LC-MS에 의한 사철쑥에 존재하는 페놀성 화합물의 정성분석 (Qualitative Analysis of Phenolic Substances in Artemisia capillaris by LC-MS)

  • The herb of Artemisia capillaris in Chinese medicine is used to treat hepatic diseases. In this research, qualitative analysis was performed using a UPLC/Q-TOF-ESI-MS/MS method for rapid identification of phenolic substances from A. capillaris: three caffeoylquinic acids (chlorogenic acid, 3,5-di-O-caffeoylquinic acid, and 4,5-di-O-caffeoylquinic acid), three flavonoids (hyperoside, isorhamnetin 3-O-robinobioside and quercetin) and three prenylated coumarins (6,8-diprenylumbelliferone, cedrelopsin and osthol) were identified. The three prenylated coumarins have not been reported from A. capillaris.

곰취차로부터 라디칼 소거능을 갖는 Caffeoylquinic Acid류 화합물의 추출조건 (Extraction Conditions of Radical Scavenging Caffeoylquinic Acids from Gomchui (Ligularia fischeri) Tea)

  • 본 연구에서는 항산화 기능이 잘 알려진 곰취로부터 곰취의 소비 및 활용도 향상을 위하여 곰취차를 제조한 후 주요 항산화물질을 탐색하고, 이들 성분의 추출율 변화를 분석하였다. 우선 온라인 항산화 장치를 통하여 곰취 추출물의 주요 항산화 성분이 5-CQA, 3,4-DCQA, 3,5-DCQA 및 4,5-DCQA로 구성된 caffeoylquinic acid류의 화합물임을 확인할 수 있었다. 곰취차에서 이들 성분의 추출율 변화를 추출 온도, 시간, 용매 및 추출 방법 등에 따라 비교 분석하였는데, 물에서의 추출율은 어느 온도에서든지 10분의 분석시간 동안 증가하는 것을 관찰할 수 있었다. 특히 $8^{\circ}C$에서의 추출율도 매우 우수하였는데, $80^{\circ}C$ 추출율의 약 60%의 유효성분이 10분 동안 추출되는 것을 확인할 수 있었다. 에탄올 함량에 따른 곰취차의 추출율에서도 추출시간에 따라 추출율이 증가하였지만, 에탄올의 함량이 많아질수록 추출율은 감소하였는데, 특히 100% 에탄올 조건에서는 추출율이 현저히 감소하였다. 곰취차의 제조 과정 중 블렌칭 단계에서는 곰취분말로부터 약 60% 이상의 항산화성분이 물로 유출되는 것이 확인되었다. 3,5-DCQA의 경우에는 90% 이상의 손실을 나타내어, 곰취분말에서는 3,5-DCQA가 가장 많이 추출되는 반면 곰취차에서는 3,4-DCQA가 가장 추출율이 높은 항산화성분이었다. 한편 초음파 추출법은 일반 추출법에 비해 항산화 성분의 추출율에는 큰 영향을 주지 않았다. 본 연구를 통해서 곰취가 차로 만들어졌을 경우에 항산화 성분을 다량 함유하고 있고, 찬물에서도 쉽게 이용할 수 있는 건강기능차로서 그 효용성과 편리성이 매우 좋을 것으로 기대된다.

목통의 페놀성 화합물의 항산화 활성 및 HPLC-UV 동시 함량분석 (Anti-oxidant Activity of Phenolic Compounds and Simultaneous Contents Determination using HPLC-UV from the Akebiae caulis)

  • Akebiae caulis have been used in folk medicines for diuretic, menstrual pain, and diuretic pain. It has been also resolved nephritis and cystitis. In this study, we isolated three phenolic compounds from 70% methanol extract using the open column chromatography. These isolated compounds which were 2-(3,4-dihydroxyphenyl)-ethyl-${\beta}$-D-glucopyranoside, 5-O-caffeoylquinic acid, and syringoylglycerol 2-O-${\beta}$-D-glucoside were determined by physico-chemical apparatus. Furthermore, we conducted DPPH and ABTS assay in order to screen the antioxidant activity of isolated three compounds. Also, we developed a rapidly HPLC-UV analysis method of two phenolic compounds (2-(3,4-dihydroxyphenyl)-ethyl-${\beta}$-D-glucopyranoside, 5-O-caffeoylquinic acid) for evaluating the Akebiae Caulis collected 30 samples from different regions. From the experiments, all three isolated compounds showed the significant antioxidant activity. We suggested that the content criteria of marker compounds were shown by a simple and rapid HPLC-UV method. The contents respectively were 0.009% (2-(3,4-dihydroxyphenyl)-ethyl-${\beta}$-D-glucopyranoside) and 0.036% (5-O-caffeoylquinic acid).

