제주도 동북해역의 해수에서 한천분해활성을 보어는 해양성 세균을 분리하였으며 16S rDNA유전자 염기서열분석과 형태학적, 배양적 및 생화학적 특징을 조사하여 해양기원의 Agarivorans 속에 속하는 균주임을 확인하고 Agarivorans sp. JA-1으로 명명하였다. Agarivorans sp. JA-1이 생성하는 한천 분해효소(agarase)는 한천의 존재유무에 상관없이 발현되며 성장의존성인 것으로 확인되었고 효소활성을 위한 최적 pH는 pH 8.04 (50 mM glycine NaOH 완충용액)이고 최적 온도는 $40^{\circ}C$로 나타났다. 한천분해효소는 기능성올리고당 생산이 가능한 베타-한천분해효소이며 내열성을 보여 $60^{\circ}C$까지 $80\%$ 이상 그리고 $80^{\circ}C$까지 $70\%$의 잔존활성을 보이는 것으로 나타났다. 분리한 Agarivorans sp. JA-1 균주가 나타내는 한천분해효소를 이용하여 $40^{\circ}C$ 이상에서 액체상태로 있는 한천을 이용한 기능성올리고당 생산에 유용한 것으로 생각되었다.
본 연구진은 청국장에서 수종의 미생물을 분리하였으며, 이중 소화 효소능이 뛰어난 균주를 선별하였으며, 유전학적 분석결과를 통해 본 균주가 Bacillus 속 균주임을 확인하고 Bacillus subtilis BC-P1이라 명명하였다. Bacillus subtilis BC-P1을 starter로 사용하여 청국장을 제조하였으며, protease, xylanase, chitinase activity, 혈전 분해능, 영양성분 분석 및 아미노산 분석을 통하여 B. subtilis BC-P1를 starter로 활용한 청국장 제품의 개선 가능성을 타진하였다. Bacillus subtilis BC-P1를 starter로 제조한 청국장은 기존 제품에 비해 우수한 소화효소 생성능을 보였으며, 특히 혈전분해효소 생성능이 우수함을 확인하였고, 대조군에 비해 증가된 환원당 및 총 아미노산 함량을 확인 할 수 있었다. 따라서 본 균주를 starter로 하여 청국장을 제조할 경우 기능성분, 생리활성의 증가, 품질 향상을 기대할 수 있을 것으로 판단되며, 향후 대규모 생산시 균주, 제조 조건에 관한 연구가 필요할 것으로 사료된다.

The purpose of this study was to isolate probiotic lactic acid bacteria (LAB) that produce bile salt hydrolase (BSH), and to evaluate its effects on serum cholesterol level. One-hundred-twenty bacterial colonies were initially isolated from human feces, and five strains were selected after screening based on their resistance to acids, tolerance against bile salts, and inhibitory activity on Escherichia coli. The Lactobacillus plantarum strain with the highest level of BSH activity was identified using 16S rRNA sequences, and was named L. plantarum CK 102. L. plantarum CK 102 at a level of 1.36$\times$10$^8$cfu/ml survived in pH 2 buffer for 6 h and exhibited excellent tolerance for bile salt. Coculturing the strain with E. coli in MRS broth resulted in strong inhibition against growth of E. coli at 18 h. Furthermore, the potential effect of CK 102 on serum cholesterol level was evaluated in rats. Thirty-two rats [Sprague-Dawley (SD) male, 129$\pm$l g, 5 weeks old] were divided into four groups of eight each. For six weeks, Group 1 was fed a normal diet (negative control); Group 2 was fed a cholesterol-enriched diet (positive control); Group 3 was fed a cholesterol-enriched diet plus L. plantarum CK 102 at 1.0$\times$10$^7$cfu/ml; and Group 4 was fed a cholesterol-enriched diet plus L. plantarum CK 102 at 5.0$\times$10$^7$cfu/ml. Blood samples were collected, serum lipids were analyzed, and weights of the organs were measured. Total blood cholesterol level, triglyceride, LDL-cholesterol, and free-cholesterol values were lower in rats that were fed 1. plantarum CK 102 than in those not fed L. plantarum CK 102. This cholesterol lowering effect implies that L. plantarum CK 102 could be utilized as an additive for health-assistance foods. In conclusion, these results suggest that the 1. plantarum CK 102 isolated could be used commercially as a probiotic.
Objectives: The genus Legionella is common in aquatic environments. Some species of Legionella are recognized as potential opportunistic pathogens for human, notably Legionella pneumophila that causes, Legionellosis. Thus, we investigated the contamination of Legionella pneumophila on water supply systems in Seoul, including cooling towers, public baths, hospitals and fountains. Methods: The existence of 16S rRNA and mip gene of L. pneumophila was confirmed in the genome of the isolated strains by PCR. Results: During the summer season of 2010 and 2011, Legionella pneumophila were detected from 163 samples (21.1%) out of 772 samples collected. Among the 163 strains of L. pneumophila, eighty one isolates belonged to serogroup 1 (57.4%), 23 isolates were serogroup 5 (16.3%), 21 isolates were serogroup 6 (14.9%), 8 isolates were serogroup 2 (5.79%), and 8 isolates were identified in serogroup 3 (5.7%). Through PFGE (pulsed-field gel electrophoresis) analysis using Sfi I, genetic types of L. pneumophila were classified into five (A to E) patterns by the band similarity with excess of 70% from public baths. Conclusions: The PFGE patterns of the serotypes showed a tendency for diversity of L. pneumophila. Our results suggest the existence of serological and genetic diversity among the L. pneumophila isolates.
