Inhibition of MicroRNA-15a/16 Expression Alleviates Neuropathic Pain Development through Upregulation of G Protein-Coupled Receptor Kinase 2

  • There is accumulating evidence that microRNAs are emerging as pivotal regulators in the development and progression of neuropathic pain. MicroRNA-15a/16 (miR-15a/16) have been reported to play an important role in various diseases and inflammation response processes. However, whether miR-15a/16 participates in the regulation of neuroinflammation and neuropathic pain development remains unknown. In this study, we established a mouse model of neuropathic pain by chronic constriction injury (CCI) of the sciatic nerves. Our results showed that both miR-15a and miR-16 expression was significantly upregulated in the spinal cord of CCI rats. Downregulation of the expression of miR-15a and miR-16 by intrathecal injection of a specific inhibitor significantly attenuated the mechanical allodynia and thermal hyperalgesia of CCI rats. Furthermore, inhibition of miR-15a and miR-16 downregulated the expression of interleukin-$1{\beta}$ and tumor-necrosis factor-${\alpha}$ in the spinal cord of CCI rats. Bioinformatic analysis predicted that G protein-coupled receptor kinase 2 (GRK2), an important regulator in neuropathic pain and inflammation, was a potential target gene of miR-15a and miR-16. Inhibition of miR-15a and miR-16 markedly increased the expression of GRK2 while downregulating the activation of p38 mitogen-activated protein kinase and $NF-{\kappa}B$ in CCI rats. Notably, the silencing of GRK2 significantly reversed the inhibitory effects of miR-15a/16 inhibition in neuropathic pain. In conclusion, our results suggest that inhibition of miR-15a/16 expression alleviates neuropathic pain development by targeting GRK2. These findings provide novel insights into the molecular pathogenesis of neuropathic pain and suggest potential therapeutic targets for preventing neuropathic pain development.

  • The egg development and early life history of short barbel gudgeon Squalidus japonicus coreanus were investigated. For the experiments, the mature adults were collected at the stream Jicheon in Korea. The eggs from the females were obtain by injecting 10 IU/g of human chorionic gonadotropin and inseminated by wet method in the laboratory. The fertilized eggs were 1.12±0.03 mm (1.10~1.16 mm, n=30) in diameter. The embryo began to hatch about 49 hrs after fertilization under water temperature of 23±1℃. The newly-hatched larvae (Yolksac larva) were 3.7±0.1 mm (3.4~3.8 mm, n=16) in total length, and they haven't Melanophore. 5 days after hatching, the Preflexion larva were 5.3±0.2 mm (5.0~5.5 mm, n=16) in total length, and they began to eat a Rotifer. 19 days after hathing, the Flexion larva were 6.0±0.3 mm (5.4~6.5 mm, n=16) in total length, and they began to eat a Brine shrimp. 29 days after hatching, the Postflexion larva were 9.6±0.5 mm (8.3~10.5 mm, n=16) in total length, and dorsal fin rays are were formed. 44 days after hatching, the juvenile were 15.5±1.0 mm (13.5~17.0 mm, n=16) in total length, and all their fin-rays were formed.

Taxonomic characteristics of novel Flavobacteriumsp. B1 from a freshwater pond

  • The genus Flavobacterium, type genus of the family Flavobacteriaceae and a member of the phylum Bacteriodetes includes gram-negative and yellow-pigmented rods. Those bacteria have been isolated from various environments of the earth. A yellow-pigmented, gram-negative rod was isolated from a pond in the campus of the Changwon University, Changwon, Kyeongnam and designated as strain B1. Strain B1 was further analyzed physiologically, biochemically and phylogenetically, and concluded to be a member of genus Flavobacterium. BLAST search of the 16S rRNA gene sequence of strain B1 shows homology no higher than 99.0% with those sequences of other bacteria. The major fatty acids of strain B1 are iso-C15:0 (19.6%), summed feature 3(C16:1 ω7c and/or C16:1 ω6c, 16.1%), iso-C17:0 3OH(10.2%), iso-C15:0 3OH(8.4%) and iso-C15:1 G(6.6%) showing significant differences in fatty acid compositions between strain B1 and the other known Flavobacterium species. DNA sequence of 16S rRNA gene of strain B1 was deposited in genbank under accession number OP060681.

