양송이 버섯의 생물활성 다당류에 관한 연구 (Studies on Bioactive Polysaccharide Isolated from Agaricus bisporus)

  • 10종의 식용 버섯 자실체에 대한 항보체 활성 검색에서 가장 높은 활성을 나타낸 양송이버섯 자실체를 대상으로 항보체 활성물질의 추출조건을 조사하였다. 최적조건으로서 5% urea가 함유된 0.1 N NaOH 용액으로 $65^{\circ}C$에서 2시간동안 양송이버섯 자실체를 추출한 후 methanol 가용획분이 제거된 조다당 획분 AB-0를 얻었다. AB-0는 1 mg/ml에서 90% 이상의 높은 항보체활성을 나타내었으며 마우스에 이식된 sarcoma-180에 대하여 74%의 저지효과를 보였다. AB-0를 acetone 농도에 따른 침전 분획을 실시하여 AB-20, AB-40, AB-60, AB-80, AB-A의 5개의 획분을 얻었다. 이중 가장 높은 항보체활성과 수율을 나타낸 AB-20 획분은 탄수화물 39%, 단백질 46%를 함유하였으며, 구성당은 glucose, arabinose, xylose, galactose, mannose가 6.49 : 1.98 : 1.24 : 1.00 : 0.71의 비율로 존재하였고 구성아미노산은 isoleucine(12.60%), glutamic acid+glutamine(12.45%), valine(11.79%), alanine(11.46%), leucine(10.19%)와 aspartic acid, asparagine(10.56%) 등 이었다. Pronase 처리한 AB-20에서는 78.5%의 항보체활성을 나타내고 대조군에 비해 14.5% 감소하였으나, AB-20의 periodate 산화물에서는 활성이 42.6%로 50% 이상 크게 감소하였다. 따라서 양송이 버섯의 알칼리 추출물중 가장 강한 활성을 보인 AB-20의 주요 활성부위는 다당류이며 단백질도 일부 관여하는 것으로 생각되었다.

운동강도의 차이가 흰쥐의 심근 세포기질 및 미토콘드리아 LDH 동위효소 변화에 미치는 영향 (Effects of Different Exercise Intensities on Cytosolic and Mitochondrial LDH Isozymes of Cardiac Muscle in Rats)

  • 생후 7주령 흰쥐를 대상으로 저강도집단, 중강도집단, 고강도집단, 통제집단으로 분리하여 8주 동안 트레드밀 운동을 실시한 후 심장을 적출하여 조직의 세포기질을 분리하고 전기영동을 실시하여 세포기질 LDH 동위효소의 분포양상을 확인하였고, 활성을 측정하였다. 또한 미토콘드리아를 순수분리한 뒤 분리된 미토콘드리아를 외막 내막 막간물질, 매트릭스로 각각 분리하여 각 분획을 전기영동을 실시하고 효소활성을 측정하였다. 운동 강도에 따른 장기간의 지구성훈련으로 심장조직의 세포기질 LDH 동위효소 활성은 운동집단과 비 운동집단에서 모든 LDH 동위효소의 활성을 나타냈다. 세포기질 LDH 동위효소의 운동 강도에 따른 활성변화는 모든 동위효소가 운동강도의 증가에 따라 증가하는 결과를 나타냈으며 모든 강도에서 $AB_3$ 동위효소의 활성이 가장 크게 나타났다. 심장조직 미토콘드리아 각 분획에서 운동 강도에 따른 LDH 등위효소의 분포와 활성은 외막의 경우 다른 분획에 비하여 동위효소의 분포 및 활성이 매우 약하게 나타났다. 운동강도가 증가하면서 모든 분획의 동위효소가 $B_4$쪽으로 활성이 강해지는 현상을 보이며, 외막에서 내막, 매트릭스로 이동하면서 각 분획에 LDH 동위효소의 분포도 많아지고 활성도 또한 증가하는 현상을 나타냈다.

$AB^2$ 연산을 위한 세미시스톨릭 구조 설계 (Design of Semi-Systolic Architecture for $AB^2$ Operation)

  • 본 논문에서는 $GF(2^m)$상에서 $AB^2$ 연산을 위한 세미시스톨릭 구조를 제안한다. 먼저 기존의 세미시스톨릭 구조를 통하여 문제점을 제시하고, 이러한 문제점을 해결하기 위한 AOP(All One Polynomial)에 기반 한 새로운 $AB^2$ 알고리즘을 제안하고 이를 위한 새로운 구조를 제안한다. 본 논문에서 제안한 구조는 기존의 구조들보다 효율적인 구성을 가진다 제안된 구조는 공개키 암호의 핵심이 되는 지수기의 구현을 위한 효율적인 기본구조로 사용될 수 있다.

