Anti-oxidant, Anti-coagulation, and Anti-platelet Aggregation Activities of Black Currant (Ribes nigrum L.) (블랙커런트의 항산화, 항응고 및 혈소판 응집저해 활성)

  • The black currant (Ribes nigrum L.) is belong to the Grossulariaceae family, and has piquant berries, which can be eaten as raw or as processed foods, such as jams, jelly, juice and syrups. In this study, the fresh juice of black currant (FJBC) from Austria and its subsequent organic solvent fractions, such as hexane fraction, ethylacetate (EA) fraction, butanol fraction and water residue, were prepared and their in-vitro anti-oxidant, anti-coagulation and anti-platelet aggregation activities were evaluated. The FJBC and EA fraction, which has concentrated polyphenol and flavonoid, showed strong radical scavenging activities and reducing power. The $RC_{50}s$ of EA fraction against DPPH anion, ABTS cation, nitrite were 136.3, 66.2 and $115.5{\mu}g/ml$, respectively, those are 1/10, 1/16, and 1/7.7 of $RC_{50}s$ of vitamin C. In anti-coagulation assay, the FJBC, EA and butanol fraction showed significant inhibitory activities against thrombin, prothrombin and coagulation factors. Furthermore, the anti-platelet aggregation activities of EA and butanol fraction were the stronger than that of aspirin. The concentrations required for 50% platelet aggregation inhibition of aspirin, EA and butanol fraction were 0.395, 0.192 and 0.261 mg/ml, respectively. The EA and butanol fraction have no hemolysis activities up to 0.5 mg/ml against human red blood cells. The results suggest that the FJBC and its EA and butanol fraction have high potentials as novel anti-thrombosis agents. This report provides the first evidence of anti-thrombosis activity of black currant.

Extractive Butanol Fermentation Using Pervaporation and a Low Acid Producing Strain (투과증발과 유기산 저생성 균주를 이용한 부탄올 추출발효)

  • An extractive fermentation process using pervaporation was studied in a 7 liter fermentor. Pervaporation was performed using a silicone membrane module and a low-acid-producing strain Clostridium acetobutylicu, B18 was used to produce butanol. In batch culture without pervaporation pH 5.5 and initial glucose concentration of 60 g/L resulted in the highest butanol productivity (0.216 g/L$.$h) with butanol yield of 0.261 Butanol flux through the membrane was best at 2.0 L/min-tubing of air flow rate In batch and fed-batch fermentation glucose consumption rate increased by 1.3 times with pervaporation.

Effect of Butanol Extracts from Prunus mume on the Growth of Salmonella typhimurium (오매의 Butanol추출물이 Salmonella typhimurium의 생육에 미치는 영향)

  • The antimicrobial properties of extracts of Prunus mume were tested on Salmonella paratyphimurium and S. typhimurium. First, the Prumus mume was extracted with methanol at several temperatures, and then fractionation of the methanol extracts from Prunus mume was carried out by using petroleum ether, chloroform, ethyl acetate or butanol. Secondly, absorption column chromatoraphy (using a Diaion HP 20) was conducted to eliminate some water soluble materials that might inhibit the antimicrobial activity of some extracts. The antimicrobial activitry of each of the Prunus mume extracts was determined using a paper disc method against several food-borne pathogens, The growth inhibition curve was determine using butanol extracts of Prunus mume against Salmonella typhimurium. The extraction temperature did not have any significant effect on the yield of the extract or on the level of antimicrobial activity. The butanol extract of Prunus mume showed strong antimicrobial activity against Salmonella paratyphimurium and S. typhimurium; a 1,000 ppm of butanol extract of Prunus mume retared the growth of S. typhimurium up to 36 hours. Impacts of Various Solvent Extracts from Wild Haw(Crataegus pinnatifida Bunge) Pulpy on the Antioxidative Activities (야생산사(Crataegus pinnatifida Bunge)과육 용매별 추출물의 항산화 효과)

