소 수정란의 간이 동결기법 개발에 관한 연구 II. 내동제의 평형시간, 융해온도, 융해시간 및 1단계 Straw법이 체외발생에 미치는 영향 (Studies on the Development of Easy Cryopreservation Technique of Bovine Embryos II. Effects of Equilibration of Cryoprotectants, Temperature and Time of Thawing and 1 Step Straw Method on In Vitro Developmental Rates of Embryos)

  • The studies on the carried out to investigate to determine the optimum thawing temperature and equilibration time and 1 step straw method of frozen bovine embryos. The follicular oocytes were cultured in TCM-199 medium containing 10 IU/ml PMSG(Sigma, USA), 10 IU/ml hCG(Sigma, USA), 1 $\mu\textrm{g}$/ml $\beta$-estradiol(Sigma, USA) and 10% FCS for 24~48 hrs in incubator with 5% CO2 in air at 38.5$^{\circ}C$ and then matured oocytes were again cultured for 12~18 hrs with motile capacitated sperm by preincubation of heparin. The bovine embryos following dehydration by cryoprotective agents and various concentration of sucrose were directly plunged into liquid nitrogen and thawed in 3$0^{\circ}C$ water. Survival and in vitro developmental rate was defined as developmental rate on in vitro culture or FDA-test. The results are summarized as followes : 1. The equilibration time on in vitro developmental rates of bovine embryos was attained after short period of time(2.5~5 min.) in the freezing medium higher than long period of time (10~20 min.). 2. The temperature thawed at 3$0^{\circ}C$ after rapid freezing of bovine embryos resulted in a significantly higher in vitro developmental rate than did at 2$0^{\circ}C$ and 35$^{\circ}C$. 3. The thawing time on in vitro developmental rates of bovine embryos was attained after short period of time(1~5 min.) in the freezing mediuim higher than long period of time(10min.). 4. The in vitro developmental rates of bovine embryos after rapid frozen-thawing by 1 step straw method in the freezing medium added 1.5M, 2.0M glycerol, DMSO, propanediol and 0.25M, 0.50M, 0.75M, 1.00M sucrose were 12.5~19.4%, 10.0~15.6%, 9.1~13.8% and 6.7~12.9%, respectively.

전자선 조사와 4-메틸모포린-N-옥사이드 용액을 이용한 볏짚의 전처리 방법 (A Facile Pretreatment Method for Rice Straw using Electron Beam Irradiation and 4-methylmorpholine-N-oxide Solution)

  • 2세대 바이오매스 볏짚을 이용하여 전자선 조사 후 NMMO 처리 공정을 수행하고 이를 분석하였다. 볏짚은 전자선 500 kGy 조사 후 NMMO 처리를 한 경우 72시간의 효소당화를 거쳐 글루코스 60.8%와 자일로스 9.7% 포함 약 83.8% 정도의 당수율을 보였다. 이 전처리 공정은 유해한 화학물질을 사용하지 않아서 친환경적이며 전자선 조사와 NMMO 처리의 시너지 효과가 있음을 알 수 있었다. 그러나 전자선 조사량이 너무 높다는 단점이 있으며 추후에는 낮은 전자선 조사량과 여러 가지 단일공정을 통합한 새로운 복합전처리 방법 연구의 필요성이 있다. 따라서 이 단점을 보완한다면 전자선 조사라는 친환경적 방법과 더불어 NMMO 용액의 재활용성을 이유로 바이오에탄올 생산 등의 산업현장에의 적용 가능성이 높다는 것을 알 수 있었다.

