Clinical Analysis of Postoperative Acute Renal Failure in the Patients undergoing Cardiovascular Operation with CPB (개심술 후 발생한 급성 신부전의 임상적 고찰)

  • From May 1, 1993 to May 31 1995, the authers studied retrospectively 211 patients who underwent cardiovascular operation with cardiopulmonary bypass(CPB). Because we were interested in new development of ARF(prevalence, mortality rate, and main risk factors), we performed a multivariate statistical analysis about data of patients with preoperative serum creatinine values of less than 1.5 mg/dL. Normal renal function before operation(serum creatinine level less than 1.5 mg/dL) was registered in 198(74%) patients. Of these, 27(14%) patients showed postoperative renal complication, including 20(10%) patients classified as renal dysfunction(serum creatinine level between 1.5 and 2.5 mg/dL) and 7(4%) patients as acute renal failure(serum creatinine level higher than 2.5 mg/dL). The mortality rate was 5.8% in normal patients, 5% in patients with renal dysfunction, and 43% when acute renal failure developed(p=0.036). Indeed, the renal impairment proved to be an independent predictor of mortality(odd ratio 2.52∼11.25), along with cardiovascular(odd ratio 4.20) and respiratory(odd ratio 2.18) complications. Multivariate analysis identified the following variables as independent risk factors for postoperative renal impairment : advanced age(odd ratio 1), need for emergency operation(odd ratio 3.78), low-output syndrome(odd ratio 3.66), respiratory complication(odd ratio 1.30), need for deep hypothermic circulatory arrest(odd ratio 1.4). The 13 patients(7%) with preoperative renal failure showed a significantly higher morbidity and mortality rate than those without renal complications before operation. We concluded that the likelihood of severe renal complications is resonably low in the patients undergoing cardiac operation without preexisting renal dysfunction, but associated mortality remains high. A prominant role of hemodynamic factor in the development of postoperative acute renal failure must be recognized during preoperative, intraoperative, and postoperative periods.

  • New seventeen imazapyr derivatives, 2-(4-isopropyl-4-methyl-5-oxo-2-imidazolin-2-yl)-3-(N-methyl-N-X(sub.)-phenylaminooxoacetyl)pyridine, 6 were synthesized and their pre-emergence herbicidal activity$(pI_{50})$ in vivo against Corn (Zea mays L.) and Pigweed (Amaranthus viridis L.) were studied by the pot test under paddly conditions. Quantitative structure activity relationships (QSARs) were analyzed using the physicochemical parameters of substituent(X) on the phenyl ring of 3-(N-methyl-N-X(sub.)-phenylaminooxoacetyl) group and regression analysis. The herbicidal activities were related to the steric effect of X-substituent. The effect was rationalized by paraholic function of MR and $L_1$, where the optimal values were MR=5.56 (Zea mays L.) and $L_1=3.34\;{{\AA}}$ (Amaranthus viridis L.). Among them, 2,5-difluoro substituted compound, 6i showed good herbicidal activity against Pigweed with excellent tolerance to Corn.

Biogas Production from Sewage Sludge in 30L Microbial Electrolysis Cell (30L 미생물전기분해전지의 하수슬러지로부터 바이오가스 생산 특성)

  • Operating characteristics of a 30 L microbial electrolysis cell (MEC) for producing biogas from sewage sludge was studied. During the 32-day inoculation period, carbon dioxide concentration decreased and methane concentration increased with operating time, and the overall methane content of biogas was 69.1% with a production rate of 171.6 mL CH4/L·d. In fed-batch experiments for 6 operating cycles, CH4 concentration of 66.5~77.2% was obtained at a production rate of 184.9~372.9 mL CH4/L·d, COD, TS and VS removal efficiency ranged from 28.2 to 42.1%, 20.7 to 37.5% and 18.5 to 36.9%, respectively. The MEC system was observed to be stabilized as operating cycles were repeated after inoculation. In the last operating cycle, 5221 mL/L of methane was produced with CH4 yield of 316.7 L CH4/kg CODrem, and the energy recovery was 73%.

