Effects of Tributyltin Chloride on L-DOPA-Induced Cytotoxicity in PC12 Cells

  • Tributyltin chloride (TBTC) at concentrations of $0.5-1.0\;{\mu}M$ inhibits dopamine biosynthesis in PC12 cells. In this study, the effects of TBTC on L-3,4-dihydroxyphenylalanine (L-DOPA)-induced cytotoxicity in PC12 cells were investigated. TBTC at concentrations up to $1.0\;{\mu}M$ neither affected cell viability, nor induced apoptosis after 24 or 48 h in PC12 cells. However, TBTC at concentrations higher than $2.0\;{\mu}M$ caused cytotoxicity through an apoptotic process. In addition, exposure of PC12 cells to non-cytotoxic (0.5 and $1.0\;{\mu}M$) or cytotoxic $(2.0\;{\mu}M)$ concentrations of TBTC in combination with L-DOPA (20, 50 and $100\;{\mu}M$) resulted in a significant increase in cell loss and the percentage of apoptotic cells after 24 or 48 h compared with TBTC or L-DOPA alone. The enhancing effects of TBTC on L-DOPA-induced cytotoxicity were concentration- and treatment time-dependent. These data demonstrate that TBTC enhances L-DOPA-induced cytotoxicity in PC 12 cells.

Monitoring of Perfluorinated Compounds (PFCs) in the Seawater of the East and West Coast in Korea (동·서해안지역 해수에서 과불화 화합물의 모니터링)

  • Perfluorinated compounds (PFCs) as environmental pollutants are an important environmental issue. However, little is known on the PFCs monitoring of sea waters around estuaries on the East and West Coasts of Korea. This study shows the monitoring results of PFCs in sea waters in these Coasts. Among 10 PFCs selected in this study, concentrations of perfluorooctanesulfonate (PFOS), perfluorooctanoate (PFOA), perfluorobutane sulfonate (PFBS), and perfluorohexane sulfonate (PFHxS) in the East Coast were 9.6-50.7 ng/L (total mean${\pm}$standard deviation: $26.14{\pm}12.66ng/L$), 13.79-44.58 ng/L ($27.95{\pm}11.41ng/L$), limit of quantification (LOQ)-2.6 ng/L ($0.96{\pm}1.15ng/L$), and 2.95-11.05 ng/L ($4.25{\pm}2.57ng/L$), respectively. In the West Coast, concentrations of PFOS, PFOA, PFBS, and PFHxS were 27.66-51.71 ng/L ($36.27{\pm}7.79ng/L$), 8.97-22.53 ng/L ($14.47{\pm}4.25ng/L$), LOQ-2.27 ng/L ($1.63{\pm}0.93ng/L$), and 3.0-7.66 ng/L ($4.27{\pm}1.49ng/L$), respectly. Other PFCs were below LOQ. The result of this study provides the distribution pattern of PFCs for assessing environmental pollution in two coastal areas of Korea.

Antioxidative Activities and Tyrosinase Inhibition of Water Extracts from Ailanthus altissima (가죽나무(Ailanthus altissima) 물 추출물의 항산화 활성과 Tyrosinase 저해)

  • Water extracts from root, stem and leaf of Ailanthus altissima were utilized to determine antioxidant properties such as electron donating ability (EDA), nitrite scavenging ability, superoxide dismutase (SOD)-like activity, and the inhibitory activities of xanthine oxidase (XO) and tyrosinase. The EDA of root extract was the highest as 77.33% at 0.5 mg/mL concentration and that of stem extract was 70.01% at 1.0 mg/mL. The nitrite scavenging ability of leaf extract revealed the highest effect as 95.18% at pH 1.2, 1.0 mg/mL while those of stem and root extracts were 55.17% and 33.33%, respectively. The leaf extract showed the highest SOD like activity as 26.77% at 1.0 mg/mL, the measurement of root extract was 3.82% and that of stem extract was not effective. All kinds of extracts had strong inhibitory activities on XO of over 92% at 1.0 mg/mL. The highest activity on tyrosinase inhibition was obtained from leaf extract of 16.33% at 2.0 mg/mL. The results indicated that among the three extracts, the leaf extract has a strong and extensive antioxidant activity.

