Triterpenoids and Flavonoids Isolated from the Leaves of Alnus firma (사방오리 잎의 Triterpenoid 및 Flavonoid 화합물)

  • In this study, three triterpenoids, two steroids and nine flavonoids were isolated from the leaves of Alnus firma Sieb. et Zucc. On the basis of spectroscopic evidences, the structures of these compounds were established as ${\beta}$-amyrin acetate, ${\beta}$-amyrin, ${\beta}$-sitosterol, alnustic acid methyl ester, ${\beta}$-sitosterol glucoside, pinocembrin, alnustinol, quercetin, quercetin-3-O-${\alpha}$-L-arabinofuranoside, quercetin-3-O-${\alpha}$-L -rhamnopyranoside, quercetin-3-O-${\beta}$-D-glucopyranoside, myricetin-3-O-${\beta}$-D-galac-topyranoside, (+)-catechin and (-)-epicatechin.

  • Two phenylpropanoids and one flavan 3-ol were isolated from Lepisorus thunbergianus (Polypodiaceae, fern), which is used as folkmedicine. Phenylpropanoids were identified as caffeic acid and chlorogenic acid, and flavan 3-ol was elucidated as (-)-epicatechin 7-O-${\beta}$-D-glucoside by physico-chemical and spectral evidences (HMQC, NOESY).

  • A chemical examination of the aerial parts of Euphorbia pekinensis $R_{UPRECHT}$. (Euphorbiaceae) has led to the isolation of seven hydrolyzable tannins and ten fl avonoid glycosides. The former ones have been identified as gallic acid, methylgallate, 3-O-galloyl shikimic acid, 1,3,4,6-tetra-O-galloyl-${\beta}-_D$-glucose, 1,2,3,4,6-penta-O-galloyl-${\beta}-_D$-glucose, corilagin, geraniin and the latter ones as isoquercitrin, quercitrin, astragalin, afzelin, prunin, rutin, kaempferol-3-O-rutinoside, quercetin-3-O-(2"-O-galloyl)-${\beta}-_D$-glucoside and quercetin-3-O-(2"-O-galloyl)-${\alpha}-_L$-rhamnoside on the basis of chemical and spectroscopic evidence.

A New Coumestan Glucoside from Eclipta prostrata

  • Eclipta prostrata is an annual herb, belonging to Asteraceae family, and has been traditionally used to improve immunity and treat hepatitis and bacterial disease in Korea. In this study, a new coumestan glucoside (1) along with ten known compounds (2 - 11) was isolated from E. prostrata. The chemical structures of isolates were elucidated to be wedelolactone-9-O-β-D-glucopyranoside (1), wedelolactone (2), demethylwedelolactone (3), apigenin (4), apigenin-7-sulfate (5), luteolin (6), luteolin-7-sulfate (7), luteolin-7-O-β-D-glucopyranoside (8), pratensein-7-O-β-D-glucopyranoside (9), 3,4-di-O-caffeoylquinic acid (10) and 3,5-di-O-caffeoylquinic acid (11) based on the spectroscopic evidence.

  • To search for inhibitory activities for lens aldose reductase, twenty two medicinal plants were tested by the method of bovine lens aldose reductase inhibition test. In this test Rubia akane(68%), Artemisia selengensis(61%), Lespedeza cuneata (58%), Ligustrum lucidum(58%), Viola patrinii(58%) extracts were shown to have aldose reductase inhibitory activity. From the radix of Rubia akane, $2-methyl-1,\;3,\;6-trihydroxy-9,\;10-anthraquinone-3-O-(6'-O-acetyl)-{\alpha}-rhamnosyl-(1{\rightarrow}2)-{\beta}-D-glucoside$ was isolated and the $IC_{50}$ of this compound in the aldose reductase inhibition test was about $1.2{\times}10^{-6}M$.

Glucosylation of Resveratrol Improves its Immunomodulating Activity and the Viability of Murine Macrophage RAW 264.7 Cells (당화된 레스베라트롤의 대식세포 RAW 264.7세포의 생존능력과 레스베라트롤의 면역제어 활성을 증가)

  • Effects of resveratrol glucosylation on the immunomodulation properties of resveratrol and on the viability of macrophage cells have been studied by using murine macrophage RAW 264.7 cells. Nitric oxide (NO) and interleukin 6 (IL-6) expression in macrophages in vitro were studied after treatment with different concentrations of (E)-resveratrol, (E)-resveratrol 3-O-${\beta}$-${\small{D}}$-glucoside (R-3-G), or (E)-resveratrol 4'-O-${\beta}$-${\small{D}}$-glucoside (R-4'-G). In vitro viability of RAW 264.7 cells after treatment with the aforementioned three compounds was also studied. As demonstrated by macrophage cell viability assays, two different resveratrol monoglucosides, R-3-G and R-4'-G, exhibited 50-80% reduced cytotoxicity in comparison to (E)-resveratrol in A549 and HepG2 cells. Compared to the resveratrol aglycon, both glucosylated resveratrol derivatives positively modulated NO and IL-6 production in macrophages positively via transcriptionally up-regulating IL-6 and iNOS expression. Conjugation of a glucose moiety on resveratrol was found to enhance the immunomodulating activity of resveratrol and the viability of RAW 264.7 cells.

