Properties of Doenjang (Soybean Paste) Prepared with Different Types of Salts

  • Samples of doenjang (a fermented soybean paste) were prepared with different types of salts; purified salt (PS), 3-year-aged solar salt (SS3), 1-year-aged solar salt (SS1), and bamboo salt (BS, 3rd processing product). For starter doenjang samples, selected starters comprising two bacilli, one yeast, and one fungus were inoculated, whereas for non-starter doenjang samples, microorganisms present in rice straw were inoculated after enrichment. The doenjang samples were fermented for 13 weeks at 25℃. During the fermentation period, SS and BS doenjang samples showed higher bacilli counts as well as much lower yeast counts than PS doenjang. At 13 weeks, yeast counts of starter doenjang samples were 7.75, 5.69, 6.08, and 4.74 log CFU/g for PS, SS3, SS1, and BS doenjang, respectively. For non-starter doenjang samples, counts were 7.17, 5.05, 5.92, and 4.54 log CFU/g for PS, SS3, SS1, and BS doenjang, respectively. SS and BS promoted growth of bacilli but inhibited growth of yeasts compared with PS. Debaryomyces hansenii was the dominant yeast in PS doenjang, whereas Candida guilliermondii and Pichia sorbitophila were dominant in SS and BS doenjang. In the sensory evaluation, SS and BS doenjang scored better than PS doenjang. In conclusion, SS and BS seem better than PS for production of high-quality doenjang.

고로슬래그미분말의 치환율 변화에 따른 굳지않은 모르타르의 레올로지 특성 검토 (Effect of Blast Furnace Slag on Rheological Properties of Fresh Mortar)

  • 본 연구에서는 고로슬래그미분말의 분말도 종류 및 치환율이 시멘트 페이스트 및 모르타르에 미치는 영향에 대하여 검토하였다. 그 결과는 다음과 같다. 전반적으로 BS1 및 BS2의 경우 치환율이 증가함에 따라 항복치도 증가하는 경향을 보였다. 반면, BS 3의 경우 치환율이 증가함에 따라 전단응력이 감소하며 항복치도 감소하는 경향을 나타 내었다. 위 실험 결과를 참조하여 고로슬래그미분말을 사용한 현장배합 설계 시 유동성 파악의 기초적 자료로 참고할 수 있을 것으로 판단된다.

산소와 염농도가 한국전통 발효식품에서 생장하는 혐기성 세균과 Bifidus균의 생육에 미치는 영향 (Effects of Oxygen and Salt on the Growth of Bifidus and Anaerobic Bacteria Isolated from Korean Traditional Fermented Foods)

  • 한국 전통 발효식품의 숙성에 절대 혐기성 세균이 관여하는지를 보기 위해 김치류와 장류 14종의 시료를 복합 BL 배지 혹은 bifidus 세균 선택 배지인 BS agar에 접종하여 절대 혐기적으로 배양한 결과 복합 BL 배지에서 $10^7$ ~ $10^8$/g 시료의 세균이 생장하였으며, BS agar에서는 $10^3$ ~ $10^6$/g 시료의 colony가 형성되었다. 이들은 모두 호기성 조건에서도 생장 할 수 잇었으며, BS 배지에서 생장하는 세균은 fructose-6-phosphate phosphoketolase를 합성하지 않는 것으로 나타났다. 또한 표준 Bifidobacterium속 세균은 3% NaCl을 함유하는 배지에서는 생장할 수 없었다. 이상의 결과로부터 김치류와 장류 등 염 농도가 높은 발표식품에는 bifidus균은 생장할 수 없다고 판단된다.