Pharmacokinetics of Procainamide and N-acetylprocainamide (Procainamide와 그 대사산물(N-acetylprocainamide)의 약동학적 분석에 관한 연구)

  • To evaluate disposition characteristics of procainamide and its active metabolite, N-acetylprocainamide (NAPA), cross-over study for procainamide and NAPA was performed in 5 male adult dogs. After single administration of 10 mg/kg procainamide over 15 minutes, the range of measured plasma NAPA concentrations during experimental period were 0.03 to 0.124 ug/ml and early 'dip' phenomenon was distinct on NAPA concentration to time curve in all 5 dogs. Volume of distribution (Vss) and central compartment volume (Vc) of procainamide were $1.20{\pm}0.27\;L/kg$ of body weight and $0.36{\pm}0.08\;L/kg$, respectively. Vss and Vc of NAPA were $1.21{\pm}0.21\;L/kg$ and $0.26{\pm}0.07\;L/kg$, respectively. Intercompartmental clearance (Clint) of procainamide was 3.44 L/kg/hr and that of NAPA was 1.62 L/kg/hr. Total body clearance (Cl) of procainamide and NAPA were $0.47{\pm}0.08$ and $0.35{\pm}0.08\;L/kg/hr$. The half-life $(t_{1/2{\beta}})$ of procainamide and NAPA were 2.85 hrs and 2.77 hrs, respectively. Metabolic clearance (Clm)of procainamide by N-acetylation was $18.24{\pm}6.22\;ml/kg/hr$, which corresponded to 3.9% of total procainamide clearance.

Effect of Jasmonic Acid on the Root Growth and the Eleutheroside Accumulation in the Adventitious Root Culture of Eleutherococcus senticosus (가시오갈피나무 부정근(不定根) 배양(培養)에서 부정근(不定根)의 생장(生長)과 Eleutheroside유(類)의 생산(生産)에 미치는 Jasmonic acid처리(處理)의 영향)

  • This study was carried out to investigate the dose-dependent effect of jasmonic acid on both the adventitious root growth and the accumulation of various eleutherosides in the Eleutherococcus senticosus bioreactor culture. The highest biomass production (4.6 g DW/L) was observed in the absence of jasmonic acid and the root growth was significantly decreased by increasing the jasmonic acid concentration. However, jasmonic acid stimulated the production of both eleutheroside B, E and $E_1$. The highest level of eleutheroside B and E ($476.3{\mu}g/g\;DW$ and $676.0{\mu}g/g\;DW$) was obtained by 0.5 mg/L jasmonic acid treatment, while eleutheroside $E_1$ was accumulated at the highest level by 0.01 mg/L jasmonic acid treatment. The highest content of total eleutheroside was $2468.6{\mu}g/L$ when 0.01 mg/L jasmonic acid was applied. In addition, when the adventitious roots were cultured with 0.01 mg/L jasmonic acid, the highest levels of eleutheroside B, E and $E_1$ were observed at the 6th, 8th and 4th day of culture, respectively.

Effects of Plant Growth Regulators on In Vitro Germination and Organ Formation of Wild Angelica gigas N. (야생 참당귀(Angelica gigas N.)의 기내발아 및 기관유도에 미치는 PGRs의 영향)

  • This study was conducted to establish the in vitro optimal condition for seed germination and organogenesis of wild Angelica gigas. The experiment was evaluated the effects of $GA_3$ for pre-treatment with different periods of time (0h, 24h, 48h, 72h) and followed the treatment of seeds by control, scarification and methanol-heating method. As a result, the highest rates (15%) of seed germination was shown under the treatment without soaking of $GA_3$ and methanol-heating treatment. The seed germination was highly increased 60% under the condition of treatment on ultrasonic waves (frequency 80 KHz) with methanol-heating treatment including 0.1 mg/L $GA_3$. The highest callus induction rate was obtained from in vitro germinated stem, root and hypocotyl on the MS medium with 1.0 mg/L NAA and 0.5 mg/L BA. The highest percentages of shooting (50%) and rooting (85%) induction were observed in hypocotyl and root cultured on PGRs free medium and 0.1 mg/L NAA, respectively. In addition, somatic embryogenesis was observed from stem (1.0 mg/L 2,4-D) and hypocotyl (0.1 mg/L NAA).

Isolation and Identification of Lactic Acid Bacteria from Commercial Kimchi (시판김치로부터 젖산균의 분리 및 동정)

  • This study was carried out to identify lactic acid bacteria isolated from commercial Kimchi. Twelve lactic acid bacteria strains were isolated from Chinese cabbage kimchi (Baechu kimchi) that was fermented for 4 days at room temperature after making kimchi, 6 strains from pickled ponytail radishes (Chongkak kimchi) that was fermented for 2 days, and 15 strains in radish cube kimchi (Kaktugi) that was fermented for 5 days, and 23 strains were isolated in pickled Wakegi (Pa kimchi) that was fermented for 4 days. Eight strains among the lactic acid bacteria of 12 strains isolated from Baechu kimchi (pH 4.0) were identified as Lactobacillus plantarum, 1 strain as Leuconostoc lactis, 2 strains as Lactobacillus casei subsp. pseudoplantarum, and 1 strain as Lactobacillus sake. Three strains among the lactic acid bacteria of 6 strains isolated from Chongkak kimchi (pH 4.5) were identified as Leuconostoc paramesenteroides, 2 strains as Leuconostoc mesenteroides subsp. mesenteroides, and 1 strain as Lactobacillus plantarum. Two strains among the 15 strains isolated in Kaktugi (pH 4.0) were identified as Leuconostoc lactis, 3 strains as Leuconostoc mesenteroides subsp dextranicum, 4 strains as Lactobacillus casei subsp. pseudoplantarum, and 4 strains as Lactobacillus coryniformis subsp. torquens. Twenty-two strains among the 23 strains isolated from Pa kimchi (pH 4.1) identified as L. plantarum and 1 strain was as Lactobacillus sake. From the results above, the dominant species of Baechu kimchi was confirmed as L. plantarum, Chongkak kimchi as L. paramesenteroides, Kaktugi as L. casei subsp. pseudoplantarum and L. coryniformis subsp. torquens, and Pa kimchi as L. plantarum.