개 정액의 정제화동결법과 Straw 동결법에 관한 비교실험 (A Comparison between Pellet and Straw Methods in Canine Semen Freezing)

  • Pellet and straw methods in canine semen freezing are compared with respect to motility, viability and acrosome demage of sperm during each of the two major processing steps, to prior-freezing and to frozen-thawing. Senen was extended with a tris-buffered egg yolk contained 4% glycero1 Pellet freezing in the hole of dry ice and straw freezing on the surface of liquid nitrogen were carried out, respectively. The frozen semen 10 days after storage in liquid nitrogen container. wao thawed. In the comparison of two freezing methods, the straw freezing method with 42.7% in motility. 49.2% in viability and 0.186 acrosome score after thawing seems to be superior to the pellet freezing method with 31.2%, 34.5% and 0.314%, respectively. Sperm motility of processing step to frozen-thawing against decrease rate 12.67% to Prior freezing appeared of 33.84% and 49.37% in straw and pellet freezing and increase of 0.02 in acrsomal score to prior freezing appeared of 0.08 and 0.21 in straw and pellet freezing method to frozen-thawing

  • This study were carried out to investigate the effective concentration of cryoprotective agents and sucrose by one-step straw method, and to determine the optimum thawing temperature and equilibration time of frozen porcine embryos. The porcine embryos foflowing dehydration by cryoprotective agents and a various concentration of sucrose were directly plunged into liquid nitrogen and thawed in 3$0^{\circ}C$ water. Survival rate was defined by FDA test. The results are sunnnarized as follows : 1. The survival rates of porcine embryos after ultrarapid frozen4hawing in the freezing medium with a various concentration of glycerol, DMSO and propanediol added 0.25M sucrose were higher survival rate than those of sucrose concentration of 0.50M. 2. The survival rates of porcine embryos after ultrarapid ftozen4hawing in the freezing medium added 0.25M and 0.SOM sucrose were higher survival rate than those of sucrose concentration of 0.75M and 1.00M. 3. The temperature thawed at 2$0^{\circ}C$ and 3$0^{\circ}C$ resulted in a significantly higher embryos survival rate after 72 hrs in culture than did at 35$^{\circ}C$. 4. The equilibration time on the survival rate of porcine embryos was attained after short period of time(2.5~5 min.) in the freezing medium higher than long period of time(10~20 min.).

체외수정과 체외배양에서 생산된 생쥐 배반포기배의 초자화 동결에 있어서 Straw Loading 방법의 개선 (Improvement of Straw Loading Method on Survival of Mouse IVF/IVC Blastocysts Cryopreserved by Vitrification)

  • 본 실험은 체외수정에 의해 생산된 생쥐 배반포기배를 초자화 동결하였을 때 straw 제작방법 및 융해조건이 배반포기배의 생존성에 미치는 효과를 검토하고자 실시하였다. 융해시 각 straw내 동결보존액이 초자화 상태로 유지되는지를 확인하기 위하여 동결보존액 충전과 봉인방법이 다른 세 종류의 straw I, II, III가 제작되었는데, 이러한 모든 straw는 실온에 1~10초까지 노출된 후 $25^{\circ}C$ 수조에 침지되었다. 수정란은 20% ethylene glycol에 5분, EFS 40 용액에 1분간 노출된 후, straw내로 옮긴 다음 straw I과 II의 경우는 straw powder로, straw III는 straw powder와 열처리로 봉인하여 액체질소에 침지하여 다음과 같은 결과를 얻었다; 1) Straw I embryo column은 융해시 3~6초의 실온 노출을 제외한 대부분의 노출시간에서 불투명한 반초자화 상태로 변화되었다. 그러나 Straw II embryo column을 사용하였을 때 다소 개선되었으나 완전히 초자화 상태를 유지하지는 못하였다. 반면, Straw III embryo column의 경우는 융해시 완전한 초자화 상태를 유지할 수 있었다. 2) Straw III loading 방법을 사용하여 초자화 동결, 융해된 난자의 생존율은 Straw I loading 방법을 사용하였을 때에 비해 유의하게 높았으며(P<0.05), 표준오차 범위는 낮게 나타났다. 3) 초자화 동결보존된 난자를 실온에 3, 5, 10초간 노출한 후 $25^{\circ}C$ 수조내에 침지하여 융해하였을 때, 배양 24시간째 생존율은 72.7~87.1%이었고, 배양 48시간째 탈출배반포기배까지의 발달율은 34.0~48.4%이었으며, 융해시 각 처리간 유의차는 인정되지 않았다. 본 연구 결과, 초자화 동결 융해 후 높은 생존율 및 낮은 오차범위는 Straw III loading 방법과 double 봉인방법 및 적절한 2단계 융해법을 사용함으로서 얻을 수 있었다.

