Objective: Our present studies were conducted to examine more effective isolating method of preantral follicles from mouse ovaries. Methods: ICR mice (3-6 weeks old) were sacrificed through cervical dislocation and their ovaries were removed and put into watch glasses containing Hams F-10 supplemented with 10% fetal bovine serum (FBS). Preantral follicles were isolated by three different methods; 1) enzymatical method and 2) mincing method, and 3) scraping method. Enzymatical method was carried out as following. Ovaries were bisected with a pair of fine 30G needles. Bisected ovaries were incubated at $37^{\circ}C$ and 5% $CO_2$ incubator in 2-well dish containing Hams F-10 supplemented with collagenase 600 lU/ml and DNAse 20 lU/ml. After 20 min., follicles were isolated by repeated pipetting. Isolated preantral follicles were collected, and the remnant of tissues was placed in incubator and previous procedure was repeated. Mincing method was carried out with a pair of fine 30G needles attached to 1 ml syringes and minced ovary. Scraping method was carried out with a pair of fine 30G needles and scratched to surface of ovary. The differences between isolating methods were analyzed using Student's t-test and Chi-square. Results were considered statistically significant when ${\rho}$ value was less than 0.05. Results: In handling time, mincing or scraping method ($28{\pm}3.42$ min or $16{\pm}1.58$ min) were significantly (p<0.00001) shorter than enzymatical method ($72{\pm}1.69$ min), and scraping method was significantly (p<0.01) shorter than mincing method. Total number of isolated follicles was significantly (p<0.0001) higher in enzymatical method ($49.8{\pm}3.91$) than in mincing or scraping method ($25.3{\pm}2.33$ or $20.5{\pm}1.75$). Isolated follicles in ${\leq}$90${\mu}m$ were significantly (p<0.005) higher in enzymatical method ($15{\pm}1.71$) than in mincing or scraping method ($7.8{\pm}0.98$ or $8.1{\pm}1.31$). In 91~130 ${\mu}m$, isolated follicles were significantly (p<0.0005) higher in enzymatical method ($33{\pm}3.27$) than in mincing or scraping method ($16.3{\pm}1.82$ or $10.7{\pm}1.38$). In ${\geq}$ 131 ${\mu}m$, isolated follicles were not significantly differences between all groups. In equal sizes, the rate of isolated follicles in ${\leq}$ 90 ${\mu}m$ was highest in scraping method (39.6% vs. enzymatical method: 30.1%, p<0.05; mincing method: 30.9%, p=0.11719, NS). Rate of follicles in $91{\sim}130$${\mu}m$ was significantly (p<0.05) lower in scraping method (52.7%) than in enzymatical or mincing method (66.3% or 64.5%). Rate of follicles in ${\geq}$131 ${\mu}m$ was highest in scraping method (8.3% vs. enzymatical or scraping method: 3.6%, p<0.05 or 4.6%, p=0.19053, NS). Conclusions: This study suggests that scraping method is simple and useful for isolation of preantral follicles, because this method reduced handling time and recovered enough follicles. The recovered rate of isolated follicles in diameter of 91 ~ 130 ${\mu}m$ was highest in all methods.
To establish an efficient herbicide screening method for purple nutsedge(Cyperus rotundus) control, its sprouting characteristics, tuber production and responses on several herbicides were investigated under greenhouse condition. The tubers stored at $4^{\circ}C$ after sterilization with the diluted prochloraz(Spotac) solution showed higher sprouting than the non-sterilized did. The harvested tubers were not dormant, and the sterilized tubers which stored at low temperature had a sprouting capability of about 80% after 6 months. If the fresh weight of purple nutsedge tubers was decreased to below 48%, they could not sprout. However, the tubers soaked in water and then stored at low temperature could sprout by 88% even 6 months later. Sprouting and initial growth of tuber were much better at $35^{\circ}C$-day/$25^{\circ}C$-night than at 30/$20^{\circ}C$ or 25/$15^{\circ}C$. The half-sected tubers, which were prepared by setting the intact tuber of above 1.2g latitudinally, were shown similar initial growth to the intact but those sected crucifically were not. These results suggest that the half-sected tuber itself can be used as a material on herbicide screening. About 1000 tubers could be harvested when 10 tubers planted in a pot($56{\times}35{\times}16cm$) filled with the artificial soil were cultivated in greenhouse of $35^{\circ}C$-day/$25^{\circ}C$-night for 3 months(April-July, 1993). Chlorimuron, Bentazon and Norflurazon were selected as the standards for the screening because of providing relatively effective control on purple nutsedge in both soil-surface and foliar spray treatment.