엄지발가락 벌림을 강조한 PNF 하지 패턴이 엄지발가락가쪽휨증을 지닌 대상자의 발의 내재근 근활성도에 미치는 영향 (The Impact of PNF Leg Patterns Hallux Abduction on the Intrinsic Foot Muscles of Participants with Hallux Valgus)

  • Purpose: This study aimed to compare the impact of proprioceptive neuromuscular facilitation leg patterns emphasizing hallux abduction (PNF-LPHA) on the intrinsic foot muscles of participants with hallux valgus (HV) using the toe-spread-out exercise (TSO). Methods: The present study recruited 12 individuals with HV. All the participants voluntarily agreed to participate in the study after hearing explanations of its purpose and process. All participants performed the TSO, PNF-LPHA 1, and PNF-LPHA 2. The participants' abductor hallucis (AbH), adductor hallucis (AdH), extensor hallucis longus (EHL), and flexor hallucis brevis (FHB) activity was measured, and the ratio of AbH:AdH was measured during the three interventions using electromyography. Additionally, the participants' AbH thickness was measured by ultrasonography. An intraclass correlation coefficient (ICC) was used to verify the intra-rater reliability of ultrasonography at rest and during contraction. Results: The intra-rater reliability was excellent at rest and during contraction ($ICC_{3,1}=0.90$ and $ICC_{3,1}=0.83$, respectively). There were no statistically significant differences in the activity of the AbH, the ratio of AbH: AdH, and the thickness of AbH between the TSO and PNF-LPHA2 groups. Additionally, EHL activity was significantly higher in the PNF-LPHA2 group than in the TSOgroup. Conclusion: PNF-LPHA 2 can be recommended as a method to optimize AbH and EHL activity, the ratio of AbH:AdH, and the thickness of AbH in individuals with HV.

The MAP Kinase Kinase Gene AbSte7 Regulates Multiple Aspects of Alternaria brassicicola Pathogenesis

  • Mitogen-activated protein kinase (MAPK) cascades in fungi are ubiquitously conserved signaling pathways that regulate stress responses, vegetative growth, pathogenicity, and many other developmental processes. Previously, we reported that the AbSte7 gene, which encodes a mitogen-activated protein kinase kinase (MAPKK) in Alternaria brassicicola, plays a central role in pathogenicity against host cabbage plants. In this research, we further characterized the role of AbSte7 in the pathogenicity of this fungus using ${\Delta}AbSte7$ mutants. Disruption of the AbSte7 gene of A. brassicicola reduced accumulation of metabolites toxic to the host plant in liquid culture media. The ${\Delta}AbSte7$ mutants could not efficiently detoxify cruciferous phytoalexin brassinin, possibly due to reduced expression of the brassinin hydrolase gene involved in detoxifying brassinin. Disruption of the AbSte7 gene also severely impaired fungal detoxification of reactive oxygen species. AbSte7 gene disruption reduced the enzymatic activity of cell walldegrading enzymes, including cellulase, ${\beta}$-glucosidase, pectin methylesterase, polymethyl-galacturonase, and polygalacturonic acid transeliminase, during host plant infection. Altogether, the data strongly suggest the MAPKK gene AbSte7 plays a pivotal role in A. brassicicola during host infection by regulating multiple steps, and thus increasing pathogenicity and inhibiting host defenses.

Synthesis and Photoaffinity Labeling of 3'(2')-O-(p-azidobenzoyl) ATP

  • A photoactive analog of ATP, 3'(2')-O-(p-azidobenzoyl)-adenosine 5-triphosphate (AB-ATP) was synthesized by chemically coupling N-hydroxysuccinimidyl-4-azidobenzoate (NHS-AB) and ATP. The utility of AB-ATP as an effective active-site-directed photoprobe was demonstrated using catalytic subunit of protein kinase A as a model enzyme. Photoincorporation of AB-ATP was saturated with apparent dissociation constant of $30{\mu}m$ and protected completely by $100{\mu}m$ of ATP. When the enzyme was covalently modified by photolysis in the presence of saturating amounts of photoprobe, about 60% inhibition of enzyme activity was observed. These results demonstrate that AB-ATP has potential application as a probe to characterize ATP-binding proteins including protein kinases.

