Antioxidant Activity of Chinese Mung Bean (중국산 녹두의 항산화 활성)

  • The aim of this study was to investigate the applicability of Chinese mung bean as a natural antioxidant agent. This study evaluated the phenolic compounds content and antioxidative activity of methanol extract from Chinese mung bean. Antioxidative activities were measured by in vitro models such as 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging activity, and 2,2-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid(ABTS) radical scavenging activity. The contents of total phenolics and total flavonoids of Chinese mung bean extract were $174.83{\pm}2.90GAE\;mg/g$ and $68.87{\pm}2.84QE\;mg/g$, respectively. The antioxidative activities of Chinese mung bean extract were significantly increased in a dose dependent manner on DPPH radical scavenging and ABTS radical scavenging(p<0.05). The concentration of Chinese mung bean extract that reduces the free radical ABTS about 50%($IC_{50}$) was 2.85 mg/mL. These results suggest that Chinese mung bean may have great potential as a natural antioxidant source linked with health benefits.

The Chemical Constituents and their Antioxidant Activity of the Stem of Rhododendron mucronulatum

  • From the n-BuOH soluble traction of the 70% aqueous acetone extract of Rhododendron mucronulatum stem, twelve compounds were isolated. On the basis of spectral data, they were identified as scopoletin (1), (+)-taxifolin (2), quercetin (3), (-)-catechin (4), (+)-epicatechin (5), scopolin (6), lyoniside (7), ssioriside (8), fraxin (9), $(+)-lyoniresinol-3{\alpha}-O-{\beta}-D-glucopyranoside$ (10), $(+)-taxifolin-3-O-{\alpha}-L-arabinopyranoside$ (11), and astragalin (12), respectively. All isolated compounds were tested antioxidant activity against 1, 1-diphenyl-2-picrylhydrazyl (DPPH) radical. Compounds 2 and 3 showed the potent antioxidant activity, and compounds 5, 8, and 11 showed moderate activity.

Antioxidant and Hepatoprotective Effects of the Ethanol Extract of Dendropanax morbifera Leveille on the t-Butyl Hydroperoxide-Induced HepG2 Cell Damages (황칠나무 추출물의 항산화 및 간세포보호효과)

  • Dendropanax morbifera Leveille, an endemic species in Korea, is best known as a tree that produces a resinous sap. Although D. morbifera is used in folk medicine for various diseases, its active ingredients are largely unknown. In this study, we investigated antioxidative activities of ethanolic extracts of three parts of this plant including leaves, debarked stems, and roots. The root extracts exhibited strong 1,1-diphenyl-2-picrylhydrazyl radical (DPPH) scavenging activity compared with leaf or stem extracts. The root extracts showed hepatoprotective activity against t-butyl hydroperoxide-induced HepG2 cells, and reduced the ROS level in the cells. The root fractions lowered the mRNA level of COX-2 on lipopolysaccharide-stimulated Raw246.7 cells. These results suggest that ethanolic root extracts of D. morbifera are a source of antioxidant and hepatoprotective compounds, which indicate a potential for a botanical drug.

In vitro antioxidant and free radical scavenging activities of stem extract of Euphorbia trigona Miller

  • Antioxidative and free radical scavenging properties of different stem extracts of Euphorbia trigona were evaluated and correlated with its total phenolic content. Aqueous, acetone and methanolic extracts of shade dried stem were obtained and were concentrated in vacuo. The antioxidant and free radical scavenging activities of stem extracts was determined by 2,2-Diphenyl-1-picrylhydrazyl (DPPH) scavenging assay, reducing power assay, deoxyribose degradation assay and $Fe^{2+}$ chelating assay. Total phenolic contents (TPC) were evaluated using Folin-Ciocalteu reagent. The results confirmed that the plant is a rich source of polyphenolic compounds which are invariably higher compared to other herbs. All extracts showed TPC in the range of 146.6 - 168.6 mg/g gallic acid equivalents at $300{\mu}g/ml$ of extract. Among the three extracts ME showed highest scavenging activity as evidenced by maximum scavenging of DPPH (83.2%), $OH{\bullet}$ radicals (94.81%), $Fe^{2+}$ chelating activity (88.59%) and a high reducing power 0.623 at $300{\mu}g/ml$. Our results demonstrate that Euphorbia trigona, an unexplored xerophytic plant could be potential source of natural antioxidants and phytotherapeutic agents. The plant possess invariably high amount of polyphenolic compounds with a broad spectrum of antioxidant properties and could be further used for food, feed and pharmaceutical applications.