참느릅나무(Ulmus parvifolia) 잎 추출물 분석 및 안정성 평가 (Analysis and stability test of the extracts from Ulmus parvifolia leaves)

  • Ulmus parvifolia 잎의 약리학적 지표 성분을 제시하기 위해 간단하고 신뢰할 수 있는 HPLC 분석방법을 개발하였다. 지표 성분들은 NMR 및 UPLC-QTof-MS 분석에 의해 neochlorogenic acid (trans-5-O-caffeoylquinic acid, 5-CQA)과 chlorogenic acid (trans-3-O-caffeoylquinic acid, 3-CQA)로 구조 동정 되었다. HPLC를 이용하여 두개 화합물을 포함한 잎 추출물의 정성/정량 분석 방법을 평가하였다. 잎 30% 에탄올 추출물과 지표 성분의 안정성 테스트는 6개월 동안 평가되었다. 그러나 안정성 조사 기간 동안 추출물과 지표 성분들 각각의 함량에 대한 유의한 변화는 관찰되지 않았다.

한국산 및 Canada산 High Bush Blueberry 열매의 항산화 활성 비교 (Comparative Anti-oxidant Activity of Korean and Canadian High Bush Blueberry Fructus)

  • Blueberries (Ericaceae) are cultivated worldwide, and are used not only as foodstuff but also for relievement of eyestrain. Bluberry species representatively includes highbush blueberry (V. corymbosum L.), lowbush blueberry (V. angustifolium $A_{IT}$.), rabbiteye blueberry (V. ashei $R_{EADE}$), and bilberry blueberry (V. myrtillus L.). All of these species contain large amounts of phenolics and anthocyanins. In this regard, we isolated six compounds from Korea cultivated blueberry and identified as 3-O-caffeoylquinic acid (1), 5-O-caffeoylquinic acid (2), myricetin-3-O-${\beta}$-D-galactoside (3), quercetin-3-O-rutinoside (4), ethyl-3-O-caffeoylquinic acid ester (5), ethyl-5-O-caffeoyl quinic acid ester (6) by $^1H$-NMR, $^{13}C$-NMR and MS. Anti-oxidative activities of six compounds were verified by anti-oxidant assay such as DPPH, ABTS and Hypoxanthine/xanthine oxidase system. And then, anti-oxidant activities of Korea blueberry and Canadian were compared with each other. These results support that Korean blueberry has also the possibility to be potential supplementary material as healthy food like Canadian blueberry. Therefore, Korean blueberry can be used as a substitute of Canadian blueberry.

Analysis of polyphenolic metabolites from Artemisia gmelinii Weber ex Stechm. and regional comparison in Korea

  • Artemisia species are widely used as food ingredients and raw material in traditional medicine. However, to date, the secondary metabolites of Artemisia gmelinii Weber ex Stechm. have not been sufficiently investigated. The secondary metabolites of A. gmelinii, which was collected from representative regions in Chungbuk, Gangwon, and Gyeongbuk, were analyzed using ultra-performance liquid chromatography-quadrupole time-of-flight mass spectrometry (UPLC-QTof MS) combined with an unsupervised principal component analysis (PCA) multivariate analysis. In the loading scatter plot of PCA, significant changes in metabolites were observed between the regions, ten metabolites (3: 5-O-caffeoylquinic acid, 4: 4-O-caffeoylquinic acid, 8: trans-melilotoside, 12: quercetin 3-O-hexoside, 15: 3,4-O-dicaffeoylquinic acid, 17: 3,5-O-dicaffeoylquinic acid, 18: 4,5-O-dicaffeoylquinic acid, 19: syringaldehyde, 20: caffeoylquinic acid derivative, and 23: icariside II) were evaluated as key markers among twenty-five identified metabolites. Interestingly, the contents of the identified marker significantly differed between the three groups. This is the first study to report the presence of marker metabolites and their correlating geographical cultivation in A. gmelinii.

