  • Objectives : Diabetic nephropathy is one of the most common chronic complications of diabetes and a leading cause of end-stage renal failure in the world. Mesangial cell proliferation is known as the major pathologic features such as glomerulosclerosis and renal fibrosis. Wiryeongtang (WRT) is a well-known traditional herbal formula as therapeutic agents for chronic edema and dysuresia of renal homeostasis. In the present study, we investigated whether WRT inhibits high glucose (HG)-induced renal dysfunction by TGF-β/Smads signal regulation in cultured mesangial cells. Methods : Inhibitory effect of WRT (10-50 ㎍/ml) on HG-stimulated mesangial cells proliferation and dysfunction were evaluated by [3H]-thymidine incorporation, Western blot, and RT-qPCR.Results : WRT significantly decreased HG-accelerated thymidine incorporation in human renal mesangial cell in a dose-dependent levels. WRT induced down-regulation of cyclins/CDKs and up-regulation of CDK inhibitor, p21waf1/cip1 and p27kip1 expression. In addition, HG enhanced expression of dysfunction biomarker such as collagen IV and CTGF, which was markedly attenuated by WRT. WRT decreased TGF-β1 and Smad-2/Smad-4 expression, whereas increased Smad-7 expression under HG. Furthermore, WRT inhibited HG-induced inflammatory factors level such as ICAM-1 and MCP-1 as well as NF-κB p65 nuclear translocation and intracellular ROS production. Conclusions : These results suggested that WRT may alleviate mesangial proliferation and inflammation possibly involved in renal fibrotic process, further diabetic nephropathy through disturbing TGF-β1/Smad signaling and NF-κB/ROS pathway. Thus, WRT might prove to be effective in the treatment of renal dysfunction leading to diabetic nephropathy.

  • High levels of extracellular xylanase activity (211.79 IU/mg) produced by Paenibacillus sp. NF1 were detected when it was submerged-cultured. After three consecutive purification steps using Octyl-Sepharose, Sephadex G75, and Q-Sepharose columns, a thermostable xylanase (XynNF) was purified to homogeneity and showed a molecular mass of 37 kDa according to SDS-PAGE. The specific activity of the purified XynNF was up to 3,081.05 IU/mg with a 14.55-fold purification. The activity of XynNF was stimulated by $Ca^{2+}$, $Ba^{2+}$, DTT, and ${\beta}$-mercaptoethanol, but was inhibited by $Fe^{2+}$, $Zn^{2+}$, $Fe^{2+}$, $Cu^{2+}$, SDS, and EDTA. The purified XynNF displayed a greater affinity for oat spelt xylan with the maximal enzymatic activity at $60^{\circ}C$ and pH 6.0. XynNF, which was shown to be cellulose-free, with high stability at high temperature ($70^{\circ}C-80^{\circ}C$) and low pH range (pH 4.0-7.0), is potentially valuable for various industrial applications. The enzyme hydrolyzed oat spelt xylan to yield mainly xylooligosaccharides (95.8%) of 2-4 degree of polymerization (DP2-4). Moreover, the majority of the xylooligosacharides (DP2-4) products was xylobiose (61.5%). The thermostable xylanase (XynNF) thus seems potentially usefull in the production of xylooligosaccharides.

  • Background: The geographical origin of Panax ginseng Meyer, a valuable medicinal plant, is important to both ginseng producers and consumers in the context of economic profit and human health benefits. We, therefore, aimed to discriminate between the cultivation regions of ginseng using the stable isotope ratios of C, N, O, and S, which are abundant bioelements in living organisms. Methods: Six Korean ginseng cultivars (3-yr-old roots) were collected from five different regions in Korea. The C, N, O, and S stable isotope ratios in ginseng roots were measured by isotope ratio mass spectrometry, and then these isotope ratio profiles were statistically analyzed using chemometrics. Results: The various isotope ratios found in P. ginseng roots were significantly influenced by region, cultivar, and the interactions between these two factors ($p{\leq}0.001$). The variation in ${\delta}^{15}N$ and ${\delta}^{13}C$ in ginseng roots was significant for discriminating between different ginseng cultivation regions, and ${\delta}^{18}O$ and ${\delta}^{34}S$ were also affected by both altitude and proximity to coastal areas. Chemometric model results tested in this study provided discrimination between the majority of different cultivation regions. Based on the external validation, this chemometric model also showed good model performance ($R^2=0.853$ and $Q^2=0.738$). Conclusion: Our case study elucidates the variation of C, N, O, and S stable isotope ratios in ginseng root depending on cultivation region. Hence, the analysis of stable isotope ratios is a suitable tool for discrimination between the regional origins of ginseng samples from Korea, with potential application to other countries.