  • Haw(Crataegus pinnatifida Bunge) belongs to the rose family and has beneficial health effects such as anti-obesity and gastroprotective activities. This study compared the antioxidative activities of various solvent extracts from haw. After removing seeds, pulpy was extracted with 70% methanol, 70% ethanol, chloroform :methanol(CM, 2:1, v/v), n-butanol, and ethyl acetate(EA). Total phenol contents were 71.46, 70.41, 47.63, 47.49 and 45.95 mg/g in the 70% methanol, 70% ethanol, CM, n-butanol and EA extracts, respectively. Total flavonoid contents of CM and n-butanol extracts were higher compared to other extracts at 1.63 mg/g and 1.59 mg/g, respectively, whereas EA extract had the lowest flavonoid content at 1.12 mg/g. Corresponding to total phenol contents, among $NO_2$ radical scavenging activity, antioxidant activity by ${\beta}$-carotene bleaching assay, and superoxide dismutase(SOD)-like ability, 70% methanol and 70% ethanol extracts showed higher antioxidative activities than the other extracts. However, the lowest activities were n-butanol extract in $NO_2$ radical scavenging activity as well as EA extract in antioxidant activity by ${\beta}$-carotene bleaching assay and SOD-like ability. These results suggest that haw extracts are available as a new natural antioxidant, and its activities are attributed to antioxidant substances such as phenolic compounds and flavonoids.

Pervaporation of Butanol from their Aqueous Solution using a PDMS-Zeolite Composite Membrane (PDMS-Zeolite 복합막을 이용한 부탄올 투과증발)

  • Pervaporation is known to be a low energy consumption process since it needs only an electric power to maintain the permeate side in vacuum. Also, the pervaporation is an environmentally clean technology because it does not use the third material such as an entrainer for either an azeotropic distillation or an extractive distillation. In this study, Silicalite-1 particles are hydrothermally synthesized and polydimethylsiloxane(PDMS)-zeolite composite membranes are prepared with a mixture of synthesized Silicalite-1 particles and PDMS-polymer. They are used to separate n-butanol from its aqueous solution. Pervaporation characteristics such as a permeation flux and a separation factor are investigated as a function of the feed concentration and the weight % of Silicalite-1 particles in the membrane. A 1,000 $cm^3$ aqueous solution containing butanol of low mole fraction such as order of 0.001 was used as a feed to the membrane cell while the pressure of the permeation side was kept about 0.2~0.3 torr. When the butanol concentration in the feed solution was 0.015 mole fraction, the flux of n-butanol significantly increased from 14.5 g/ $m^2$/hr to 186.3 g/$m^2$/hr as the Silicalite-1 content increased from 0 wt% to 10 wt%, indicating that the Silicalite-1 molecular sieve improved the membrane permselectivity from 4.8 to 11.8 due to its unique crystalline microporous structure and its strong hydrophobicity. Consequently, the concentration of n-butanol in the permeate substantially increased from 0.07 to 0.15 mole fraction. This composite membrane could be potentially appliable for separation of n-butanol from insitu fermentation broth where n-butanol is produced at a fairly low concentration of 0.015 mole fraction.

The Effect of Ginseng Butanol Fraction on the Acetaldehyde Metabolism in Mice (마우스의 아세트알데히드 대사에 미치는 인삼 부탄올 분획의 영향)

  • The present study was undertaken to investigate the possible effect of ginseng butanol fraction on the hepatic acetaldehyde metabolism. Experimental animals were used for the subject of the study. When, in case of mitochondrial aldehyde dehydrogenase (Ald DH), ginseng butanol fraction was added, enzyme activity was increased in a small dose, while, in a large dose, it showed inhibitory effect. In terms of kinetic aspect, ginseng butanol fraction has the effect to decrease the Km values of Ald DH. In vivo studies, the activity of Aid DH increased by induction of acute intoxication of ethanol was further increased through pretreatment with ginseng butanol fraction. When ginseng butanol fraction was given to mice fed with 5% ethanol instead of water for 60 days, the activity of Ald DH in mitochondrial fraction decreased to about 35% in chronic alcoholism, but after pretreatment of ginseng butanol fraction the activity was restored to the control level. By the pretreatment with disulfiram, the Ald DH activity was inhibited in normal and alcohol-treated groups, but after the treatment with ginseng butanol fraction the activity was restored to the control level. The results suggest that ginseng butanol fraction enhance the Ald DH activity inhibited by the treatment of disulfiram with no relation to NAD. It was observed that ginseng butanol fraction markedly decrease the acetaldehyde levels in plasma and liver. All these observations suggested that reduction of acetaldehyde in blood and liver should be dependent upon increased activity of mitochonclrial Ald DH. It is concluded that the recovery from alcohol intoxication should be prompted by treatment with ginseng.