This study was conducted to determine the effects and optimum concentrantion of chemical mutagens, colchicine, EMS (ethyl methan sulfonate), MNU (1-methyl-3-1-nitrosoguanidinenitro), sodium azide $(NaN_3)$ for induction of mutant plants. In order to induce the mutants of Dianthus superbus L, immature seed were pre-soaked in the warter adding each mutagens and concentration of EMS, colchicine, MNU, and sodium azide $(NaN_3)$. Comparision of morphological characteristic and seed germination in each mutant plants differed depending on mutagen sources and their concentrations. When 0.2% EMS were treated on seed, germination decreased to 12% while untreated control was germinated 76.6% for twenty days. Treatments of colchicine appeared higher germination than other mutagen but not survived. The survival rate was extremely decreased in MNU treatment at 0.5mM and chlorophyll-mutant plantlets were obtained by sodium azide treatment at 0.2mM. Chlorophyll mutants were produced by pre-soaking the immature seed of Dianthus superbus L. with mutagen, sodium azide. The control plants appeared normal green leaf color, while mutant plant after mutagenic treatment of immature seed results in yellow-green stripes and albino in normal green leaf tissue. RAPD was carried out to check the genetic modification of regenerated plants by mutagen treatments at 0.2mM sodium azide. Three polymorphic DNA fragments out of thirty-seven obtained by RAPDs were observed in regenerated plants using five decamer primers.

Adsorption onto the surfaces of solid particles is a well known phenomenon that causes the retardation effect of heavy metals in soils. For adequate remediation of soil and groundwater contamination, it is important to investigate the mobility of heavy metals that largely depends on pH conditions in the soil water since adsorption of heavy metals is pH-dependent. In this study, we investigated the transport of Zn ion under various pH conditions in a sandy soil by conducting batch and column tests. The batch test was performed using the standard procedure of equilibrating fine fractions collected from the soil with eleven different initial $ZnCl_2$ concentrations, and analysis of Zn ion in the equilibrated solutions using ICP-AES. The column test consisted of monitoring the concentrations of soil solutions exiting the soil column with time known as a breakthrough curve (BTC). We injected respectively $ZnCl_2$ and KCl solutions with the concentration of 10 g/L as a tracer in a square pulse type under three different pH conditions (7.7, 5.8, 4.1) and monitored the flux concentration at the exit boundary using an EC meter and ICP-AES. The resident concentration was also monitored at the 10cm-depth by Time Domain Reflectometry (TDR). The results of batch test showed that ion exchange process between Zn and other cations (Ca, Mg) was predominant. The retardation coefficients obtained from adsorption isotherms (Linear, Freundlich, Langmuir) resulted in the various values ranging from 1.2 to 614.1. No retardation effect but ion exchange was found for the BTCs under all pH conditions. This can be explained by the absence of other cations to desorb Zn ion from soil exchange sites under the conditions of ETC experiment imposing blank water as leachate in steady-state flow. As pH decreased, the peak concentration of Zn increased due to the competition of Zn with hydrogen ions ($H^+$) and the concentrations of other cations decreased. The peak concentration of Zn was increased by 12.7 times as pH decreased from 7.7 to 4.1.