Natural Anthraquinone Derivatives from a Marine Mangrove Plant-Derived Endophytic Fungus Eurotium rubrum: Structural Elucidation and DPPH Radical Scavenging Activity

  • There is considerable interest in the isolation of potent radical scavenging compounds from natural resources to treat diseases involving oxidative stress. In this report, four new fungal metabolites including one new bisdihydroanthracenone derivative (1, eurorubrin), two new seco-anthraquinone derivatives [3, 2-O-methyl-9-dehydroxyeurotinone and 4, 2-O-methyl-4-O-(${\alpha}$-D-ribofuranosyl)-9-dehydroxyeurotinone], and one new anthraquinone glycoside [6,3-O-(${\alpha}$-D-ribofuranosyl)-questin], were isolated and identified from Eurotium rubrum, an endophytic fungal strain that was isolated from the inner tissue of the stem of the marine mangrove plant Hibiscus tiliaceus. In addition, three known compounds including asperflavin (2), 2-0-methyleurotinone (5), and questin (7) were also isolated and identified. Their structures were elucidated on the basis of spectroscopic analysis. All of the isolated compounds were evaluated for 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging activity.

Antioxidant Effects of Tocotrienol in Rice Bran (미강 함유 Tocotrienol의 항산화 효과)

  • The pharmaceutical function of tocotrienol in rice bran was evaluated. Distinctive antioxidative effects by 1,1-diphenyl-2-picrylhydrazyl(DPPH) could be observed. Also, Superoxide Dismutase(SOD) and Glutathione Peroxidase(GPX) activities of the cultured cells such as human firbroblast and hepatocyte, were increased up to 2 fold by the treatment of tocotrienol. The effects on GPX activity were more evident than SOD activity, and the stimulation was up to 2 fold. The changes of gene expression patterns were examined by applying the cell extracts of fibroblast treated with the increasing concentrations of tocotrienol on two-dimensional gel electrophoresis(2-D gel electrophoresis). As the concentrations increasing, many proteins began to appear with the increasing amounts, while several proteins diminished or disappeared. From these results, tocotrienol was clearly shown to have abilities on protecting any oxidizing damages and stimulating anti-oxidizing activities of the organisms.

  • Objective: The objectives of this study were to investigate the direct antioxidative effect of 90 Kda heat shock protein (Hsp90) obtained from duck muscle. Methods: The interaction of Hsp90 with phospholipids and oxidized phospholipids was studied with surface plasmon resonance (SPR), and their further oxidation in the presence of Hsp90 was evaluated with thiobarbituric acid reactive substances (TBARS) assay. The scavenging effect on the 1,1-diphenyl-2-picrylhydrazyl (DPPH) and 2,2'-azinobis (3-ethylbenzthiazoline-6-sulfonic acid (ABTS) was measured, and the electron paramagnetic resonance (EPR) spectroscopy in combination with 5-tert-Butoxycarbonyl-5-methyl-1-pyrroline-N-oxide and 2-phenyl-4,4,5,5,-tetramethylimidazoline-1-oxyl-3-oxide (PTIO) was utilized to determine the abilities of Hsp90 in scavenging hydroxyl and PTIO radicals. Results: SPR showed Hsp90 could bind with both phospholipids and oxidized phospholipids, and prevent their further oxidation by the TBARS assay. The DPPH and ABTS scavenging activity increased with Hsp90 concentration, and could reach 27% and 20% respectively at the protein concentration of 50 μM. The EPR spectra demonstrated Hsp90 could directly scavenge ·OH and PTIO· radicals. Conclusion: This suggests that Hsp90, a natural antioxidant in meat, may play an important role in cellular defense against oxidative stress, and may have potential use in meat products.