여러 식물의 근권에서 세균 균주들을 분리하여 이들의 항진균성 물질 생성능을 조사하였다. 분리균주 중 Fusarium oxysproum을 포함한 5가지 주요 식물병원성 진균에 대한 생장저해가 우수한 7개 균주를 16S rRNA 유전자 염기서열로 동정하였다. 이들 중 Paenibacillus peoriae RhAn32, Pseudomonas otitidis TK1과 Bacillus cereus TK2가 가장 넓은 생장 저해범위를 나타내었으며 항진균성 물질 중 siderophore 생성능은 P. otitidis TK1이 3.21 mM/ml로 가장 높았고 HCN은 Pseudomonas koreensis Rh2와 Rh7 균주만이 생성하였으며 Brevibacterium frigoritolerans EnA9와 P. peoriae RhAn32는 각각 1558.9와 $1436.7{\mu}M$ glucose/min/mg protein의 높은 chitinase 활성을 나타내었다. P. otitidis TK1과 P. peoriae RhAn32의 항진균능을 진균과의 공동배양을 통해 정량분석한 결과 세 종류의 Fusarium oxysproum의 생장을 유의성 있게 저해하였다. P. otitidis TK1은 식물생장을 촉진하는 호르몬인 auxin의 생성능도 가장 높았는데 이 균주를 4주 자란 토마토 유묘에 F. oxysporum f. sp. lycopersici와 동시에 접종하여 8주간 재배하였을 때 줄기와 뿌리 길이 및 습윤 중량이 비접종 대조군에 비해 각각 45.7, 64.9와 118%로 유의성 있게 증가하였다. 따라서 이 균주들은 유기합성농약을 대체하면서 식물생장을 촉진하는 미생물제로서의 적용이 가능할 것으로 기대된다.
A novel Gram-positive, motile, non-spore forming aerobic marine bacterium, designated 107-1T was isolated from tidal mud collected in Gyehwa-do, South Korea. Cells of strain 107-1T were short rod or coccoid, oxidase negative, catalase positive and grew at 10-40℃ (with optimum growth at 25-30℃). It utilized menaquinones MK-7 and 8 as its respiratory quinones and its major fatty acids were anteiso-C15:0 (37.9%), iso-C16:0 (14.9%), and iso-C14:0 (10.8%). Phylogenetic analysis based on 16S rRNA gene sequences revealed a distinct clade containing strain 107-1T and close species Planococcus ruber CW1T(98.52% sequence similarity), P. faecalis KCTC 33580T(97.67%), P. kocurii ATCC 43650T(97.48%), P. donghaensis DSM 22276T(97.47%), and P. halocryophilus DSM 24743T(97.37%). Strain 107-1T contains one circular chromosome (3,513,248bp in length) with G+C content of 44.6 mol%. Estimated ranges for genome to genome distance, average nucleotide identity, and average amino acid identity comparing strain 107-1T with close taxa were 20.3-34.8%, 77.9-86.9%, and 73.6-92.8%, respectively. Based on polyphasic analysis, strain 107-1T represents a novel species belonging to the genus Planococcus.
Objectives: The aim of this study is to investigate microbial contamination in the school food service environment for the assessment of microbial food safety. Methods: We collected both swab samples from tables and desks and airborne bacterial samples from an elementary school (School A) and a high school (School B). Heterotrophic plate count, total coliform, Staphylococcus aureus, and Bacillus cereus were measured with selective media to quantify microbial concentration. PCR assay targeting 16S rRNA genes was performed to identify the strains of S. aureus and B. cereus isolated. In addition, we made a food service checklist for the locations to evaluate the food service environment. A Wilcoxon test was employed to examine the differences in microbial concentration between before lunchtime and afterwards. Results: Heterotrophic plate counts showed higher levels after-lunch compared to before-lunch at School B. However, levels of S. aureus were higher in the after-lunch period (p<0.05) in both classrooms and in the cafeteria in School A. B. cereus was only sparsely detected in School B. Several samples from food dining carts were found to be contaminated with bacteria, and facilities associated with food delivery were found to be vulnerable to bacterial contamination. Although microbial concentrations in the air showed little difference between before- and after-lunchtime in the cafeteria in School A, those in classrooms were greater after-lunchtime at both schools. Conclusion: Our results suggested that the microbial safety in schools after lunchtime of concern. Necessary preventive measures such as hygiene education for students and food handlers should be required to minimize microbial contamination during food service processes in schools.
