Polymerization and Optical Properties of Polymers with High Tensile Strength Added Isocyanate Group

  • Polyurethane resin containing isocyanate is marked by excellent tensile and mechanical strengths and this test aims to gauge its applicability as a medical high polymer. Tris [2-(acryloyloxy)ethyl]isocyanurate and hexamethylenediisocyanate were added to a basic mixing ratio of HEMA (2-hydroxyethyl methacrylate), MMA (methyl methacrylate), NVP (n-vinyl-2-pyrrolidone) and crosslink agent, EGDMA (ethylene glycol dimethacrylate) with increasing proportions and copolymerized respectively. Also, the basic physical properties of the polymerized high polymers including refraction rate, tensile strength, light transmission and water content were measured to confirm that they are appropriate as hydrogelcontact lenses. After measuring the physical properties of high performance polymers produced by adding tris [2-(acryloyloxy) ethyl]isocyanurate, it was found that the average tensile strengths of sample TRIS1 to TRIS10 were between 0.285 and 0.612 kgf, while the average values of refractive index were ranged from 1.441 to 1.449 with water content from 30.00 to 37.35%.The measurement of physical properties of the copolymers generated by adding hexamethylenediisocyanate showed that the average tensile strength of sample HEXA1 to HEXA10 ranged from 0.267 to 1.742 kgf, the refractive index ranged from 1.443 to 1.475 and water contents were in the range of 21.22 to 35.58%. In all combinations the transmission rates satisfied the transmittance of general hydrogel contact lenses. From theresults, it is possible to conclude that the produced copolymers can be used as contact lens materials with excellent tensile strength.

Transport Characteristics of Alcohol Solutes through Copolymer Hydrogel Membranes Containing Poly(2-Hydroxyethylmethacrylate) (Poly(2-Hydroxyethylmethacrylate)를 포함한 공중합체 수화겔막에 대한 알콜용질의 투과특성)

  • Three kinds of hydrogel membranes were prepared by the copolymerization of 2-hydroxyethylmethacrylate (HEMA) with acrylamide, N, N-dimethylamide and methylmethacrylate in the presence of solvent and crosslinker respectively. The equilibrium water content, relative permeability and partition coefficient of the membranes for alcohol solutes were measured. It has been found that the permeation of organic solute occurs through the water-filled regions in the hydrogel membrane, and that the gpermeability coefficient of organic solute depends on the molecular size. But the permeability of organic solute was controlled by the interaction of solute-membrane at the low water content. By the partition data, it has been shown that the partition of solute is only controlled by hydrophobic interaction between solute and membrane. The diffusion coefficient data were interpreted on the basis of water-solute interaction. It has been found that the diffusion of organic solute is determined by the free volume of water in the membrane, and that hardly depends on polarity-polarizability and hydrogen bonding ability between water and solute.

The effect of dentin desensitizers and Nd:YAG laser pre-treatment on microtensile bond strength of self-adhesive resin cement to dentin

  • PURPOSE. The purpose of this study is to evaluate if pre-treatment with desensitizers have a negative effect on microtensile bond strength before cementing a restoration using recently introduced self-adhesive resin cement to dentin. MATERIALS AND METHODS. Thirty-five human molars' occlusal surfaces were ground to expose dentin; and were randomly grouped as (n=5); 1) Gluma-(Glutaraldehyde/HEMA) 2) Aqua-Prep F-(Fluoride), 3) Bisblock-(Oxalate), 4) Cervitec Plus-(Clorhexidine), 5) Smart protect-(Triclosan), 6) Nd:YAG laser, 7) No treatment (control). After applying the selected agent, RelyX U200 self-adhesive resin cement was used to bond composite resin blocks to dentin. All groups were subjected to thermocycling for 1000 cycles between $5-55^{\circ}C$. Each bonded specimen was sectioned to microbars ($6mm{\times}1mm{\times}1mm$) (n=20). Specimens were submitted to microtensile bond strength test at a crosshead speed of 0.5 mm/min. Kolmogorov-Smirnov, Levene's test, Kruskal-Wallis One-way Analysis of Variance, and Conover's nonparametric statistical analysis were used (P<.05). RESULTS. Gluma, Smart Protect and Nd:YAG laser treatments showed comparable microtensile bond strengths compared with the control group (P>.05). The microtensile bond strengths of Aqua-Prep F, and Cervitec Plus were similar to each other but significantly lower than the control group (P<.05). Bisblock showed the lowest microtensile bond strength among all groups (P<.001). Most groups showed adhesive failure. CONCLUSION. Within the limitation of this study, it is not recommended to use Aqua-prep F, Cervitec Plus and Bisblock on dentin when used with a self-adhesive resin cement due to the decrease they cause in bond strength. Beside, pre-treatment of dentin with Gluma, Smart protect, and Nd:YAG laser do not have a negative effect.