Antinocicepetive Effects of 3,4-Dicaffeoyl Quinic Acid of Ligularia fischeri var. spiciformis

  • The plant Ligularia fischeri var. spiciformis (Compositae) is a candidate for available functional foods. It has been used to treat diabetes mellitus and rheumatoid arthritis. We have reported the isolation of a new eremophilanolide named 6-oxoeremophilenolide and cytotoxic intermedeol together with the isolation of hydrophilic constituents, chlorogenic acid, 3,4-di-O-caffeoylquinic acie (3), and 5-O-[1-butyl]-3,4-di-O-caffeoylquinic acid. Compound 3 was again isolated by combination of silica gel- and ODS column chromatography for the anti-nociceptive action. Compound 3 and 4 were assayed in hot plate- and writhing tests in the rat. Although the three derivatives of caffeic acid exhibited significant anti-nociceptive effects at 10 mg/kg dose (i.p.),(activity potency: 4>3). These results suggest that compound 3 is responsible for at least rheumatoid arthritis, and caffeic acid moiety is the active moiety of dicaffeoylquinic acid.

Antioxidant Activities of Phenolic Derivatives from Dipsacus asper Wall. (II)

  • The six phenolic compounds isolated from the root of Dipsacus asper, 3,4-di-O-caffeoylquinic acid (1), methyl 3,4-di-O-caffeoyl quinate (2), 3,5-di-O-caffeoylquinic acid (3), methyl 3,5-di-O-caffeoyl quinate (4), 4-5-di-O-caffeoylquinic acid (5), methyl 4,5-di-O-caffeoyl quinate (6) were continuously evaluated for their antioxidant activity using superoxide radical scavenging and AAPH-mediated (LDL) oxidation assay. The results demonstrated that compounds 1 - 6 had remarkable antioxidant activities with the $IC_50$ values ranging from 12.0 to $2.8{\mu}M$ in superoxide radical scavenging. They also inhibited AAPH-mediated low-density lipoprotein LDL oxidation by the generation of thiobarbituric acid reactive substances (TBARS) with $IC_50$ ranging from 6.7 to $8.7{\mu}M$.

괴불나무(Lonicera maackii) 줄기의 성분 (Phytochemical Constituents of Lonicera maackii Stems)

  • Two phenylpropanoids, three caffeoylquinic acid deravatives and an iridoid were isolated from the stem of Lonicera macckii. On the basis of physico-chemical data, these compounds were identified as caffeic acid methyl ester(1), 5-caffeoylquinic acid n-butyl ester(2), methyl 3,4-dicaffeoyl quinate(3), 3,5-dicaffeoyl quinic acid n-butyl ester(4), loganin (5) and caffeic acid(6). All compounds were isolated for the first time from the stem of this plant.

향일규 잎의 에탄올 추출물에 대한 주요 성분 동시 정량분석 (Simultaneous Quantitative Analysis of Major Constituent of Ethanol Extract from Leaves of Helianthus annuus L.)

  • Helianthus annuus L. has been reported with various pharmacological activities such as antibacterial, antidiabetic, and antioxidant effects. According to recent studies, H. annuus L. is known to contain components such as phenolic compounds, flavonoids, alkaloids, sesquiterpenoids, and lignans. The seeds of H. annuus L. have been reported to contain chlorogenic acid and di-O-caffeoylquinic acid as major components. However, studies on the main components and content of leaves of H. annuus L. are still incomplete. Therefore, in this study, the contents of four major components of H. annuus L. were evaluated by simultaneous quantitative analysis with high performance liquid chromatography (HPLC)-diode array detector (DAD). The isolated four compounds Caffeoylquinic acid(CQA), 3,4-dicaffeoylquinic acid(3,4-DCQA), 3,5-dicaffeoylquinic acid(3,5-DCQA) and 4,5-dicaffeoylquinic acid(4,5-DCQA) were shown in a large linearity with a correlation coefficient (R2) of 0.99. In addition, as a result of intra-inter day analysis of four major compounds by the analysis method of this study, the accuracy of 88.46% or more and less than 112.85% and excellent precision of less than 3% were shown, the content analysis showed CQA (0.383±0.018 mg/g), 3,4-DCQA (0.282±0.017 mg/g), 3,5-DCQA (1.109±0.068 mg/g), and 4,5-DCQA (0.673±0.020 mg/g).
