Immunomodulating Activity of Fungal $\beta$-Glucan through Dectin-1 and Toll-like Receptor on Murine Macrophage

  • $\beta$-Glucan is a glucose polymer that has linkage of $\beta$-(1,3), -(1,4) and -(1,6). As exclusively found in fungal and bacterial cell wall, not in animal, $\beta$-glucans are recognized by innate immune system. Dendritic cells (DC) or macrophages possesses pattern recognition molecule (PRM) for binding $\beta$-glucan as pathogen-associated molecular pattern (PAMP). Recently $\beta$-glucan receptor was cloned from DC and named as dectin-l which belongs to type II C-type lectin family. Human dectin-1 is consisted of 7 exons and 6 introns. The polypeptide of dectin-1 has 247 amino acids and has cytoplasmic, transmembrane, stalk and carbohydrate recognition domains. Dectin-1 could recognize variety of beta-1,3 and/or beta-1,6 glucan linkages, but not alpha-glucans. In our macrophage cell line culture system, dectin-1 mRNA was detected in RA W264.7 cells by reverse transcription-polymerase chain reaction (RT-PCR). Dectin-1 was also detected in the murine organs of spleen, thymus, lung and intestines. Treatment of RA W264.7 cells with $\beta$-glucans of Ganoderma lucidum (GLG) resulted in increased expression of IL-6 and TNF-$\alpha$ in the presence of LPS. However, GLG alone did not increase IL-6 nor TNF-$\alpha$. These results suggest that receptor dectin-1 cooperate with CD14 to activate signal transduction that is very critical in immunoresponse.

Tissue Factor Inhibitory Flavonoids from the Fruits of Chaenomeles sinensis

  • Tissue factor (TF, tissue thromboplastin or coagulation factor III) accelerates the blood clotting, activating both the intrinsic and the extrinsic pathways to serve as a cofactor. In order to isolate TF inhibitors from the fruits of Chaenomeles sinensis, an activity-guided purification utilizing a bio-assay method of prothrombin time prolongation, was carried out to yield five active flavoniods such as hovetrichoside C (1) ($IC_{50}$ = 14.0 $\mu$g), luteolin-7-Ο-$\beta$-D-glucuronide (3) ($IC_{50}$ = 31.9$\mu$g), hyperin (4) ($IC_{50}$ = 20.8 $\mu$g), avicularin (6) ($IC_{50}$ = 54.8 $\mu$g) and quercitrin (10) ($IC_{50}$ = 135.7 $\mu$g), along with other inactive compounds such as ($\pm$)-(2E,4E)-Ο-$\beta$-D-glucopyranosyl-4'-hydroxy-$\beta$-ionylideneacetic acid ester (2), genistein-7-Ο-$\beta$-D-glucopyranoside (5), luteolin-3'-methoxy-4'-Ο-$\beta$-D-glucopyranoside (7), luteolin-7-Ο-$\beta$-D-glucuronide methyl ester (8), tricetin-3'-methoxy-4'-Ο-$\beta$-D-glucopyranoside (selagin-4'-Ο-$\beta$-D-glucopyranoside) (9), (-)-epicatechin (11), luteolin-4'-Ο-$\beta$-D-glucopyranoside (12) and apigenin-7-Ο-$\beta$-D-glucuronide methyl ester (13). The structures of the isolated compounds were elucidated through spectral analysis. Among them, compounds 1 to 9, 12 and 13 were isolated for the first time from the fruits of this plant and the compound 9 is a new flavonoid.

Exploration of β-Glucosidase Activity of Lactic Acid Bacteria Isolated from Kimchi (김치에서 분리된 젖산균의 β-glucosidase 활성 탐색)

  • The ${\beta}$-glucosidase (E.C. 3.2.1.21) production capabilities of lactic acid bacteria isolated from a variety of kimchi (fermented vegetables) were examined. When grown in a medium containing cellobiose as carbon source, most lactic acid bacteria showed significantly higher intracellular levels of ${\beta}$-glucosidase than the extracellular levels. A maximum intracellular ${\beta}$-glucosidase activity of 3.7${\pm}$0.5 (unit/mg protein) was obtained in the case of Weissella cibaria KFRI88010 isolated from kimchi. The optimum reaction conditions for W. cibaria KFRI88010 ${\beta}$-glucosidase activity were pH 5.0 and ${37^{\circ}C}$, and addition of divalent cations to the reaction mixture resulted in a notable decrease in enzyme activity. The ${\beta}$-glucosidase activity was enhanced twofold when W. cibaria KFRI88010 was grown in a medium containing fructose as compared with to a medium containing glucose or cellobiose.

