Study on the Relationship between the Structure and Antioxidant Activities of Chalcones

  • The purpose of this study is to examine the relationship between antioxidant activities and chemical structures of various chalcones. Twenty-two chalcones were assessed for their radical scavenging activity using the 1,1-diphenyl-2-picrylhydrazyl (DPPH) assay. Of 22 compounds tested, the most active on DPPH radical was 2',4-dihydroxy-3,3'5-trimethoxy-5'-propylchacone (4) (72.6% at 100 ppm). It was followed by 3',4'-dihydroxy-3,4,5-trimethoxy-6'-methylchalcone (6), 2',4,4'-trihydroxy-3-methoxy-5-propenylchalcone (7) and 2',4,4'-trihydroxy-3,5-dimethoxychalcone (13). Based on the results, we concluded that the scavenging activity is controlled by the number and the position of the substitution in the compound.

Radical Scavenging Activities of Korean Traditional Rice Wine, Takju

  • The principal objective of this study was to assess the radical scavenging activities and total polyphenols, flavonoids, and flavonol contents of takju, a Korean traditional rice wine. The antioxidant properties of the wine and takju samples were evaluated using five distinct assays: specifically, 1,1-diphenyl-2-picrylhydrazyl (DPPH), 2,2-azino-di-(3-ethylbenzothiazoline-6-sulfonic acid) (ABTS); hydroxyl; superoxide anion; and nitric oxide (NO) radical-scavenging activity assays. In this study, the takju evidenced strong scavenging activities against the hydroxyl, superoxide anion, and NO radicals. Furthermore, the total polyphenol contents of T-2 were similar to those previously observed in white wine (p<0.05). However, the flavonoids and flavonol contents of all takju samples were significantly (p<0.05) lower than that of white wine. The results of this study show that takju possesses a powerful radical scavenging activity against a variety of oxidative systems. The findings of this study also indicate that takju should be considered a useful antioxidant, and that their functional compound reduces oxidative stress.

Antioxidant Activity-of Curcuma Longa L., Novel Foodstuff

  • The potential antioxidant activities of different fractions from methanolic extract of Curcuma longa L. were assayed in vitro. All of the fractions exception of n-hexane and $H_2O$ showed a strong antioxidant activity, especially the ethylacetate (EtOAc) fraction, which showed the highest 1,1-diphenyl-2-picrylhydrazyl (DPPH) free radical scavenging activity ($IC_{50}=9.86\;{\mu}g/mL$). The results of 2,2'-azinobis (3-ethylbenzothiazoline-6-sulfonic acid) diammonium salt (ABTS) radical scavenging activity and ferric reducing antioxidant power (FRAP) assay showed concentration dependency, the EtOAc fraction demonstrating a better result than the other fractions at the same concentration in this studies. Additionally, when the total phenolic contents of fractions were measured, the EtOAc fraction contained the highest level. Meanwhile, correlation analysis indicated a high correlation between the antiradical activity and the total phenolic contents, suggesting that fractions obtained from the methanolic extract of Curcuma longa L. have wide potential for use as sources of antioxidant material.

Natural Anthraquinone Derivatives from a Marine Mangrove Plant-Derived Endophytic Fungus Eurotium rubrum: Structural Elucidation and DPPH Radical Scavenging Activity

  • There is considerable interest in the isolation of potent radical scavenging compounds from natural resources to treat diseases involving oxidative stress. In this report, four new fungal metabolites including one new bisdihydroanthracenone derivative (1, eurorubrin), two new seco-anthraquinone derivatives [3, 2-O-methyl-9-dehydroxyeurotinone and 4, 2-O-methyl-4-O-(${\alpha}$-D-ribofuranosyl)-9-dehydroxyeurotinone], and one new anthraquinone glycoside [6,3-O-(${\alpha}$-D-ribofuranosyl)-questin], were isolated and identified from Eurotium rubrum, an endophytic fungal strain that was isolated from the inner tissue of the stem of the marine mangrove plant Hibiscus tiliaceus. In addition, three known compounds including asperflavin (2), 2-0-methyleurotinone (5), and questin (7) were also isolated and identified. Their structures were elucidated on the basis of spectroscopic analysis. All of the isolated compounds were evaluated for 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging activity.

Biological activities of extracts from Caryopteris incana Miq. (층꽃나무(Caryopteris incana Miq.) 추출물의 생리활성)

  • In this study, extracts from Caryopteris incana Miq. (C. incana) were investigated to assess anti-oxidation, skin-whitening and anti-wrinkle activity. The total phenolic compounds of C. incana extracts with water and 80 % ethanol showed 7.69 and 12.50 mg/g respectively. Antioxidation activity of C. incana extracts was measured by using 1,1-diphenyl-2-picrylhydrazyl (DPPH), 2,2-azinobis(3-ethylbenzothiazoline-6-sulfonic acid (ABTS), protection factor (PF), and Thiobarbituric acid reactive substances (TBARs). At concentration of $200{\mu}g/mL$, the DPPH free radical scavenging activity of water and ethanol extracts were 84 and 92 %, respectively. ABTS radical scavenging activity of water and ethanol extracts were both at approximately 99 %. Antioxidant PF of water and ethanol extracts were 1.56 PF and 1.67 PF, respectively. The TBARs of water and ethanol extracts were 62 and 82 %, respectively. In anti-wrinkle and skin-whitening activity, 80 % ethanol extract had more outstanding effect than water extract at concentration of $200{\mu}g/mL$. The levels of elastase and collagenase inhibitory activity related with anti-wrinkle were 58 and 89 % in ethanol extract. The tyrosinase inhibitory activity related with skin-whitening was 13 % in ethanol extract. The astringent effect of ethanol extract was 50 %. Throughout the results, C. incana extracts showed an excellent effect on anti-oxidation, skin-whitening and anti-wrinkle activity. Therefore, C. incana extracts can be used as a new material for cosmetics.

