3T3-L1 지방세포에서 초과 에탄올 추출물의 항비만 효과 (Anti-obesity effect of Amomum taso-ko ethanol extract in 3T3-L1 adipocytes)

  • 본 연구에서는 초과 에탄올 추출물을 이용하여 3T3-L1 지방전구세포에서 지방세포로의 분화 연구를 통해 항비만 활성을 확인하고자 하였다. 초과 에탄올 추출물에 의한 지방세포 분화 억제 활성 및 지방형성에 미치는 영향을 확인하기 위해 3T3-L1 지방전구세포에 분화를 유도하면서 추출물을 농도별로 처리하였고, 그 결과 초과 에탄올 추출물은 3T3-L1 지방전구세포에서 지방세포로의 분화를 농도에 의존적으로 감소시켰다. 이 같은 활성에 대한 작용기전을 알아보기 위해 지방세포 분화에 중요한 역할을 담당하고 있는 peroxisome proliferator-activated receptor ${\gamma}$ ($PPAR{\gamma}$)와 CCAAT-enhancer-binding protein ${\alpha}$ ($C/EBP{\alpha}$), 그리고 지방세포 분화에 관여하는 유전자들의 활성을 확인해 보았다. 실험 결과 초과 에탄올 추출물은 $PPAR{\gamma}$$C/EBP{\alpha}$의 유전자 및 단백질 발현을 유의적으로 감소시켰고 지방세포 분화에 관여하는 adipogenesis marker 유전자들의 발현을 억제 시켰다. 따라서 초과 에탄올 추출물은 지방분화 및 adipogenesis 억제 활성을 통해 항비만 소재로의 가능성이 있을 것으로 기대한다.

통영바다목장에 서식하는 조피볼락, Sebastes schlegeli의 연령과 성장 (Age and Growth of Black Rockfish, Sebastes schlegeli in the Tongyeong Marine Ranching Area in Korea Waters)

  • 본 연구는 2001년 7월부터 2004년 5월까지 29개월동안 매월 경상남도 통영 바다목장해역에서 채집된 총 1,173마리의 조피볼락의 이석을 사용하여 연령과 성장을 연구하였다. 윤문은 불투명대에서 투명대로 이행되는 7월에 형성되었다. 주성숙기는 1월이며 산출시기는 2~5월 사이로 조피볼락의 초륜이 형성되는 시간은 주성숙기인 1월부터 이듬해 7월까지 약 1.5년으로 사료된다. 추정된 조피볼락의 von Bertalaffy length식과 weight growth equation식은 암컷은 $L_t=48.45(1-e^{-0.2139(t+0.4313)})$, $W_t=1,837.93(1-e^{-0.2139(t+0.4313)})^{3.02}$이었으며 수컷은 $L_t=49.32(1-e^{-0.1775(t+0.7403)})$, $W_t=1,887.83(1-e^{-0.1775(t+0.7403)})^3$였다.

Acinetobacter marinus sp. novo and Acinetobacter seohaensis sp. nov., Isolated from Sea Water of the Yellow Sea in Korea

  • Two Gram-negative, nonmotile, coccobacilli, SW-$3^T$ and SW-$100^T$, were isolated from sea water of the Yellow Sea in Korea. Strains SW-$3^T$ and SW-$100^T$ contained ubiquinone-9 (Q-9) as the predominant respiratory lipoquinone and $C_{18:1}\;{\omega}9c$ and $C_{16:0}$ as the major fatty acids. The DNA G+C contents of strains SW-$3^T$ and SW- $100^T$ were 44.1 mol% and 41.9 mol%, respectively. A neighbor-joining tree based on l6S rRNA gene sequences showed that the two isolates fell within the evolutionary radiation enclosed by the genus Acinetobacter. Strains SW-$3^T$ and SW-$100^T$ exhibited a l6S rRNA gene similarity value of 95.7% and a mean DNA-DNA relatedness level of 9.2%. Strain SW-$3^T$ exhibited l6S rRNA gene sequence similarity levels of 93.5-96.9% to the validly described Acinetobacter species and fifteen Acinetobacter genomic species. Strain SW-$100^T$ exhibited l6S rRNA gene sequence similarity levels of less than 97.0% to the other Acinetobacter species except Acinetobacter towneri DSM $14962^T$ (98.0% similarity). Strains SW-$3^T$ and SW-$100^T$ exhibited mean levels of DNA-DNA relatedness of 7.3-l6.7% to the type strains of some phylogenetically related Acinetobacter species. On the basis of phenotypic, phylogenetic, and genetic data, strains SW-$3^T$ and SW-$100^T$ were classified in the genus Acinetobacter as two distinct novel species, for which the names Acinetobacter marinus sp. novo (type strain SW-$3^T$=KCTC $12259^T$=DSM $16312^T$) and Acinetobacter seohaensis sp. novo (type strain SW-$100^T$=KCTC $12260^T$=DSM $16313^T$) are proposed, respectively.