  • The error probability of QPSK and DPSK signals with diversity reception technique in m-distribution fading plus impulsive noise environments has been derived and the error probability is evaluated and compared with that in Gaussian noise environment. The error performance degrades as impulsive noise becomes strong and degree of degradation of signal performance in QDPSK signal is larger than that in QPSK signal. The diversity reception technique can improve the error performance not only in fading plus Gaussian noise environment but in fading plus impulsive noise environment. When diversity reception technique is used, the improvement of error performance attains about 10dB to 15dB in terms of CNR as compared with that in non diversity reception. Among diversity techniques the maximal ratio combining is must effective. When diversity reception and coding techniques are used together in impulsive noise plus Rayleigh fading environments, the improvement of error performance attains about 12dB to 15dB in terms of CNR as compared with that of only diversity reception technique case and the improvement of error perform- ance in RS coding attains about 2dB in terms of CNR as compared with that of BCH coding case.

  • Toothache is a common complaint in the dental office. Most toothaches have their origin in the pulpal tissues or periodontal structures1). These odontogenic pains are managed well and predictably by dental therapies. One of the most frequent encounters and most confusing phenomena with which the dental diagnostician must deal is the problem of referred pain. The most important step toward proper management of a toothache is to consider that the pain may not be of dental origin. And Patients with orofacial pain, especially those in chronic pain, present a diagnostic and management challenge for the practitioners. There are many structures in the head and neck that can produce heterotopic pains felt in the teeth and other structures. Once referred pain is suspected, the true source of the pain must be located in order to render effective therapy. With increased interest in temporomandibular disorders and orofacial pain, many studies of accurate diagnosis and differential diagnosis about orofacial pain have been established. The purpose of this paper is to present a case for pansinusitis which produced pain referral in teeth and mimicked the symptoms of migraine.

  • When routine behavior control is impossible, midazolam is often used for sedation, because it has wide margin of safety, relatively few side-effects and multiple route of administration. Although intramuscular administration of all administration route is frequently used, it is a major source of anxiety, discomfort, and trauma in children. To the contrary, oral administration of midazolam is easily administered and accepted by children. But, it's therapeutic drug concentration has not been established. The purpose of this study was to compare sedation effect and physiologic parameter of oral midazolam which palliate demerits of intramuscular administration in sedating young pediatric patients with intramuscular midazolam Twelve negative children, mean age 62.5 months, who needed at least two separate restorative visits, requiring local anesthesia participated in this study On every visit, one of the following 2 different sedative regimen was given : (1) 0.30mg/kg midazolam by intramuscular administration (2) 0.75mg/kg by oral administration. Physiologic parameter(oxygen saturation, heart rate) was recorded by ten procedure and behavior was videotaped and rated using Ohio State University Behavior Rating Scale and Automated Counting System by one investigator, blind to administration route The analyzed sedative effect of oral midazolam resulted in good sedative effect, comparing to intramuscular route, And there is no statistically difference between oral and intramuscular administration of midazolam (p>0.05).

  • To investigate and analyze the change in the forest community structure between non-attacked time(in 1980) and severly damaged time(in 1987) by pine gall midge, Thecodoplosis japonensis, four plots with five subplots were sampled at Cheongwon-gun of Chungcheungbuk-do. The total number of woody species per 0.2ha changed 32 in 1980 to 53 in 1987. According to the damage increasing, the importance value of the genus Quercus as a substituting species has sprung up. Especially the importance value of Quercus serrata and Q. aliena singnificantly increased in the heavily insect infested forest. As to changes of forest community structure during 7 years, diversity of investigation structure has increased. The similarity indices between 1980 and 1987 were 52.9% and 49.7% in the understory and shrub stratum, respectively.

  • In order to inspect growth responses of Fraxinus rhynchophylla Hance, Koelreuteria paniculata Laxmann, Quercus acutissima Carruther and Ulmus parvifolia Jacquin to NaCl treatment, NaCl solution was treated for four months with 0, 25, 50, 100 and 200 mM concentrations, then survival rate, change of relative growth rate, weight, dry weight and pigment content of leaf were investigated. According to NaCl treatment, pH and EC (electrical conductivity) of soil increased, and growth rates of four tree species fell apparently as treatment time became longer. U. parvifolia had the highest survival rate with 15% in the 200mM treatment, and the other three species withered in the treatment. Relative growth rate, weight and dry weight decreased when NaCl treatment time grew longer. The total chlorophyll declined after it rose to 60 days, and the total chlorophyll and carotenoid of the all species according to NaCl treatment did not change very much. With the result from anlayzing growth responses of four tree species to oxidative stress which occurs during NaCl treatment, U. parvifolia has the highest tolerance, followed by K. paniculata, F. rhynchophylla and Q. acutissima.