Inflammatory Effect of Rheum undulatum L. (대황추출물의 염증반응 제어효과)

  • Objectives : The present study investigated inflammatory effect of Rheum undulatum L. in lipopolysaccharide-exposed rats and Raw 264.7 cells. Methods : Male rats weighting $185.39{\pm}8.21g$ fed basal diet for 1 week and 32 rats were divided into a control group and 3 experimental groups. We fed a control group of rats a basal diet and administered normal saline(100 mg/kg, 1time/1day) for 6 weeks. And we fed basal diet and administered an extract of Rheum undulatum L.(100 mg/kg, 200 mg/kg, 300 mg/kg, 1time/1day) to each experimental group of rats. We measured the plasma concentration of $IL-1{\beta}$($interleukin-1{\beta}$), IL-6 and $TNF-{\alpha}$(tumor necrosis $factor-{\alpha}$), liver cytokines, Raw 264.7 macrophages cytokines. Results : The plasma concentration of $IL-1{\beta}$, IL-6 and $TNF-{\alpha}$ peaked at 5h(hour) after LPS(lipopolysaccharides) injection, and the values of the Rheum undulatum L. extract groups were lower than those of the control group. In the increment of these cytokines concentration at 2h and 5h after LPS injection, the Rheum undulatum L. groups were lower than that of control group. The plasma concentration of IL-10 peaked at 5h after LPS injection, and the values of the Rheum undulatum L. extract groups were higher than those of the control group. In the increment of this cytokine concentration at 2h and 5h after LPS injection, the Rheum undulatum L. groups were higher than that of control group. Liver cytokines measurement was done at 5h after LPS injection. The concentration of liver $IL-1{\beta}$ and IL-6 in the Rheum undulatum L. groups was lower than that of the control group. The concentrations of liver $TNF-{\alpha}$, and IL-10 showed no significant differences among all the treatment groups. In the studies of lipopolysaccharide-exposed Raw 264.7 cells, the concentration of $IL-1{\beta}$, IL-6 and $TNF-{\alpha}$ in the lipopolysaccharide-exposed cells groups was higher than that of control group(normal group), and in the lipopolysaccharide-exposed cells groups, these values showed a tendency to decrease in the Rheum undulatum L. groups. The concentration of IL-10 in the lipopolysaccharide-exposed cells groups was higher than that of control group(normal group), and in the lipopolysaccharide-exposed cells groups, the values showed a tendency to increase in the Rheum undulatum L. groups. Conclusions : These results indicate that the Rheum undulatum L. extracts have an functional material for inflammatory activities.

Comparison of the Chemical Components of Buckwheat Seed and Sprout (메밀종자와 메밀나물의 화학적 성분비교)

  • The chemical components of buckwheat seed and sprout were compared for predicting the usefulness of buckwheat sprout as food materials. The buckwheat sprout was harvested and lyophilized after germination for 7 days. Crude protein, lipid and ash contents of buckwheat sprout were 20.8, 1.3 and 2.6% in dry basis, respectively. Major amino acids of buckwheat sprout were glutamic acid (2,764 mg/l00 g) and aspartic acid (1,698 mg/l00 g). The contents of tryptophan, alanine, tyrosine and histidine of buckwheat sprout were about 1.7 to 1.9 times higher than those of buckwheat seed. Major fatty acids of buckwheat sprout were linoleic acid (45.9%) and oleic acid (18.4%). The contents of stearic acid (18:0) and oleic acid (18:1) were decreased by about 21% and 50%, whereas those of linoleic acid (18:2) and linolenic acid (18:3) were increased by 1.3 and 5.4 times, respectively after germination for 7 days. The mineral contents of buckwheat sprout were 152.0 mg/l00 g for Ca, 9.9 mg/l00 g for Zn, 485.0 mg/l00 g for Mg and 5.4 mg/l00 g for Fe. Vitamin A, C and E contents of buckwheat sprout were 1,180 IU/100 g, 203 mg/l00 g and 32.1 mg/l00 g in dry basis, respectively. Especially, the content of $\alpha$ -tocopherol was increased by 27.5 times as compared to that of buckwheat seed. The rutin content of buckwheat sprout was 343.67 mg/l00 g, which was about 18 times higher than that of buckwheat seed.

The analysis of pharmaceuticals in drinking water by HPLC/ESI-MS/MS (HPLC/ESI-MS/MS에 의한 먹는물(정수) 중 의약물질의 분석)

  • The analytical method of four pharmaceuticals (virginiamycin, erythromycin, tylosin and cimetidine) in drinking water was developed. Effective simultaneous sample clean-up and extraction by solid-phase extraction (SPE) using HLB cartridge prior to LC/ESI-MS/MS analysis were performed. A linear correlation observed in the calibration curves for drinking water in the range of 0.01~2.0 ng/mL showed above $r^2$=0.995. Absolute recovery was in the range of 64.7~118.1% (except cimetidine (37.7~48.1%)). Limit of detection (LOD) and limit of quantitation (LOQ) in spiked drinking water matrix were in the range of 1.6~74.8 pg/mL and 5.5~249.7 pg/mL, respectively. The established method can be used to determine low pg/mL levels of pharmaceuticals in the drinking water.