미생물 제제와 살균제에 의한 인삼 점무늬병의 방제 (Control of Alternaria Leaf Blight of Ginseng by Microbial Agent and Fungicides)

  • Bacillus subtilis QST713(BS QST713)의 단독, 살균제와 혼합 및 교호처리하여 인삼 점무늬병의 방제효과를 검정하였다. BS QST713를 10일 간격으로 단독으로 처리했을 때, 인삼 점무늬병에 대한 방제가는 83.3%, azoxystrobin, chlorothalonil copper sulfate basic, copper sulfate basic, kresoxim-methyl, difenoconazole, mancozeb 단독 처리구는 $80.4{\sim}83.7%$로 나타났다. BS QST713와 mancozeb를 혼합하여 14일 간격으로 처리하였을 때의 방제가는 83.6%, BS QST713와 azoxystrobin 혼합 처리구는 82.6%이었으나, difenoconazole, kresoxim-methyl, copper sulfate basic 및 chlorothalonil copper sulfate basic와 혼합 처리구는 $61.1{\sim}76.4%$로 방제효과가 약간 감소하였다. 대전에서 BS QST713와 copper sulfate basic를 10일 간격으로 교호처리시 방제가가 85.9%이었으나, 다른 살균제와의 교호처리 시는 $55.5{\sim}78.2%$로 낮아 졌다. 그러나 무주에서 모든 교호처리에서 방제가가 90% 이상으로 나타났다. 이상의 결과에 의하면 BS QST713의 단독, 혼합 및 교호처리는 살균제의 사용량을 줄일 수 있는 방제법으로 이용될 것이다.

사료내 맥반석과 BAISM 복합첨가가 치어기 뱀장어 Anguilla japonica의 성장과 내병성에 미치는 영향 (Effects of Dietary Quartz Porphyry and Feed Stimulants, BAISM Supplementation on Growth Performance and Disease Resistance of juvenile eel Anguilla japonica)

  • 이 연구는 치어기 뱀장어 Anguilla japonica의 사료 내 맥반석(quartz porphyry, QP)과 BAISM을 단독 및 혼합 첨가가 성장과 내병성에 미치는 영향을 확인하고 첨가제로써 이용성을 평가하였다. 사료 내 첨가 물질로 맥반석(Quartz porphyry, QP)과 실험실에서 제작한 섭취 촉진 물질인 BAISM(BS)을 기초 사료에 각각 0% QP+0% BS($Q_0B_0$), 0.7% QP+0% BS($Q_{0.7}B_0$), 0.7% QP+0.3% BS($Q_{0.7}B_{0.3}$), 0.7% QP+0.5% BS($Q_{0.7}B_{0.5}$), 0.7% QP+0.75% BS($Q_{0.7}B_{0.75}$), 0.7% QP+1.0% BS($Q_{0.7}B_{1.0}$)의 6가지 수준으로 첨가하였다. 실험어는 4주간 예비사육 후, 평균 어체중 $15{\pm}0.3g(mean{\pm}SD)$인 치어기 뱀장어를 60 L 사각수조에 실험구별 30 마리씩 3반복으로 무작위 배치하여 8주간 사육실험을 진행하였다. 사육실험 종료 후, Edwardsiella tarda 복강 주사에 의한 누적 생존율을 조사하였다. 증체율, 일간성장률, 사료효율, 단백질전환효율에서 맥반석 단독 첨가구($Q_{0.7}B_0$)와 맥반석 BAISM 복합 첨가구($Q_{0.7}B_{0.3},\;Q_{0.7}B_{0.5},\;Q_{0.7}B_{0.75},\;Q_{0.7}B_{1.0}$)는 대조구($Q_0B_0$)에 비하여 유의하게 높았다(P<0.05). 특히, 맥반석 BAISM 복합 첨가구들 중 BAISM 0.5% 이상의 실험구($Q_{0.7}B_{0.5},\;Q_{0.7}B_{0.75},\;Q_{0.7}B_{1.0}$)는 나머지 실험구들에 비해 유의하게 높았고(P<0.05), 이들에 대한 유의한 차이는 없었다(P>0.05). 전어체 일반성분의 분석결과 수분 함량은 전 실험구에서 유의한 차이가 없었고(P>0.05), 단백질 함량은 첨가수준에 따라 유의적으로 증가한 반면(P<0.05), 지질과 회분의 함량은 유의적으로 감소하는 경향을 나타내었다(P<0.05). E. tarda에 의한 공격실험 결과 주사 15일 후, 맥반석BAISM 0.5% 이상 혼합 첨가구의 누적 생존율은 다른 실험구에 비해 유의하게 높았고(P<0.05), 전 실험구에서 40% 이상의 누적 생존율을 나타내었다. 따라서 치어기 뱀장어 사료에 맥반석 0.7%와 BAISM 0.5%를 첨가하는 것이 성장과 사료효율 증대 및 질병 저항성 증진에 효과가 있을 것으로 판단된다.