iMBR 공정을 이용한 수산물가공폐수 처리에 관한 실증적 고찰 (The Practical Study for the Treatment of Fish Processing Saline Wastewater Using Immersed MBR)

  • 본 연구는 실제 운영되는 수산물 가공 산업폐수의 immersed MBR (iMBR)공정을 이용한 폐수처리시설 운영 결과에 대한 실증적 고찰을 수행한 것이다. 수산물 가공 산업의 특성상 일별, 월별 유량 변동이 심하여 유량조정조의 설계 및 운전방법이 중요하며, 유량조정조 교반시 포기식 교반을 적용하면 산발효 방지를 통하여 후속 응집/부상 공정의 약품비 절감이 가능하다. 동 현장은 유량조정조, 가압부상조, iMBR을 거쳐 방류하며, 이때 가압부상조를 거쳐 iMBR로 유입되는 BOD, $COD_{Mn}$, SS, T-N, T-P의 농도는 2,291 mg/L, 530 mg/L, 38 mg/L, 256.8 mg/L, 13.5 mg/L으로 나타났다. 수산물 가공 폐수와 같이 고농도의 염이 함유된 폐수의 생물학적 처리는 슬러지의 침강성과 관계없는 침지식 중공사막을 이용한 iMBR 공법을 적용하는것이 바람직한 것으로 나타났다. iMBR 공정의 주요 에너지 소모 요인인 공기세정에 대한 운영 값의 검토 결과 SADm값이 $0.31m^3/hr{\cdot}m^2$ membrane area이었으며, SADp값은 $26.5m^3/hr{\cdot}m^3$ permeate으로 상용화된 평막 대비 월등히 에너지 효율이 우수한 것으로 나타났다. 무산소, 혐기, 호기 탈기조로 구성된 침지식 중공사막이 결합된 iMBR 공정에서 막오염 지표인 Normalized TMP와 온도, MLSS 등을 비교 분석한 결과 F/M비가 0.08~0.10 gBOD/gMLSS에서 임계 F/M 값을 나타냈다. 생물반응조에서의 BOD, $COD_{Mn}$, SS, T-N, T-P의 처리수질은 각각 1.8 mg/L, 11.0 mg/L, 1.1 mg/L, 11.0 mg/L, 0.24 mg/L으로 운전되었으며, 제거율은 99.9%, 97.9%, 96.3%, 95.7%, 97.8%으로 나타났다.

Effect of Sodium Selenite and Zinc-L-selenomethionine on Performance and Selenium Concentrations in Eggs of Laying Hens

  • The objective of this study was to determine the effect of sodium selenite and zinc-L-selenomethionine on performance and egg Se concentration in laying hens. Two hundred and twenty-four CP Browns aged 71 weeks were divided according to a $2{\times}3$ factorial in a completely randomized design. One more group without additional Se supplementation was used as a negative control. Each treatment consisted of four replicates and each replicate contained eight laying hens. The dietary treatments were T1: basal diet; T2, T3 and T4: basal diets plus 0.3, 1.0 and 3.0 mg Se from sodium selenite/kg, respectively; T5, T6 and T7: basal diets plus 0.3, 1.0 and 3.0 mg Se from zinc-L-selenomethionine/kg, respectively. The findings revealed that feed conversion rate/kg eggs, egg production, egg weight, Haugh units and eggshell thickness were not affected by source and level of Se (p>0.05). Increasing level of dietary Se significantly increased (p<0.05 the Se content of eggs. Zinc-L-selenomethionine markedly increased p<0.05 egg Se concentration as compared with sodium selenite. The results indicated that Se source did not influence performance of laying hens. However, zinc-L-selenomethionine increased p<0.05 egg Se concentration more than sodium selenite.

