Biogenic amines are produced primarily by microorganisms found in fermented foods and are often implicated in poisoning incidents in humans. In this study, 620 strains of microorganisms were isolated from traditional Korean fermented food in Sunchang in order to screen for non-biogenicamine-producing microorganisms present in these foods. One strain was identified and named Bacillus subtilis SCJ1, by using 16S rRNA sequencing and biochemical characterization. We investigated the cell growth of this organism in order to understand its potential for industrial application. To this end, we optimized the culture medium constituents by using the response surface methodology. The Plackett-Burman experimental design was used for screening of the medium constituents, such as molasses, yeast extract and peptone, for improving cell growth. In order to determine the optimal concentration of each constituent, we used a central composite design. Consequently, the optimized concentrations of molasses, yeast extract and peptone were predicted to be 27.5 g/l, 7.5 g/l and 17.5 g/l, respectively. By model verification, we confirmed that a 1.49-fold increase in dry cell weight compared to the basal medium-from 1.32 g/l, to 1.9722 g/l-was achieved.
This study was conducted to increase grape quality by treating plant growth regulator (PGR) in 'Campbell Early' grape. Foliar application of gibberellic acid ($GA_3$) at $5mg{\cdot}L^{-1}$ on flower cluster of 'Campbell Early' grape at 3-5 unfolded leaf stage effectively increased columella length, berry weight, soluble solid contents and promoted skin color development. Foliar application of $20mg{\cdot}L^{-1}$ abscisic acid (ABA) mixed with $5mg{\cdot}L^{-1}$ of $GA_3$ on flower cluster of 'Campbell Early' grape at 3-4 unfolded leaf stage effectively increased skin anthocyanin contents without any detrimental effects on berry enlargement and columella growth. Foliar application of $2.5mg{\cdot}L^{-1}$ thidiazuron mixed with $5mg{\cdot}L^{-1}$ of $GA_3$ on flower cluster of 'Campbell Early' grape at 3-4 unfolded leaf stage effectively increased fruit quality indices such as higher soluble solid contents and less titratable acidity.
As one of the fundamental studies for the breeding of marine algae, this paper deals wit_the effects of plant hormone on the growth of microscopic filamentous gametophyte of Undaria pinnatifida. The results obtained are summarized as follows: (1) All zoospores were settled on slide glass, germinated and developed into gametophytes, without the growth of germination tubes when treated with 0.1 mg/l of 2.4-D, or 0.1, 1.0, or 5.0 mg/1 of Keinetin. (2) The best growth effect in total average was observed at 0.1 mg/1 of Keinetin, when the growth-rate was $248.9\%$ in contrast with control, and was followed by 1.0 mg/1 of IAA ($243.3\%$), and 0.05 mg/1 of 2.4-D ($205.6\%$). (3) It was certain that the growth-effect by each plant hormone had some differences between male and female gametophytes. IAA was very effective in the growth of male gameto-phytes but Keinetin in that of females. Especially in females, the efficiency of Keinetin was recorded best at $239.0\%$ at 5.0 mg/l and $222.0\%$ at 0.1 mg/1. On the other hand, it was $195.1\%$ in its best at 0.5 mg/1 of IAA, and $146.6\%$ was recorded best in 2.4-D at 0.05 mg/1.
Age and growth of the yellow goosefish, Lophius litulon, were studied using samples collected from the southwestern waters of Korea. Vertebrae of the fish had relatively clear annuli on their surface. The opaque zone of vertebrae was formed once a year between March and April. The oldest fish observed in this study was 8 years old for females and 5 years old for males. The relationship between the radius (R) of vertebral centrum and total length (L) was as follows: L=12.7+4.8R for females and L=9.8+5.6R for males. The relationship between total length and body weight (W) was as follows : $W=0.0089L^{3.0311}$ for females and $W=0.0329L^{2.7752}$ for males. Growth in length of the fish was expressed by the von Bertalanffy's equation as $L_t=127.60(1-e^{-0.1228(t+0.3851)})$ for females and $L_t=82.23(1-e^{-0.1832(t+0.6431)})$ for males.
Breast milk samples collected from 8 areas of 59 lactating women were analysed by atomic absorption spectrophotometry for copper, zinc and manganese concentrations. The results were as follows; 1. The mean levels of Cu, Zn and Mn in the total samples were $342.6\pm138.9{\mu}g/l$ of Cu, $2.01\pm1.44mg/l$ of Zn and $8.67\pm5.12{\mu}g/l$ of Mn. 2. (p<0.01) 3. The mean Zn levels were $3.50\pm2.10mg/1$ in 1-4 weeks, $2.08\pm0.79mg/1$ in 5-12 weeks, $1.38\pm0.91mg/l$ and showed the decline trend by the lactation periods. (p< 0.01) 4. The mean levels of Mn were $8.964\pm3.313{\mu}g/l$ in 1-4 weeks, $7.971\pm6.066{\mu}g/l$ in 5-12 weeks, $8.357\pm4.966{\mu}g/$ in 13-36 weeks and didn't indicate significant decreasing concentrations by the lactation periods.
