Optimization for the Production of Mono- and Di-acylglycerols from Corn Oil by Enzymic Glycerolysis Using Response Surface Methodology (반응표면분석에 의한 옥수수유 유래 monoacylglycerol과 diacylglycerol 합성 조건의 최적화)

  • Response surface methodology was used to optimize production conditions of monoacylglycerol (MAG) and diacylglycerols (DAG) from corn oil by enzymic glycerolysis. Contents of $1,3-DAG\;(Y_1),\;1,2-DAG\;(Y_2),\;total\;DAG\;(Y_3),\;MAG\;(Y_4)$, and total $DAG+MAG\;(Y_5)$ were obtained. Conditions were optimized using central composite design with incubation temperature $(35-75^{\circ}C,\;X_1)$, incubation time (1-11 hr, $X_2$), and amount of hexane added (0-2 mL, $X_3$) as three variables. Content of 1,3-DAG was maximized by 20.43 area% at incubation temperature of $44.92^{\circ}C$, incubation time of 10.24 hr, and hexane content of 1.16 mL, whereas that of 1,2-DAG (26.78 area%) was maximized at $56.32^{\circ}C$, 6.95 hr, and 1.04 mL, respectively. Predicted maximum total DAG content was 45.09 area% at $53.82^{\circ}C$, 8.03 hr, and 1.08 mL, while production conditions of MAG (9.57 arae%) were $64.14^{\circ}C$, 7.00 hr, and 0.13 mL. At variables of $54.07^{\circ}C$, 7.98 hr, and 1.02 mL, maximum content of total DAG+MAG predicted by RSM was 53.54 area%.

  • Purpose The glomerular filtration rate (GFR) test is an important indicator of glomerular filtration and has been used to test renal function and the extent of its function. The GFR test is performed by intravenous injection of radioactive medicines made of $^{51}Cr$-EDTA, and blood concentration is measured by taking blood according to the elapsed time. also, PET-CT, bone scan, transfusion and so on will affect the outcome. Therefore, we will improve the quality of the test by providing guidelines for the GFR test for more accurate testing. Materials and Methods 5 mL of physiological saline solution and 2 mL of $^{51}Cr$-EDTA solution are used to make 5 mL of the radiopharmaceutical solution to be injected into the patient. First, the syringe weight is measured before the injection, and then the radioactive medicine is injected into the patient's vein and the syringe weight is measured after the injection. Blood sampling is performed twice in total. In adults, blood is collected 3 hours / 5 hours after injection and in children 2 hours / 5 hours after injection. The blood sample is centrifuged at 3300 rpm for 5 minutes. Standard solution is prepared by filling diluent water up to the scale indicated in the 200-mL volumetric flask, discarding $500{\mu}L$, injecting $500{\mu}L$ of GFR reagent and mixing well. $500{\mu}L$ each of the standard solution is dispensed into two test tubes, and $500{\mu}L$ of each of the plasma samples collected in time is dispensed into two test tubes and measured with a Cobra Counter. Results At present, the reference range applied in this study is $119.5{\pm}30.3ml/min/1.73m2$ for males and $125.2{\pm}28.2ml/min/1.73m^2$ for females. Conclusion The GFR test is conducted using radioactive medical products. GFR testing is performed as a scheduled test, but PET-CT, dialysis and transfusion, which may affect GFR testing, may be scheduled during GFR testing. Therefore, we could get accurate GFR test results by notifying the ward and department beforehand when booking.

  • When a commercial prealloyed Fe-powder(Fe-5Cr-lMo-2Cu-0.5P-3C) is liquid phase sintered at 116$0^{\circ}C$, liquid precipitates with various shapes form within solid grains during the initial stage of sintering. The shape of a liquid precipitate changes pith the increment of their size from sphere(with radius<0.3$\mu\textrm{m}$), a transient polyhedron with more than 7 faces(1~2 $\mu\textrm{m}$), cuboid(3~5 $\mu\textrm{m}$), and finally to sphere(>5 $\mu\textrm{m}$). The shapes of liquid precipitates closely resemble the growth shapes predicted on the basis of solid-liquid interfacial energy and the coherency strain energy with anisotropic elastic constants in the diffusion zone around the precipitates.