  • Chitosan, a natural polymer, has been importantly considered as biomedical materials due to its good biocompatibility and various bio-active characteristes. Water soluble chitosan was then copolymerized EGDMA(ethylene glycol dimethacrylate; used as a cross-linking agent for the free-radical copolymerization), MMA (methylmethacrylate), MA (methacrylic acid) in the presence of AIBN (azobisisobutyronitrile) as a radical initiator. The water content and visible transmissibility, ultimate strength of copolymerized ophthalmic polymer were measured to be 24$\sim$59%, 88$\sim$89% and 0.1$\sim$2.4 Kgf, respectively. And also, we tested for antimicrobial activities against staphylococcus aureus, Pseudomonas aeruginosa. They showed that in case of antimicrobial activities, the values including chitosan were much higher than that of the polymers of no including chitosan, suggesting that the copolymer can be used as a novel ophthalmic material of high performance.

The genomic landscape associated with resistance to aromatase inhibitors in breast cancer

  • Aromatase inhibitors (AI) are drugs that are widely used in treating estrogen receptor (ER)-positive breast cancer patients. Drug resistance is a major obstacle to aromatase inhibition therapy. There are diverse reasons behind acquired AI resistance. This study aims at identifying the plausible cause of acquired AI resistance in patients administered with non-steroidal AIs (anastrozole and letrozole). We used genomic, transcriptomic, epigenetic, and mutation data of breast invasive carcinoma from The Cancer Genomic Atlas database. The data was then separated into sensitive and resistant sets based on patients' responsiveness to the non-steroidal AIs. A sensitive set of 150 patients and a resistant set of 172 patients were included for the study. These data were collectively analyzed to probe into the factors that might be responsible for AI resistance. We identified 17 differentially regulated genes (DEGs) among the two groups. Then, methylation, mutation, miRNA, copy number variation, and pathway analyses were performed for these DEGs. The top mutated genes (FGFR3, CDKN2A, RNF208, MAPK4, MAPK15, HSD3B1, CRYBB2, CDC20B, TP53TG5, and MAPK8IP3) were predicted. We also identified a key miRNA - hsa-mir-1264 regulating the expression of CDC20B. Pathway analysis revealed HSD3B1 to be involved in estrogen biosynthesis. This study reveals the involvement of key genes that might be associated with the development of AI resistance in ER-positive breast cancers and hence may act as a potential prognostic and diagnostic biomarker for these patients.

Changes of Lens Morphology and TBUT by Dehydration of Soft Contact Lens (소프트콘택트렌즈 건조로 인한 렌즈 형태 및 TBUT의 변화)

  • Purpose: This study was performed to evaluate the changes of lens morphology and tear stability during wearing soft contact lenses (SCLs) which were kept in drying condition like dry eye or became to be dried due to heedless care. Method: SCLs having different water content, thickness or material were rehydrated after being dehydrated artificially 2 or 4 times, and estimated their diameter and radius. Furthermore, the changes of tear film break-up time (TBUT) during SCL wearing were also measured. Result: Due to the dryness, the diameter of both 70% water content SCL and 59% water content SCL decreased, but the decrement was larger in 59% water content SCL. The more 59% water content SCL was dehydrated, the more its radius changed. However, the radius of 70% water content SCL did not change by 2 times dehydration and increased greatly by 4 times dehydration. The reduction of diameter of -1.00 D SCL was greater than that of -9.00 D SCL. Moreover, the radius of -1.00 D SCL increased depending on the frequency of dehydration but that of -9.00 D SCL did not changed. The diameter and radius changes of lotrafilcon B, silicone hydrogel lens, were less than those of hilafilcon B, copolymer of HEMA and N-vinyl pyrrolidone. TBUT during wearing SCLs decreased by wearing dehydrated SCLs. Conclusion: The diameter and radius of dehydrated SCLs as well as TBUT during wearing them were changed in spite of rehydration, which would be the important cause of uncomfortable feeling when people wore dehydrated SCL. The changes of SCL morphology and TBUT differed according to the water content, lens thickness and material.

Comparison of Contact Lenses and Storage Solution for Cytotoxic Potential using a Cell Growth Inhibition Assay (소프트 콘택트렌즈 용출액과 보존 용액의 세포 독성 비교)

  • Human corneal epithelial cells and mouse fibroblast L929 cells were grown to 60-70% confluency in 96 well plates. 24 hours after the well plates are inoculated, the medium on the test plates is aspirated and replaced with an extract supplemented medium prepared from the materials to be tested. The contact lenses manufactured by 7 companies were collected from optical shops and used for this study. The exactracts having been prepared by autoclaving 8 lenses in 2.5ml saline. The cell monolayer is then cultured for a further 48 hour period. MTT and SRB assys were performed for cytotoxic effect on cultured cells An inhibition of 30% is considered clear indication of cytotoxic potential in the test material. All the materials were not cytotoxic, but 3 storage solutions of them inhibited growing L929 cells.

