ZAS3 represses NFκB-dependent transcription by direct competition for DNA binding

  • $NF{\kappa}B$ and ZAS3 are transcription factors that control important cellular processes including immunity, cell survival and apoptosis. Although both proteins bind the ${\kappa}B$-motif, they produce opposite physiological consequences; $NF{\kappa}B$ activates transcription, promotes cell growth and is often found to be constitutively expressed in cancer cells, while ZAS3 generally represses transcription, inhibits cell proliferation and is downregulated in some cancers. Here, we show that ZAS3 inhibits $NF{\kappa}B$-dependent transcription by competing with $NF{\kappa}B$ for the ${\kappa}B$-motif. Transient transfection studies show that N-terminal 645 amino acids is sufficient to repress transcription activated by $NF{\kappa}B$, and that the identical region also possesses intrinsic repression activity to inhibit basal transcription from a promoter. Finally, in vitro DNA-protein interaction analysis shows that ZAS3 is able to displace $NF{\kappa}B$ by competing with $NF{\kappa}B$ for the ${\kappa}B$-motif. It is conceivable that ZAS3 has therapeutic potential for controlling aberrant activation of $NF{\kappa}B$ in various diseases.

Presence of Foxp3-expressing CD19(+)CD5(+) B Cells in Human Peripheral Blood Mononuclear Cells: Human CD19(+)CD5(+)Foxp3(+) Regulatory B Cell (Breg)

  • Foxp3 is a transcript factor for regulatory T cell development. Interestingly, Foxp3-expressing cells were identified in B cells, especially in CD19(+)CD5(+) B cells, while those were not examined in CD19(+)CD5(-) B cells. Foxp3-expressing CD5(+) B cells in this study were identified in human PBMCs and were found to consist of $8.5{\pm}3.5%$ of CD19(+)CD5(+) B cells. CD19(+)CD5(+)Foxp3(+) B cells showed spontaneous apoptosis. Rare CD19(+)CD5(+) Foxp3(+) regulatory B cell (Breg) population was unveiled in human peripheral blood mononuclear cells and suggested as possible regulatory B cells (Breg) as regulatory T cells (Treg). The immunologic and the clinical relevant of Breg needs to be further investigated.

메밀가루와 밀가루 복합분의 건면 제조시험 (Dried Noodle Making of Composite Flours Utilizing Buckwheat and Wheat Flour)

  • 메밀을 제분하여 내층부터 외층까지 $B_1,B_2,B_3$$B_0$$(B_1:B_2:B_3=1:1:1)$으로 나누고 이들에게 제면적성을 향시키기 위하여 강력밀가루를 혼합하고 여기에 결착성개량제인 sodium alginate (SA)와 xanthan gum(XG)를 첨가하여 전면을 제조할 때의 제면적성을 알아보았다. 1) 메밀분 $B_1$에서 $B_2,B_3$로 될 수록 조회분, 조단백, 조지방의 함량이 현저하게 높았다. 2) Amylograph에 의한 메밀분 $B_1$의 최고 점도는 밀가루에 비하여 $4{\sim}5$배 정도로 높으며, $B_3$$B_1$보다는 낮으나 역시 밀가루보다 2배이상 높다. 또한 SA와 XG는 원료분의 최고점도를 더 높여주었다. 3) <메밀분 40+강력밀가루60>수준부터 밀가루 혼합수준을 높여야 상온에서의 전면제조가 가능하였으며, 이들의 조리특성도 메밀분의 혼합수준이 낮아질수록 밀가루면에 접근하였다. 4) 메밀건면의 제조에서 XG, SA를 $1.0{\sim}1.5$% 수준으로 첨가하면 면대형성, 조리면의 제성질등 제면적성을 개선하였다.

Bacillus subtilis와 Bacillus megaterium에서의 $\beta$-1,3-glucanase 유전자의 발현 (Expression of a $\beta$-1,3-Glucanase Gene from Bacillus circulans in B. subtilis and B. megaterium)