3T3-L1 지방세포주에서 포도당 수송에 미치는 $CdCl_2$의 영향 (Effects of Cadmium on Glucose Transport in 3T3- L1 adipocytes)

  • Cadmium is well known as a toxic metal and has insulin mimicking effects in rat adipose tissue. This study was undertaken to investigate the effect of CdCl₂ on glucose transport and its mechanism in 3T3 - L1 adipocytes. CdCl₂ exhibits respectively 2.2 and 2.8 fold increases in the 2-deoxyglucose uptake when exposed to 10 and 25 μM of CdCl₂ for 12 hr. To investigate the stimulating mechanism of glucose transport induced by CdCl₂. Wortmannin and PD98059 were used respectively as PI3K inhibitor and MAPK inhibitor, which did not affect 2-DOG uptake. This results suggest that induced 2-deoxy-(l-3H)-D-glucose (2-DOG) uptake by CdCl₂ may not be concerned with the insulin signalling pathway. Whereas nifedipine, a calcium channel blocker inhibited the 2- DOG uptake stimulated by CdCl₂. In addition, we also measured the increased production of Reactive oxygen substances (ROS) and glutathione (GSH) level in 3T3-L1 adipocytes to investigate correlation between the glucose uptake and increased production of ROS with H2DCFDA. CdCl₂ increased production of ROS. Induced 2-DOG uptake and increased production of ROS by CdCl₂ were decreased by N-acetylcystein (NAC). And L-buthionine sulfoximine (BSO) a potent inhibitor of γ-GCS, decreased of 2-DOG uptake. Also NAC and BSO changed the cellular GSH level, but GSH/GSSG ratio remained unchanged at 10, 25 μM of CdCl₂.

Effects of Panicum miliaceum L. extract on adipogenic transcription factors and fatty acid accumulation in 3T3-L1 adipocytes

  • The dietary intake of whole grains is known to reduce the incidence of chronic diseases such as obesity, diabetes, cardiovascular disease, and cancer. To investigate whether there are anti-adipogenic activities in various Korean cereals, we assessed water extracts of nine cereals. The results showed that treatment of 3T3-L1 adipocytes with Sorghum bicolor L. Moench, Setaria italica Beauvois, or Panicum miliaceum L. extract significantly inhibited adipocyte differentiation, as determined by measuring oil red-O staining, triglyceride accumulation, and glycerol 3-phosphate dehydrogenase activity. Among the nine cereals, P. miliaceum L. showed the highest anti-adipogenic activity. The effects of P. miliaceum L. on mRNA expression of peroxisome proliferator-activated receptor-${\gamma}$, sterol regulatory element-binding protein 1, and the CCAAT/enhancer binding protein-${\alpha}$ were evaluated revealing that the extract significantly decreased the expression of these genes in a dose-dependent manner. Moreover, P. miliaceum L. extract changed the ratio of monounsaturated fatty acids to saturated fatty acids in adipocytes, which is related to biological activity and cell characteristics. These results suggest that some cereals efficiently suppress adipogenesis in 3T3-L1 adipocytes. In particular, the effect of P. miliaceum L. on adipocyte differentiation is associated with the downregulation of adipogenic genes and fatty acid accumulation in adipocytes.

Effect of Rare Earth Elements on Proliferation and Fatty Acids Accumulation of 3T3-L1 Cells