전자선 조사를 이용한 볏짚의 친환경 전처리 공정 (Environmentally-Friendly Pretreatment of Rice Straw by an Electron Beam Irradiation)

  • The autoclaving assisted by an irradiation pretreatment method was developed without toxic chemicals to produce fermentable sugars for their conversion to bioethanol. In the first step, electron beam irradiation (EBI) of rice straw was performed at various doses. The electron beam-irradiated rice straw was then autoclaved with DI water at $120^{\circ}C$ for 1 h. A total sugar yield of 81% was obtained from 300 kGy electron beam-irradiated rice straw after 72 h of enzymatic hydrolysis by Cellulase 1.5L (70 FPU/mL) and Novozyme-188 (40 CbU/mL). Also, the removal of hemicellulose and lignin was 32.0% and 32.5%, respectively. This result indicates that the environmentally-friendly pretreatment method of rice straw by an electron beam irradiation could be applied for bioethanol production in plant.

수정란의 급속동결융해법에 관한 연구. III. 소 동결수정란에 대한 1단계 Straw법이 난자 생존성에 미치는 영향 (Studies on Quick Freezing and Thawing of Embryos. III. Survival Effects of Bovine Embryos Cryopreserved and Diluted by One-Step Straw Method for Handling of Frozen-Thawed Embryos)

  • The objective of this experiment was to study some possibilities to simplify freezing, thawing and transfer procedure of one-step straw method comparing with the conventional methods using bovine embryos. The previous work are also designed to investigate the thawing effect by development stage and its quality using the embryos. Results obtained were summarized as follows: 1. A total of 87 embryos from 14 donor cows were frozen-thawed and an average of frozen embryo/donor was 6.2. 2. The survival rates of morula stage(65.4%) were higher than those of blastocyst stage(57.l%) and vice versa in rate of morphological recovery (80% vs 95.4%). However. no significant difference was denoted between them. 3. In difference between the groups of good quality and poor quality. good quality was resulted in a significantly higher embryo survival rate(75%) and recovery rates(95%) than poor quality(P<0.0l). 4. In effects of non-permeable sugar dilution in added to l.0M glycerol. higher survival rates were orderd in sucrose. lactose, raffinose and xylose. But lactose-raffinose, sucrose-trehalose and xylose in added to 2.OM glycerol. 5. The highest survival rates were obtained by direct plunge into the liquid nitrogen with 3.OM concentration both of glycerol and trehalose. 6. The survival rates in vitro condition of one-step and direct plunge methods(75%-87.5%) were significantly higher than those of multiple steps (21.4-52.6%) in in vitro (P<0.0l). However, the results of single-step were critical in comparing to other steps of final pregnant conformation.

1단계 straw동결법이 소 수정란의 생존성에 미치는 영향에 관한 연구 (A Study on the Effects of Cryopreservation by One-Step Straw Method on the Survival of Bovine Embryos)

The survival rate of bovine embryos thawed after rapid freezing in the freezing medium containing a various kinds of cryoprotective agents added 0.25M and O.50M sucrose were 28.6% and 25.0%, 35.1% and 31.6%, 32.4% and 24.4%, 34.2% and 28.2%, 18.9% and 17.6%, 14.7.% and 21.6%, respectively. 2. The survival rate of bovine embryos thawed after rapid freezing in the freezing medium containing a various concentration of sucrose added 1.5M and 2.OM glycerol, i.5M and 2.OM DMSO and 1.5M and 2.OM propanediol were 22.9~37.8%, 2O.7~31. 3%, 19.2~30.0% and 17.2~25.0%, respectively. 3. The temperature thawed at 2$0^{\circ}C$ after rapid freezing of bovine embryos resulted in a significantly higher embryos survival rate than did at 3$0^{\circ}C$ and 35$^{\circ}C$. 4. The equilibration time on the survival rate of bovine embryos was attained after short period of time(2.5~5 min.) in the freezing medium higher than long period of time (1O~20 min.).

