3D Light-Sheet Fluorescence Microscopy of Cranial Neurons and Vasculature during Zebrafish Embryogenesis

  • Precise 3D spatial mapping of cells and their connections within living tissues is required to fully understand developmental processes and neural activities. Zebrafish embryos are relatively small and optically transparent, making them the vertebrate model of choice for live in vivo imaging. However, embryonic brains cannot be imaged in their entirety by confocal or two-photon microscopy due to limitations in optical range and scanning speed. Here, we use light-sheet fluorescence microscopy to overcome these limitations and image the entire head of live transgenic zebrafish embryos. We simultaneously imaged cranial neurons and blood vessels during embryogenesis, generating comprehensive 3D maps that provide insight into the coordinated morphogenesis of the nervous system and vasculature during early development. In addition, blood cells circulating through the entire head, vagal and cardiac vasculature were also visualized at high resolution in a 3D movie. These data provide the foundation for the construction of a complete 4D atlas of zebrafish embryogenesis and neural activity.

Preoperative CT Navigation of Perigastric Vessel Anatomy for Gastrectomy

  • The aim of this report is showing the case that we could give exact navigation of perigastric vessels for gastrectomy with 3D CTA. A 74-year-old male patient visited hospital with gastric cancer. Early gastric cancer, type IIb was found at stomach antrum great curvature side. Before surgery, he underwent 3D CT angiography. 3D volume rendering images and MIP images were made by post processing. He had replaced Lt. hepatic artery arising from Lt. gastric artery. Surgeon could get patient's specific vascular anatomy before surgery including surgically relevant anatomical distance and direction and could finish gastrectomy within 4 hours and just 53ml blood loss.

  • These studies were conducted in an attempt to investigate effects of 'Daeshiho-Tang' on the blood vessels, activities of liver enzyme and excretory action of bile juice in serum of $CCl_4-intoxicated$ rabbits, glucose and total cholesterol levels in serum of alloxan-induced diabetic rabbits. The result of these studies were summarized as follows; 1. Hypotensive ind vaso-dilatating actions due to vascular smooth muscle relaxation were noted in frogs, rabbits and dogs. 2. GOT, GPT and total cholesterol in serum of $CCl_4-intoxicated$ rabbits showed significant depressive effects. 3. Significantly increased biliary secretary effect was noted in $CCl_4-intoxicated$ rabbits. 4. Glucose and total cholesterol levels showed markedly decraese in alloxan-induced diabetic rabbits. Considering the experimental studies results, it is suggested that 'Daeshiho-Tang' has therapeutic efficacy to treat febrile diseses and metabolic diseases.

3차원 곡관에서의 굴절률 일치법을 이용한 맥동 유동의 PIV 측정 (PIV Measurement of Pulsatile Flows in 3D Curved Tubes Using Refractive Index Matching Method)

  • 3차원의 협착 혈관모델을 3D 프린터를 이용하여 제작하였다. 협착부는 관의 중심축에 대하여 대칭인 형태이며, 협착부가 0도인 직관과 10도로 굽어진 관인 두 가지 모델에 대하여 실험을 수행하였다. 협착모델 내부 속도장을 매질에 대한 왜곡 없이 측정하기 위하여 굴절률일치법을 이용하였다. 정량펌프를 사용하여 발생된 맥동유동은 펌프의 회전속도로 세 가지의 속도조건을 조절하였다. 비정상상태의 속도장은 time-resolved PIV 기법을 이용하여 측정되었다. 주기적인 와류의 생성과 이동은 관 내 최대속도 영역과 관련 있으며, 와류의 크기와 위치 및 대칭성은 레이놀즈수와 관의 기하학적 구조에 영향을 받음을 알 수 있었다. 곡선관에서는 협착부 하류에 재순환 영역이 관찰되며, 이는 혈류역학적 관점에서 혈전의 형성과 침착 가능성을 설명해준다.

