T cells play a key role in viral infection. However, in patients with chronic hepatitis C virus (HCV) infection, HCV-specific T cells are dysfunctional and impaired in the liver, which is the primary site for HCV replication. There are multiple potential mechanisms for HCV-specific T cell dysfunction including induction of immune inhibitory pathways (program death-1; PD-1, cytotoxic t lymphocyte associated antigen-4; CTLA-4) and immune tolerance induced specific for the liver. However, the interaction between hepatocytes and HCV-specific CD8 T cells has not clearly established. In this study, we confirmed huh (human hepatoma) 7.5 cells expressing HLA (human leukocyte antigen) A2 presented antigen to activate HCV-specific CD8 T cells in HLA A2-restricted manner and expression of PD-L (program death ligand) 1 on huh7.5 cells reduced HCV-specific CD8 T cell activation, suggesting an immune modulatory activity. Loss of HCV-specific tetramer responses following antigenic stimulation correlated with increased caspase-3 activity. In addition, PD-L1 on huh7.5 cells rescued HCV-specific CD8 T cells from apoptosis. Our results suggest that the interaction between PD-L1 and PD-1 can recover the function of HCV-specific CD8 T cells in the liver, which could be applied in therapy of HCV chronic infection.
Hydrogen producing bacterium, strain MeL 6-2 was isolated from the sludge of the factory areas in Anyang through the acclimation in basal salt medium (BSM) supplemented with 10 g/L of sucrose. Isolated strain MeL 6-2 was a facultative anaerobe which could grow in both aerobic and anaerobic environments. An aerobically grown pure culture isolated from enriched culture was analyzed by 16S rDNA sequencing and identified as Rhodopseudomonas sp. MeL 6-2. Effects of the concentrations of glucose and sucrose on the hydrogen production rate and the hydrogen production yield were investigated. When glucose in the range of 1~12 g/L was supplemented to the BSM, strain MeL 6-2 could grow without lag phase. An increased glucose concentration increased the specific hydrogen production rate linearly to $4.2\;mmol-H_2{\cdot}L^{-1}{\cdot}h^{-1}$ at 10 g/L, and $60\;mmol-H_2{\cdot}mg-DCW^{-1}{\cdot}h^{-1}$, but decreased slightly as the concentration increased to 12 g/L. The hydrogen production yield was maintained over a range from 2.6 to $3.1\;mol-H_2{\cdot}mol-glucose^{-1}$. When sucrose in the range of 1~12 g/L was supplemented to the BSM, strain MeL 6-2 could grow after ten hours. An increased sucrose concentration increased the specific hydrogen production rate and the hydrogen production yield to $163\;mmol-H_2{\cdot}mg-DCW^{-1}{\cdot}h^{-1}$ and to $4.5\;mol-H_2{\cdot}mol-sucrose^{-1}$, respectively.
Stability-enhanced mutants, H44, 11-94, 5A2-84, and F8, of L-threonine aldolase(L-TA) from Streptomyces coelicolor A3(2)(SCO1085) were isolated by an error-prone PCR followed by a high-throughput screening. Each of these mutant, had a single amino acid substitution: H177Y in the H44 mutant, A169T in the 11-94 mutant, D104N in the 5A2-84 mutant and F18I in the F8 mutant. The residual L-TA activity of the wild-type L-TA after a heat treatment for 20 min at $60^{\circ}C$ was only 10.6%. However, those in the stability-enhanced mutants were 85.7% for the H44 mutant, 58.6% for the F8 mutant, 62.1% for the 5A2-84 mutant, and 67.6% for the 11-94 mutant. Although the half-life of the wild-type L-TA at $63^{\circ}C$ was 1.3 min, those of the mutant L-TAs were longer: 14.6 min for the H44 mutant, 3.7 min for the 11-94 mutant, 5.8 min for the 5A2-84 mutant, and 5.0 min for the F8 mutant. The specific activity did not change in most of the mutants, but it was decreased by 45% in the case of mutant F8. When the aldol condensation of glycine and 3,4-dihydroxybenzaldehyde was studied by using whole cells of Escherichia coli containing the wild-type L-TA gene, L-threo-3,4-dihydroxyphenylserine(L-threo-DOPS) was successfully synthesized with a yield of 2.0 mg/ml after 20 repeated batch reactions for 100 h. However, the L-threo-DOPS synthesizing activity of the enzyme decreased with increased cycles of the batch reactions. Compared with the wild-type L-TA, H44 L-TA kept its L-threo-DOPS synthesizing activity almost constant during the 20 repeated batch reactions for 100 h, yielding 4.0 mg/ml of L-threo-DOPS. This result showed that H44 L-TA is more effective than the wild-type L-TA for the mass production of L-threo-DOPS.