항생제 및 항진균제 개발에 있어 미생물 유래 천연 생리활성물질 분리를 통한 선도물질을 확보하는 일은 매우 중요하며, 신규 물질을 확보하기 위해 꾸준한 연구가 필요할 것으로 사료된다. 따라서 본 연구에서는 천연 항생물질의 개발을 위한 연구의 일환으로 태백산 일대의 토양에서, 인체에 다양한 감염증을 유발하는 효모와 곰팡이에 대하여 강한 항진균 활성을 나타내는 BCNU 2003 균주를 분리하여 항진균 활성 물질의 이용가능성에 대해 연구하였다. BCNU 2003은 계통적으로는 B. amyloliquefaciens와 B. vallismortis의 subcluster에 속하는 균주로 동정되어 Bacillus sp. BCNU 2003으로 명명하였다. 항균물질 분리를 위해 ethyl acetate (EA) 추출물과 펩타이드 추출물로 나누어 그람양성 세균, 그람음성 세균 및 진균에 대한 항균활성을 측정한 결과, EA 추출물이 6종의 인체병원성 진균에 대해 모든 높은 항진균 활성을 나타내었다. 특히 기회성 감염을 유발하는 A. niger, C. albicans 그리고 Sa. cerevisiae에 대해 높은 억제 활성을 나타냈으며, 균배양액에서 낮은 저해율을 보였던 Ep. floccosum에 대해서도 EA 추출물은 높은 활성을 나타내었다. 따라서 다양한 인체 병원성 진균에 대해 넓은 항균스펙트럼을 가지는 Bacillus sp. BCNU 2003 균주의 활성물질 분리를 통해 특정 항균 및 항진균 물질의 대량생산 조건 등의 추가적인 연구를 수행한다면, 인체 감염증을 포함한 광범위한 피부치료제의 응용개발이 가능하리라 사료된다.
본 연구에서는 김치로부터 분리된 L. brevis SM61을 생화학적 특성과 16s rRNA 염기서열 분석을 통해 분리 및 동정하였으며, 흰점박이꽃무지 유충 열수추출물과 산양삼 분말을 L. brevis SM61을 이용하여 48시간동안 발효를 수행하여 발효물을 제조하였다. 그리고, 흰점박이꽃무지 유충 열수추출물과 L. brevis SM61을 사용한 발효물에 대해 농도에 따른 세포 독성은 없는 것으로 판단되었다. 그리고, 흰점박이꽃무지 유충 열수추출물과 L. brevis SM61을 사용한 발효물에 대해 발효 시간에 따른 폴리페놀 함량을 분석한 결과 발효 초기에는 약 $1.49{\mu}g/mL$ 비해 48시간 이후 약 $5.09{\mu}g/mL$ 수준으로 증가하였다. 또한, 항산화능은 흰점박이꽃무지 유충 열수추출물이 약 60%였으나, L. brevis SM61을 이용한 발효물이 약 98%로 높아졌다. 항균 활성의 경우 L. brevis SM61을 이용한 발효물에서만 항균 효과가 확인되었다. 따라서, 본 연구를 통해 분리된 신규 L. brevis SM61을 이용한 흰점박이꽃무지 유충의 열수추출물에 대한 발효물은 식용 곤충의 기능성을 강화시킬 수 있을 것으로 판단되며, 해당 유산균을 활용하여 다양한 식용 곤충에도 적용이 가능할 것으로 사료된다.

Purpose: The aims of this study were to examine the salivary microbiota in conditions of periodontal health and disease and to explore microbial changes following nonsurgical periodontal treatment. Methods: Non-stimulated saliva samples were collected from 4 periodontally healthy participants at baseline and from 8 patients with chronic periodontitis at baseline and 3 months following nonsurgical periodontal therapy. The V3 and V4 regions of the 16S rRNA gene from the DNA of saliva samples were amplified and sequenced. The salivary microbial compositions of the healthy participants and patients with periodontitis prior to and following nonsurgical treatment of periodontitis were compared based on the relative abundance of various taxa. Results: On average, 299 operational taxonomic units were identified in each sample. The phylogenetic diversity in patients with periodontitis was higher than that in healthy participants and decreased following treatment. The abundance of the phylum Spirochaetes and the genus Treponema in patients with periodontitis was 143- and 134-fold higher than in the healthy control group, respectively, but decreased significantly following treatment. The species that were overabundant in the saliva of patients with periodontitis included the Peptostreptococcus stomatis group, Porphyromonas gingivalis, the Fusobacterium nucleatum group, Parvimonas micra, Porphyromonas endodontalis, Filifactor alocis, and Tannerella forsythia. The phylum Actinobacteria, the genus Streptococcaceae_uc, and the species Streptococcus salivarius group were more abundant in healthy participants than in those with periodontitis. There was a trend toward a decrease in disease-associated taxa and an increase in health-associated taxa following treatment. Conclusions: Our results revealed differences in the taxa of salivary microbiota between conditions of periodontal health and disease. The taxa found to be associated with health or disease have potential for use as salivary biomarkers for periodontal health or disease.
