Investigation on Lead-Borosilicate Glass Based Dielectrics for LTCC (Lead-Borosilicate Glass계 LTCC용 유전체에 대한 고찰)

  • The effects of lead-borosilicate glass frits on the sintering behavior and microwave dielectric properties of ceramic-glass composites were investigated as functions of glass composition of glass addition ($10{\sim}50vol%$), softening point (Ts) of the glass, and sintering temperature of the composites ($500{\sim}900^{\circ}C$ for 2 h). The addition of 50 vol% glass ensured successful sintering below $900^{\circ}C$. Sintering characteristics of the composites were well described in terms of Ts. PbO addition in to the glass enhanced the reaction with $Al_{2}O_3$ to form liquid phase and $PbAl_{2}Si_{2}O_8$, which was responsible to lower Ts. Dielectric constant(${\epsilon}_r$), $Q{\times}f_0$ and temperature coefficient of resonant frequency (${\tau}_f$) of the composite with 50 vol% glass contents ($B_{2}O_{3}:PbO:SiO_{2}:CaO:Al_{2}O_3$ = 5:40:45:5:5) demonstrated 8.5, 6,000 GHz, $-70\;ppm/^{\circ}C$, respectively, which is applicable to substrate requiring a low dielectric constant. When the same glass composition was applied sinter $MgTiO_3\;and\;TiO_2,\;at\;900^{\circ}C$ (50 vol% glass in total), the properties were 23.8, 4,000 GHz, $-65ppm/^{\circ}C$ and 31.1, 2,500 GHz, $+80ppm/^{\circ}C$ respectively, which is applicable to filter requiring an intermidiate dielectric constant.

QTL Identification Using Combined Linkage and Linkage Disequilibrium Mapping for Milk Production Traits on BTA6 in Chinese Holstein Population

  • Milk production traits are important economic traits for dairy cattle. The aim of the present study was to refine the position of previously detected quantitative trait loci (QTL) on bovine chromosome 6 affecting milk production traits in Chinese Holstein dairy cattle. A daughter design with 918 daughters from 8 elite sire families and 14 markers spanning the previously identified QTL region were used in the analysis. We employed a combined linkage and linkage disequilibrium analysis (LDLA) approach with two options for calculating the IBD probabilities, one was based on haplotypes of all 14 markers (named Method 1) and the other based on haplotypes with sliding windows of 5 markers (named Method 2). For milk fat yield, the two methods revealed a highly significant QTL located within a 6.5 cM interval (Method 1) and a 4.0 cM interval (Method 2), respectively. For milk protein yield, a highly significant QTL was detected within a 3.0 cM interval (Method 1) or a 2.5 cM interval (Method 2). These results confirmed the findings of our previous study and other studies, and greatly narrowed down the QTL positions.

Metabolism of Rutin and Poncirin by Human Intestinal Microbiota and Cloning of Their Metabolizing α-L-Rhamnosidase from Bifidobacterium dentium

  • To understand the metabolism of flavonoid rhamnoglycosides by human intestinal microbiota, we measured the metabolic activity of rutin and poncirin (distributed in many functional foods and herbal medicine) by 100 human stool specimens. The average α-L-rhamnosidase activities on the p-nitrophenyl-α-L-rhamnopyranoside, rutin, and poncirin subtrates were 0.10 ± 0.07, 0.25 ± 0.08, and 0.15 ± 0.09 pmol/min/mg, respectively. To investigate the enzymatic properties, α-L-rhamnosidase-producing bacteria were isolated from the specimens, and the α-L-rhamnosidase gene was cloned from a selected organism, Bifidobacterium dentium, and expressed in E. coli. The cloned α-L-rhamnosidase gene contained a 2,673 bp sequcence encoding 890 amino acid residues. The cloned gene was expressed using the pET 26b(+) vector in E. coli BL21, and the expressed enzyme was purified using Ni2+-NTA and Q-HP column chromatography. The specific activity of the purified α-L-rhamnosidase was 23.3 µmol/min/mg. Of the tested natural product constituents, the cloned α-L-rhamnosidase hydrolyzed rutin most potently, followed by poncirin, naringin, and ginsenoside Re. However, it was unable to hydrolyze quercitrin. This is the first report describing the cloning, expression, and characterization of α-L-rhamnosidase, a flavonoid rhamnoglycosidemetabolizing enzyme, from bifidobacteria. Based on these findings, the α-L-rhamnosidase of intestinal bacteria such as B. dentium seem to be more effective in hydrolyzing (1 →6) bonds than (1 →2) bonds of rhamnoglycosides, and may play an important role in the metabolism and pharmacological effect of rhamnoglycosides.