Isolation and Characterization of Antioxidative Compounds from the Aerial Parts of Angelica keiskei

  • Ethyl acetate-soluble neutral fraction of hot water extracts from the aerial parts of Angelica keiskei showed a 1, 1-diphenyl-2-picrylhydrazyl (DPPH) radical-scavenging activity. Six antioxidative compounds were purified and isolated by various chromatographic procedures. Based on the analyses of FAB-MS and NMR, the isolated compounds were structurally elucidated as luteolin 7-O-${\beta}$-D-glucopyranoside (1), quercetin 3-O-${\beta}$-D-galactopyranoside (2), quercetin 3-O-${\beta}$-D-glucopyranoside (3), quercetin 3-O-${\alpha}$-D-arabinopyranoside (4), kaempferol 3-O-${\alpha}$-D-arabinopyranoside (5), and luteolin 7-O-rutinoside (6). The glycosides of flavonols and luteolin showed DPPH radical-scavenging activity. One molecule of 2, 3, 4, 6, 1, and 5 scavenged 4.2, 4.2, 4.1, 2.5, 2.2, and 1.4 molecules of DPPH radical, respectively.

H2O2 Pretreatment Modulates Growth and the Antioxidant Defense System of Drought-stressed Zoysiagrass and Kentucky Bluegrass

  • This study investigated the effect of exogenous hydrogen peroxide ($H_2O_2$) on the antioxidant responses and growth of warm-season turfgrass (Zoysia japonica Steud.) and cool-season turfgrass (Poa pratensis L.) subjected to drought stress. Compared with control plants that were not pretreated with $H_2O_2$, plants pretreated with $H_2O_2$ had significantly greater fresh and dry weights of shoots and roots, and increased water content. $H_2O_2$ pretreatments before drought stress significantly decreased the concentrations of malondialdehyde and $H_2O_2$. DPPH radical scavenging and glutathione activities were significantly increased. The responsive activities of the antioxidant enzymes superoxide dismutase, ascorbate peroxidase, catalase, and peroxidase were also significantly enhanced. Our results suggest that exogenous $H_2O_2$ could improve the growth of warm-season and cool-season turfgrass under drought stress by increasing the activity of their antioxidant enzymes, while decreasing lipid peroxidation.

Radical-Scavenging Activities of Fermented Cactus Cladodes (Opuntia humifusa Raf.) (천년초 발효물의 라디칼 소거능)

  • The aim of this work was to select suitable fermentation treatments for the efficient bioconversion of cactus (Opuntia humifusa Raf.) bioactive components with an improved radical scavenging activity for use as a nutraceutical. To obtain microorganisms for the microbial conversion of cactus, Leuconostoc mesenteroides ATCC8294, Lactobacillus plantarum KCTC 3099, Lactobacillus plantarum KERI 236 and Monascus pilosus KCCM 60029 (ATCC 22080) were used for fermentation. Fermentation by Lac. plantarum KCTC 3099 was the most effective at scavenging 1,1-diphenyl-2-picrylhydrazyl hydrate (DPPH) and 2,2-azino-bis-(3-ethylbenzothiazoline-6-sulfonic acid) (ABTS) radicals and reducing iron (III). In particular, uronic acid levels showed a remarkable increase in fermentation. The polyphenol and quercetin content of the fermented cactus showed large increases from $108.65{\mu}g/mL$ and $2.71{\mu}g/mL$ to $227.83{\mu}g/mL$ and $9.73{\mu}g/mL$, respectively, showing a maximum level at 36 h of fermentation with Lac. plantarum KCTC 3099. Thus, cactus fermentation with Lac. plantarum is an useful process for the enhancement of antioxidant contents and activity of fresh cactus.