  • The present study including two experiments was designed to determine the effect of media containing different rare earth elements (REE) on proliferation and fatty acids accumulation in 3T3-L1 cell cultures. In Experiment 1, 3T3-L1 preadipocytes in 96-well plates ($1.5{\times}10^4cells/ml$) were cultured with Dulbecco's modified Eagle's medium (DMEM) containing 10% fetal bovine serum (FBS) for 24 h. Then the media were changed to the following 10 different media for 48 h: DMEM containing 10% FBS for the control; the above media containing $5{\mu}M$, $10{\mu}M$ or $15{\mu}M$ of $LaCl_3$, $CeCl_3$ or the mixture of these REE chlorides. The proliferation rate of the cells was measured and compared by a non-isotope method-XTT method. In Experiment 2 the cells in 24-well plates ($1.5{\times}10^4cells/ml$) were cultured in DMEM containing 10% FBS for 7 days until confluent and then were changed to above DMEM containing dexamethasone, methyl-isobutylxanthine and insulin (DMI) for two days. Afterwards the media were changed to the 10 different media with REE supplements as in Experiment 1 and cultured for 6 days. The cells were then harvested for fatty acids analysis by gas chromatography. It was found that supplementation of La (5, 10 and $15{\mu}M$), Ce ($5{\mu}M$ and $15{\mu}M$) and the mixture REE (5, 10 and $15{\mu}M$) stimulated (p<0.05) the proliferation of 3T3-L1 preadipocytes (Experiment 1). In the differentiating 3T3-L1 cells supplementation of La ($5{\mu}M$ and $10{\mu}M$), Ce ($5{\mu}M$) and the mixture REE ($5{\mu}M$ and $15{\mu}M$) decreased (p<0.05) the concentration of monounsaturated fatty acids (MUFA) per $10^5cells$, while the supplementation of La ($5{\mu}M$), Ce ($5{\mu}M$) and the mixture REE ($15{\mu}M$) increased (p<0.05) the ratio of saturated fatty acids (SFA) to MUFA. These results indicate that the supplementation of REE to the media may affect proliferation, differentiation and lipogenesis rates of 3T3-L1 cells. However, the effect may depend upon the level or type of REE applied.

Phanerochaete chrysosporium PSBL-1을 이용한 축산폐수와 하수의 연계처리 (Combined Treatment of Livestock Wastewater with Sewage Using Phanerochaete chrysosporium PSBL-1)

  • 본 연구에서는 축산폐수와 하수의 연계처리 가능성을 연구하였다 축산폐수와 하수를 2:1로 혼합하고 응집제인 BF02와 응집보조제인 양이온 polymer인 C-210EL를 각각 2 mL, 100 mL씩 주입시 SS 97.6%, T-P 95%로 최적 제거효율을 보였다. Phanerochaete chrysosporium PSBL-1의 처리 특성을 살펴보기 위해서 전처리수를 축산폐수 원수에 대해 10배 희석되도록 하수와 혼합($\fallingdotseq$전처리수:하수=3:17)하였다. 이렇게 혼합된 폐수의 NBDCOD, $NH_3-N$, T-N의 제거율은 pH가 증가할수록 증가하였다. 즉, T-N 농도는 pH 6.7(5일 경과시), pH 8.0(3일 경과시), pH 10.0(1일 경과시)에서 각각 35 mg/L, 51 mg/L, 33 mg/L으로 축산폐수공공처리시설의 방류수허용기준 60 mg/L을 만족하였다. 또한 모든 pH(1일 경과시)에서 $COD_{Mn}$의 방류수허용기준 40 mg/L을 만족했다. V.A.(veratryl alcohol) 첨가시 V.A.를 첨가하지 않은 조건보다 유출수의 유기물 및 질소농도가 높게 측정되었다. $COD_{Mn}$은 C/N비(3:1)를 조절하지 않은 경우 1일 이후, T-N은 C/N비를 $4{\sim}6$으로 조절한 경우 2일 후에 축산폐수공공처리시설 방류수수질기준을 만족하였다.

Cottonseed Extract Improves the Function of Osteoblastic MC3T3-E1 Cells

  • We have investigated the effects of cottonseed extract on the proliferation, differentiation and lipopolysaccharide (LPS)-induced production of local factors in murine clonal osteoblastic MC3T3-E1 cells. Ethanol extract of cotton seed ($4{\sim}63{\mu}g/mL$) significantly increased the proliferatin of MC3T3-E1 cells (p<0.05). Moreover, cottonseed extract ($10{\sim}50{\mu}g/mL$) caused a significant elevation of alkaline phosphatase (ALP) activity and collagen content in the cells. Lipopolysaccharide (LPS) is a potent stimulator of bone resorption in inflammatory diseases. We examined the effect of cottonseed extract on the LPS-induced production of tumor necrosis factor a (TNF-$\alpha$) and nitric oxide (NO) in MC3T3-E1 cells. Treatment with cottonseed extract ($10{\sim}50{\mu}g/mL$) decreased the $5{\mu}g/mL$ LPS-induced production of TNF-$\alpha$ and NO in osteoblasts, suggesting that the antiresorptive action of cottonseed extract may be mediated by decrease in these local factors. This study suggests that cottenseed may contribute to antiresorptive action against osteoblastic cells, resulting in a beneficial effect in promoting the function of osteoblastic cells.
