Investigation of the Hydrolysis Characteristics of Fish Oil by Means of Aspergillus oryzae Lipase Lipolase-100T (Aspergillus oryzae 유래의 리파제 Lipolase-100T에 의한 물고기 기름의 가수분해 특성 규명)

  • Fish oil was hydrolyzed with Aspergillus oryzae lipase, Lipolase-100T. The hydrolysis characteristics of Lipolsae-100T were investigated. Lipolase-100T showed 1,3-positional specificity which hydrolyzed acyl chains combined on the 1 or 3 position of triglyceride into free fatty acids. Lipolase-100T represented another property that the saturated fatty acids composing the triglyceride were hydrolyzed more easily that the polyunsaturated fatty acids(PUFAs). n-3 PUFAs, such as C16:4, C20:5 and C22:6, were hardly hydrolyzed, so that the concentrations of those in the mixture of glycerides were increased according to hydrolysis time. Especially docosahexaenoic acid(DHA), C22:6 showed the highest increase in the concentration. This result explained that n-3 PUFAs were combined on 2-position of triglyceride. When the hydrolysis of fish oil with Lipolase-100T 0.4 wt% was performed for 120 hr, n-3 PUFAs wt% was increased to 50 wt% in the mixture of glycerides. This result was obtained due to the 1,3-positional specificity of Lipolase-100T and positional specificity of n-3 PUFAs.

Properties of Barley for Extrusion Processing (보리의 Extrusion 가공적성)

  • Using piston type extruder, barley flour was extruded at various processing conditions, The used variables were three shear rates (apparent shear rate 118, 534, $1169sec^{-1}$), four extrusion temperatures(90, 120, 150, $180^{\circ}C$) and three moisture contents (15, 25, 35%). The rheological properties and the extrudate quality were monitored in extrusion. Barley flour showed pseudoplastic behavior having average power law index 0.28 in used shear rate range. When viewed from general appearance, die swell, density, water uptake, rehydration swell and gelatinization degree of extrudate, $25{\sim}35%$ moisture and $120^{\circ}C$ temperature was suitable processing condition for noodle-like product, and 25% moisture and $150^{\circ}C$ temperature was good for snack or flake product. Moisture content of the extrudate can be pretty well estimated from energy balance at higher temperature and higher moisture content.

  • In this study, the experiment conditions for the variation of heat release rate in compartment space were constructed to analyze the effects of fire spread and the operation time of sprinkler in accordance with the heat loss of the sprinkler's heat element. The compartment composed of fire board (width = 0.3 m, height = 0.5 m, length = 3.0 m), are manufactured to measure the temperature distributions in the inner space, the mass loss rate and heat release rate during the experiment of N-heptane pool fire test. Also, the operation time of sprinkler is analyzed with the installation of sprinkler and C-factor using Fire Dynamics Simulator Ver.6 under the experiment conditions. The results show that the operation time of sprinkler, which has RTI $100(m{\cdot}s)^{0.5}$ operating temperature $70^{\circ}C$, is 30 s~60 s for C-factor = 0 and 1, 62 s~92 s for C-factor = 3, and 120 s over for C-factor = 5, respectively.

Effects of Extraction Temperature and Time on Antioxidant Activities of Rhus verniciflua Extract (추출 온도와 시간이 옻나무 추출물의 항산화활성에 미치는 영향)

  • Rhus verniciflua was extracted at different temperatures and times, and the extracts were evaluated for contents of total phenolics and flavonoids, and antioxidant activities using DPPH and ABTS radical scavenging activities. Extraction temperatures and times were 80, 100, 120, 140 and $160^{\circ}C$, and 1, 3 and 5 hr. The total phenolic and flavonoid contents as well as antioxidant activities by DPPH and ABTS increased as the extraction temperature increased. The total phenolic contents did not significantly increase at less than $100^{\circ}C$ with different extraction times. However, they were increased at 3 hr and decreased at more than $120^{\circ}C$. The total flavonoid contents reached a high value of 8.04 mg CE/g at $140^{\circ}C$. The antioxidant activity by DPPH increased as the extraction temperature increased and reached high values of 34.28 mg TE/g at $140^{\circ}C$. The total antioxidant activity by ABTS was similar to DPPH. These results presumed that the optimum extraction temperature and time in the process of Rhus verniciflua extraction are $140^{\circ}C$ for 3 hr.

Study on the Electromagnetic Property of (Mn, Zn)-Ferrite as the Condition of Heat-treatment (열처리 조건에 따른 (Mn, Zn)-ferrite의 전자기적 특성에 관한 연구)

