Toxic Effects of Heavy Metal (Cd, Cu, Zn) on Population Growth Rate of the Marine Diatom (Skeletonema costatum) (중금속(Cd, Cu, Zn)이 해산규조류(Skeletonema costatum)의 개체군 성장률에 미치는 독성영향)

  • In this study, we evaluated the toxic effects of heavy metals (Cd, Cu, Zn) on the population growth rate (r) of the marine diatom, Skeletonema costatum. S. costatum. The population growth rate (r) of the species was determined after 96 hrs. of exposure to Cd (0, 0.63, 1.25, 2.50, 5.00, 10.00 ppm), Cu (0, 0.25, 0.50, 0.75, 1.00, 1.25, 1.50 ppm) and Zn (0, 0.31, 0.63, 1.25, 2.00, 2.50, 5.00 ppm). It was observed that 'r' in the control (absence of Cd, Cu and Zn) were greater than 0.05, however suddenly decreased with increased heavy metal concentrations. Cd, Cu and Zn reduced 'r' in a dose-dependent manner and a significant reduction were occurred at concentration of greater than 1.25, 1.25 and 2.50 ppm, respectively. Based on the toxicity, the heavy metal were ranked as Cu>Zn>Cd, with EC50 values of 1.11, 2.13 and 6.84 ppm, respectively. The lowest-observed-effective-concentration (LOEC) of 'r' in exposed to Cd, Cu and Zn were 1.25, 1.00, 2.00 ppm, respectively. Precisely, a concentration of greater than 1.25 ppm of Cd, 1.00 ppm of Cu and 2.00 ppm of Zn in marine ecosystems induced toxic effect on the 'r' of S. costatum. Based on our results, we suggested that the 'r' of S. costatum might be a useful bio indicator for the toxicity assessment of heavy metals in marine ecosystems.

Biological Control of Aphids on Pepper in Greenhouses Using Aphidius gifuensis (싸리진디벌을 이용한 하우스고추에서 발생하는 진딧물의 생물적 방제)

  • The life-cycle of the aphid-parasite, Aphidius gifuensis, took 11.9 days at $25^{\circ}C$ and 12.1 days at $30^{\circ}C$. The female appearance rate of A. gifuensis was the best at $20{\sim}25^{\circ}C$ compared to the other temperatures. Optimum rearing temperature of Aphidius gifuensis was $20{\sim}25^{\circ}C$ Optimum temperatures for A. gifuensis emergence were $20^{\circ}C$ and $25^{\circ}C$ with 99.1% and 98.2%, respectively. However the emergence rate of Aphidius gifuensis was 58.8% at $10^{\circ}C$. There was a positive correlation between A. gifuensis adult activity and temperature, but there was a negative correlation between developmental period of Myzus persicae nymph and temperature. Among the pests occurring in pepper greenhouse, dominant species was M. persicae and its density was 6~1,024 per plant. The damaged fruit rate by Helicoverpa assulta was 3.3~53.3%. The number of aphid and mummy in the A. gifuensis released plot were 173.5 and 10 in June, 1.8 and 17 in July, 2000, respectively. The numbers of Aphidius gifuensis were 7.5 and 0.4 in May, 27.1 and 2.1 in June, 2001, respectively. The suppressive effects on M. persicae in A. gifuensis released plot was better than the control plot, but there was no significant difference compared to the pesticide-sprayed plot. The first leaf length and the stem width in the A. gifuensis released plot, the control plot, and the pesticide-sprayed plot were not significantly different, but the A. gifuensis released plot showed the highest yield among the plots.

Effect of Starter Culture and Temperature on the Flavor and Sensory Characteristics of Dry-Cured Ham

  • This study focused on understanding the effects of yeast and mold on the sensory properties of dry-cured ham aged at 20℃ and 25℃. Debaryomyces hansenii isolated from Doenjang and fermented sausages, and Penicillium nalgiovense isolated from fermented sausages were utilized. The CIE a* tended to increase in all treatments as the aging period increased. At 6 weeks of aging, DFD25 showed a significantly higher CIE a* value than other treatments. The shear force tended to increase in all treatments as the aging period increased. At 6 weeks of aging, among the treatments aged at 25℃, DFD25 showed a low tendency to shear force. The PC1 of the electronic nose was 42.872%. At 25℃, the hexane content was higher and levels of ethanol, propan-2-one, 2,4,5-trimethylthiazole, and limonene were lower than that at 20℃. DFD25 showed significantly higher hexane content and significantly lower limonene content than other treatments. The PC1 of the electronic tongue was 84.529%. All treatments, except for the C starter, exhibited higher salt and lower sour levels at 25℃ compared to 20℃ when the same starter was used. The DFD25 showed the lowest sour taste and a higher tendency of umami than the other treatments. Sensory evaluation revealed that DFD25 had significantly higher scores for texture than C25, whereas no significant differences were observed in other aspects. Therefore, the used starters are considered suitable for aging at 25℃; among them, the DFD starter demonstrates superior qualities and enhanced commercial potential compared to the control.