Proposal for a gingival shade guide based on in vivo spectrophotometric measurements

  • PURPOSE. The purpose of this study was to propose and assess a shade guide for pink gingival aesthetics using a Spanish population sample. MATERIALS AND METHODS. The $L^*$, $C^*$, h, $a^*$ and $b^*$ coordinates of 259 participants were measured using a spectrophotometer in 3 standardized points along the attached gingiva of the maxillary central incisors. A hierarchical clustering analysis was applied to obtain separate solutions regarding the number of shade tabs. For each of the solutions obtained, color differences (${\Delta}E^*$) were calculated using the CIELab and CIEDE2000 formulas, and the proposed shade guide was selected considering (1) the color differences between tabs and (2) the coverage error of each of the solutions. RESULTS. The proposed shade guide consisted of 8 gingival shade tabs and achieved CIELab and CIEDE2000 coverage errors of less than the respective 50:50% acceptability thresholds (${\Delta}E^*=4.6$ units and ${\Delta}E_{00}=4.1$). The coordinates for the various gingival shade tabs were as follows: Tab 1: $L^*43.3$, $a^*21.9$, $b^*12.3$ (1.6); Tab 2: $L^*42.9$, $a^*34.1$, $b^*19.1$; Tab 3: $L^*46.5$, $a^*25.8$, $b^*10.9$; Tab 4: $L^*46.5$, $a^*27.3$, $b^*15.1$; Tab 5: $L^*49.6$, $a^*23.5$, $b^*16.8$; Tab 6: $L^*51.5$, $a^*19.7$, $b^*13.6$; Tab 7: $L^*55.9$, $a^*22.0$, $b^*15.0$; and Tab 8: $L^*56.0$, $a^*19.9$, $b^*18.8$. CONCLUSION. The CIELab and CIEDE2000 coverage errors for the 8 shade tabs of the proposed gingival shade guide were significantly lower than those of other guides. Therefore, despite the limitations of this study, the proposed guide is more appropriate for matching gingival shade in the Spanish general population.

Diagnostic Value of ADA Multiplied by Lymphocyte to Neutrophil Ratio in Tuberculous Pleurisy (결핵성 흉막염에서 ADA 활성도와 림프구/중성구 비의 곱의 진단적 유용성)

  • Background: Many diagnostic approaches for defining the definitive cause of pleurisy should be included due to the large variety of diseases resulting in pleural effusion. Although ADA is a useful diagnostic tool for making a differential diagnosis of pleural effusion, particularly for tuberculous pleural effusion, a definitive diagnostic cut-off value remains problematic in Korea. It was hypothesized that ADA multiplied by the Lymphocyte/Neutrophil ratio(L/N ratio) might be more powerful for making a differential diagnosis of pleural effusion. Methods: One hundred and ninety patients, who underwent thoracentesis and treatment in Chung-Ang University Hospital from January, 2005 through to February 2006, were evaluated. The clinical characteristics, radiologic data and the examination of the pleural effusion were analyzed retrospectively. Results: 1. Among the 190 patients, 59 patients (31.1%) were diagnosed with tuberculous pleurisy, 45 patients(23.7%) with parapneumonic effusion, 42 patients(22.1%) with malignant effusions, 36 patients(18.9%) with transudate, and 8 patients(4.2%) with empyema. One hundred and twenty one patients were found to have an ADA activity of 1 to 39 IU/L(63.7%). Twenty-nine were found to have an ADA activity of 40 to 75 IU/L(15.3%) and 40 were found to have an ADA activity of 75 IU/L or greater(21.0%). 2. Among the patients with tuberculous pleurisy, 5(8%), 18(30%) and 36 patients(60%) had an ADA activity ranging from 1 to 39 IU/L, 40 to 75 IU/L, and 75 IU/L or greater, respectively. In those with an ADA activitiy 40 to 75 IU/L, 18 patients(62%) had tuberculous pleurisy, 9(31%) had parapneumonic effusion and empyema, and 1(3.4%) had a malignant effusion. 3. In those with an ADA activity of 40 to 75 IU/L, there was no significant difference between tuberculous pleurisy and non-tuberculous pleural effusion(tuberculous pleurisy : 61.3 ${\pm}$ 9.2 IU/L, non-tuberculous pleural effusion : 53.3${\pm}$10.5 IU/L). 4. The mean L/N ratio of those with tuberculous pleurisy was 39.1 ${\pm}$ 44.6, which was significantly higher than nontuberculous pleural effusion patients (p<0.05). The mean ADA x L/N ratio of the tuberculous pleurisy patients was 2,445.7 ${\pm}$ 2,818.5, which was significantly higher than the non-tuberculous pleural effusion patients (level p<0.05). 5. ROC analysis showed that the ADA x L/N ratio had a higher diagnostic value than the ADA alone in the group with an ADA between 40-75 IU/L. Conclusion: The ADA multiplied by the lymphocyte-to-neutrophil ratio might provide a more definitive diagnosis of tuberculous pleurisy.