Shoot clusters were induced from bulb scales of Lilium longiflorum 'Geogia', and proliferated on medium containing 0.5 mg/L BA and 0.5 mg/L IAA. Thereafter, these shoot clusters were cultured in 5 L air-lift bioreactors to form and grow normal bulblets. Number of bulblets increased on medium with 30 g/L sucrose, but growth of bulblets was effective on medium with 60 g/L sucrose. The number of bulblets from shoot clusters had no differences, though bulblet growth was very effective on medium with between full and double strength of MS salts. The inoculation of 100 g shoot clusters as a cultural material was suitable for formation and growth of bulblets in 5 L bioreactors. Air-lift type was more effective for the formation and growth of bulblets than that in ebb and flood one, and 200∼300 mL$.$min$^{-1}$ injection of air was suitable in growth of bulblets.
Paper electrophoretic separation of octahedrally bonded (Ruc $l_{6}$ )$^{3-}$ has been carried out by using the specially designed migration apparatus. The supporting electrolyte solutions are as follows: 0.1M-HCl $O_4$, 0.05 M-HCl+0.09M-KCl, 0.1M-HCl, 5$\times$10$^{-3}$ M-NTA, 0.01M-HCl, 0.01M-HCl $O_4$, 0.01M-citric acid, 0.01M-K $H_2$P $O_4$+0.01M-N $a_2$HP $O_4$, 0.05M-borax, 0.025M-N $a_2$C $O_3$+0.025M-NaHC $O_3$, 0.01M-N $a_3$P $O_4$, 0.01M-NaOH and 0.1 M-NaOH. This study was conducted to establish mass propagation system from the tissue culture using immature embryos in Eleutherococcus senticosus. Immature embryos from seeds were removed under the microscope and placed on MS, $MSB_5\;and\;B_5$ medium containing several plant growth regulators. While the calli were well formed on media containing 2 mg/l of 2, 4-D, 2 mg/l of 2, 4-D and 0.7 mg/l of TDZ, shoot regeneration was better on MS medium with combinations of high concentrations of TDZ and low concentrations of 2, 4-D. Treatment of 2, 4-D alone was better than treatment of TDZ alone in callus induction, but plant regeneration was reversed. The results of callus formation and shoot regeneration on $MSB_5\;and\;B_5$ media were similar to those of MS media. The rate of callus formation was nearly 100% when 2, 4-D was added to $B_5$, medium on concentration of 2 mg/l or 0.7 mg/l. TDZ showed very significant effect on the formation of multiple shoots.
Callus was induced from the petioles of scented-geranium (Pelargonium graveolense) in MS medium containing various concentrations of plant growth regulators. The highest frequency of more than 70% of callus was induced in 2 mg/l NAA and 0.5 mg/l BAP or 2 mg/l 2,4-D and 0.5 mg/l BAP combined treatment, while not in 2,4-D, NAA or BAP alone. When the callus was transferred to the MS medium containing 0.05 mg/l NAA and 0.5 mg/l BAP, were highest intensity of shoot formation, 14 shoots/callus, was induced after 5 weeks. The highest rooting was observed on hormone-free rooting media from the regenerated shoots after 3 weeks, indicating that the regeneration from geranium callus might be possible by changing the hormone ratios. Peroxidase (POD) specific activities in callus induced from 2 mg/l NAA and 0.5 mg/l BAP were higher than those of 2 mg/l 2,4-D and 0.5 mg/l BAP callus during the entire culture periods. POD isozyme C3 was the main cathodic POD isozyme expressed in NAA and BAP callus, while C1 was the main in 2,4-D and BAP callus. However, anodic POD isozymes, A1, A2 and A3 were expressed with similar activities in both hormone combinations.
An adenine-guanine doubless and $\beta$-alanine requiring mutant, D-1550-40, which had been derived from Brevibacterium ammoniagenes ATCC 6872, produced a copious amount of xanthosine-5'-monophosphate (XMP). The optimum concentration of adenine and guanine for maximal accumulation of XMP was about 75 ml/l and 100 ml/l for growth. Concentrations higher than 100 mg/l of adenine and guanine inhibited cell growth and XMP accumulation strongly. The inhibition, however, could be recovered by adding $100{\mu}g$ of biotin per liter or 0.3% of casamino acids to the culture solution. High concentrations of phosphate and magnesium salts (1.0 to 1.5%(w/v) in media) were found to be indispensable for XMP accumulation, and the presence of manganese in the culture medium stimulated both growth of cells and accumulation of XMP leaving 5'-inosinic acid unaffected. The maximal accumulation of XMP reached to 60.5 mg/l after 4 days of fermentation which had been started with a medium containing 100 mg of adenine-guanine, 5 mg of $MnSO_4{\cdot}H_2O$ and $100{\mu}g$ of biotin per liter. The specific XMP synthesis(mg of XMP/mg of cells) was increased with the increase of the cell growth rate.