Mycelial Growth Enhancement in Liquid Cultivation of Hericium erinaceus

  • Erinacine which is produced from Hericium erinaceus mycelia is known to have a stimulating activity of nerve growth factor(NGF) synthesis. Thus, this work was concentrated on the maximum production of Hericium erinaceus mycelia. In order to maximize cell growth, the cultivation was performed with varying the agitation rate and applying the additional medium. As the results, the mycelium concentration was 24.2g/L.

  • This experiments were conducted to investigate growth regulator concentrations and a period of chilling treatment for increasing the size of bulblets of Narcissus pseudo-narcissus cv King Alfred. Combination of 2.5 mg/L BA and 2.5 mg/L NAA remarkedly increased the diameter and growth of regenerated bulblets. Chilling period favorable for the growth of regenerated bulblets was found to be 8 weeks, and addition of 1.0 mg/L NAA alone to MS medium was the most effective for increasing the size of bulblet for the culture after the chilling treatment.

Studies on the Enzymes Produced by Basidiomycetes -Part II. Properties of Cellulase and Xylanase- (담자균(擔子菌)이 생산(生産)하는 효소(酵素)에 관(關)한 연구(硏究) -제2보(第二報) Cellulase 및 Xylanase의 성질(性質)-)

  • Some properties of cellulase and xylanase produced from Pleurotus ostreatus 301 and Lentinus edodes 3-1 during its growth in rice straw medium were investigated. The cellulase activities of P. ostreatus 301 and L. edodes 3-1 were increased in proportion to substrate concentration within 0.6% and 0.8%, respectively, and xylanase activities of two strains were increased within 1%. The reducing sugar production of cellulase and xylanase in two strains were proportionaly increased until 30 min. and 60 min. respectively. The opium pH for cellulase activities of P. ostreatus 301 and L. edodes 3-1 were pH 4.0 and pH 4.5, respectively, and xylanase activities of two strains were pH 5.0. The stable pH range for cellulase activities of P. ostreatus 301 was within 4.0 to 6.0 and L. edodes 3-1 was within 3.0 to 5.0, Xylanase activities of P. ostreatus 301 was within 4.5 to 6.0 and L. edodes 3-1 was within 3.5 to 6.0. The optium temperature for cellulase activities of P. ostraeatus 301 and L. edodes 3-1 were $40^{\circ}C$ and $50^{\circ}C$, respectively, but xylanase activities of P. ostreatus 301 and L. edodes 3-1 were $50^{\circ}C$ and $45^{\circ}C$, respectively. Thermal stability of enzymes were below of optimum temperature and these were mostly inactivate at $70^{\circ}C$ for 10 min of the metalic ions tested, cellulase activities of L. edodes 3-1 was increased by $Co^{++},\; Mg^{++}$ at the concentration of $10^{-2}M$, but were greatly inhibited by $Hg^{++},\;Cu^{++}$ in two strains. Xylanase activities were increased by $Ca^{++},\;Co^{++},\;Mg^{++}$ and $Cd^{++}$ but was greatly inhibited by $Hg^{++}$.

${\beta}-galactosidase$ Activity of Lactobacillus spp. from Pickles (젓갈에서 분리한 Lactobacillus spp.의 ${\beta}-galactosidase$ 특성)

  • Two strains of Lactobacillus(L.) casfi and one strain of L. Pentosus, which were isolated from pickles, were used to investigate in studing their characteristics of ${\beta}-galactosidase$. The preferable carbon sources and pH of the MRS media for enzyme production from L. casei No.10 was found to be 1.0% lactose and pH 7.5, from L. Pentosus No.63 was 1.0% galactose and pH 7.5, and from L. casei No.36 was 1.0% lactose and pH 6.5, respectively. The maximum enzyme production from each strain was found after 48 hours culture at $30^{\circ}C$ in a medium with preferable carbon source. The optimum reaction temperature with substrate for ${\beta}-galactosidase$ activity was found at $60^{\circ}C$ for all three strains . The stability of enzyme from L. casei No.36 was found to be at $45^{\circ}C$, from L. Pentosus No.63 was found at $55^{\circ}C$. This stability from L. casei No.36 was found at $40^{\circ}C$, but it was reduced to 60% at $55^{\circ}C$. These stabilities of enzymes remained about 90% at $40^{\circ}C$ for all three strains. The optimal pH for enzyme activities was found to be pH 6.5 for all three strains. Enzyme activity remained over 90% for L. casei No.10 at $pH\;5.0{\sim}6.0$, for L. casei No.36 at $pH\;5.0{\sim}8.0$, and for L. pentosus No.63 at $pH\;6.0{\sim}7.0$.
