Pantoea Bacteria Isolated from Three Thrips (Frankliniella occidentalis, Frankliniella intonsa, and Thrips tabaci) in Korea and Their Symbiotic Roles in Host Insect Development

  • Gut symbionts play crucial roles in host development by producing nutrients and defending against pathogens. Phloem-feeding insects in particular lack essential nutrients in their diets, and thus, gut symbionts are required for their development. Gram-negative Pantoea spp. are known to be symbiotic to the western flower thrips (Frankliniella occidentalis). However, their bacterial characteristics have not been thoroughly investigated. In this study, we isolated three different bacteria (BFoK1, BFiK1, and BTtK1) from F. occidentalis, F. intonsa, and T. tabaci. The bacterial isolates of all three species contained Pantoea spp. Their 16S rRNA sequences indicated that BFoK1 and BTtK1 were similar to P. agglomerans, while BFiK1 was similar to P. dispersa. These predictions were supported by the biochemical characteristics assessed by fatty acid composition and organic carbon utilization. In the bacterial morphological analysis, BFoK1 and BTtK1 were distinct from BFiK1. All these bacteria were relatively resistant to tetracycline compared to ampicillin and kanamycin, in which BFoK1 and BTtK1 were different from BFiK1. Feeding ampicillin (100,000 ppm) reduced the bacterial density in thrips and retarded the development of F. occidentalis. The addition of BFoK1 bacteria, however, rescued the retarded development. These findings indicate that Pantoea bacteria are symbionts to different species of thrips.

  • Several bacterial etiological agents of streptococcal disease have been associated with fish mortality and serious global economic loss. Bacterial identification based on biochemical, molecular, and phenotypic methods has been routinely used, along with assessment of morphological analyses. Among these, the molecular method of 16S rRNA sequencing is reliable, but presently, advanced genomics are preferred over other traditional identification methodologies. This review highlights the geographical variation in strains, their relatedness, as well as the complexity of diagnosis, pathogenesis, and various control methods of streptococcal infections. Several limitations, from diagnosis to control, have been reported, which make prevention and containment of streptococcal disease difficult. In this review, we discuss the challenges in diagnosis, pathogenesis, and control methods and suggest appropriate molecular (comparative genomics), cellular, and environmental solutions from among the best available possibilities.

상악동염 병소 부위에서 세균의 분리 동정 및 항생제 감수성에 대한 연구 (ANTIBIOTIC SUSCEPTIBILITY OF BACTERIA ISOLATED FROM MAXILLARY SINUSITIS LESION)

  • The purpose of this study was to isolate and identify the bacteria in chronic maxillary sinusitis (CMS) lesions from 3 patients and to determine the antimicrobial susceptibility of them against 10 antibiotics. One of them was odontogenic origin and the others were non-odontogenic origin. Pus samples were collected by needle aspiration from the lesions and examined by culture method. Bacterial culture was performed in three culture systems (anaerobic, CO2, and aerobic incubator). Identification of the bacteria was performed by 16S rRNA gene (16S rDNA) nucleotide sequencing method. To test the sensitivity of the bacteria isolated from the maxillary sinusitis lesions against seven antibiotics, penicillin G, amoxicillin, tetracycline, ciprofloxacin, cefuroxime, erythromycin, clindamycin, and vancomycin, minimum inhibitory concentration (MIC) was performed using broth dilution assay. Our data showed that enterobacteria such as Enterobacter aerogenes (30%), Klebsiella pneumoniae (25%), and Serratia marcescens (15%) were predominately isolated from the lesion of non-odontogenic CMS of senile patient (70 year old). Streptococcus spp. (40.3%), Actinomyces spp. (27.4%), P. nigrescens, M. micros, and P. anaerobius strains were isolated in the lesion of odontogenic CMS. In the lesion of non-odontogenic CMS, Streptococcus spp. (68.4%), Rothia spp. (13.2%), and Actinomyces sp. (10.5%) were isolated. The susceptibility pattern of 10 antibiotics was determined according to the host of the bacteria strains ratter than the kinds of bacterial species. Even though the number of CMS was limited as three, these results indicate that antibiotic susceptibility test must be accompanied with treatment of CMS. The combined treatment of two or more antibiotics is better than single antibiotic treatment in the presence of multidrug-resistant bacteria in the CMS lesions.