METAL ION RESISTANCE OF THE BACTERIOCIN PRODUCING ENTEROCOCCI

  • Ten bacteriocin-producing Enterococcus faecium strains with urease activity ($1.10-6.2nkat.mL^{-1}$) were isolated from the rumen of 2-8 weeks old calves. All strains were resistant aginst disodium arsenate at a minimal inhibition concentration - MIC $5g.L^{-1}$ and mercury chloride ($MIC=10-20mg.L^{-1}$). Eight strains were resistant against silver nitrate ($MIC=40-50mg.L^{-1}$) and three against antibiotics used. The resistance against six antibiotics was found in A23 strain. Values of adherence index ranged from 5.02 to 20.4 enterococci adhered per one epithelial cell of rumen wall. All isolates produced bacteriocins which inhibited the growth at least of one of five indicator organisms. The EF1 strain with a good affinity to the epithelial cell ($15.2{\pm}1.2$) produced bacteriocin substance with antimicrobial activity against grampositive and gramnegative indicator bacteria.

Seasonal Variations in Nutrients and Principal Ions Contents of the Han River Water, and its Water Characteristics (한강의 영양염류 및 주요이온류의 년변화와 그 수질적고찰)

  • Nutrients and principal ions contents in downstream water of the Han River, near Seoul, measured at somewhat three week intervals over a period of two years were shown its characteristic seasonal cycles. The annual ranges of nutrients were 0.5-7.7 $\mu\textrm{g}$ at/L of ammonia, 0.12-0.90 $\mu\textrm{g}$ at/L of nitrite, 2.9-35.9 $\mu\textrm{g}$ at/L of nitrate, 8.8-42.0 $\mu\textrm{g}$ at/L of tatal soluble nitrogen, 0.09-0.57 $\mu\textrm{g}$ at/L of phosphorus, and 3.0-15.3 mg/L of silicate, and these were lower in spring and fall closely correlate with phytoplankton blooms and higher in winter except ammonia and silicate contents. Annual variations of BOD and COD were 0.10-2.14 ml/L, 0.88-6.82 mg/L, respectively. The ionic concentrations of Cl, Ca and Mg were not high of which Cl 5.0-13.1 mg/L, Ca 3.7-9.1 mg/L and Mg 1.5-10.4 mg/L of annual ranges. Fe ranged 18.5-82.8 $\mu\textrm{g}$/L, Mn 15-58 $\mu\textrm{g}$/L, Cu 0.3-4.7 $\mu\textrm{g}$/L annually In general, the Han River water is low in nutrients, mineral contents, and may be said not yet polluted, excellent quantity and quality of river water.

Adhesion Ability and Inhibition of Enterohemorrhagic E. coli O157:H7 Adhesion to Intestinal Epithelial Cells in Lactobacillus acidophilus (Lactobacillus acidophilus의 장 상피세포에 대한 부착능력 및 장 출혈성 대장균의 부착 억제 능력)

  • The ability of probiotics containing Lactobacillus acidophilus to adhere to the intestinal epithelium may play an important role in colonization of the gastrointestinal tract and preventing enteric pathogen such as enterohemorrhagic E. coli(EHEC O157:H7. In the study, we investigated the adhesion to human intestinal epithelial cells(HT-29) of strains of L. acidophilus(3 from human, 2 from pig, and 1 from calf). All of the tested strains of L. acidophilus were highly observed adhesion ability(from 10$\^$6/ to 10$\^$7/ cfu/mL), compared to L. rhamnosus GG as control. Also, adhered strains of L. acidophilus were significantly preserved in serial wash-out steps. However, no correlation could be observed between cell surface hydrophobicity and adhesion abilities of the tested strains of L. acidophilus. Inhibition of adhesion of EHEC O157:H7 was also examined, a 2 log cycle reduction was observed by all of the tested strains of L. acidophilus. These results suggest that the strains of L. acidophilus with high adhesion ability are resistant to wash-out and adhesion ability inhibition by selected strains of L. acidophilus helps to prevent adhesion of EHEC O157:H7 to intestinal epithelial cells.