돼지 정액의 간편 동결 방법 확립과 동결 정액의 융해 후 생존성 평가 (Establishment of the Convenient Boar Semen Freezing Method and Assessment of Viability in Frozen/Thawed Boar Semen)

  • 본 연구는 돼지정액의 보다 간편하고 손쉽게 동결시킬 수 있은 방법을 확립하기 위하여 수행하였다. 돼지정액은 3시간에 걸쳐 $5^{\circ}C$까지 냉각 후 Straw에 봉입하고 다양한 방법 및 step에 의해 스티로폼 용기 내에 들어있는 $LN_2$ 중에서 동결하였다. 정자의 생존성은 $LN_2$ 표면으로부터 10 cm 위에서 10분간 정치 후 침적할 경우 가장 높게 나타났다(54.0%). Straw를 $-102^{\circ}C$에서 10분간 정치시킨 처리구가 여타구보다 높은 생존성이 얻어졌다(74.0%, P<0.05). 응해 방법에 따른 동결정액의 생존성 실험에서는 $37^{\circ}C$의 융해구가 $52^{\circ}C$ 융해구보다 유의적으로 높은 결과를 나타냈다(P<0.05). 1단계 동결 방법과 3단계 동결방법으로 돼지 정액을 동결시킨 결과 정액의 일반적 특성 및 첨체의 이상 유무를 평가하는 CTC 검사에서는 커다란 차이가 인정되지 않았다. 동결정액을 이용한 체외수정 결과에서는 상실배기 이상 발육율에서 1-step이 3-step보다 높은 발육율을 나타내었다(27.5 vs 14.7%, P<0.05). 본 실험의 결과 돼지정액 동결 시 $-102^{\circ}C$에서 10분간 정치시키는 1단계 동결방법이 간편하면서 유리한 동결 방법으로 활용될 수 있음을 보여준다.

Estimation of Ruminal Degradation and Intestinal Digestion of Tropical Protein Resources Using the Nylon Bag Technique and the Three-step In vitro Procedure in Dairy Cattle on Rice Straw Diets

  • The experiment was carried out using fistulated multiparous Holstein Friesian crossbred (75% Holstein Friesian and 25% Red Sindhi) dairy cows in their dry period fed on untreated rice straw to evaluate the nutritive value of local protein feed resources using the in sacco method and in vitro pepsin-pancreatin digestion. Experimental feeds were cottonseed meal (CSM); soybean meal (SBM); dried brewery's grains (DBG); palm kernel meal (PSM); cassava hay (CH); leucaena leaf meal (LLM). Each feedstuff was weighed into duplicate nylon bags and incubated in each of the two rumen fistulated cows for 0, 2, 4, 8, 16, 24, and 48 h. Rumen feed residues from bags of 16 h incubation were used for estimation of lower gut digestibility by the technique of in vitro pepsin-pancreatin digestion. Ruminal ammonia-nitrogen ($NH_3-N$) concentrations did not differ between treatments or time with a mean of 5.5 mg%. Effective degradability of DM of CSM, SBM, DBG, PSM, CH and LLM were 41.9, 56.1, 30.8, 47.0, 41.1 and 47.5%, respectively. Effective degradabilities of the CP in feedstuffs were 49.6, 59.2, 40.9, 33.5, 47.3 and 65.0% for the respective feedstuffs. The CP in vitro pepsin-pancreatin digestibility as ranked from the highest to the lowest were SBM, CSM, LLM, CH, DBG, PSM, respectively. The intestinal and total tract digestion of feedstuffs in the current study were relatively lower than that obtained from previous literature. The results of this study indicate that SBM and LLM were highly degradable in the rumen, while CH, CSM and DBG were less degradable and, hence resulted in higher rumen undegradable protein. Soybean meal and LLM could be used to improve rumen ecology whilst CH, CSM and DBG could be used as rumen by-pass protein for ruminant feeding in the tropics.