멕시코 B 광산 슬러지에 존재하는 생물학적 광물화 미생물의 특성에 관한 연구 (Isolation and Identification of Bacteria Involved with Biomineralization at B Mine Sludge in Mexico)

  • Microbial processes that bind heavy metals and form minerals are widespread, and they represent a basic aspect of biogeochemistry. Some microorganisms can crystallize minerals by secreting a specific enzyme. In particular, calcite ($CaCO_3$) precipitation is an important part of biomineralization, and has been studied extensively because of its wide application in civil engineering technology. This process provides an effective way to stabilize heavy metals within a relatively stable crystal phase. In this study, biomineralization of calcite by three urea-hydrolyzing indigenous bacterial strains was investigated by microbiological analyses. Three bacterial strains were isolated from the sludge of B mine in Mexico and each bacterial strain was identified by the cellular fatty acid composition and 16S rRNA partial sequencing analysis. The results of the identification analysis showed that these strains were closest to Sporosarcina pasteurii, Kurthia gibsonii, and Paenibacillus polymyxa. We found that the optimum conditions for growth of these indigenous bacteria were $30-40^{\circ}C$ and pH range of 7-8. Microbiological analyses showed the possibility that the bioaccumulated heavy metals ions were deposited around the cell as crystalline carbonate minerals under the optimum conditions. The findings of our study suggest that the indigenous bacterial strains play an important role in heavy metal immobilization.

돼지 장내로부터 분리된 항생제 저항성 Pediococcus pentosaceus의 특성 (Characterization of Pediococcus pentosaceus Isolated from Porcine Intestine)

  • A lactic acid bacterial strain resistant to several antibiotics such as oxytetracycline, tylosin, neomycin and sulfathiazole, which have been often used as a therapeutic agent in livestock, was isolated from the porcine gastrointestinal tract. The isolate YB-55 was identified as belonging to the genus Pediococcus with the highest similarity to P. pentosaceus on the basis of its 16S rRNA sequence and biochemical properties. The isolated strain showed viability of over 85% at pH 3.0 and was resistant to bile salt. The strain produced lactic acid of 12.3 g/L by jar fermentation and maintained its viability in the presence of antibiotics at dosage related therapeutic effect, suggesting P. pentosaceus YB-55 may of ffr potential as a probiotics for livestock.

First Report of Pectobacterium aroidearum Causing Soft Rot on Ficus carica in Korea

  • In July 2021, symptoms of soft rot were observed on the stems of Ficus carica in Yeongam, Jeollanamdo, Korea. To accurately diagnose the cause, infected stem was collected and bacterial strain was isolated. Among these, the pathogenic strain KNUB-08-21 was identified as Pectobacterium aroidearum through 16S rRNA gene sequencing and phylogenetic analysis based on the concatenated sequences of the dnaX, leuS, and recA genes. The affiliation of the isolate with this bacterial species was also confirmed by its biochemical characteristics obtained using API ID 32 GN system. Artificial inoculation confirmed the strain's pathogenicity in figs, causing significant damage to both stems and fruits. To our knowledge, this is the first report of P. aroidearum causing soft rot disease in F. carica in Korea.

Identification and Characterization of Pseudomonas syringae pv. syringae, a Causative Bacterium of Apple Canker in Korea

