^{\circ}$ Dwihuryeochagi in Taekwondo. Twenty Taekwondo athletes: ten success group (S, age: $22.3{\pm}1.8$ yrs, height: $172.1{\pm}5.4$ cm, body mass: $64.4{\pm}4.2$ kg) and ten failure group (F, age: $22.3{\pm}1.8$ yrs, height: $172.1{\pm}5.4$ cm, body mass: $64.4{\pm}4.2$ kg) participated in this study. Three-dimensional motion analysis using a system of 3 video cameras with a sampling of 60 fields/s was performed during the competition of $540^{\circ}$ Dwihuryeochagi. Motions were divided into five events: pivot foot landing (E1), pivot foot toe off (E2), COM max height (E3), kick impact (E4) and landing (E5). At E1, the stride width was greater for S than for F (p<.05) while the time was greater for S than for F during P4 (p<.05). At E4, knee angle was greater for S than for F (p<.05). At E5, hip angle was greater for S than for F (p<.05) while kick distance was greater for S than for F (p<.05). Furthermore, at P3, the time would be related to kicking velocity (p<.05), while at P4, the time, range of hip angle and knee angle would be related to kick distance (p<.05). At P1, COM horizontal velocity would be related to COM vertical velocity of P1 and P2 (p<.05). Based on the findings, success factors of $540^{\circ}\acute{y}$ Dwihuryeochagi were COM horizontal velocity of P1, COM vertical velocity of P2, the time, kick distance, velocity, angle of lower extremities and coordination of P3-P4.