Pilot-scale Optimization of Parameters Related to Dissolved Oxygen for Mass Production of Pullulan by Aureobasidium pullulans HP-2001 (Aureobasidium pullulans HP-2001 균주를 사용한 풀루란의 대량 생산을 위한 파이롯트 규모에서 용존산소와 관련된 조건의 최적화)

  • Parameters related to dissolved oxygen for the production of pullulan by Aureobasidium pullulans HP-2001 were optimized in 7 l and 100 l bioreactors. The optimal concentrations of glucose and yeast extract for the production of pullulan were 50.0 and 2.5 g/l, respectively, and its conversion rate from glucose was 37% at a flask scale. The optimal initial pH of the medium and temperature for cell growth were 7.5 and $30^{\circ}C$, whereas those for the production of pullulan were 6.0 and $25^{\circ}C$. The optimal agitation speed and aeration rate for cell growth were 600 rpm and 2.0 vvm in a 7 l bioreactor, whereas those for the production of pullulan were 500 rpm and 1.0 vvm. The production of pullulan with an optimized agitation speed of 500 rpm and aeration rate of 1.0 vvm was 18.13 g/l in a 7 l bioreactor. Maximal cell growth occurred without inner pressure, whereas the optimal inner pressure for the production of pullulan was 0.4 kgf/$cm^2$ in a 100 l bioreactor. The production of pullulan under optimized conditions in this study was 22.89 g/l in a 100 l bioreactor, which was 1.38 times higher than that without inner pressure.

Effect of MS Medium Strength, Sucrose Concentration, and Light Condition on Bulblet Formation and Growth of Muscari armenicum In Vitro (MS 배지내 무기물 농도, 당 농도 및 광 조건이 무스카리의 기내 자구 형성과 비대에 미치는 영향)

  • The influences of MS medium strength, sucrose concentration, and light condition on bulblet formation and growth were studied in leaf tissue culture of Muscari armenicum 'Early Giant'. Bulblet formation from leaf segments were the most effective on MS medium supplemented with $0.01mg{\cdot}L^{-1}$ NAA, $0.2m{\cdot}L^{-1}$ kinetin, $30g{\cdot}L^{-1}$ sucrose, and $8g{\cdot}L^{-1}$ gelrite under darkness for 2 weeks followed by 16 hr photoperiod with a photosynthetic photon flux density of $50mol{\cdot}m^{-2}{\cdot}s^{-1}$. However the compactness of bulblets formed in vitro was promoted in the MS medium with $60gL^{-1}$ sucrose. Acclimatized plants flowered during the second year of the growing period without any phenotypic variations and formed average 1.5 bulblets per mother bulb.

Physicochemical Properties and Antioxidant Activities of Green Tea with Reference to Extraction Conditions (추출조건에 따른 녹차음료의 이화학적 특성 및 항산화활성)