Chemical Constituents from the Stems of Lagerstroemia indica and Their Anti-oxidant Effect (배롱나무의 항산화 활성 성분)

  • Phytochemical investigation of the 80% MeOH extract from the stems of Lagerstroemia indica resulted in the isolation of eighteen compounds; four norsesquiterpenes, fourteen phenolic derivatives. Their chemical structures were characterized by spectroscopic methods to be tachioside (1), isotachioside (2), 2,4,6-trimethoxyphenyl ${\beta}$-D-glucopyranoside (3), gallic acid 4-methyl ether (4), protocatechuic acid (5), gallic acid (6), vanillic acid (7), vanillin (8), 2-methoxy-5-hydroxymethyl-phenyl-1-O-(6"-galloyl)-${\beta}$-D-glucopyranoside (9), 2,4,6-trimethoxyphenol-1-O-${\beta}$-D-(6'-O-galloyl)-glucopyranoside (10), 4-hydroxy-3-methoxyphenyl-1-O-(6'-O-galloyl)-${\beta}$-D-glucopyranoside (11), vomifoliol (12), vomifoliol 9-O-${\beta}$-D-glucopyranoside (13), 6R,9R-3-oxo-${\alpha}$-ionol-9-O-${\beta}$-D-glucopyranoside (14), dihydrophaseic acid 4'-O-${\beta}$-D-glucopyranoside (15), ${\beta}$-hydroxypropiovanillone 3-O-${\beta}$-D-glucopyranoside (16), myrciaphenone A (17), and coumaric acid (18). Compounds 1-5 and 7-18 were isolated for the first time from this plant. Compounds 1-18 were investigated for their antioxidant properties using DPPH and ABTS radical scavenging capacity assay, $Fe^{2+}$ chelating, and FRAP assay. It was found that 4, 6, and 11 possessed the highest antioxidant capacities.

Changes of Volatile Compounds in the Pericarp of Chopi (Zanthoxylum piperitum DC.) During Maturation (초피 과피의 성숙정도에 따른 향기성분의 변화)

  • This study was conducted to estimate volatile compounds in pericarp of Zanthoxylum piperitum DC (Chopi). Chopi which harvested on lune 2, July 14 and September 11 in 2001 was dried at room temperature for one week. Fifty-two, 47, and 44 volatile compounds were analyzed with GC-MS in pericarp harvested on lune 2, July 14 and September 11, respectively. Eight terpenes including myrcene, ${\gamma}$-terpinene, $\alpha$-terpinolene, $\alpha$-phellandrene and $\beta$-caryophyllene were detected in pericarp harvested on tulle 2 and July 14, but not $\alpha$-phellandrene and $\beta$-caryophyllene in pericarp harvested on September 11. Thirteen alcohols or terpene alcohols including linalool L and citronellol were detected in pericarp harvested on lune 2, and added cis-linallol oxide and piperitol isomer in pericarp harvested on July 14 and September 11. Three aldehydes or terpene aldehydes were not affected by degree of maturation, but citronellal was increased in pericarp harvested on September 11. Five volatile compounds of ketones containing cryptone and piperitone were detected, and their concentration was changed during maturation. Six esters including lavandulyl acetate and $\alpha$-terpinenyl acetate were detected in pericarp harvested on lune 2, and [(E)-6,7-ephoxy-3,7-dimethyl-2-octenyl]ester of acetic acid was added in pericarp harvested on July 14 and September 11. Seven hydrocarbons including $\delta$-cadinene and neopentylidene cyclohexane were detected in pericarp harvested on June 2 and $\alpha$-muurolene was newly added in pericarp harvested on July 14 and September 11. We suggest that kinds and concentration of volatile compounds in pericarp were remarkably different from those in mature stage.

The Preparation of Crystalline ${\beta}$-1,4-Mannotriose from Poonac Using the Enzyme System and Yeast Fermentation

  • Beta-1,4-mannotriose was prepared by the enzymatic hydrolysis of poonac and the subsequent elimination with yeast of monosaccharides and disaccharide from the resultant hydrolysate. The enzyme system hydrolyzed poonac and produced monosaccharides, disaccharide and ${\beta}$-1,4-mannotriose without other oligomers at the final reaction stage. Poonac (50 g) was hydrolyzed at $50^{\circ}C$ and pH 6 for 48 hr with the crude enzyme solution (500 mL) from Trichoderma harzianum. The elimination of monosaccharides and disaccharide from the hydrolysis products with a yeast (Candida guilliermondii) produced 10.5 g of crystalline [${\beta}$-1,4-mannotriose without the use of chromatographic techniques. After 48 hr of yeast cultivation, the total sugar content fell from 4.8% to 3.4%, and the average degree of polymerization (D.P) rose from 2.5 to 3.2. The preparation method presented was confirmed to be suitable for the preparation of mannotriose from poonac.

