유방암 세포주에서는 우수한 항암활성을 가진 것으로 알려진 indole-3-carbinol을 HepG2세포주에 시간과 농도별로 처리한 결과 cell growth inhibition을 확인하였으며, $IC_{50}$ 값은 48시간배양에서 $446\mu$M 72시간 배양에서 444$\mu$M로 나타났다. $400\mu$M의 I3C을 투여하고, 24, 48, 72시간에 HePG2 세포주의 cell cycle pattern을 분석한 결과, G1 phase에서 P21의증가와 함께 Cdk 6와 cyclin D의 확연한 감소와 Pb protein의 hypo-phosphorylation을 확인하였다. 반면 G2 phase에서는 I3C의 직접적인 억제로 인해 24시간 후부터 Cdc2와 cyclin B1가 급격히 감소하는것을 확인하였다. Flow cytomery 분석결과 I3C 처리 24시간 뒤 G2 arrest (25%)가 발생하였으며, 72시간이 지난후 G1 arrest (53%)가 발생하였다. 이러한 I3C의 간암세포주인 HePG2 cell의 cell cycle arrest가 apoptosis를 유발하는지를 알고자 caspase 3 Bcl2 Bax protein의 발현양상을 확인한 결과 아무런 변화가 보이지 않았다. 즉 I3C은 간암세포주인 HepG2 cell에서 apoptosis를 유도하지 못한다는것을 확인하였따. 결론적으로 I3C은 HepG2 세포주에서 G1와 G2 phase에서 cell cycle arrest는 발생시키나, 특이적으로 apoptosis 와는 연관되지 않는다는 사실을 확인하였다.
Phytochemical investigation on dragon's blood of Dracaena cambodiana led to the discovery of two new flavonoid derivatives, cambodianin G (1) and cambodianin H (2). Their structures were elucidated on the basis of detailed spectroscopic analysis, including 1D and 2D NMR techniques and chemical methods. The two compounds were observed to exhibit antibacterial activities against Staphylococcus aureus, and compound 1 showed cytotoxicities against K562 and SGC-7901 cell lines.

New $\alpha$-pyrones (1 and 2) and cyclohexenones (13 and 14) were isolated along with known analogues (3, 5−12) from the ethyl acetate extract of the whole broth of the fungus Paecilomyces lilacinus, a strain derived from a marine sponge Petrosia sp. Their structures were established by interpretation of 1D and 2D NMR, and FABMS data. It is interesting to isolate cyclohexenone derivatives from the genus Paecilomyces (family Trichocomaceae, order Eurotiales), since these cyclohexenones were previously reported only from far distinct genera, Phoma and Alternaria (family Pleosporaceae, order Pleosporales). Compounds 6, 7, and 9 were evaluated for cytotoxicity against a small panel of human solid tumor cell lines. Their cytotoxicity was insignificant upto a concentration of 30 ${\mu}g/mL$.
강원도 산지에서 채취한 만병초 잎의 70% ethanol 추출물과 다른 유기용매 분획 과정을 통한 3가지의 분획물에 대한 in vitro에서의 정상세포에 대한 세포독성과 유전독성, 여러 암세포에 대한 항암 기능, 면역세포에 대한 면역기능 등을 조사하여 다음과 같은 결과를 얻었다 1. 만병초 잎의 EtOH 추출물과 각 용매 분획물 들의 사람의 정상 세포인 HEL 299와 Chang에 대한 세포 독성을 SRB assay로 측정한 결과, 대부분의 시료 0.5 mg/ml 이하의 농도에서 35% 이하의 세포 독성이 나타났다. 2. 만병초 잎의 70% EtOH 추출물 및 각 다른 용매 분획물만 처리한 정상 세포에 대한 유전독성을 comet assay에서 측정한 결과, 양성 대조구 $(10^{-2}M,\;H_2O_2)$과 비교시, 가장 높은 농도인 1 mg/ml의 농도에서 1/3 이하의 낮은 DNA damage가 관찰되었다. 이것은 세포독성과 관련하여 대부분의 각 시료가 세포 DNA에 대하여 매우 약한 손상만 일으키는 것이 확인되었다. 3. 사람의 암세포에 대한 SRB 및 MTT assay를 이용한 만병초의 잎의 모든 시료의 1 mg/ml의 농도의 70% EtOH의 추출물은 실험대상인 모든 사람의 암세포에 대하여 80% 이상의 항암 기능을, EtOAc fr.는 모든 암세포에 대하여 가장 높은 농도인 1 mg/ml에서 70% 이상의 높은 억제 활성이 보인 결과로 부터 유효한 성분이 역시 EtOAc fr.에 함유되어 있을 것을 시사하였다. 4. 만병초 잎 추출물의 면역세포 생육에 대한 조사에서 사람의 B-cell과 T-cell에 대하여 0.5 mg/ml의 농도에서 배양 3일째에 BuOH fr.에서 대조구와 비교시 1.6배 정도의 생육 촉진활성을 보였다. 5. SCGE를 통해 $H_2O_2$로 유도된 산화적 손상에 대한 만병초 잎 추출물과 분획물들의 유전독성 억제 기능은 EtOAc fr.이 $100\;{\mu}g/ml$ 농도에서 대조구 대비 68.5%의 가장 높은 산화손상 억제율을 보였으며 BuOH fr.에서도 강한 유전독성 억제기능이 있음이 확인되었다. 그러나 수용액 분획은 매우 약한 유전독성 억제 활성을 보여 각 분획의 특성이 다양함을 알 수 있었다.