  • We investigated the physicochemical properties and antioxidant activities of green tea with respect to extraction conditions. The values of pH, and the L, a, and b Hunter parameters of green tea beverage 1 (GTB 1), green tea beverage 2 (GTB 2), and commercial green tea beverage (CGTB) were 6.22, 96.91, -1.06, and 7.77 5.40, 96.39, -1.73, and 13.68 and 6.20, 95.40, -4.75, and 25.51, respectively. The total free amino acid content of GTB 1 and 2, and CGTB, were 253.21, 262.65, and 58.36 mg/100 mL, and the major free amino acids were aminoadipic acid (102.56, 136.29, and 27.02 mg/100 mL), arginine (23.32, 30.75, and 7.31 mg/100 mL), and serine (18.22, 17.96, and 5.94 mg/100 mL). The levels of total phenolics and caffeine were higher in GTB 2 (852.58 and $225.51\;{\mu}g/mL$) than in GTB 1 (500.65 and $317.34\;{\mu}g/mL$) or CGTB (387.14 and $164.53\;{\mu}g/mL$). The catechin content of GTBs 1 and 2, and CGTB, were 294.8, 415.7, and $130.99\;{\mu}g/mL$, respectively. The major catechins of GTB 1 and 2, and CGTB were epigallocatechin, catechin, and epigallocatechin gallate, in that order, and the epigallocatechin contents were 186.50 in GTB 1, 268.10 in GTB 2, and $82.26\;{\mu}g/mL$ in CGTB. GTB 1 and 2 and CGTB showed substantial dose-dependent antioxidative activities. The DPPH radical-scavenging activities of GTB 1 and 2, and CGTB, were 85.48, 87.09, and 87.03%, respectively at a concentration of $125\;{\mu}g/mL$. The ferric reducing/antioxidant activities (FRAPs) of GTB 1 and 2 and CGTB were 2.66, 2.70 and 2.67 absorbance at a concentration of $1,000\;{\mu}g/mL$. Sensory evaluation tests revealed no significant differences among the three green tea beverages.

Changes in Agricultural Irrigation Water Quality in Nam River (남강 농업용수의 시기별 수질변화)

  • To investigate chemical changes of agricultural water in Nam river used for the basic information. Samples were collected from seven sites along the Nam river and were analyzed for inorganic content from April to September in $1994{\sim}1995$. Average value of analyzed inorganic concentrations at seven sampling sites were pH 7.9, COD 7.3mg/l, $NO_3-N$ 1.2mg/l, $Na^+$ 6.2mg/l, $Cl^-$ 14.8mg/l, EC 0.13dS/m, $PO_4\;^{3-}$ 0.21mg/l, $K^+$ 2.6mg/l, $Ca^{2+}$ 10.8mg/l,$Mg^{2+}$ 2.9mg/l, $SO_4\;^{2-}$ 10.5mg/l, $Fe^{3+}$ and $Zn^{2+}$ 0.02mg/l. The monthly average value of COD, $NO_3-N$, $Na^+$ and $Cl^-$ showed highest peak in July $8.4{\sim}11.6$, $1.1{\sim}1.7$, $5.4{\sim}13.1$ $18.9{\sim}27.9mg/l$. The highest region of average COD, $NO_3-N$, $Na^+$ and $Cl^-$ were Weola pumping station, $8.8{\sim}11.3$, $1.6{\sim}2.4$, $9.0{\sim}10.2$ and $21.7{\sim}23.0mg/l.$ The ionic $copmposition({\Sigma}A/{\Sigma}C)$ : ratio between total equivalant of anions and canon) of Nam river was higher at Weola pumping station than other topography. The EC was positively correlated with $K^+$, $Ca^{2+}$, $Mg^{2+}$, $Na^+$, $Cl^-$ and $SO_4\;^{2-}$.

Synthesis, Molecular and Microstructural Study of Poly-N-Vinylpyrrolidone Oximo-L-Valyl-Siliconate with IR, 1H-NMR and SEM

  • By reducing PVP with $H_2NOH$.HCl and NaOH 2:2:1 mass ratios in aqueous ethanol, poly-N-vinyl pyrrolidone oxime [PVPO] was prepared with 92% yield. Applying the sol-gel concept, orthosilicic acid [OSA] was made by hydrolyzing TEOS with ethanol in 1:0.5 molar ratios using 1 N KOH aqueous solution as a catalyst. The OSA + PVPO + $_L$-Valine ($\alpha$-amino acid) were mixed with pure ethanolic medium in 1:2:2 mass ratios and refluxed at $78^{\circ}C$ and 6 pH for 6.5 h. A white residue of poly-N-vinyl pyrrolidone oximo-L-valyl-siliconate [POVS] appeared after 5 h. The heating of reaction mixture was stopped and the contents were brought to NTP. The residue formation of POVS was intensified with lowering a temperature and completely solidified within 5 h, was filtered using a vacuum pump with Whatmann filter paper no. 42. The residue of POVS was washed several times with 20% aqueous cold ethanolic solution and dried in vacuum chamber at $25^{\circ}C$ for 24 h. The MP was noted above $350^{\circ}C$. Structural and internal morphology were analyzed with IR and $^1H$-NMR, and SEM respectively. A drug loading and transporting ability of the POVS in water and at pH = 5 and 8 was determined chromatographically.
