소랄렌의 광화학 반응에 대한 Ab initio 계산 (Ab initio Calculation for Photochemistry of Psoralens)

  • 소랄렌과 피리디민 염기와의 $C_4$-고리화 부가반응을 통한 가닥내 교차 결합(interstrand crosslinking) 에 의해 만들어진 소랄렌 복합체를 ab initio 방법인 HF와 DFT 6-31G 방법을 이용하여 연구하였다. 소랄렌과 피리디민 염기에 의해 만들어진 광생성물인 8-MOP< >Thy, Ps< >Cyt, Ps< >Thy, Ps< >Ps, Thy< >(3, 4)Ps(12, 13)< >Thy의 최적화된(optimized) 구조를 알 수 있으며, 8-MOP< >Thy은 (trans-syn) 구조, Ps(3, 4)< >Cyt은 (trans-anti) 구조, Ps(12, 13)< >Cyt은 (trans-anti) 구조, Ps(3, 4)< >Thy은 (trans-syn) 구조, Ps(12, 13)< >Thy은 (trans-syn) 구조가 가장 유리하다. Ps(3, 4)< >Thy과 Ps(12, 13)< >Thy의 Gibbs 자유 에너지 변화(${\Delta}{G^{\circ}}$)를 비교하면 Ps(12, 13)< >Thy의 광생성물(단일부가 생성물)이 이루어진 뒤에 Ps(3, 4)< >Thy의 광고리화 부가반응 생성물(이중부가 생성물)을 형성한다는 사실을 알 수 있다. Bispsoralen(psoralen dimer)에서는 Ps(12, 13)< >Ps(12, 13)(trans-anti) 구조가 가장 유리하며, Thy< >(3, 4)Ps(12, 13)< >Thy에서는 (cis syn)(cis anti) 형태가 가장 유리하다.

Up-regulation of Thy-1 Promotes Invasion and Metastasis of Hepatocarcinomas

  • Increasing evidence has revealed that thy-1 was a potential stem cell marker of liver cancer, but no data have been shown on how thy-1 regulates the pathophysiology of liver cancer, such as proliferation, apoptosis, invasion and migration. We previously demonstrated that thy-1 was expressed in about 1% of hepg2 cells, thy-1+hepg2 cells, but not thy-1-, demonstrating high tumorigenesis on inoculation $0.5{\times}10^5$ cells per BACA/LA mouse after 2 months. In the present study, our results showed that higher expression of thy-1 occurs in 72% (36/50 cases) of neoplastic hepatic tissues as compared to 40% (20/50 cases) of control tissues, and the expression of thy-1 is higher in poorly differentiated liver tumors than in the well-differentiated ones. In addition, thy-1 expression was detected in 85% of blood samples from liver cancer patients, but none in normal subjects or patients with cirrhosis or hepatitis. There was a significant negative correlation between thy-1expression and E-cadherin expression (a marker of invasion and migraton), but not between thy-1 expression and AFP expression in all the liver cancer and blood samples. We further investigated the relationship between thy-1 and E-cadherin in liver cancer hepg2 cell line which was transfected with pReceiver-M29/thy-1 eukaryotic expression vector followed by aspirin treatment. Lower expression of E-cadherin but higher expressions of thy-1 were detected in hepg2 cells transfected with pReceiver-M29/thy-1. Taken together, our study suggested that thy-1 probably regulates liver cancer invasion and migration.

폐흡충(Paragonimus westermani) 감염이 흰쥐의 IgE 생성 및 비장림프구 아군분포에 미치는 영향 (Changes of IgE production, splenic helper and suppressor T lymphocytes in mice infected with Paragonimus westermani)

