Application for Degradation of 2,6-dichlorophenol by in-situ Synthesized Liquid Ferrate(VI) (원위치 제조 액상 Ferrate(VI)를 이용한 2,6-dichlorophenol의 분해적용 연구)

  • Degradation of 2,6-Dichlorophenol (DCP) using liquid ferrate(VI) synthesized by wet oxidation method has been studied. Several parameters such as pH (acid, base and neutral), DCP initial concentration, ferrate dosage, and temperature have been examined to determine the optimal experimental conditions. The ferrate(VI) has useful properties such as strong oxidizing power and selectivity and generates a non-toxic end product, Fe(III). Ferrate ion reduced rapidly to Fe(III) and oxygen in acidic and neutral conditions. The experimental results showed the higher DCP degradation efficiency in the neutral condition than in the acidic and basic conditions. The oxidation of DCP strongly depended on the dosage of ferrate added to the reactor and DCP initial concentration. With increasing of ferrate dosage the degradation efficiency of DCP increased, while the degradation efficiency of DCP decreased with increasing of DCP initial concentration. The effect of temperature has been tested at 4 different levels (10, 25, 35, and $50^{\circ}C$). The optimal temperature was obtained in $25^{\circ}C$ and degradation efficiency decreased as the temperature increased in the range from $25^{\circ}C$ to $50^{\circ}C$. The DCP degradation pathways were studied and proposed based on the intermediate products identified by GC/MS analysis.

A Fiber Optic Sensor for Determination of 2,4-Dichlorophenol Based on Oxygen Oxidation Catalyzed by Iron(III) Tetrasulfophthalocyanine

  • A new fiber optical sensor was developed for the determination of 2,4-dichlorophenol (DCP). The sensor was based on DCP oxidation by oxygen with the catalysis of iron(III) tetrasulfophthalocyanine (Fe(III)PcTs). The optical oxygen sensing film with $Ru(bpy)_3Cl_2$ as the fluorescence indicator was used to determine the consumption of oxygen in solution. A lock-in amplifier was used for detecting the lifetime of the oxygen sensing film by measuring the phase delay change of the sensor head. The different variables affecting the sensor performance were evaluated and optimized. Under the optimal conditions (i.e. pH 6.0, $25^{\circ}C$, Fe(III)PcTs concentration of 0.62 mg/mL), the linear detection range and response time of the sensor are $1.0{\times}10^{-6}-9.0{\times}10^{-6}$ mol/L and 250 s, respectively. The sensor displays high selectivity, good repeatability and stability, and can be used as an effective tool in analyzing DCP concentration in practical samples.

A Comparison of the IAS and Langmuir Models for Multisolute Adsorption of Organic Cowlpounds in Soil (유기화합물들이 혼합상태에서 토양입자에 흡착하는 정도를 IAS와 Langmuir Model을 이용한 예측비교연구)

  • The Langmuir competitive model and the IAS(ideal adsorption solution) model were eveluated and compared in a multisolute adsorption study using five organic compounds (phenol, 2, 4-dichlorophenol, 2, 4, 6-trichlorophenot brucine, and thiourea) and two soils. The chemicals were evaluated individually and in mixtures. In general, the IfS model predicted the equilibrium concentration of a chemical in a mixture better than the Langmuir model. The Langmuir model underestimated the sorption of phenol when the concentration of another compound in a mixture with phenol was high. Neither of the models predicted satisfactorily the equilibrium concentration of thiourea in the mixtures. Thiourea is an aliphatic compound while the other four chemicals are aromatic compounds.

Purification and Characterization of 2,4-Dichlorophenol Oxidizing Peroxidase from Streptomyces sp. AD001

  • Streptomyces sp. AD001 is a Gram-positive soil actinomycetes secreting an uncharacterized 2,4-dichlorophenol (DCP) oxidizing enzyme, whose activity is similar to the previously known Actinomycetes lignin-peroxidase (ALiP). This extracellular peroxidase was purified from Streptomyces sp. AD001 as a single protein band on an SDS-PACE by ammonium sulfate fractionation, Q-sepharose, concanavalin A, and Bio-Gel HTP column chromatographies. The molecular mass of the purified peroxidase was determined by SDS-PAGE to be 45.2 kDa, and 49.7 kDa with MALDI-TOF-MS, respectively. The highest level of peroxidase activity was observed at pH 7.5 and $30^{\circ}C$. The amino terminal sequence of the purified peroxidase (G-E-P-E-E-G-N-V-D-G-T-L) showed no significant homologies to my known proteins, suggesting that Streptomyces sp. AD001 may secrete a novel kind of bacterial peroxidase Initial rate kinetic data of the 2,4-DCP oxidation were best modeled with a random-binding bireactant system.

The Effect of Electron Donor on Reductive Dechlorination of Chlorophenols (염소계페놀의 환원적 탈염소화에서의 전자공여체의 영향)

  • Batch experiments were conducted to investigate the effect of electron donor on reductive dechlorination of 2,4,5-trichlorophenol by a methanogenic consortium. The methanogenic consortium was obtained from the anaerobic digester of a municipal wastewater treatment plant. The batch reactor containing methanogenic consortium was spiked with 2,4,6-trichlorophenol at 10 mg/$\ell$. Acetate, ethanol, glucose of methanol, each was added as an electron donor for methanogenic consortium. During the course of the experiments liquid samples were taken from the batch reactor to measure dechlorination rate and find the dechlorination pathway of 2,4,6-trichlorophenol. After incubation 2,4,6-trichlorophenol was first dechlorinated to 2,4-dichlorophenol and then to 4-chlorophenol. Phenol was not detected in the batch reactor the highest rate of dechlorination of 2,4,6-trichlorophenol was observed when ethanol was used as an electron donor.