  • 높은 투자율과 낮은 손실특성을 지닌(Mn, Zn)ferrite의 전자기적 특성은 소결공정과 소결분위기에 따라 상당히 다양하게 나타난다. 본 연구에서는(Mn, Zn)ferrite를 소결시킬 때의 승온속도와 냉각속도에 따른 초투자율 특성과 미세구조의 관계에 관하여 고찰하였다. 승온속도의 영향을 고찰하기 위하여 spinel ferrites 생성 및 결정입 성장 거동에 가장 중요한 온도인 800~120$0^{\circ}C$의 범위에서 속도를 변화시켰고, 냉각속도 영향을 살펴보기 위하여 1100~80$0^{\circ}C$ 온도범위를 택하였다. 열처리 온도범위 800~120$0^{\circ}C$에서 승온속도가 증가함에따라 초투자율이 7$^{\circ}C$/min까지는 증가했으나 그 이상의 승온속도에서는 오히려 초투자율이 감소하였다. 또한 1100~80$0^{\circ}C$ 온도범위에선 냉각속도가 증가될수록 초투자율이 감소하는 경향을 나타내었다. 치밀화 및 투자율을 높이기 위하여 첨가된 CaO, SiO2, V2O5의 분포상태를 분석한 결과, 이들 첨가물들은 입자와 입계를 따라 균일하게 분포하는 것으로 나타났다.

Phenolic Contents of Different Parts of Rhus verniciflua Stokes according to Extraction Conditions (추출조건에 따른 옻나무 부위별 Phenolics 함량)

  • This study investigated changes in phenolic contents of different parts of Rhus verniciflua Stokes (RV) according to extraction conditions. Bark and xylem parts of RV were extracted at 80, 100, 120, 140, and $160^{\circ}C$ for 1, 3, and 5 h, respectively. Major phenolic compounds (gallic acid, protocatechuic acid, fustin, fisetin, sulfuretin, and butein) of RV were analyzed. The gallic acid, fisetin, sulfuretin, and butein contents significantly increased as extraction temperature increased. Protocatechuic acid and fustin contents increased as increasing extraction temperature to $120^{\circ}C$ and decreased afterward. The gallic acid, protocatechuic acid, and butein contents of bark were higher than those of xylem extracts. The optimal extraction conditions of gallic acid, protocatechuic acid, fustin, fisetin, sulfuretin, and butein were $160^{\circ}C/3h$ (380.22 mg%), $120^{\circ}C/1h$ (9.25 mg%), $100^{\circ}C/3h$ (206.97 mg%), $140^{\circ}C/5h$ (93.84 mg%), $140^{\circ}C/5h$ (16.07 mg%) and $160^{\circ}C/5h$ (1.49 mg%), respectively. These results suggest that the optimum extraction temperature and time considering RV extraction yield and cost are $140^{\circ}C$ and 3 h, respectively.

Fffect of Wearing Diving Suit on Energy Metabolism and Diving Time Period in Korean Women Divers (한국 해녀에서 잠수복 착용이 작업시간 및 열대사에 미치는 영향)

  • Rectal temperature, skin temperature and oxygen consumption were measured in five women divers during diving under two conditions a) wearing a cotton bathing suit and b) wearing a diving suit and the following results were obtained. 1. The duration of the diving period was 30 min with the cotton bathing suit whereas it was above 120 min with the diving suit. 2. The average rectal temperature fell to $34.5^{\circ}C$ after 30 min of diving with the cotton bathing suit. On the other hand, with the diving suit, immediately after submersion, the rectal temperature rose about $3^{\circ}C$ and remained at about $39^{\circ}C$ throughout the diving period of 120 min. 3. Average mean skin temperature during diving was $11^{\circ}C$ and $21^{\circ}C$ with cotton bathing suit and with the diving suit, respectively. The mean body temperature fell to $25^{\circ}C$ with the cotton bathing suit while it reached $32^{\circ}C$ with the diving suit at the end of each diving period. 4. Oxygen consumption during diving was three fold of the pre-dive level with the cotton bathing suit but it was two fold with the diving suit. 5. Total extra heat loss was 323 kcal during 120 min of diving with the diving suit. On the contrary, with the cotton bathing suit, it was 528 kcal for 30 min, These results may suggest that the prolongation of the diving period with the diving suit is primarily due to maintenance of core temperature at more or less physiological level as a result of reduction in conductive heat loss from the body surface to the water and the Preferential distribution of blood in the core area.