Mucosubstance Histochemistry of the Epidermis in Yellowtail, Striped Beakperch, Brown Spotted Grouper, Sea Chub, and Multicolorfin Rainbowfish

  • To investigate the properties of mucosubstances of the epidermis in various teleostean species, conventional histochemical stainings were used on the skin in five species of order Perciformes, i. e., yellowtail, Seriota quinqueradiat, striped beakperch, Oplegnathus fasciatus, brown spotted grouper, Epinephelus chlorostigma, sea chub, Ditrema temmincki and multicolorfin rainbowfish, Halichoeres poecilopterus. The following methods were used: periodic acid Schiff (PAS), alcian blue (AB) pH at 2.5, AB pH at 1.0, AB pH at 2.5-PAS, AB pH at 1.0-PAS, aldehyde fuchsin (AF) pH at 1.7-AB pH at 2.5 and high iron diamine (HID)-AB pH at 2.5. The epidermis of all five species consisted of three layers: superficial, middle, and basal layer. The superficial layer was comprised of rather flattened cells. In particular, the outermost layer of striped beakperch and middle layer of sea chub consisted of mucus-secreting cells. Mucous cells, the unicellular glands, were found in epidermis but varied in number in different body regions and species. Although there was a slight difference in the amount in various species and body regions, the secretory contents of the mucous cells in the five teleostean species contained acidic mucopolysaccharides. In yellowtail, striped beakperch, and multicolorfin rainbowfish, the property of mucosubstances was identified as sialomucin, while it was sulphomucin in brown spotted grouper and sea chub.

Activin A/BMP2 Chimera (AB204) Exhibits Better Spinal Bone Fusion Properties than rhBMP2

  • Objective : To compare the spinal bone fusion properties of activin A/BMP2 chimera (AB204) with recombinant human bone morphogenetic protein (rhBMP2) using a rat posterolateral spinal fusion model. Methods : The study was designed to compare the effects and property at different dosages of AB204 and rhBMP2 on spinal bone fusion. Sixty-one male Sprague-Dawley rats underwent posterolateral lumbar spinal fusion using one of nine treatments during the study, that is, sham; osteon only; $3.0{\mu}g$, $6.0{\mu}g$, or $10.0{\mu}g$ of rhBMP2 with osteon; and $1.0{\mu}g$, $3.0{\mu}g$, $6.0{\mu}g$, or $10.0{\mu}g$ of AB204 with osteon. The effects and property on spinal bone fusion was calculated at 4 and 8 weeks after treatment using the scores of physical palpation, simple radiograph, micro-computed tomography, and immunohistochemistry. Results : Bone fusion scores were significantly higher for $10.0{\mu}g$ AB204 and $10.0{\mu}g$ rhBMP2 than for osteon only or $1.0{\mu}g$ AB204. AB204 exhibited more prolonged osteoblastic activity than rhBMP2. Bone fusion properties of AB204 were similar with the properties of rhBMP2 at doses of 6.0 and $10.0{\mu}g$, but, the properties of AB204 at doses of $3.0{\mu}g$ exhibited better than the properties of rhBMP2 at doses of $3.0{\mu}g$. Conclusion : AB204 chimeras could to be more potent for treating spinal bone fusion than rhBMP2 substitutes with increased osteoblastic activity for over a longer period.

개구리(Rana nigromaculata) 발생에 따른 위점막 점액분비세포의 조직학적 및 조직화학적 연구 (Histological and Histochemical Studies on the Mucous Secreting Cells of the Gastric Mucosae according to the Development of Frog, Rana nigromaculata)

  • 양서류 발생에 따른 위점막 점액세포의 조직학적 및 조직화학적인 변화를 밝히고저 개구리(Rana nigromaculata)변태 각 단계의 위체부 조직을 10% formalin 완충액에 고정($4^{\circ}C$), paraffin에 포매, 4 $\mu$m 두께로 절편한후 periodic acid-Schiff(PAS) 및 alcian blue(AB) pH 2.5, pH 1.0에 반응시켜 다음과 같은 결과를 얻었다. 1. 위 표면점액세포내 점액질은 변태과정에 따라 특이한 변화없이 PAS에 강한 반응을 AB pH 2.5 및 pH 1.0에서는 약한 alcianophilia를 보였으며, 변태 XXIV와 XXV에서 세포내 중성 점액질의 함량이 현저히 증가하였다. 2. 위소와점액세포는 변태 XXI이후에서 볼 수 있었는데 PAS에 강한 반응을, AB pH 2.5 및 pH 1.0에서 약한 alcianophilia를 보였으나, AB pH 1.0에서는 AB pH 2.5에서 보다 강하였고, 변태 XXIV와 XXV에서 세포내 중성 점액질의 함량이 현저히 증가하였다. 3. 분비능을 보이는 점액질세포는 변태 XXIV이후인데 변태 XXIV에서는 PAS에 강양성을, AB pH 1.0에서는 약한 alcianophilia를 보였고, 변태 XXV 이후에는 PAS에 약한 반응을, AB pH 2.5에는 약하고 AB pH 1.0에는 중등도의 alcianiphilia를 보였는데 이들 점액질은 전 세포질에 비교적 고르게 분포하였다.