  • Bacillus circulans KCT3004 기원의 $\beta$-1,3-glucanase 유전자를 함유한 재조합 플라스미드 pLM460과 pUB110을 이용하여 shuttle 플라스미드 pMLS1180을 제작하고 Bacillus 세포에 이동.발현시켰다. pLMS1180으로 형질전환된 B. subtilis와 B. megaterium은 효율적으로 $\beta$-1,3-glucanase를 생산하였고, 이 효소들은 세포의 증식과 비례하여 생산되었다. 형질전환체가 생산하는 $\beta$-1,3-glucanase의 최대 활성을 유전자 공여 균주인 B. circulans와 비교하여 보니, B. subtilis는 14배, B. megaterium은 5배 정도의 높은 활성을 나타내었다. 그리고 대장균 형질전환체는 분비율이 7% 정도인데 반하여 B. subtilis 형질전환체는 생산된 효소를 전부, B. megaterium 형질전환체는 약 97%를 세포 외로 분비하는 것을 알 수 있었다. SDS-PAGE를 통해 대장균과 B. subtilis, B. megaterium에서 발현된 효소의 분자량을 분석해 보니 약 38,000으로 추정되었다. 또한, 이들 형질전환체가 생산하는 $\beta$-1,3-glucanase는 laminarin에 작용하여 주된 산물로서 laminaribiose (G2), laminaritriose (G3) 이상의 다양한 laminarioligosaccharide들을 생산함이 확인되었다. pLMS1180의 각 숙주 내에서이 안정성을 살펴본 결과 B.megaterium에서는 88%, 대장균에서는 75%, B. subtilis에서는 48%로 나타났다.

Superconducting properties of MgB2 superconductors in-situ processed using various boron powder mixtures

  • In this study, the effect of the size of B powder on the critical current density (Jc) of MgB2 prepared by an in situ reaction process was investigated. Various combinations of B powders were made using a micron B, ball-milled B and nano B powders. Micron B powder was reduced by ball milling and the milled B powder was mixed with the micron B or nano B powder. The mixing ratios of the milled B and micron or nano B were 100:0, 50:50 and 0:100. Non-milled micron B powder was also mixed with nano powder in the same ratios. Pellets of (2B+Mg) prepared with various B mixing ratios were heat-treated to form MgB2. Tc of MgB2 decreased slightly when the milled B was used, whereas the Jc of MgB2 increased with increasing amount of the milled B or the nano powder. The used of the milled B and nano B power promoted the formation MgB2 during heat treatment. In addition to the enhanced formation of MgB2, the use of the powders reduced the grain size of MgB2. The use of the milled and nano B powder increased the Jc of MgB2. The highest Jc was achieved when 100% nano B powder was used. The Jc enhancement is attributed to the high volume fraction of the superconducting phase (MgB2) and the large grain boundaries, which induces the flux pinning at the magnetic fields.

죽염된장의 항산화 및 HT-29 암세포에서 in vitro 항암 효과 (Antioxidative and Anticancer Effects of Doenjang Prepared with Bamboo Salt in HT-29 Human Colon Cancer Cells)

  • 죽염된장의 발효기간(3년, 6년)과 죽염의 구운 횟수($3{\times}$, $9{\times}$)에 따른 항산화 및 HT-29 암세포에서 in vitro 항암 효과를 측정하였다. 죽염된장의 발효 기간에 따른 pH 및 아미노태 질소, 암모니아태 질소 함량은 3B-S($3{\times}$, 삶은 콩)를 제외하고는 큰 차이가 없는 것으로 나타났다. 총 폴리페놀 함량은 6Y9B-D(6년, $9{\times}$)가 유의적으로 가장 함량이 높았다(P<0.05). DPPH free radical 소거 효과에서는 6Y3B-D(6년, $3{\times}$)는 $34.1{\pm}0.4%$, 6Y9B-D는 $38.2{\pm}2.5%$, hydroxyl radical 소거 효과 역시 6Y3B-D는 $80.8{\pm}0.9%$, 6Y9B-D는 $86.2{\pm}2.0%$로 높은 소거 효과를 나타내었다. HT-29 인체 대장암세포를 이용한 암세포 성장 억제 효과 역시 6Y3B-D는 $65.9{\pm}2.4%$, 6Y9B-D는 $72.8{\pm}1.7%$로 가장 높은 성장 억제율을 나타내었다. TNF-${\alpha}$, IL-6과 iNOS, COX-2 염증 유발 인자에서 6Y9B-D군이 유의적인 차이를 보이며 mRNA의 발현을 가장 많이 억제하는 것으로 나타났다. 장기간 숙성된 6년 죽염된장이 3년 죽염된장보다 항산화, 항암 효과가 뛰어났고, 3회 죽염보다 9회 죽염을 첨가한 된장이 천일염과 정제염을 혼합해 제조한 시판된장보다 항산화, 항암 효과가 더 우수했다.