강릉연안 토종 다시마의 양식 연구

  • 동해안의 강릉 연안에 자생하는 토종 다시마인 개다시마는 산업적으로 개발의 여지가 매우 크다. 그러나 천연 자원량에 의존하고 있는 가공산업은 해황에 따라 원료 공급이 좌우되는 불안정성을 가지고 있어서 이 식물의 산업적 생산량을 확보하기 위한 방안으로 대량양식의 필요성이 대두되고 있다. 본 연구는 동해안 토종 다시마인 개다시마의 종묘생산기술 개발을 위해서 항온배양기와 실내수조배양을 실시하고 바다 가이식 양성에 의한 환경적응성 실험을 수행하였다. 연구 방법은 2001년 10월에 강릉연안 수심 20m에서 분포하는 개다시마 군락의 모조로부터 유주자액을 받아 온도 5, 10, 15, 20, $25^{\circ}C$, 조도 40, 80, 120 $\mu$mol m$^{-2}$ s$^{-1}$의 조건에서 6주간 정치배양하였다. 유주자의 발아 및 아포체 생장 실험 이후 바다 가이식 및 양성실험은 2002년 1~2월까지 1개월간 실시하였다. 유주자의 발아 및 아포체 형성은 1$0^{\circ}C$, 120$\mu$mo1 m$^{-2}$ s$^{-1}$에서 배양 2주만에 가장 먼저 아포체의 발아가 관찰되었으며, 이후 15$^{\circ}C$와 5$^{\circ}C$에서 순차적으로 아포체의 발아가 이루어졌다. 2$0^{\circ}C$는 80, 120$\mu$mol m$^{-2}$ s$^{-1}$에서 소수의 아포체만이 관찰되었으며, $25^{\circ}C$에서는 아포체가 형성되지 않았다. 아포체는 1$0^{\circ}C$에서 가장 좋은 생장을 보였고, 15$^{\circ}C$와 1$0^{\circ}C$에서는 비슷한 생장을 나타냈으며, 2$0^{\circ}C$에서는 80$\mu$mol m$^{-2}$ s$^{-1}$에서만 아포체의 형성이 관찰되어 가장 낮은 생장을 보였다. 동일 온도에서는 조도에 따른 생장의 차이가 많이 났는데, 40$\mu$mol m$^{-2}$ s$^{-1}$에서 가장 좋은 생장을 나타냈다. 80과 120$\mu$mol m$^{-2}$ s$^{-1}$에서는 120$\mu$mol m$^{-2}$ s$^{-1}$이 약간 좋은 생장을 보였지만 큰 차이는 나지 않았다. 생장이 가장 좋은 조건은 1$0^{\circ}C$, 40$\mu$mol m$^{-2}$ s$^{-1}$으로 엽장 1.28mm까지 생장했다. 전체적으로 수심이 깊은 곳에 서식하는 특성상 저수온과 저조도에서 좋은 생장 경향을 보였다. 실내배양을 통해 얻은 엽장 약 1~2mm의 종묘를 2002년 1월말에 삼척 호산에 위치한 바다 양성장에서 종묘들과 함께 2주간 수심별로 가이식(수온 12$^{\circ}C$)을 실시한 결과, 수심 1m에서 좋게 나타나 엽장이 약 5mm정도 생장하였다. 수심 0m에서는 강한 조도에 의해 엽상체가 사그라드는 기형이 나타났으며, 수심 5m에서는 엽상체의 기형이나 생장의 차이는 없었지만, 색택이 약간 바랜 것을 볼 수 있었다. 종합적으로 볼 때 종묘의 관리나 생장에는 수심 1~2m가 양호할 것으로 생각된다. 바다 가이식을 통해 엽장 5mm의 종묘로 생장시킨 후 바로 본양성장에 씨줄감기를 하여 양성을 실시하였다 양성 1개월 후 엽상체는 약 2~3cm, 최대 4~5cm까지 생장하였다. 이를 통해 강릉연안 토종 다시마인 개다시마에 대한 종묘 생산과 대량 양성의 가능성을 확인하였다.

  • 광섬유 Fabry-Perot 간섭계를 센서로 하는 TDM 다중화 광섬유 압력/온도 센서시스템을 개발하고, 이 시스템을 이용하여 수위와 온도 측정실험을 행하였다. 측정시스템의 측정속도는 측정데이타를 저장하지 않는 경우 최대 초당 4500회이며, 센서의 응답속도는 ~1 ms로 추정된다. 압력센서와 온도센서의 특성은 이론적 추정치와 비교하여 각각 +13.7%,-18%의 차이를 보였으며, 반복실험을 통하여 선형화한 후의 선형화 오차는 1%이내, 온도의 변화가 0.1$^{\circ}C$이내 일 때 수위측정의 오차는 $\pm$0.3cm이며, 수위측정에 대한 시스템 잡음은 측정하지 않았다. 온도센서의 시스템 잡음은 0.1$^{\circ}C$이내였으며, 이 시스템을 이용하여 수위 및 온도 변화량에 대한 고속 측정실험을 수행할 결과 예상된 결과를 얻을 수 있었다.

Effects of Die Temperature and Repeated Extrusion on Chemical Components and Antioxidant Properties of Extruded White Ginseng (사출구 온도와 반복 압출성형이 백삼압출성형물의 화학적 조성 및 항산화 활성에 미치는 영향)

  • This study investigated the effects of die temperature and repeated extrusion on the chemical components and antioxidant properties of extruded white ginseng (EWG). Die temperature was adjusted to 100, 120, and followed by repeated extrusion under the same conditions with corresponding samples. Secondary extruded white ginseng (SEWG) at a die temperature of $120^{\circ}C$ had the highest acidic polysaccharide content of all extrudates. Increasing die temperature and repeated extrusion both increased crude saponin content of the extrudate. Ginsenoside Rh1 was detected in the EWG ($140^{\circ}C$) and SEWGs, whereas ginsenosides Rg3s and Rg3r were only detected in SEWG ($140^{\circ}C$). The highest total phenolic content, DPPH radical scavenging activity, and reducing power obtained from SEWG ($140^{\circ}C$) were $8.55{\pm}0.03$ mg/g, $72.05{\pm}0.63%$, and $0.80{\pm}0.004$, respectively. In conclusion, repeated extrusion increases antioxidant activity and crude saponin contents for the development of improved ginseng products.