  • 폐흡충 피낭유충을 BALB/c 마우스에 경구 감염시킨 후 마우스를 시기별(3일, 1주, 2주, 3주, 4주, 5주, 7주, 9주, 13주 및 23주)로 희생시켜 말초혈액내 호산구수와 혈청내 IgE 항체가를 측정하는 한편 비장 림프구를 분리하여 림프구 아군(helper T, cytotoxic / suppressor T, pan T)에 대한 단세포군 항체를 결합시켜 EPICS C fluorocytometer로 분석하였다. 폐흡충에 대한 혈청내 총 IgE 항체가는 감염 3주부터 증가되어 감염 4주에 높게 증가되었으며 13주 이후 23주까지 높은 항체가가 유지되었다. 말초혈액내 호산구수는 감염 2주 부터 증가되어 감염 9주에 $664.1/\textrm{mm}^3$으로 가장 증가 되었으며 그 이후에는 감소하여 23주에는 정상보다 약간 증가되었다. 폐흡충을 감염시키고 감염 3일, 1주, 2주, 3주, 4주, 7주 및 23주에 T 림프구 아군을 분석한 결과 L3T4 helper T 림프구와 Lyt-2 cytotoxic / suppressor T 림프구의 비율은 감염 군에서 감염 초기에 대조군에 비해 약간 감소된 경향을 보였으나 대조군과 유의한 차이를 보이지 않았다. Thy 1.2 pan T 림프구의 비율은 감염 2주와 4주에 대조군에 비해 유의하게 감소되었고(P<0.05) 7주 이후에는 대조군과 비슷하였다. 이러한 결과들로 보아 마우스의 폐흡충감염에서 특이 항원이나 마이토젠의 자극이 없이는 비장킬프구의 아군분포에 큰 변화가 없는 것으로 생각된다. 생각된다.

  • A rapid and accurate method is described for the determination of nucleo-bases in rumen micro-organisms. A procedure to satisfactorily hydrolyse the micro-organisms involving reaction with a mixture of readily volatile organic acids (acetic and formic acids) under high pressure, is proposed, and optimal conditions for an analytical procedure with reversed phase HPLC is described. The following nucleobases contents (mmol/kg DM) of rumen micro-organisms were found: Adenine (Ade), 82.62; Guanine (Gua), 61.34; Cytosine (Cyt), 84.61; Thymine (Thy), 35.74; Uracil (Ura), 68.62; Hypoxanthine (Hxn), 13.06; Xanthine (Xn), 8.35. Total purine-N content (g/kg N) of rumen micro-organisms were 99.60. The nucleic acid N content (g/kg N) of microbial isolates were: RNA-N, 109.9; DNA-N, 50.9.

Glycyrrhizin이 Mel/ret transgenic mice에서의 melanoma 세포의 apoptosis에 미치는 영향 (Effect of Glycyrrhizin on the Aoptosis of Melanoma Cells in Mel/ret Transgenic Mice)

  • The effect of glycyrrhizin on melanoma cells was investigated. DNA fragmentation in cultured melanoma cells was promoted by the addition of glycyrrhizin in a dose dependent manner. Administration(i.m.) of glycyrrhizin to Mel/ret transgenic mice resulted in apoptosis induction, reduction of mitochondrial transmembrane potential in melanoma cells. Decreased B220+ B cells were recovered by the treatment of glycyrrhizin in splenocytes and mesenteric lymph node cells, while Thy-1+ T cells were not influenced. Results suggested that glycyrrhizin acted as an inducer of apoptosis of melanoma cells and an immuno-potentiator via recovered B lymphocyte population in Mel/ret transgenic mice.

소음인(少陰人) 승양익기탕(升陽益氣湯)의 면역조절작용(免疫調節作用) (Immunoregulatory Action of Soeumin Seungyangikkitang)

  • The purpose of this research was to investigate the effects of Seungyangikkitang (SIT) on the immune cells in BALB/c mice. SIT (500mg/kg) was administerd p.o. once a day for 7 days. SIT enhanced the proliferation of thymocytes, but decreased the proliferation of splenocytes. SIT enhanced the subpopulation of cytotoxic T cells in thymocytes and helper T cells in splenocytes, but did not affect the subpopulation of B220/Thy1 cells. SIT enhanced the production of γ-interferon and interleukin-2 in thymocytes, splenocytes and serum, but did not affect the production of interleukin-4. SIT suppressed the production of nitric oxide, but enhanced the lucigenin chemiluminescence and the engulfment of FITC-conjugated E. coli particles in peritoneal macrophages. These results suggest that SIT has a potent activity on the specific immunity via the cytokine secretion of Th1 cells and the non-specific immunity via the phagocytic activity of macrophages in vivo.