The effects of media, growth regulators, low-temperature treatments, culture temperature and light were investigated to improve the callus induction and growth in the anther culture of Comus officinalis Sieb. et Zucco. The frequency of callus induction was more effective on WPM medium than MS medium, and it was highest as 54% in WPM medium supplemented with Img/L NAA. Callus growth was stimulated on MS medium supplemented with 2mg/L NAA. Effect of temperature and light on the callus induction and growth was highest as 62% in the treatment for 16/8 hrs. (light/dark) at $25^{\circ}C$ Effect of low - temperature treatment on callus induction was highest as 19. 5% in the treatment for 36 hrs. at $4^{\circ}C$. For organization, green cells and rootings were promoted on MS medium supplemented with O. 5mg/L 2,4-D and 1 mg/L kinetin. The prevention of callus browning was effective on the medium containing $3{\sim}5mg/L$ ABA or 5mg/L $AgNO_3$. The supplement of ABA or $AgNO_3$,were maintained callus activity for 4-5 weeks and they were promoted the development of green cells.
Various structures of navigation messages in Global Navigation Satellite System (GNSS) signals can be categorized into fixed, packetized, and mixed types, each with different broadcast intervals and sequences. In the fixed format, the broadcast intervals and sequences of navigation messages of Global Positioning System (GPS) L1 C/A and BDS B1I are strictly defined in the Interface Control Document (ICD). Navigation messages of GPS L2C, L5, Quasi-Zenith Satellite System (QZSS) L2C, L5, and BeiDou Navigation Satellite System (BDS) B1C are in a packetized format, and their broadcast sequence is not explicitly specified in the ICD; only the maximum broadcast intervals are described. In the mixed format, signals such as GPS L1C, QZSS L1 C/A, L1C, and BDS B2a exhibit both fixed and packetized patterns. Sequences of certain parts of the message are not defined. Therefore, in this study, experimental-based investigations were additionally conducted alongside an ICD investigation to analyze the current broadcast intervals and sequences of GNSS navigation messages.
A method for induction of multiple shoots using cotyledonary nodes and shoot tips of Macrotyloma uniflorum (Lam.) Verdc. was described. The experiment was conducted in which shoot induction was noticed on Murashige and Skoog (MS) medium supplemented with different concentrations and combinations of four cytokinins (KIN, 2iP, Ads, BAP). These multiple shoots were later developed into normal shoots. The highest rate of shoot proliferation came from MS medium added with BAP 1.5 mg/L. The multiple shoot buds were subcultured into MS medium with BAP (0.5-1.5 mg/L) along with Ads (1.0 mg/L) and GA$_3$ (0.5 mg/L), which gave rise to the highest frequency of shoot proliferation and elongation. The shoots were rooted on MS medium supplemented with 1.75 mg/L IBA.
Recently, GPGPU has been widely used for general-purpose processing as well as graphics processing by providing optimized hardware for parallel processing. Memory system has big effects on the performance of parallel processing units such as GPU. In the GPU, hierarchical memory architecture is implemented for high memory bandwidth. Moreover, both memory address coalescing and memory request merging techniques are widely used. This paper analyzes the GPU performance according to various memory organizations. According to our simulation results, GPU performance improves by 15.5%, 21.5%, 25.5%, 30.9% as adding 8KB L1, 16KB L1, 32KB L1, 64KB L1 cache, respectively, compared to case without L1 cache. However, experimental results show that some benchmarks decrease performance since memory transaction increases due to data dependency. Moreover, average memory access latency is increased as the depth of hierarchical cache level increases when cache miss occurs significantly.

Burkholderia sp. D5, a polyaromatic hydrocarbons(PAHs)-degrading bacterium, was isolated from oil-contaminated soil. The bacterium could utilize phenanthrene (Phe) as a sole carbon source but could not use pyrene (Pyr). However, the strain could degrade Pyr when a cosubstrate such as yeast extract (YE) was supplemented. The PAH degradation rate of the bacterium was enhanced by the addition of other organic materials such as YE, peptone and glucose. YE was a particularly effective additive in stimulating cell growth as well as PAH degradation. When 1 g-YE/L was supplemented into the basal salt medium (BSM) with 215 mg-Phe/L, the specific growth rate (0.28 h-1) and Phe-degrading rate (29.30 μmol/L/h) were enhanced approximately ten and two times more than those obtained in the BSM with 215 mg-Phe/L, respectively. Through kinetic analysis, the maximum specific growth rate (μmax) and PAH degrading rate (Vmax) for Phe were obtained as 0.34/h and 289 ${\mu}mol$/L/h, respectively. Also, μmax and Vmax for Pyr were 0.27 h-1 and 50 ${\mu}mol$/L/h, respectively. The degradation rates for each Phe (2.20 μmol/L/h) and Pyr (2.18 μmol/L/h) were lower in mixture substrates than in a single substrate (29.30 ${\mu}mol$/L/h and 9.58 ${\mu}mol$/L/h, respectively).