Infrared and visible image fusion based on Laplacian pyramid and generative adversarial network

  • An image with infrared features and visible details is obtained by processing infrared and visible images. In this paper, a fusion method based on Laplacian pyramid and generative adversarial network is proposed to obtain high quality fusion images, termed as Laplacian-GAN. Firstly, the base and detail layers are obtained by decomposing the source images. Secondly, we utilize the Laplacian pyramid-based method to fuse these base layers to obtain more information of the base layer. Thirdly, the detail part is fused by a generative adversarial network. In addition, generative adversarial network avoids the manual design complicated fusion rules. Finally, the fused base layer and fused detail layer are reconstructed to obtain the fused image. Experimental results demonstrate that the proposed method can obtain state-of-the-art fusion performance in both visual quality and objective assessment. In terms of visual observation, the fusion image obtained by Laplacian-GAN algorithm in this paper is clearer in detail. At the same time, in the six metrics of MI, AG, EI, MS_SSIM, Qabf and SCD, the algorithm presented in this paper has improved by 0.62%, 7.10%, 14.53%, 12.18%, 34.33% and 12.23%, respectively, compared with the best of the other three algorithms.

Studies of the Properties of Commercial Woods Grown in the Southern Part of Korea (한국산(韓國産) 유용목재(有用木材)의 기초재질(基礎材質)에 관(關)한 연구(硏究))

  • Five species, Abies koreana Wilson (A. koreana), Castanopsis cuspidata var. Sieboldii Nakai (C. Cuspidata). Machilus thunbergii Sieb. et Zucc. (M. thunbergii), Styrax japonica (S. japonica), and Quercus acuta Thunberg(Q. acuta) growing in the southern part of Korea were selected for the investigation of wood properties. In order to evaluate the wood properties of these five species, anatomical, physical, mechanical, chemical and pulping characteristics were investigated. And this study also covered wood technological problems related to the drying, gluing, debarking, flooring, and wood workability so that these species might serve to the best advantage. The results obtained were summarized as follows: 1. The trunk of A. koreana with many knots was straight. However, the trunks of S. japonica and C. cuspidata were crooked. 2. A. koreana showed the longest and the widest ill the fiber morphology; 2.97mm in length, 39.3${\mu}$ in width. In general, fiber width of all the species investigated were greater than those of other Korean hardwoods. 3. The specific gravity of Q. acuta was 0.74${\pm}$0.03, and that of A.koreana was 0.34${\pm}$0.02. The range of specific gravity of the other species was 0.47-0.52. 4. The adsorption of water was propotioned inversely with the specific gravity, but the adsorption of humidity was proportioned with the specific gravity. In spite of their medium density, S. japonica showed the greatest adsorption, and M. thunbergii the least. The water adsorption of cross section was twice greater than that of lateral direction, and there was a slight difference in between the radial and the tangential direction. 5. Shrinkage for tested five species was ranged from 5.36 to 10.24% in tangential direction, and 2.83~6.13% in radial direction. Q. acuta recorded the greatest shrinkage rate, and A. koreana the least. The greater was the specific gravity, the larger was the shrinkage rate. 6. The mechanical properties of Q. acuta were similar to those of Quercus mongolica which grow in Kangwon-Do. Strength properties of C. cuspidata, M. thunbergii, A. koreana were equivalent to those of other Korean commercial woods with similar specific gravity, except S. japonica which showed slightly higher strength than that of other species with similar density. 7. Higher glue joint strength for urea and phenol adhesieves was recorded in the species of M. thunbergii and C. cuspidata, however, high-density species(Q. acuta) and even low-density species(A. koreana) did not show good joint strength. 8. The attractive figure of M. thunbergii in texture seemed to he appreciated for decoration. And the grain and texture of other species were proper for furniture and building materials. 9. All of the species except Q. acuta were considered good for wood workability. 10. The denser the specific gravity was, the longer the drying time took. However, severe drying defects were formed in M. thunbergii whose density was medium. 11. All the species were considered suitable for the flooring wood expect A. koreana whose density was light. 12. Pentosan component in all the species was great, and the amount of extractives in Q. acuta was worth noticing. 13. Yield in kraft pulp was above the level of economic pulp yield, i.e. 45% in all species. 14. Debarking was easy in the species of A. koreana and M. thunbergii, and debarking after being boiled in water was the most efficient in all species.