Three Hydroxylated Ginsenosides from Heat Treatmented Ginseng (인삼의 열처리 과정 중 생성되는 3종의 수산화진세노사이드에 대한 연구)

  • Ginsenosides are considered to be the most important ingredients in ginseng. They are chemically converted by endogenous organic acids contained in ginseng and the heat applied during red ginseng processing. During this procedure, various converted ginsenosides are produced through hydrolysis of substitute sugars of ginsenosides and forming double bonds through dehydration in the dammarane skeleton. In order to study the conversion mechanism of protopanaxadiol-type ginsenosides during the heat treatment process of ginseng, we purified the three final converted ginsenosides by heating fresh ginseng for a long time. The three isolated ginsenosides were identified as 25(OH)-ginsenoside Rg5, 25(OH)-ginsenoside Rz1 and 25(OH)-ginsenoside Rg3 through NMR spectrum analysis. As a result of quantification of ginseng heated at 100 ℃ for 0 to 6 days by HPLC/UV and TLC methods, the content of 25(OH)-ginsenosides tended to increase in proportion to the time exposed to heat. In particular, the content of 25(OH)-ginsenosid Rg5 was confirmed to be noticeably increased.

LIN-23, an E3 Ubiquitin Ligase Component, Is Required for the Repression of CDC-25.2 Activity during Intestinal Development in Caenorhabditis elegans

  • Caenorhabditis elegans (C. elegans) utilizes two different cell-cycle modes, binucleations during the L1 larval stage and endoreduplications at four larval moltings, for its postembryonic intestinal development. Previous genetic studies indicated that CDC-25.2 is specifically required for binucleations at the L1 larval stage and is repressed before endoreduplications. Furthermore, LIN-23, the C. elegans ${\beta}$-TrCP ortholog, appears to function as a repressor of CDC-25.2 to prevent excess intestinal divisions. We previously reported that intestinal hyperplasia in lin-23(e1883) mutants was effectively suppressed by the RNAi depletion of cdc-25.2. Nevertheless, LIN-23 targeting CDC-25.2 for ubiquitination as a component of E3 ubiquitin ligase has not yet been tested. In this study, LIN-23 is shown to be the major E3 ubiquitin ligase component, recognizing CDC-25.2 to repress their activities for proper transition of cell-cycle modes during the C. elegans postembryonic intestinal development. In addition, for the first time that LIN-23 physically interacts with both CDC-25.1 and CDC-25.2 and facilitates ubiquitination for timely regulation of their activities during the intestinal development.

Determining the Required Vitamin D Level for Bone Health Based on Bone Turnover Markers

  • To date, no clear threshold that has been established for defining an adequate store of vitamin D for bone health. Therefore, this study aims to determine the required level of vitamin D to maintain a healthy skeleton based on bone remodelling process among healthy adult population. This was a cross sectional study, involving a healthy adult population in Kota Bharu, Malaysia, aged 18~50 years. We measured serum 25(OH)D (vitamin D), serum parathyroid hormone (PTH), serum C-terminal telopeptide of type 1 collagen (CTX), and Procollagen 1 Intact N-Terminal (P1NP) in 120 healthy adults selected via multi stage sampling (64 males, 56 females) from 6 subdistricts in Kota Bharu. The mean level of 25(OH)D was 23.50 (${\pm}8.74$) nmol/L. There was a significant difference of the vitamin D level between genders ($26.81{\pm}8.3nmol/L$ vs $19.72{\pm}7.68nmol/L$ in males and females respectively) (p value<0.001). More than 50% of female subjects had 25(OH)D less than 20 nmol/L, while only 20.3% of male subjects had 25(OH)D below 20 nmol/L. Based on the LOESS plot, the bone turnover markers showed a plateauing result, at the 25(OH)D level of 35 nmol/L for CTX and 20 nmol/L for P1NP. Contrastingly, PTH showed a step rise in the 25(OH)D level of 20 nmol/L. Based on the LOESS plot for CTX, P1NP and PTH versus 25(OH)D, level of vitamin D between 20 to 35 nmol/L is recommended to maintain healthy skeleton.