Lipid N-formylation Occurs During Fixation with Formalin

  • Human tissues and organs can be preserved intact by fixation with formalin for the future analysis of biomolecules of interest. With the advances in high-throughput methods, numerous protocols have been developed and optimized to attain the most pathophysiological information out of biomolecules, including RNA and proteins, in formalin-fixed samples. However, there is no systematic study to examine the effects of formalin fixation on the lipidome of biological samples in a global fashion. In this study, we conducted a mass spectrometry-based analysis to survey the alteration in the lipidome of mice brains by fixation methods. A total of 308 lipids were quantitatively measured using triple quadrupole mass spectrometry. We found that most were unchanged after formalin fixation except for a few lipid classes such as phosphatidylethanolamine.

Mycobacterium tuberculosis Derived Epitope Peptide Specific CD8+T Cell Responses in Tuberculous Pleurisy

  • Cell-mediated immune response (CMI) is a major immune protective mechanism against tuberculosis (TB) infection. Among several components involved in CMI, recent studies suggest that CD8+ T cells are important in controlling TB infection. In our previous report, we defined four Mycobacterium tuberculosis (MTB) derived epiotpe peptides specific for HLA-A*0201-restricted CD8+ T cells. These four peptides are $PstAl_{75-83}$, $ThyA_{30-38}$, $RpoB_{127-135}$ and $85B_{15-23}$. In this study, these epitope peptides specific CD8+ T cell responses in tuberculous pleurisy were investigated using ex vivo $IFN-\gamma$ elispot assay and intracellular $IFN-\gamma$ staining method. As a result, we observed these epitope peptide specific CD8+ T cell responses are induced in all three patients with tuberculous pleurisy suggesting that CD8+ T cells are involved in protective immune mechanism against MTB infection in tuberculous pleurisy. However, the CMI to mitogens and MTB antigens from pleural fluids of patients with tuberculous pleurisy does not seem to correlate with that from peripheral blood, although the sample size is too small to make any conclusion. In sum, the MHC I restricted CD8+ T cell responses seem to be induced efficiently in the pleural fluids, at the site of TB infection, in which the CMI is actively induced. In addition, these experiments suggest that MHC I restricted CD8+ T cell mediated immune responses are also involved in protective mechanism against MTB infection in extra-pulmonary TB.

코레옵시스(Coreopsis spp.)와 석죽(Dianthus spp.)의 삽목시 마디수, 적심 횟수 및 식물생장억제제 처리가 생장에 미치는 영향 (Effect of Node size, Pinching and Growth Retardants on Growth of Coreopsis spp. and Dianthus spp.)

  • 본 연구는 Dianthus spp. 및 Coreopsis spp.을 분화용으로 개발하기 위하여 줄기 마디수, 적심 횟수 및 왜화제 처리가 생장에 미치는 영향을 조사하였다. Coreopsis spp. 삽목시 마디 수와 육묘일수에 관한 실험 결과 정식 후 한 달 이후에는 도장의 우려가 있어 육묘 기간은 4주를 넘기지 않는 것이 좋다고 판단되었다. 또한, Coreopsis spp. 분화용으로는 2마디 처리에서 초장, 분지수 등의 및 관상가치가 적당하고 분화 상품으로서의 가치가 가장 좋았다. 적심횟수는 Coreopsis spp.는 2회 적심하는 것이 초장 신장 억제 및 측지 분화에 효과적인 것으로 나타났다. Coreopsis spp.의 식물생장억제제 처리결과에 있어서, B-9 처리는 식물 생육이나 분화 및 꽃수 형성에 효과적인 것으로 관찰되었으며, 특히 4,000 ppm 2회 처리한 것이 가장 좋았다. 반면에 CCC 처리는 큰 차이를 보이지 않아 Coreopsis spp.의 분화 상품 생산에는 B-9이 효과적인 것으로 판단되었다. Dianthus spp.는 적심 및 식물생장억제제를 처리하지 않아도 생장이 양호하며 초장신장이 적당하여 분화용으로 적합한 품종으로 판단되었다. 그러나 B-9 4,000ppm 2회 처리한 것이 엽수가 월등히 증가하였다.