Expression of Human SOD1 and Mutant SOD1 (G93A) in E. coli and Identification of SOD1 as a Substrate of HtrA2 Serine Protease (대장균에서의 human SOD1과 mutant SOD1 (G93A) 단백질의 발현과 HtrA2의 기질 여부 확인에 관한 연구)

  • Superoxide dismutase (SOD) is physiologically important in regulating cellular homeostasis and apoptotic cell death, and its mutations are the cause of familial amyotrophic lateral sclerosis (FALS). Mitochondrial serine protease HtrA2 has a pro-apoptotic function and has known to be associated with neurodegenerative disorders. To investigate the relationship between genes associated with apoptotic cell death, such as HtrA2 and SOD1, we utilized the pGEX expression system to develop a simple and rapid method for purifying wild-type and ALS-associated mutant SOD1 proteins in a suitable form for biochemical studies. We purified SOD1 and SOD1 (G93A) proteins to approximately 90% purity with relatively high yields (3 mg per liter of culture). Consistent with the result in mammalian cells, SOD1 (G93A) was more insoluble than wild-type SOD1 in E. coli, indicating that research on the aggregate formation of SOD1 may be possible using this pGEX expression system in E. coli. We investigated the HtrA2 serine protease activity on SOD1 to assess the relationship between two proteins. Not only wild-type SOD1 but also ALS-associated mutant SOD1 (G93A) were cleaved by HtrA2, resulting in the production of the 19 kDa and 21 kDa fragments that were specific for anti-SOD1 antibody. Using protein gel electrophoresis and immunoblot assay, we compared the relative molecular masses of thrombin-cleaved GST-SOD1 and HtrA2-cleaved SOD1 fragments and can predict that the HtrA2-cleavage sites within SOD1 are the peptide bonds between leucine 9-lysine 10 (L9-K10) and glutamine 23-lysine 24 (Q23-K24). Our study indicates that SOD1 is one of the substrate for HtrA2, suggesting that both HtrA2 and SOD1 may be important for modulating the HtrA2-SOD1-mediated apopotic cell death that is associated with the pathogenesis of neurodegenerative disorder.

Effect of Temperature on Growth and Related Gene Expression in Alternative Type Wheat Cultivars (양절형 밀 생장에 대한 온도의 영향과 유전자 발현 양상)

  • We have investigated the effects of ambient temperature on the growth of wheat in Korea. The differences in the growth phase of wheat were compared according to the temperature treatment. The productive tiller number and dry weight were decreased in a plot under a higher temperature treatment. We found that the growth of Jinpum was different from that of the alternative wheat cultivars, which were bred in Korea, at 50 days after treatment. While the Jinpum wheat grown at 17℃ showed vegetative stage growth, that grown in the 23℃ growth chamber entered the heading and flowering stage. The differences in the expression of 16 genes known to be involved in high-temperature responses were checked by using Jinpum wheat 50 days after two temperature treatments (17℃ and 23℃), which showed apparent differences in expression between the higher and lower temperatures during the growth phase. In the 23℃ treatment samples, the genes with increased expression were HSP70, HSP101, VRN2, ERF1, TAA1, YUCCA2, GolS, MYB73, and Histone H2A, while the genes with decreased expression were VRN-A1, DREB2A, HsfA3, PIF4, PhyB, HSP17.6CII, rbcL, and MYB73. YUCCA2, HSP101, ERF1, and VRN-A1 showed a significant difference in gene expression between lower- and higher-temperature conditions. Overall, combining the means of the expression of various genes involved in thermosensing, vernalization, and abiotic stresses, it is possible to conclude that different sets of genes are involved in vernalization and summer depression of wheat under long term, high ambient temperature conditions.