The Influence Analysis of Support Working Expenses for Yongdam Dam Area Considering the Resolution of Digital Topographic Map (수치지형도 해상도를 고려한 용담댐 주변지역 지원사업비 영향 분석)

  • A dam is effective in stable supply of water required in daily life and reduced damage from floods, but there are problems as a lot of land or houses are submerged. Therefore many projects have been conducted in order to improve and support daily life environment surrounding a dam. This study has focused on analyzing how to calculate support working expenses for Dam area by using GIS spatial overlay in addition to effects of scale of a topographic map and reached the following conclusion. First, as a result of areal error in submerged area by scale based on a 1/3,000 digital topographic map, it has been found that a 1/5,000 digital topographic map is 9.5 times more accurate than a 1/25,000 digital topographic map in the total of areal error. Second, as a result of areal error in area surrounding a dam, it has been found that a 1/5,000 digital topographic map is 7.4 times more accurate than a 1/25,000 digital topographic map in the total of areal error. Third, as a result of error of support expense for submerged area, it has been found that a 1/5,000 digital topographic is 15.9 times, 14.7 times and 15.9 times more effective than a 1/25,000 digital topographic map in terms of the total error of support expense, standard error and the total support expense error on the entire project costs in submerged area. In addition, as a result of analysis on error of support expense for area surrounding a dam, it has been found that a 1/5,000 digital topographic map was 10.7 times, 9.6 times and 10.6 times more effective, respectively, in the total error of support expense, standard error and the total error of support expense for the entire project costs in area surrounding a dam compared to a 1/25,000 digital topographic map. Lastly, as a result of error of the entire project costs for area surrounding a dam, it has been found that a 1/5,000 digital topographic map was 1.4 times, 1.3 times and 1.4 times more effective, respectively, in the total error of support expense, standard error and the total error of the entire project costs compared to a 1/25,000 digital topographic map, but it was not much different from the result of calculating areal error in submerged area or area surrounding a dam because population item didn't consider areal concept.

Effects of Temperature and Irrigation Intervals on Photosynthesis, Growth and Growth Analysis of Pot-grown Cucumber Seedlings (온도와 관수 주기가 오이 포트 묘의 광합성, 생육 및 생장 해석에 미치는 영향)

  • This study was conducted in an indoor cultivation room and chamber where environmental control is possible to investigate the effect of temperature and irrigation interval on photosynthesis, growth and growth analysis of potted seedling cucumber. The light intensity (70 W·m-2) and humidity (65%) were set to be the same. The experimental treatments were six combinations of three different temperatures, 15/10℃, 25/20℃, and 35/25℃, and two irrigation intervals, 100 mL per day (S) and 200 mL every 2 days (L). The treatments were named 15S, 15L, 25S, 25L, 35S, and 35L. Seedlings at 0.5 cm in height were planted in pots (volume:1 L) filled with sandy loam and treated for 21 days. Photosynthesis, transpiration rate and stomatal conductance at 14 days after treatment were highest in 25S. These were higher in S treatments with a shorter irrigation interval than L treatments. Total amount of irrigation water was supplied evenly at 2 L, but the soil moisture content was highest at 15S and lowest at 25S > 15L > 25L, 35S and 35L in that order. Humidity showed a similar trend at 15/10℃ (61.1%) and 25/20℃ (67.2%), but it was as high at 35/25℃ (80.5%). Cucumber growth (plant height, leaf length, leaf width, chlorophyll content, leaf area, fresh weight and dry weight) on day 21 was the highest in 25S. Growth parameters were higher in S with shorter irrigation intervals. Yellow symptom of leaf was occurred in 89.9% at 35S and 35L, where the temperature was high. Relative growth rate (RGR) and specific leaf weight (SLA) were high at 25/20℃ (25S, 25L), RGR tended to be high in the S treatment, and SLA in the L treatment. Water use efficiency (WUE) was high in the order of 25S, 25L > 15S > 15L, 35S, and 35L. As a result of the above, the growth and WUE were high at the temperature of 25/20℃.

Isolation of Aspergillus niger K-25 Prroducing Acid-stable ${\alpha}-amylase$ (내산성(耐酸性) 아밀라제를 생산하는 Aspergillus niger 균주의 분리)

  • One strain of Aspergillus niger K-25 producing an acid-stable ${\alpha}-amylase$ was isolated from the soil. The optimum culture conditions were investigated. The production of the acid-stable ${\alpha}-amylase$ was enhanced when the strain was incubated in a medium containing soluble starch 3.5%, peptone 2%, $KH_2PO_4$ 0.5%, $MaSO_4{\cdot}7H_2O$ 0.25% and $FeCI_3$ 1.0% at pH 3 for 7 days. However, higher activity of acid-stable ${\alpha}-amylase$ was demonstrated on wheat bran culture. Amylase production was doubled when A. niger K-25 was incubated on the wheat bran supplemented with fumaric acid buffer (pH 3).