Avirulence Gene Diversity of Xanthomonas axonopodis pv. glycines Isolated in Korea

  • The hybridization patterns with the avrBs3 gene that is known to determine the recognition of host specificity were used to study the diversity of Xanthomonas axonopodis pv. glycines causing bacterial leaf pustule in soybean. A total of 155 strains were isolated from diverse tissues of soybean cultivars collected in Korea and were classified into six different type strains of OcsF, SL1017, SL1018, SL1045, SL1157, and SL2098 according to the patterns of avrBs3-homologous bands. When these type strains were inoculated on various cultivars, most of the Korean strains mildly induced disease symptoms on the resistant CNS1 cultivars. Unlike other type strains, strain SL2098, which appeared not to contain any avrBs3 homolog, induced only a few pustules on even highly susceptible cultivars. When a plasmid carrying the 3.7-kb avrBs3-homologous gene from strain SL1045 was introduced into SL2098, the transformant could not recover the pathogenicity in susceptible host plants. However, when avrBs3-homologous genes of strain SL1018 were mutated by transposon mutagenesis, one of the mutants in which a 5.2-kb chromosomal band homologous to avrBs3 was disrupted could not induce the hypersensitive response on resistant cultivars such as William82 or CNS2. Our results suggest that the avrBs3 homologs may play important roles in the pathogenicity of Xanthomonas axonopodis pv. glycines and the recognition of soybean cultivars.

Properties of Cheonggukjang Fermented with Bacillus Strains with High Fibrinolytic Activities

  • We previously isolated Bacillus strains with high fibrinolytic activities (FAs) from cheonggukjang prepared by traditional ways. To test their potential as starters for cheonggukjang, soybean was fermented for 72 hr at $37^{\circ}C$ with each isolate and a control lab strain: B. subtilis CH3-25 (BS3-25), B. amyloliquefaciens CH51 (BA51), B. amyloliquefaciens CH86-1 (BA86-1), and B. subtilis 168 (BS168, control, lab strain). Viable cell numbers of all cheonggukjang samples rapidly increased and reached about $10^9$ CFU/g after 6 hr. During 72 hr, the initial pH of 6.3 rapidly increased to 8.1$\sim$8.2 for cheonggukjang fermented with BS3-25 or BA86-1, and 7.3 for those with BA51 or BS168. FAs and protease activities (acid, neutral, and alkaline) rapidly increased in cheonggukjang fermented with BS3-25, BA51, or BA86-1 during the first 12 hr. On the other hand, those of cheonggukjang fermented with BS168 slightly increased during the first 36 hr. There were significant changes in acid and neutral protease activities in cheonggukjang fermented with BA51 or BA86-1 during the 24 hr. Rapid increases of $\beta$-glucosidase activity corresponded well with rapid increases of $\alpha$-amylase and $\alpha$-galactosidase activities in addition to increases in antioxidant activities and the TPCs (total phenolic contents). The highest increase in the TPCs was observed in cheonggukjang fermented with BA86-1 while the least was that fermented with BS168.