Antioxidant Properties and Ubiquinone Contents in Different Parts of Several Commercial Mushrooms (시판버섯의 부위별 항산화능과 유비퀴논 함량)

  • Antioxidant properties and antioxidant compound contents in different parts of 14 commercial mushrooms were evaluated. Methanolic extracts from the entire mushroom, the pileus and the stipe, separately, were screened for their DPPH and ABTS free radical scavenging activities. Total ubiquinones (Coenzyme Qs; CoQs), total phenolic, and flavonoid contents were determined, in order to assess the extract's antioxidant activity. The portion of the mushroom selected had an effect on the results, with pileus methanolic extract exhibiting the greatest antioxidant effect (p<0.05). The analyzed mushrooms contained powerful antioxidants such as phenols (144.5~536.6 mg of gallic acid equivalents, mg GAE/100 g of dried weight, dw), flavonoids (3.7~31.2 mg of quercetin equivalents, mg QE/100 g dw) and ubiquinones (65.6~485.1 ${\mu}g$/100 g dw). Content of CoQ9 and CoQ10 in the 14 commercial mushrooms varied from 23.1 to 256.2 ${\mu}g$/100 g and from 16.1 to 238.3 ${\mu}g$/100 g, respectively. Phellinus linteus showed the highest antioxidant activity among all species due to the contribution of antioxidants such as phenols (530.5 mg GAE/100 g dw) and ubiquinones (308.8 ug/100 g dw). A positive linear correlation was demonstrated between free radical scavenging activity and total phenolic ($R^2=0.79$) and ubiquinone ($R^2=0.59$) contents in the pileus of mushrooms (p<0.05). Our data indicate that commercial mushrooms have potential as dietary sources of CoQs and phenolic antioxidants.

Estimation of Soil CO2 Efflux from an Apple Orchard (사과 과수원에서의 토양 CO2 발생량 평가)

  • This study was conducted to quantify the soil respiratory $CO_2$ emission (SR) in an apple orchard and to determine its relationship with key environmental factors such as air temperature, soil temperature and soil moisture content. Experiment was made over the period from 23 April 2007 to 31 March 2008 in 'Fuji' apple orchard of National Institute of Horticultural and Herbal Science in Suwon, Gyeonggi-do, Korea. The SR was measured by using the automatic opening/closing chamber system based on a closed method. Diurnal variations in SR showed an increase around 0700 hours with increasing soil temperature, its peak between 1400 and 1500 hours, and then a gradual decrease thereafter. Daily variations in SR depended largely on soil and air temperatures over the year, ranging from 0.8 to 13.7 g $CO_2$ $m^{-2}d^{-1}$. During the rainy spell in summer (July$\sim$Autumn) with higher temperature and more precipitation, the SR was lower than that in the spring (May$\sim$June) with moderate temperature. The SR showed a significant exponential relationship with soil temperature ($r^2=0.800$) and air temperature ($r^2=0.805$), but not with soil moisture content ($r^2=0.160$). The $Q_{10}$ values of SR with annual soil temperature and air temperature were 2.0 and 1.9, respectively. The annually integrated SR was 19.6 ton $CO_2$ $ha^{-1}$.

Effect of Resistance Training on Skeletal Muscle Gene Expression in Rats: a Beadarray Analysis (저항성 운동이 골격근 유전자 발현에 미치는 영향: Beadarray 분석)

  • The aim was to examine resistance exercise-related genes after 8 weeks of resistance training. Thirty-two male Sprague-Dawley rats were divided into four groups: 4 weeks sedentary (4 wks CON, n=8), 8 weeks sedentary (8 wks CON, n=8), 4 weeks exercise training (4 wks REG, n=8), and 8 weeks exercise training (8 wks REG, n=8). The rats were trained to climb a 1-m vertical incline (85-degree), with weights secured to their tails. They climbed 10 times, 3 days per week, for 8 consecutive weeks. Skeletal muscle was taken from the flexor halucis longus after the exercise training. After separating the total RNA, large-scale gene expression was investigated by beadarray (Illumina RatRef-12 Expression BeadChip) analysis, and qPCR was used to inspect the beadarray data and to analyze the RNA quantitatively. The detection p-value for the genes was p<0.01, the M-value {M=$log_2$(condition)-$log_2$(reference)} was >1.0, and the DiffScore was >20. In total, the expression of 30 genes significantly increased 4 weeks after the exercise training, and the expression of six genes decreased. At 8 weeks, the expression of five genes significantly increased and that of 12 decreased. Several genes are potentially involved in resistance exercise and muscle hypertrophy, including 1) regulation of cell growth (IGFBP1, PLA2G2A, OKL38); 2) myogenesis (CSRP3); 3) tissue regeneration and muscle development (MUSTN1, MYBPH); 4) hypertrophy (CYR61, ATF3, NR4A3); and 5) glucose metabolism (G6PC, PCK1). These results may help to explain previously reported physiological changes of the skeletal muscle and suggest new avenues for further investigation.