The Effects of Vitamin B6 Deficiency on Stored Fuel Utilization During 3 days Fasting or 6 days underfeeding in Rats

  • The effects of vitamin B6 deficiency on energy utilization during fasting or underfeeding were studied in rats. Fifteen rats were fed a vitamin B6 deficient(-B6) diet and another 15 rats wee fed a control (+B6) diet. These rats were fed for 5 weeks with respective diet, and then subdivided into 3 groups : non-fasted group, fasted group, underfed group. Rats of the fasted group were fasted for 3 days and those of underfed group for 6 days. At the respective time (non-fast, 3 day-fast, 6 day-underfeed at 5 weeks), animals were sacrificed. Feed efficiency ratio of - B6 rats was significantly lower than that of +B6 rats. In - B6 rats, the liver and kidney weights were significantly heavier than those of +B6 rats but spleen and heart weights were not. In non-fasted group, liver protein and triglyceride level of - B6 rats were significantly higher than that of +B6 rats. After - B6 rats were fasted for 3 days, plasma free fatty acid level was significantly lower but liver glycogen level was higher than that of +B6 rats and muscle protein level of +B6 was decreased while that of - B6 was not changed. Vitamin B6 deficiency had little effect on the energy utilization with 6 days underfeeding. These results suggest that vitamin B6 deficiency may impair the utilization of stored fuel during fasting.

고압 자전 고온반응 합성법에 의한 B4C-Al2O3복합분말 제조 (Preparation of B4C-Al2O3 Composite Powder by Self-propagation High-temperature Synthesis(SHS) Process under High Pressure)

  • 정량의 B$_2$O$_3$/A1/C의 혼합분말을 화학노 대신 고압의 알곤 분위기를 사용하여 SHS 공정으로 B$_4$C-A1$_2$O$_3$ 복합분말을 제조하였다. 2B$_2$O$_3$+4A1+C=B$_4$C-2A1$_2$O$_3$의 반응식에 해당하는 B$_2$O$_3$(-100 메쉬), Al(-200 메쉬), C(-200 메쉬)의 분말을 2시간 건식 볼밀로 혼합한 후, 고온 고압의 SHS 반응기에 넣고 약 10기압의 알곤 분위기에서 점화하여 SHS을 일으켰다. 반응 생성물은 XRD 분석으로 안과 겉이 균일하게 반응이 일어났으며 반응 생성물로 화학노 사용시 동반되는 부산물 AlB$_{12}$C$_2$가 없는 B$_4$C-A1$_2$O$_3$ 복합 분말을 얻었다. 이 복합 분말은 SEM으로 보면 약 0.3~l $mu extrm{m}$ 크기의 결정이 모인 약 60~100$\mu\textrm{m}$ 크기이었다. 그러나 약 15기압을 사용하였을 때는 부분 소결이 일어나 15~25$\mu\textrm{m}$ B$_4$C 분말에 0.1~0.2$\mu\textrm{m}$의 알루미나가 분산되어 있는 고강도의 복합 분말이 생성되었다.

Pb(${Mg_{1/3}}{Nb_{2/3}}$)$O_3$계 고용체의 B자리 양이온 질서배열구조 (Ordering Structures of B-Site Cations in Pb(${Mg_{1/3}}{Nb_{2/3}}$)$O_3$-Based Solid Solutions)

  • Single phae Pb(Mg1/3Nb2/3)O3-based solid solutions, the Mg2+ of which are replaced by 20mol% of Ni2+, Zn2+, Cd2+, and the Pb2+ of which are replaced by 0∼20 mol% of La3+, were synthesized and their ordering structures of B-site cations were investigated by XRD and TEM. The B'-site cations (Mg2+, Ni2+, Zn2+, Cd2+) are disordered while these B'-site cations and the B"-site cations (Nb5+) are nonstoichiometrically 1:1 ordered within the ordered nano-domains dispersed in the Nb5+-rich disordered matrix. The charge imbalance between the B'-rich ordered nano-domains and the B"-rich disordered matrix are compensated by the doping of electron donor such as La3+, which enhances the degree of nonstoichiometric 1:1 ordering. For a given La3+ content, the degree of nonstoichiometric 1:1 ordering increases as the average ionic size difference between the B'-and B"-site cations increases, Ni2+

Moieties of Complement iC3b Recognized by the I-domain of Integrin αXβ2

  • Complement fragment iC3b serves as a major opsonin for facilitating phagocytosis via its interaction with complement receptors CR3 and CR4, also known by their leukocyte integrin family names, αMβ2 and αXβ2, respectively. Although there is general agreement that iC3b binds to the αM and αX I-domains of the respective β2-integrins, much less is known regarding the regions of iC3b contributing to the αX I-domain binding. In this study, using recombinant αX I-domain, as well as recombinant fragments of iC3b as candidate binding partners, we have identified two distinct binding moieties of iC3b for the αX I-domain. They are the C3 convertase-generated N-terminal segment of the C3b α'-chain (α'NT) and the factor I cleavage-generated N-terminal segment in the CUBf region of α-chain. Additionally, we have found that the CUBf segment is a novel binding moiety of iC3b for the αM I-domain. The CUBf segment shows about a 2-fold higher binding activity than the α'NT for αX I-domain. We also have shown the involvement of crucial acidic residues on the iC3b side of the interface and basic residues on the I-domain side.