Satsuma mandarin(Citrus unshiu Marc. var. miyagawa) was stored at $3^{\circ}C$ and 85% relative humidity, and then the changes of firmness, pectin- degrading enzymes activity and other physicochemical properties of citrus fruits during storage were investigated. Firmness of fruits with 2 m probe was decreased quickly from 1,176.8 g-force to 503.6 g-force, and moisture of peel and flesh were decreased from 75.3% to 74.9%, and from 91.8% to 90.7% during maturation, respectively. Decay ratio was increased to 18.75% after 90 days' storage, and after then it was increased rapidly. Weight loss was increased gradually to 24.5% during long-term storage. Firmness with 2 mm probe were decreased from 538.9 g-force to 336.9 g-force gradually during storage. Peel moisture was decreased from 75.8% to 72.6%, and flesh moisture was also decreased gradually from 90.3% to 88.3% during storage. Exopoly-galacturonase activity of peel and flesh were increased from 326.0 units/100 g to 534.9 units/100 g, and from 63.1 units/100 g to 81.0 units/100 g at 90 day's storage, respectively. After then, He enzyme activities were decreased from 394.0 units/100 g and 38.0 units/100 g, respectively. Pectinesterase activity of peel and flesh were increased from $14.4\;{\mu}mol$ to $38.8{\mu}mol$, and from $26.0{\mu}mol$ to $39.0{\mu}mol$ at 60 days' storage, respectively. After then, the enzyme activities were decreased to $6.0{\mu}mol$ and $8.2{\mu}mol$, respectively.

Remarkable winter season warming has been observed in East Asian countries during the last century. Accordingly, significant effects on dormancy and the resulting budburst of deciduous trees are expected. However phenological observations are rare and insufficient compared with the long-time climate records in the same region. A chill-day accumulation, which can be estimated from daily maximum and minimum temperature, is expected to make a reasonable proxy for dormancy depth of temperate zone fruit trees. To simulate dormancy depth during 1921-2004, a chill-day model parameterized for 'Campbell Early' grapevine, which is the major cultivar grown virtually anywhere in South Korea, was applied to daily temperature data at 8 locations in South Korea. The calculations showed that the chilling requirement for breaking endo-dormancy of this grapevine cultivar can be satisfied by mid-January to late February in South Korea, and the date was delayed going either northward or southward from the 'Daegu-Jeonju' line crossing the middle of South Korea in the east-west direction. Maximum length of the cold tolerant period (the number of days between endo-dormancy release and forced dormancy release) showed the same spatial pattern. When we divide the 83 years into 3 periods (I: 1921-1950, II: 1951-1980, and III: 1981-2004) and get the average of each period, dormancy release date of period III was accelerated by as much as 15 days compared with that of period I at all locations except Jeju (located in the southernmost island with subtropical climate) where an average15-day delay was predicted. The cold- tolerant period was also shortened at 6 out of 8 locations. As a result, budburst of 'Campbell Early' in spring was accelerated by 6 to 10 days at most locations, while inter-annual variation in budburst dates was increased at all locations. The earlier budburst after the 1970s was due to (1) warming in winter resulting in earlier dormancy release (Incheon, Mokpo, Gangneung, and Jeonju), (2) warming in early spring accelerating regrowth after breaking dormancy (Busan and Jeju), and (3) both of them (Seoul and Daegu).