  • In the present investigation, bacterial isolates from infected apple trees causing apple canker during winter were studied in the northern Gyeongbuk Province, Korea. The pathogen was identified as Pseudomonas syringae pv. syringae (Pss) through various physiological and biochemical characterization assays such as BIOLOG, gas chromatography of fatty acid methyl esters, and 16S rRNA. Bioassays for the production of phytotoxins were positive for syringopeptin and syringomycin against Bacillus megaterium and Geotrichum candidum, respectively. The polymerase chain reaction (PCR) method enabled the detection of toxin-producing genes, syrB1, and sypB in Pss. The differentiation of strains was performed using LOPAT and GATTa tests. Pss further exhibited ice nucleation activity (INA) at a temperature of -0.7℃, indicating an INA+ bacterium. The ice-nucleating temperature was -4.7℃ for a non-treated control (sterilized distilled water), whereas it was -9.6℃ for an INA- bacterium Escherichia coli TOP10. These methods detected pathogenic strains from apple orchards. Pss might exist in an apple tree during ice injury, and it secretes a toxin that makes leaves yellow and cause canker symptoms. Until now, Korea has not developed antibiotics targeting Pss. Therefore, it is necessary to develop effective disease control to combat Pss in apple orchards. Pathogenicity test on apple leaves and stems showed canker symptoms. The pathogenic bacterium was re-isolated from symptomatic plant tissue and confirmed as original isolates by 16S rRNA. Repetitive element sequence-based PCR and enterobacterial repetitive intergenic consensus PCR primers revealed different genetic profiles within P. syringae pathovars. High antibiotic susceptibility results showed the misreading of mRNA caused by streptomycin and oxytetracycline.

인천지역 대기 환경 중 배양성 세균의 특성 (Characterization of Culturable Bacteria in the Atmospheric Environment in Incheon, Korea)

  • Objectives: This study aims to provide basic data regarding the bacterial total plate count in the atmospheric environment for related studies. Methods: Total plate count and the identification of culturable bacteria in the atmospheric environment in Incheon took place in 2015 using periodic survey. Correlationship analysis was performed between the number of culturable bacteria and environmental elements. In addition, an estimation of novel bacterial species was undertaken using the similarities and phylogenetic tree based on the 16S rRNA gene. Results: The total plate count of culturable bacteria was on average $176CFU/m^3$, and did not exceed $610CFU/m^3$ in the atmospheric environment. Periodic monthly measuring of total plate count was highest in June at $293CFU/m^3$, while the lowest was in July at $125CFU/m^3$. Furthermore, as a result of the identification of culturable bacteria, the genera Arthrobacter and Kocuria were dominant, while novel bacterial taxa that belong to the genera Chryseobacterium and Herbiconiux were separated. Conclusion: The total number of culturable bacteria from the atmospheric environment in Korea is on average $176CFU/m^3$. In addition, the genera Arthrobacter and Kocuria dominate. The presence of novel bacterial taxa are expected in the atmospheric environment, such as belonging to the genera Chryseobacterium and Herbiconiux.

Norcardia 감염증에 의한 양식 가물치의 대량 폐사 (Mass Mortality Caused by Nocardial Infection in Cultured Snakehead, Channa arga in Korea)

  • 2005년 여름에 국내에서 사육되는 가물치에서 대량폐사를 수반하는 새로운 질병이 발견되었다. 감염 어류는 복부팽만과 항문 주위 출혈 이외의 특이한 외부 증상을 나타내지 않았으나 복부를 절개하면 많은 수의 백색 결절이 간, 비장 및 신장을 포함한 내부 장기에서 관찰되었다. 특히 내부 장기의 조직병리학적 관찰에서 granuloma가 관찰되었다. 이들 감염 어류에서 Gram 양성의 사상균이 분리되었다. 분리균은 Nocardia spp.와 non-Nocardia 균의 16S rRNA 염기 분석에 기초한 Nocardia 특이 primer를 사용한 PCR 방법으로 확인하였다. 이것은 어류에서 발생한 Nocardia 감염증의 국내 첫 사례이다.

Waterborne Pathogens Identification in Public Bathroom by PCR-Reverse Blot Hybridization Assay

  • A total of 30 water samples were collected from 30 different public baths in Seoul, Korea. Contamination of public bath water by waterborne pathogens can cause disease outbreaks and contribute to increase background rates of disease. Pathogens in water was filtered by nitrocellulose membrane with $0.45{\mu}m$ pore size. The membrane filters were analyzed by both cultivation and polymerase chain reaction (PCR) amplification of partial 16S rRNA gene. Various microorganisms including 4 Escherichia coli/Shigella spp. 1 Salmonella spp. 3 Pseudomonas aeruginosa and 2 Mycobacterium spp. were identified by reverse blot hybridization assay (REBA). PCR-REBA was able to identify many bacterial genera in one assay. Our results suggest that appropriate hygiene practice and continuous monitoring is needed for reducing health risk associated with public bath houses.