Quantitative Analysis of Chemical Components of Hydrolysate from Silkworm Fed with Cudrania tricuspidata Leaves

  • The purpose of this study was to develop hydrolysate from silkworm (Cudrania Silkworm Fibroin Peptide; CSFP), a food containing components for improving blood vessel health. In general, Cudrania tricuspidata leaves contain about 5 times more rutin and 25 times more dihydroquercetin than mulberry leaves. 1-Deoxynojirimycine (1-DNJ), dihydroquercetin and rutin inhibit the activity of carbohydrate-digesting enzymes, inhibit blood lipid peroxidation, and regulate insulin secretion, which helps blood vessels to be healthy. When the diet-controlled silkworm was enzymatically hydrolyzed, it was confirmed that rutin content was about 8 times higher than that of the in general silkworm as a control. In the silkworm hydrolysate, CSFP, developed as a final food material, the active ingredients were 65 mg/kg for rutin, 3,328 mg/kg for DNJ, 0.43 mg/kg for dihydroquercetin, and 82,624 mg/kg for total polyphenol, which was confirmed through LC-MS/MS analysis. In conclusion, it was found that silkworms fed with C. tricuspidata leaves as a diet had more active components that can help control blood sugar and improve blood vessel health than silkworms fed with mulberry leaves.

메추리의 발달 중 배자 생식선에서 분리한 원시생식세포의 이동능에 대한 연구 (Studies on the Migratory Ability of Primordial Germ Cells from Embryonic Gonads at Different Developmental Stages in Quail)

  • Avian primordial germ cells (PGCs) originate from the epiblast and appear in the germinal crescent. These PGCs enter the developing blood vessels during stage 10∼12 (H&H), circulate in the blood stream, migrate into the developing gonadal anlage and differentiate into germ cells. However, it is not clear until when the migratory ability of PGC is maintained. This study was conducted to examine whether migratory ability is present in PGCs from the gonad at later embryonic developmental stages. In the present study, gonads were dissected from 5-, 6- and 10-day old quail embryos and treated with trypsin-EDTA. Gonadal PGCs (gPGCs) were purified by Ficoll-density-gradient-centrifugation and labeled with PKH26 fluorescent dye. The PKH26-labeled gPGCs were microinjected into the blood vessel of the recipient quail embryo. Manipulated recipients were incubated for 3 days, embedded in paraffin and sdctioned. The foreign gPGCs were detected by fluorescent and confocal laser microscopy. As a result, quail gPGCs, from 10, 6 and 5 day old embryos could migrate through the recipient blood stream at early stage and settle in the gonads. Thus, results suggest that gPGCs from upto 10-day old embryos keep properties seen in circulating PGC. Therefore, the PGCs of 10-day old embryonic gonads can be used for the tools of genetic manipulation.

Effect of Extreme Long-Distance Running on Hepatic Metabolism and Renal Function in Middle-Aged Men

  • The aim of this study was to investigate the impact of participation in the 622 km hyper-ultra-marathon on hepatic metabolism and renal function in middle-aged men. Healthy middle-aged male amateur ultra-marathoners between the ages of 40 and 60. Blood was collected at the pre-race, immediately after 300 km, 622 km hyper-ultra marathon race, 72 hours (3 day) and 144 hours (6 day) after the race, AST (aspartate aminotransferase), ALT (alanine aminotransferase), ALP (alkaline phosphatase), ${\gamma}$-GTP (gamma glutamyl transferase), T-Bil (total bilirubin), D-Bil (direct bilirubin), T-protein (total protein), albumin, uric acid, BUN (blood urea nitrogen), creatinne were analyzed. ALP was significantly increased at 300 km, 622 km, day 3 and day 6 than the pre-race. ${\gamma}$-GTP, T-protein, albumin, uric acid, BUN and creatinine were not significantly different between the distances and the recovery period respectively. AST and ALT were significantly increased at 300 km, 622 km, day 3 and day 6 than the pre-race, respectively (P<0.05) at day 3 and day 6 they showed significant decrease from 300 km and 622 km, respectively (P<0.05). T-Bil and D-Bil increased significantly at 300 km and 622 km, respectively (P<.05) and significantly decreased at day 3 (P<0.05) compared to the pre-race, at day 3 and day 6 they were decreased significantly than 300 km and 622 km, respectively (P<0.05). In conclusion, no disturbance of renal function was observed according to the distances and between the recovery period of 622 km hyper-ultra marathon race, but reversible hepatocyte function could be degraded and some hemolysis of blood vessels was induced.