Synthesis and Characterization of Novel pH-Sensitive Hydrogels Containing Ibuprofen Pen dents for Colon-Specific Drug Delivery

  • The aim of this study was to develop novel intestinal specific drug delivery systems with pH sensitive swelling and drug release properties. The carboxyl group of ibuprofen was converted to a vinyl ester group by reacting ibuprofen and vinyl acetate as an acylating agent in the presence of catalyst. The glucose-6-acrylate-1, 2, 3, 4-tetraacetate (GATA) monomer was prepared under mild conditions. Cubane-1, 4-dicarboxylic acid (CDA) linked to two 2-hydroxyethyl methacrylate (HEMA) group was used as the crosslinking agent (CA). Methacrylic-type polymeric prodrugs were synthesized by the free radical copolymerization of methacrylic acid, vinyl ester derivative of ibuprofen (VIP) and GATA in the presence of cubane cross linking agent. The structure of VIP was characterized and confirmed by FTIR, $^1H$ NMR and $^{13}C$ NMR spectroscopy. The composition of the cross-linked three-dimensional polymers was determined by FTIR spectroscopy. The hydrolysis of drug polymer conjugates was carried out in cel-lophane membrane dialysis bags, and the in vitro release profiles were established separately in enzyme-free simulated gastric and intestinal fluids (SGF, pH 1 and SIF, pH 7.4). The detection of a hydrolysis solution by UV spectroscopy at selected intervals showed that the drug can be released by hydrolysis of the ester bond between the drug and polymer backbone at a low rate. Drug release studies showed that increasing the MAA content in the copolymer enhances the rate of hydrolysis in SIP. These results suggest that these polymeric prodrugs can be useful for the release of ibuprofen in controlled release systems.

The Effects of the Content of Isobornyl Methacrylate in Acrylate Copolymers on Physical Properties (아크릴계 공중합체에서 이소보닐 메타크릴레이트의 함량에 의한 물리적 특성 변화)

  • The acrylate copolymer having good thermal stability, coating and adhesion properties was designed and prepared. We prepared copolymers in >95% high yield using methyl methacrylate, isobornyl methacrylate and 2-hydroxyethyl methacrylate monomers by the bulk and emulsion polymerization techniques. The $^1H$-NMR spectrum was used to identify chemical structure and glass transition temperatures increased from $123^{\circ}C$ to $140^{\circ}C$ confirmed by DSC, DMA and TGA analysis. In addition, as the content of IBMA increased, storage modulus and thermal decomposition temperature increased. As the content of IBMA increased from 10% to 30% in the composition for the entire monomer, tensile strength increased from 22 to 30 MPa in both polymers prepared by bulk and emulsion techniques. The contact angle increased from 70 to up to 88 degrees due to hydrophobic property of IBMA.

Adsorption Properties of the Lysozyme and Albumin with Physicochemical Properties of the Contact Lens (콘택트렌즈의 물리화학적 특성에 따른 라이소자임과 알부민의 흡착 특성)

  • Purpose: Adsorption properties of lysozyme and albumin according to physiochemical properties of commercial contact lens classified with the FDA categories and a contact lens fabricated in the laboratory were investigated. Methods: The contact lens were prepared using HEMA(2-hydroxyethyl methacrylate) and TRIM(3-(trimethoxysilyl) propyl methacrylate) in a cast mold. Artificial tears containing lysozyme and albumin were prepared. We measured the amounts of protein adsorbed on the each lenses with varying adsorbed time (48 hour) and the pH range (6, 6.8, 7.4, 8.2, 9) of artificial tear. Amount of the proteins absorbed on the contact lenses were measured by using HPLC. Results: Time to reach the equilibrium of protein adsorption for silicone hydrogel lens was taken longer than hydrogel lens. The amount of adsorbed both lysozyme and albumin at equilibrium were greater for the hydrogel lens than the silicone hydrogel lens, and larger for the ionic lens than the non-ionic lens. Lysozyme was more adsorbed on the higher water content of contact lens, whereas albumin was more adsorbed on the lower water content of contact lens. Only lysozyme was adsorbed on the Group IV hydrogel lens of ionic higher water content. The adsorption of protein on contact lens increased with pH of artificial tears as close to the isoelectric point of each protein. Conclusions: The adsorption amount of lysozyme is more affected by the ionic strength of the contact lens surface than the water content of contact lens. Albumin adsorption is more affected by water content than the ionic strength of the contact lens surface. For the adsorption of proteins on the silicone hydrogel lens, the pore size, determined both by the number of Si atoms and the chemical structure of the silicone-containing monomers, as well as the polarity of contact lens should be also considered.