Systematic analysis of caffeine from the commercial samples of Indian tea leaves was performed by a routine method and the content of caffeine was found to be 19.0-37.4 mg/100 g leaves. The caffeine contents from coffee seeds and chicory from Indian origin were analyzed and found to be 0.6540-1.4920 g/100 g seeds. Caffeine contents of roasted Indian chicory roots were lower than either those of Indian tea leaves or Indian coffee seeds. The multidrug resistance (MDR) reversing effects were tested on a mouse leukemia cell line of L-5178 cells by methanol extracts [M1-M15] of Indian tea leaves and coffee seeds, comparing to a control of $({\pm})-verapamil$. The effects were measured by fluorescence ratio between treated and untreated group cells. Among fifteen methanol extracts, a Gemini tea [M6] (fluorescence activity ratio 5.26) had the most potent effect for L-5178 cells. The extract M6 was 0.63-fold of $({\pm})-verapamil$. We suggest that one of mechanisms of reversal by M6 might have strong affinity to dopamine $D_1$ and D_2$ receptors. Further studies with many more tumor and normal cell lines are necessary to confirm the MDR reversal specificity of coffee methanol extracts.
An antiproliferative agent, streptochlorin, was isolated from the fermentation broth of a marine actinomycete isolated from marine sediment. Phylogenetic analysis of the 16S rRNA gene sequence indicated that the strain belongs to the genus Streptomyces. Bioactivity guided fractionation of the culture extract by solvent partitioning, ODS open flash chromatography, and reversed-phase HPLC gave a pure compound, streptochlorin. Its structure was elucidated by extensive 2D NMR and mass spectral analyses. Streptochlorin exhibited significant antiproliferative activity against human cultured cell lines.
A series of pyrazoloxyphenyl benzoyl urea derivatives was designed and synthesized for cytotoxic evaluation as potential antitumor agents. The synthetic compounds were evaluated for in vitro cytotoxicity against five human tumor cell lines, including A-549, SKOV-3, SK-MEL-2, XF-498 and HCT-15. Among others, compound 11 exhibited 50∼100 times greater antitumor activities than the commercial product, Cisplatin.
Bacillus Anthracis is the causative agent of anthrax. The major virulence factors are a poly-D glutamic acid capsule and three-protein component exotoxin, which is collectively known as anthrax toxin, protective antigen (PA, 83 kDa), lethal factor (LF, 90 kDa), and edema factor (EF, 89 kDa). These three proteins individually have no known toxic activities, but in combination with PA form two toxins (lethal toxin and edema toxin), causing different pathogenic responses in animals and cultured cells. However, it remains to be elucidated for pathogenic mechanism of anthrax toxin. In this study, we constructed toxin component in bacterial overexpression system and purified the native toxin from Bacillus anthracis delta sterne F32 using FPLC system. Recombinant toxin showed high homogeneity and rapid purification processes. Also, this recombinant toxin was comparable to B. anthracis native toxin in terms of cytotoxic effects on cultured cell lines.
Silver materials may be toxic in humans because they can enter the body and accumulate, typically in the lungs. We hypothesized that the cytotoxicity of naive silver materials is affected by their size and shape. Our in vitro assays revealed that the overall toxicity was in the following order: submicro-particles>wires>micro-particles. These results contrast with previous studies, which showed that silver wires are the most toxic among the three tested materials, possibly due to differences in cell lines. Evaluations of in vivo pulmonary toxicity revealed eryptosis in the cavity lining of the lung sections. The observed eryptosis was consistent with the in vitro results. Our results indicate that silver materialinduced cytotoxicity must be measured and compared using various methods.
The aim of this work is the real-time road edge detection using the fast processing of Cellular Analogic Parallel Processing Networks(CAPPN). The CAPPN is composed of 2D analog cell way. If the dynamic programming is implemented with the CAPPN, the optimal path can be detected in parallel manner Provided that fragments of road edge are utilized as the cost inverse(benefit) in the CAPPN-based optimal path algorithm, the CAPPN determines the most plausible path as the road edge line. Benefits of the proposed algorithm are the fast processing and the utilization of optimal technique to determine the road edge lines.