PRR16/Largen Induces Epithelial-Mesenchymal Transition through the Interaction with ABI2 Leading to the Activation of ABL1 Kinase

  • Advanced or metastatic breast cancer affects multiple organs and is a leading cause of cancer-related death. Cancer metastasis is associated with epithelial-mesenchymal metastasis (EMT). However, the specific signals that induce and regulate EMT in carcinoma cells remain unclear. PRR16/Largen is a cell size regulator that is independent of mTOR and Hippo signalling pathways. However, little is known about the role PRR16 plays in the EMT process. We found that the expression of PRR16 was increased in mesenchymal breast cancer cell lines. PRR16 overexpression induced EMT in MCF7 breast cancer cells and enhances migration and invasion. To determine how PRR16 induces EMT, the binding proteins for PRR16 were screened, revealing that PRR16 binds to Abl interactor 2 (ABI2). We then investigated whether ABI2 is involved in EMT. Gene silencing of ABI2 induces EMT, leading to enhanced migration and invasion. ABI2 is a gene that codes for a protein that interacts with ABL proto-oncogene 1 (ABL1) kinase. Therefore, we investigated whether the change in ABI2 expression affected the activation of ABL1 kinase. The knockdown of ABI2 and PRR16 overexpression increased the phosphorylation of Y412 in ABL1 kinase. Our results suggest that PRR16 may be involved in EMT by binding to ABI2 and interfering with its inhibition of ABL1 kinase. This indicates that ABL1 kinase inhibitors may be potential therapeutic agents for the treatment of PRR16-related breast cancer.

  • This paper has been studied the design of 16-QAM demodulator used in broadband wireless local loop subscriber station. In B-WLL systems, transmission signal experience the inter symbol interference(ISI) due to multipath, frequency offset of RF/IF local oscillator and phase offset. In this paper, we discuss the effective data recovery algorithm for 16-QAM demodulator to compensate the distorted signal from ISI, frequency offset and phase offset.

  • We have developed $16{\mu}m{\times}16{\mu}m$ digital micromirror array suitable for seamless-picture projection display system. This structure can improve the picture quality by making seamlesspicture image when combined with high-fill-factor microlens array to focus lights onto the mirror center. The fabricated micromirror shows excellent dynamic performances including the resonant frequency of 400 kHz.

국산 F-16 전투기 힘찬 날개짓

  • 근 10여년이 걸린 한국의 차세대전투기사업이 첫 결실을 맺었다. 지난 11월 7일 삼성항공과 공군은 사천 삼성항공항공기조립장에서 F-16전투기 생산기념식을 개최함으로써 그동안의 준비기간을 마무리하고 본격적인 양산체제에 돌입했다. 여기서는 이번 F-16전투기 생산기념식을 계기로 KFP사업을 종합 정리해보고 기술획득, 국산화실적, 획득기술 관리 및 활용실태, 앞으로 우리나라 항공기산업 발전을 위한 발전방향에 대해서 알아보고자 한다.

  • 지난 1월 17일 전북 고창의 종오리 농가에서 HPAI가 발생해 현재('14.04.16)까지 총 34건의 의심신고가 접수 되었으며 이중 28건이 양성판정을 받았다. 4월 16일 기준(잠정집계)으로 총 494농가에서 12,729,000수가 매몰(4월 14일 기준, 원종계(육용) 16천수, 종계 548천수, 육계 3,438천수, 산란계 5,044천수)되었다. 이와관련 금번 AI 발생현황('14.04.16기준)과 농가지침사항에 대해 요약 정리 하였다.