Hologram Quantitative Structure-Activity Relationships Study of N-Phenyl-N'-{4-(4-quinolyloxy)phenyl} Urea Derivatives as VEGFR-2 Tyrosine Kinase Inhibitors

  • Vascular endothelial growth factor (VEGF) is an important signaling protein involved in angiogenesis, which is the formation of new blood vessels from pre-existing vessels. Consequently, blocking of the vascular endothelial growth factor receptor (VEGFR-2) by small molecule inhibitors leads to the inhibition of cancer induced angiogenesis. In this study, we performed a two dimensional quantitative structure activity relationship (2D-QSAR) study of 38 N-Phenyl-N'-{4-(4-quinolyloxy) phenyl} urea derivatives as VEGFR-2 inhibitors based on hologram quantitative structure-activity (HQSAR). The model developed showed reasonable $q^2=0.521$ and $r^2=0.932$ values indicating good predictive ability and reliability. The atomic contribution map analysis of most active compound (compound 7) indicates that hydrogen and oxygen atoms in the side chain of ring A and oxygen atom in side chain of ring C contributes positively to the activity of the compounds. The HQSAR model developed and the atomic contribution map can serve as a guideline in designing new compounds for VEGFR-2 inhibition.

뇌혈관 직경측정을 위한 3차원 회전 혈관조영술의 유용성 (Usefulness of 3D Rotational Angiography for Cerebral Vascular Diameter Measurement)

  • When measuring cerebrovascular with 3D rotational angiography, the accuracy was verified by comparing the actual size and measurement size, respectively. It is intended to help select therapeutic materials and instruments during cerebrovascular intervention by comparing the average error rates for measured values in the 3DRA and CTA methods by examining with protocols such as brain CTA, which are always performed in emergency situations. The mean error rate between the groups of measurers was ±3.655% for radiation technologist and ±3.331% for university students, and the mean error rate of the student group was within tolerance (±10%), and the independent sample T-test result t =0.879, p=0.394 (p>0.05) showed no statistically difference between the two. In addition, the average error rate measured by both groups by 3DRA was measured below ±5% within the tolerance error rate (±10%), and most of CTA was measured within the tolerance range (±10%), but showed an average error rate of up to 5.65%, and the independent sample T-test result was statistically more accurate than 3DRA. Both the 3DRA method and the brain CTA method for measuring cerebrovascular size could be accurately measured within tolerance, but it would be better to measure cerebrovascular blood vessels using a more accurate 3DRA method during cerebrovascular intervention.

장기도말법(臟器塗抹法)과 조직표본법(組織標本法)에 의(依)한 Babesia bigemina와 Babesia argentina의 형태학적감별(形態學的鑑別)에 관한 연구(硏究) (Studies on Morphological Differentiation of Babesia bigemina and Babesia argentina Infections Using Smears and Tissue Sections of Organs of Splenectomized Calves)

  • The smears and tissue sections of organs (liver, lung, kidney, heart, lymph-node, brain) were carried out to compare the detection of parasites in the splenectomized calves artificially infected with B. bigemina and B. argentina, respectively. The results obtained from this experiment were abstracted as follows: 1. In the calves infected with B. argentina the parasites were easily detected by the blood smears and tissue smears but, particularly, the detection of parasites in the kidneys and brain by tissue smears and tissue sections was greater useful. In this experiment, the packing of the parasite-infected red blood cells was found in the capillary vessels of the organs. 2. When the fresh infected-brain was kept at $5^{\circ}C$ refrigerator, Babesia argentina could be detected during 9 days from the brain smears. According to this results, we could do differentiate the parasites from the brain of the dead animal by the doubtful babesiosis in the field. 3. If we use the brain smears and brain sections in order to survey or detect Babesia infections of cattle in Korea, these methods could be worthwhile for the differential diagnosis of B. argentina and B. bigemina.