  • Polyisocyanurate has been obtained from toluene diisocyanate(TDI) in the presence of potassium octoate catalyst and the effects of water and butanol in solvent were studied for the determination of the change of viscosity, the reaction time (length of time required for 5 poise of viscosity of product), molecular dispersity of product, and TDI conversion. When butyl acetate contains 0.1% of water by weight, uretidione was formed and a higher conversion was obtained at the condition. The uretidione was not formed by adding butanol to the solvent. At a higher concentration of butanol, a higher TDI conversion and a wider molecular dispersity were obtained.

Flash Point Measurement of n-Propanol+n-Hexanol and n-Butanol+n-Hexanol Systems Using Seta Flash Closed Cup Tester (Seta Flash 밀폐식 장치를 이용한 n-Propanol+n-Hexanol계와 n-Butanol+n-Hexanol계의 인화점 측정)

  • Flash point is the important indicator to determine fire and explosion hazards of liquid solutions. In this study, flash points of n-propanol+n-hexanol and n-butanol+n-hexanol systems were obtained by Seta flash tester. The methods based on UNIFAC equation and multiple regression analysis were used to calculate flash point. The calculated flash point was compared with the experimental flash point. Absolute average errors of flash points calculated by UNIFAC equation are $2.9^{\circ}C$ and $0.6^{\circ}C$ for n-propanol+n-hexanol and n-butanol+n-hexanol, respectively. Absolute average errors of flash points calculated by multiple regression analysis are $0.5^{\circ}C$ and $0.2^{\circ}C$ for n-propanol+ n-hexanol and n-butanol+n-hexanol, respectively. As can be seen from AAE, the values calculated by multiple regression analysis are noticed to be better than the values by the method based on UNIFAC eauation.

Fermentation of Rice Bran and Defatted Rice Bran for Butanol Production Using Clostridium beijerinckii NCIMB 8052

  • We examined butanol fermentation by Clostridium beijerinckii NCIMB 8052 using various hydrolyzates obtained from rice bran, which is one of the most abundant agricultural by-products in Korea and Japan. In order to increase the amount of fermentable sugars in the hydrolyzates of rice bran, various hydrolysis procedures were applied. Eight different hydrolyzates were prepared using rice bran (RB) and defatted rice bran (DRB) with enzyme or acid treatment or both. Each hydrolyzate was evaluated in terms of total sugar concentration and butanol production after fermentation by C. beijerinckii NCIMB 8052. Acid treatment yielded more sugar than enzyme treatment, and combined treatment with enzyme and acid yielded even more sugars as compared with single treatment with enzyme or acid. As a result, the highest sugar concentration (33 g/l) was observed from the hydrolyzate from DRB (100 g/l) with combined treatment using enzyme and acid. Prior to fermentation of the hydrolyzates, we examined the effect of P2 solution containing yeast extract, buffer, minerals, and vitamins on production of butanol during the fermentation. Fermentation of the hydrolyzates with or without addition of P2 was performed using C. beijerinckii NCIMB 8052 in a 1-1 anaerobic bioreactor. Although the RB hydrolyzates were able to support growth and butanol production, addition of P2 solution into the hydrolyzates significantly improved cell growth and butanol production. The highest butanol production (12.24 g/l) was observed from the hydrolyzate of DRB with acid and enzyme treatment after supplementation of P2 solution.

  • Butanol has an ability to improve the ignition quality due to its lower latent heat of vaporization; it has an advantage to reduce a volume of a fuel tank because its energy density is higher than that of ethanol. Also, butanol-diesel oil blending quality is good because butanol has an effect to prevent the phase-separation between two fuels. Even if the blended oil contains water, it can reduce the corrosion of the fuel line. Thus, it is possible to use butanol-diesel oil blended fuel in diesel engine without modification, and it may reduce the environment pollution due to NOx and particulate and the consumption of diesel oil. Therefore, some studies are being advanced whether butanol is adequate as an alternative fuel for diesel engines, and the results of the combustion and exhaust gas emission characteristics are being presented. Though the injection and spray characteristics of butanol are more important in diesel combustion, the has not yet dealt with the matter. In this study, the influence in which differences of physical properties between butanol and diesel oil may affect the injection and spray characteristics such as injection rate, penetration, spray cone angle, spray velocity and process of spray development were examined by using CRDI system, injection rate measuring device and spray visualization system. The results exhibited that the injection and macroscopic spray characteristics of two fuels were nearly the same.