The bacteriological and physicochemical analysis of sea water and sediments in south western part of Jinhae Bay was conducted. The samples were collected from 40 stations, which were established once a month from January to December, 2000. During the study period, the range of temperature was from 5.5 to $23.7^{\circ}C$, the concentration of chemical oxygen demand ranged from 1.20 to 1.55 mg/L, dissolved oxygen ranged from 3.7 to 9.1 mg/L, dissolved inorganic nitrogen ranged from 2.79 to 7.09 $\mu$g-at/L phosphate phosphorus ranged from 0.39 to 0.57 $\mu$g-at/L, and chlorophyll-$\alpha$g-at/L ranged from 4.28 to $9.66 mg/m^{3}$. The chemical oxygen demand, acid volatile sulfide and ignition loss of sediments in south western part of Jinhae Bay ranged from 0.04 to 0.40, from 24.23 to 35.52 mg/L and from 10.49 to $11.09\%$ respectively. The coliform group and fecal coliform MPN's of sea water in south western part of Jinhae Bay ranged from <3.0 to 1,600 MPN/1003nL (means <3.0 MPN/100 mL) and from <3.0 to 93 MPN/100 mL (means <3.0 MPN/ 100 mL), respectively. The coliform group was classified with IMViC reactions were analyzed. Two hundred eighteen strains that were obtained from sea water samples in south westen of Jinhae Bay represented Escherichia coli group, $61.9\%$; Citrobacter freundii group, $12.1\%$; Enterobacter aerogenes, $14.1\%$; and unknown, $11.9\%$.
Objective of research is obtain fundamental data of carbonized wood wastes for soil condition, de-ordorization, absorption of water, carrier for microbial activity, and purifying agent for water quality of river. The carbonization technique and the properties of carbonized wood wastes(wood-based materials) were analyzed. Proximate analysis showed the wood-based materials contains 0.37~2.27% ash, 70~74% volatile matter, and 17~20% fixed carbon. As carbonization temperature was increased, the charcoal yield was decreased. However, no difference in charcoal yield was found due to time increase. The specific gravity after the carbonization decreased about 30~40% comparing to green wood. The charcoal had 1.08~4.18% ash, 5.88~13.79% volatile matter, and 80.15~90.94% fixed carbon. The pH of plywood and particleboard(pH 9 at $400^{\circ}C$, pH 10 at $600^{\circ}C$ and $800^{\circ}C$) made charcoals was higher than that of fiberboard. The water-retention capacity was not affected by the carbonization temperature and time. The water-retention capacity within 24h was about 2~2.5 times of sample weight, and the Equilibrium moisture content(EMC) became 2~10% after 24h. EMC of charcoal from the thinned trees were 9.40~11.82%($20^{\circ}C$, RH 90%), 6.87~7.61%($20^{\circ}C$, RH 65%), and 1.69~2.81%($20^{\circ}C$, RH 25%). EMC of charcoal from the wood-based materials under $20^{\circ}C$, relative humidity(RH) 90% was similar to EMC of charcoal from the thinned trees(9~11 %). However, under $20^{\circ}C$, RH 25.65%, EMC of charcoal from the wood-based materials were higher(2~3%) than EMC of charcoal from the thinned trees. Every charcoal from the wood-based materials fulfilled the criteria in JWWA K 113-1947.
Background: Central nervous system complication after coronary artery bypass grafting(CABG) is one of the major prognostic determinants and the use of the cardiopulmonary bypass(CPB) may increase the incidence of this devastating complication. In this study, the outcomes after off-pump CABG were studied and compared with those following the conventional CABG using CPB. Material and Method: Among the consecutive isolated CABG's performed in SNUH during Feb. 1995 and Jun. 1999, 338 coronary artery bypass grafting were divided into two groups. 223 patients underwent CABG using the CPB(Group I), and 115 patients underwent CABG without CPB(OPCAB)(Group II). All patients enrolled in this study received extensive preoperative examinations including thorough neurologic examination before and after surgery, transcranial doppler study, carotid duplex ultrasonography, and magnetic resonance angiography if necessary. Central nervous system(CNS) complications were defined as stroke, seizure, metabolic or hypoxic encephalopathy and transient delirium after surgery. Result: There were 61 cases(27.3%) who developed postoperative CNS complication in Group I, whereas 8 cases(7.0%) of CNS complications developed postoperatively in group II(p<0.05). Statistically significant predictors of postoperative CNS complications in group I were age and the use of cardiac assist devices perioperatively. Conclusion: This study suggested that omitting the use of CPB in CABG resulted in significant decrease of the postoperative CNS complications. OPCAB should be more widely applied especially to the elderly who have preexisting cerebrovascular disease.