지방분화가 유도된 3T3-L1 세포와 고지방식이로 유도된 마우스에서 보리순 물추출물의 항비만 효과 (Anti-obesity Effects of Barley Sprout Young Leaf on 3T3-L1 Cells and High-fat Diet-induced Obese Mice)

  • Background: An imbalance in energy intake and expenditure can cause obesity, which is a major risk factor for chronic diseases such as heart disease, type 2 diabetes, insulin resistance, cancers and hyperlipidemia. Methods and Results: In this study, we evaluated the anti-obesity effects of a water extract from the young leaves of barley sprout (BS) in 3T3-L1 cells and in high-fat diet (HFD)-induced obese mice (HF). Lipid accumulation measurement indicates that BS markedly inhibited adipogenesis by reducing lipid droplet production in a dose-dependent manner. Furthermore, the mRNA expression of adipogenic transcription factors peroxisome proliferator-activated receptor-${\gamma}$ and fatty acid synthetase, CCAAT/enhancer binding protein-${\alpha}$ and fatty acid binding protein 4 in 3T3-L1 cells was significantly inhibited by BS treatment. In an in vivo test, the BS-administered group of HFD-induced mice showed less body weight gain, and lower liver and epididymal white adipose tissue weights. The BS-treated mice showed decreased serum levels of leptin and lipids compared to untreated HFD mice and the levels of adiponectin and the HDL-cholesterol/total cholesterol ratio increased. These results indicate that BS inhibits body fat accumulation by reducing the mRNA expression of lipogenesis transcription factors and increasing serum adipokine concentration in in vitro and in vivo tests. Conclusions: BS reduced high fat diet-induced weight gain and had a positive effect on dyslipidemia.

  • Hypertension induces many of the social costs related by cardiovascular diseases. Sodium is known as a crucial factor in inducing type I hypertension. In traditional Korean medicine, bamboo salt (BS) has been used in the attenuation of salts toxic coldness and nowadays it has shown various therapeutic effects. It contains mostly sodium chloride (about 91.7% of BS); however, the effect of BS on hypertension is still not completely understood. Thus, we investigated the effect of BS on blood pressure for the first time. Two group of BS, sun-dried salt (SDS), NaCl, or distilled water (DW, vehicle control) was administrated orally for 8 weeks. Although BS had no effect on body weight and food intake, it increased water intake (p < 0.05). The BS groups, in terms of blood pressure, was similar to the DW group; whereas the SDS and NaCl groups showed significantly increased blood pressure levels (p < 0.05). BS also decreased sodium-chloride cotransporter (NCC) mRNA expression, unlike SDS or NaCl. These observations indicate that BS may be a promising strategy for the prevention of various diseases including salt-related diseases.

TCP Performance Enhancement by Implicit Priority Forwarding (IPF) Packet Buffering Scheme for Mobile IP Based Networks

  • The smooth handoff supported by the route optimization extension to the mobile IP standard protocol should support a packet buffering mechanism at the base station (BS), in order to reduce the degradation in TCP performance caused by packet losses within mobile network environments. The purpose of packet buffering at the BS is to recover the packets dropped during intersubnetwork handoff by forwarding the packets buffered at the previous BS to the new BS. However, when the mobile host moves to a congested BS within a new foreign subnetwork, the buffered packets forwarded by the previous BS are likely to be dropped. This subsequently causes global synchronization to occur, resulting in the degradation of the wireless link in the congested BS, due to the increased congestion caused by the forwarded burst packets. Thus, in this paper, we propose an implicit priority forwarding (IPF) packet buffering scheme as a solution to this problem within mobile IP based networks. In the proposed IPF method, the previous BS implicitly marks the priority packets being used for inter-subnetwork handoff. Moreover, the proposed modified random early detection (M-RED) buffer at the new congested BS guarantees some degree of reliability to the priority packets. The simulation results show that the proposed IPF packet buffering scheme increases the wireless link utilization and, thus, it enhances the TCP throughput performance in the context of various intersubnetwork handoff cases.