Carbon Monoxide Dehydrogenase in Cell Extracts of an Acinetobacter Isolate (Acinetobacter sp.1의 일산화탄소 산화효소의 특성)

  • Extracts of CO-autotrophically grown cells of Acinetobacter sp. 1 were shown to use thionin, methylene blue, or 2,6-dichlorophenol-indophenol, but not NAD, NADP, FAD, or FMN, as electron acceptors for the oxidation of CO under strictly anaerobic conditions. The CO dehydrogenase (CO-DH) in the thes bacterium was found to be an inducible enzyme. The enzyme activity was determined by an assay based on the CO-dependent reduction of thionin. Maximal reaction rates were found at pH 7.5 and $60^{\circ}C$, and the Arrhenius plot revealed an activation energy of 6.1 kcal/mol(25.5kJ/mol). THe $K_m$ m/ for CO was $154{\mu}M$. Known metalchelating agents tested had no effects on the CO-DH activity. No divalent cations tested affect the enzyme activity significantly escept $Cu^{2+}$ which suppressed the activity completely. The enzyme was inhibited by glucose and succinate. The same extracts catalyzed oxidation of hydrogen gas and formate with thionin as electron acceptor. The CO-DH of Acinetobacter sp. 1 was to have no immunological relationship with that of Pseudomonas carboxydohydrogena.

  • The contents of ascorbic acid in milk and fermented milk, yogurt added with fruits were determined by 2.6-dichlorophenol indophenol method using a Spectrophotometer. Threes sample of fruit milks and three of fruit yogurts were collected from the several markets in Busan City. The amounts of the vitamin as the reduced form were 0.16 mg/100m1 of strawberry milk, 0.31mg/100m1 of orange milk, and 0.08mg/100m1 of banana milk. The vitamin was not detected from the fermented fruit yogurt samples.

  • Chlorination of the polluted water may produce odoriferous and objectionable-tasting chlorophenols which are hazardous to health. These studies were undertaken to investigate the hazardous effects of chlorophenols to the rat. 1. The chlorophenols such as o-chlorophenol and 2,6-dichlorophenol inhibited rat growth and caused increment of the ratio between liver weight and body weight. 2. The hemoglobin content, hamatocrit ratio and A/G of rat blood were decreased by chlorophenols administration. The activities of alkaline phosphatase, lactic dehydrogenase (LDH) and glutamate oxaloacetate transaminase (GOT) in serum as well as in liver were increased provisonally and decreased after one or two weeks adminstration. 3. The liver mitochondrial respiration ($QO_{2}$) was inhibited by chlorophenols treatment in in-vivo and in-vitro test. 4. The liver microsomal cytochrome P-450 was decreased by chlorophenols administration 5. Liver tissue was degenerated with congestion, atrophy, swelling, vacuolation, dilation of rough endoplasmic reticulum and denature of mitochondrial particle with swelling, and cristal destruction by chlorophenols adminstration. 6. After one and two weeks of adminstration of chlorophenols to rat, the aberrations of bone marrow chromosome and inhibition of its mitosis were observed respectively.

Isolation and Characterization of Soil Streptomyces Involved in 2,4-Dichlorophenol Oxidation

  • Over 50 morphologically distinctive soil Streptomyces were isolated from various Jocations in the Yongin area in Korea and visually screened for dye-decoloring activities on an agar plate. Two Streptomyces species (AD001 and ND002) showed strong dye-decoloring activities on the plate containing congo-red and new-fuchin dyes, respectively. Also, the liquid culture supernatants of these species showed 2,4-dicholophenol (DCP) oxidation activities only in the presence of hydrogen peroxide, a characteristic of Actinomycetes lignin-peroxidase (ALiP)-P3 isoform found in dye-degrading S. viridosporus T7A and S. badius 252. Based on their dye-decoloring capabilities and the 2,4-DCP oxidation kinetic data, it is suggested that these Streptomyces secrete not-yet-characterized extracelluar enzyme(s), whose activities are very similar to the ALiP-P3 enzyme.

Simultaneous determination of bisphenol A, chlorophenols and alkylphenols by solid-phase extraction and HPLC

  • An analytical method for determining potential endocrine disruptors (bisphenol A, 2-chlorophenol, 2,4-dichlorophenol, 2,4,6-trichlorophenol, pentachlorophenol, p-t-butylphenol, p-pentylphenol, p-hexylphenol, p-t-octylphenol, p-heptylphenol, nonylphenol) by solid-phase extraction (SPE) and High Perfomance Liquid Chromatography(HPLC) equipped with fluorescence and variable wavelength detector has been developed. The SPE process for sample concentration was performed on a commercially available Oasis HLB cartridge packed with polymeric sorbents. The effect of elution solvent and elution volume on the recoveries of the analytes were investigated with HPLC. Average recovery of >85% was achieved with 60mg sorbents using 5mL of methanol as elution solvent. Phenolic compounds in canned drinks, beverages and water samples were surveyed by this proposed method.