The purpose of this study was to provide the fundamental information for establishing the database needed to estimate total intakes of trans fatty acids in Korea. The amounts of trans fatty acids contained in 164 samples including 25 samples of margarines, 21 samples of shortenings, 19 samples of vegetable salad and cooking oils, 53 samples of confectionery products, 18 samples of bakery products, 19 samples of dairy products, and 9 samples of animal fats and meats were analyzed by capillary gas liquid chromatography. The average amounts of trans fatty acids in those foods were calculated and expressed as gram per one serving. Then, the average daily intakes of trans fatty acids per capita were estimated using the analyzed amounts of trans fatty acids and the amount of yearly production for those foods. The amounts of trans fatty acids per 100 g of lipids were $2.11{\sim}33.83%$ (14.66% on average) in margarines, $1.47{\sim}44.48%$ (14.21% on average) in shortenings, $0.18{\sim}3.82$ (1.54% on average) in vegetable salad and cooking oils, $0{\sim}45.81%$ (10.92% on average) in confectionery products, $0{\sim}18.32%$ (7.87% on average) in bakery products, $0.90{\sim}4.54%$ (2.27% on average) in dairy products, and $0.61{\sim}6.07%$ (2.24% on average) in animal fats and meats. Major isomers of trans fatty acid in the sample foods were $C_{18:1}$ and $C_{18:2}$. As a result, the korean average daily intake of trans fatty acids in korea was estimated to be 2.3 g per capita. The amounts of trans fatty acids consumed from each selected food were as follows: 0.35 g from margarines, 0.57 g from shortenings, 0.11 g from vegetable salad and cooking oils, 0.65 g from confectionery products, 0.07 g from bakery products, 0.14 g from dairy products and 0.21 g from animal fats and meats.

② 유료서비스를 이용하려는 회원은 정해진 요금체계에 따라 요금을 납부해야 합니다.
제 5 장 계약 해지 및 이용 제한
제 15 조 (계약 해지)
회원이 이용계약을 해지하고자 하는 때에는 [가입해지] 메뉴를 이용해 직접 해지해야 합니다.
제 16 조 (서비스 이용제한)
① 당 사이트는 회원이 서비스 이용내용에 있어서 본 약관 제 11조 내용을 위반하거나, 다음 각 호에 해당하는
경우 서비스 이용을 제한할 수 있습니다.
- 2년 이상 서비스를 이용한 적이 없는 경우
- 기타 정상적인 서비스 운영에 방해가 될 경우
② 상기 이용제한 규정에 따라 서비스를 이용하는 회원에게 서비스 이용에 대하여 별도 공지 없이 서비스 이용의
일시정지, 이용계약 해지 할 수 있습니다.
제 17 조 (전자우편주소 수집 금지)
회원은 전자우편주소 추출기 등을 이용하여 전자우편주소를 수집 또는 제3자에게 제공할 수 없습니다.
제 6 장 손해배상 및 기타사항
제 18 조 (손해배상)
당 사이트는 무료로 제공되는 서비스와 관련하여 회원에게 어떠한 손해가 발생하더라도 당 사이트가 고의 또는 과실로 인한 손해발생을 제외하고는 이에 대하여 책임을 부담하지 아니합니다.
제 19 조 (관할 법원)
서비스 이용으로 발생한 분쟁에 대해 소송이 제기되는 경우 민사 소송법상의 관할 법원에 제기합니다.
[부 칙]
1. (시행일) 이 약관은 2016년 9월 5일부터 적용되며, 종전 약관은 본 약관으로 대체되며, 개정된 약관의 적용일 이전 가입자도 개정된 약관의 적용을 받습니다.