Antilipidperoxidative activity of Astragalus membranaceus (황기의 지질과산화 억제작용)

  • The root of Astragalus membranaceus (Leguminosae) has been used in traditional chinese prescriptions for strengthning the superficial resistance, promoting pus discharge and tissue regeneration, diuretis and alleviating edema. Lipid peroxidation has been suggested as a major cause of atherosclerosis, cancer, liver disease, and the aging process. In order to investigate the anti-lipid peroxidation activity, Astragali Radix was extracted with $80\%$ MeOH and fractionated with hexane, $CH_{2}Cl_2$, EtOAc, BuOH and Water. The antilipid peroxidation activities of them were determined by human erythrocyte ghost and $CCl_4$-induced lipid peroxidation. $CH_{2}Cl_2$ and EtOAc fractions, especially, isoflavonoids, 7,2'-dihydroxy-3',4'-dimethoxyisoflavan-7-O-${\beta}$-D-glucoside and calyco sin-7-O-${\beta}$-D-glucoside from EtOAc fraction showed anti-lipid peroxidation activities.

  • A tannin compound has been isolated from the underground part of Rosa rugosa Thunb. and its structure has been established to be methyl gallate $3-O-{\beta}-D-(6'-O-galloyl)-glucopyranoside(1)$ on the basis of chemical and spectroscopic evidences. This compound was the first example in this plant.

  • We isolated and identified antioxidants from acidic and neutral ethyl acetate fractions of the peel of pear (Pyrus pyrifolia N. cv. Chuhwangbae). We isolated 4 compounds from the methanol extract, by using 3 different types of column chromatography (Sephadex LH-20, silica gel, and octadecylsilane) and preparative HPLC. We identified the isolated compounds as (S)-(+)-2-cis-abscisic acid O-${\beta}$-D-glucopyranosyl ester (compound 1), 1-[4-O-${\beta}$-D-glucopyranosyl]phenyl ethanone (picroside, compound 2), ${\beta}$-sitosterol (compound 3), and ${\beta}$-sitosteryl 3-O-${\beta}$-D-glucopyranoside (compound 4) by nuclear magnetic resonance analysis. We are the first to report the identification of compounds 1, 2, and 4 from pear.

Hepatoprotective Effect of Flavonol Glycosides Rich Fraction from Egyptian Vicia calcarata Desf. Against $CCl_4$-Induced Liver Damage in Rats

  • The hepatoprotective activity of flavonol glycosides rich fraction (F-2), prepared from 70% alcohol extract of the aerial parts of V calcarata Desf., was evaluated in a rat model with a liver injury induced by daily oral administration of $CCl_4$ (100 mg/kg, b.w) for four weeks. Treatment of the animals with F-2 using a dose of (25 mg/kg, b.w) during the induction of hepatic damage by $CCl_4$ significantly reduced the indices of liver injuries. The hepatoprotective effects of F-2 significantly reduced the elevated levels of the following serum enzymes: alanine aminotransferase (ALT), aspartate aminotransferase (AST), alkaline phosphatase (ALP) and lactate dehydrogenase (LDH). The antioxidant activity of F-2 markedly ameliorated the antioxidant parameters including glutathione (GSH) content, glutathione peroxidase (GSH-Px), superoxide dismutase (SOD), plasma catalase (CAT) and packed erythrocytes glucose-6-phosphate dehydrogenase (G6PDH) to be comparable with normal control levels. In addition, it normalized liver malondialdehyde (MDA) levels and creatinine concentration. Chromatographic purification of F-2 resulted in the isolation of two flavonol glycosides that rarely occur in the plant kingdom, identified as quercetin-3,5-di-O-$\beta$-D-diglucoside (5) and kaempferol-3,5-di-O-$\beta$-D-diglucoside (4) in addition to the three known compounds identified as quercetin-3-O-$\alpha$-L-rhamnosyl- (${\rightarrow}6$)-$\beta$-D-glucoside [rutin, 3], quercetin-3-O-$\beta$-D-glucoside [isoquercitrin, 2] and kaempferol-3-O-$\beta$-D-glucoside [astragalin, 1]. These compounds were identified based on interpretation of their physical, chemical, and spectral data. Moreover, the spectrophotometric estimation of the flavonoids content revealed that the aerial parts of the plant contain an appreciable amount of flavonoids (0.89%) calculated as rutin. The data obtained from this study revealed that the flavonol glycosides of F-2 protect the rat liver from hepatic damage induced by $CCl_4$ through inhibition of lipid peroxidation caused by $CCl_4$ reactive free radicals.