Quality Control of Radiation Counting Systems and Measurement of Minimum Delectable Activity (방사선 계측기의 품질관리 및 최소검출방사능 측정)

  • Various radiation counters have been using to determine radioactivity of radwastes for disposal. A radiation counting system was set up using a radiation detector chosen in this study and its stability was investigated through the periodic determination of background and counting efficiencies in accordance with a quality control program to increase the confidence level. The average background level for the $\gamma$-spectrometer was 1.59 cps and the average counting level for the standard sample was 45248 Ops within $2{\sigma}$ confidence levels. The average alpha background level for the low background ${\alpha}{\beta}$ counting system was 0.31 cpm and the efficiency for alpha counting was 34.38%. The average beta background level for the ${\alpha}{\beta}$ counting system was 1,30 cpm and the efficiency for beta counting was 46.5%, The background level in the region of 3H and 14C for the liquid scintillation counting system was 2.52 and 3.31 cpm and the efficiency for alpha counting was 58.5 and 95.6%, respectively. The minimum detectable activity for the$\gamma$-spectrometer was found to be 3.2 Bq/$m\ell$ and 3.8 Bq/$m\ell$ for the liquid scintillation counter, and 20.5 and 23.0 Bq/$m\ell$, respectively for the $\alpha$ and $\beta$ counting system.

Di- and Sesqui-Terpenoids Isolated from the Pods of Sindora sumatrana and Their Potential to Inhibit Lipopolysaccharide-Induced Nitric Oxide Production

  • Activity-guided fractionation of the n-hexane and ${CHCl_3}-soluble$ fractions of Sindora sumatrana using a bioassay based on the inhibition of lipopolysaccharide (LPS)-induced nitric oxide (NO) production by inducible nitric oxide synthase (iNOS) in murine macrophage RAW 264.7 cells led to the isolation of the known compound, $(+)-7{\beta}-acetoxy-15,16-epoxy-3$, 13(16), 14-clero-datriene-18-oic acid (2) as an active constituent. In addition, a new trans-clerodane diterpenoid, (+)-2-oxokolavenic acid (1), together with six known compounds, (+)-3, 13-clerodadiene-16,15-olide-18-oic acid (3), $(+)-7{\beta}-acetoxy-3$,13-clerodadiene-16,15-olide-18-oic acid (4), $(+)-7{\beta}-acetoxy-16-hydroxy-3$,13-clerodadiene-16, 15-olide-18-oic acid (5), ${\beta}-caryophyllene$ oxide (6), $clovane-2{\beta},9{\beta}-diol (7),{\;}and{\;}caryolane-1,9{\beta}-diol$ (8) were isolated and found to be inactive. The structure of compound 1 was determined using physical and spectroscopic methods such as 1D and 2D-NMR experiments. The known compounds 2-8 were identified by the spectroscopic data and by comparison with the published values. Of eight isolates (1-8), only compound 2 exhibited an iNOS inhibitory activity with $IC_{50}$/ value of $51.6{\;}\mu\textrm{m}M$.

Studies on $\beta$-Glucuronidase Activities in Liver, Stomach and Small Intestinal Tissues of Rabbits Infected with Clonorchis sinensis (간디스토마 감염토끼의 소화기관에 대한 $\beta$-Glucuronidase의 활성치에 관한 연구)

  • The author has studied the $\beta$-glucuronidase activity in several tissues such as liver, stomach and small intestine of the male and female rabbits infected with different doses of metacercariae of Clonorchis sinensis. The metacercariae of Clonorchis sinensis were isolated from Pseudorasbora parva caught in Kim Hae by digestion technic. The experimental animals were sacrificed in the period of 1, 7, 14, 21, 28 and 35th days following the infection. The results obtained were summarized as follows. 1. In the groups infected with 100 metacercariae, $\beta$-glucuronidase activity was slightly increased during the entire periods than control rabbits. It was the highest in the first day with 1.535 and $1.421m{\mu}/g$, 14th days with 2.521 and $2.200m{\mu}/g$, and then lowered by the time, gradually. 2. In the groups infected with 500 metacercariae, $\beta$-glucuronidase activity was highly increased on the first day with 1.535 and $1.856m{\mu}/g$ than that 100 metacercariae groups according to each organs. It was the highest on the 7th day and 14th day. 3. In the groups infected with 1,000 metacercariae, $\beta$-glucuronidase activity was remarkably increased in the first and 14th days according to each organs, and then lowered gradually day by day. 4. $\beta$-glucuronidase activity of all organs was more increased than that of normal organs and the highest activity in the liver with $2.521m{\mu}/g$, intestine(1.612) and stomach (1.581) respectively. 5. $\beta$-glucuronidase activity of rabbits was higher in the female than in the male. On the basis of these results, it was suggested that $\beta$-glucuronidase activity was affected by the duration of infection and by the number of Clonorchis sinensis, according to the organs and sex of the rabbits.