Antioxidant capacity of crude extract and fractions from Woodfordia fruticosa flower

  • Woodfordia fruticosa Kurz. (Lythraceae), commonly known as Dhayero, is used in the treatment of various ailments in Nepal. In this study, the antioxidant capacity of crude extract and different polarity fractions of Woodfordia fruticosa flowers were assayed for their scavenging abilities against 1,1-diphenyl-2-picrylhydrazyl (DPPH) and nitric oxide radicals, competitive ${\beta}$-carotene bleaching, reducing power, metal chelating ability and total phenolic content. Crude extract and polar fractions showed stronger antioxidant capacity and contained very high level of total phenolics. They exhibited strong DPPH radical scavenging, nitric oxide scavenging and reducing power, medium ${\beta}$-carotene bleaching and poor metal chelating capacity. Positive correlation obtained between total phenolic content and antioxidant capacity assays, indicated the contribution of phenolics toards antioxidant capacity.

Antioxidant Activity of Ethanolic Extract of Potato(Solanum tuberlosum) (감자(Solanum tuberlosum) 에탄올 추출물의 황산화 활성)

  • 감자는 전세계적으로 섭취되는 주요 작물로서 우리나라에서는 감자의 생즙을 관절염 및 통증을 억제하는 민간요법으로 사용되어 왔다. 본 연구에서는 감자 에탄을 추출물의 항산화 작용을 검토하였다. 감자 에탄올 추출물(0.05~5 mg/mL) 은 1,1-diphenyl-2-picrylhydrazyl radical에 대하여 강한 free radical scavenger 활성을 나타내었다. 또한 감자 추출물(0.5 mg/mL)은 쥐간 균질액 에서 생성 된 TBARS의 양을 유의적으로 억제하였으며 (p<0.05) Fe/sup 2+//ascorbate induction system에서 감자 추출물(0.5~2 mg/mL)은 DNPH 반응으로 측정된 carbonyl 형성을 억제하였다. 항산화 활성과 마찬가지로 감자 추출물의 환원력은 0.5 mg/mL 농도에서 가장 높았고 이는 α-tocopherol의 경우와 비슷하였다. 이 결과는 감자 추출물이 항산화력을 통해 생체에서 작용할 수 있는 가능성을 제시한다.

Antioxidant Activities of Bromotopsentin from the Marine Sponge Spongosorites sp. (해면으로부터 분리된 Bromotopsentin의 항산화활성)

  • Bromotopsentin (BSM) is a bisindole alkaloid compound, which is recognized as a metabolite of the marine sponge Spongosorites sp. In this study, the antioxidant activity of BSM was investigated. The 1,1-diphenyl-2-picrylhydrazyl (DPPH) free radical scavenging assay, the trolox equivalent antioxidant capacity (TEAC) assay, the superoxide radical scavenging (NBT) assay, the lipid peroxidation and hydroxyl radical-induced DNA damage assays were carried out to evaluate the antioxidant activity of BSM. It was found that BSM had stronger scavenging activity on the stable free radical DPPH and superoxide radical than L-ascorbic acid with an $IC_{50}$ value of 62 and 64 ${\mu}M$, respectively. The TEAC value which indicated the total antioxidant capacity of BSM was about 0.8, which was also stronger than L-ascorbic acid. About 1.3 ${\mu}M$ of BSM induced 50% inhibition of lipid peroxidation. 60 nM of BSM exhibited a significant protective activity against DNA strand scission by hydroxyl radical on pBR322 DNA. Taken together, we suggest that BSM possesses strong antioxidant activity, and could be a valuable new addition to the list of anti-aging chemotherapeutic agents.

The Anti-oxidative Compounds of Smilax riparia Leaves (Smilax riparia 잎의 항산화 성분)

  • Rhizoma of Smilax china has been used as anti-inflammatory and analgesic, antiedemic agent in Korean folk medicine. In order to investigate the efficacy of anti-oxidative activity, the activity-guided fractionation and the isolation were performed. Each fractions ($H_2O$ fraction, 20%, 40%, 60%, 100% MeOH fractions and CHCl$_3$ fraction) was examined antioxidant activity by 1,1-diphenyl-2-picrylhydrazyl (DPPH) free radical scavenging potential. It was revealed that 40%, 20% MeOH fractions and $H_2O$ fractions have significant anti-oxidative activity. From 40% and 20% MeOH fractions two flavonoid glycosides and one procyanidin were isolated and elucidated apigenin-7-Ο-$\alpha$-L-rhamnopyranosyl(1\longrightarrow2)-$\beta$-D-glucopyranoside, apigenin-7-Ο-$\alpha$-L-rhamnopyranosyl(1\longrightarrow6)-$\beta$-D-glucopyranoside and catechin(4$\alpha$\longrightarrow6)epicatechin through their physicochemical data and IR, FAB-MS, $^{13}$ C-NMR, and $^1$H-NMR analysis with authentics, respectively. The isolated compounds were examined by DPPH method. Apigenin-7-Ο-$\alpha$-L-rhamnopyranosyl(1\longrightarrow2)-$\beta$-D-glucopyranoside and catechin (4$\alpha$\longrightarrow6) epicatechin showed powerful radical scavenging activities on DPPH radical among three compounds.

Antioxident components from Aralia continentalis

  • The root of Aralia continentalis Kitagawa(Araliaceae) have been used as an analgesic and fever remedy, and for treatment of rheumatism in Chinese medicine, whereas the young leaves are used for salad. An antioxidant activity of the young leaves of A continentalis was determined by measuring lipid peroxide produced when a mouse liver homogenate was exposed to the air at 37$^{\circ}C$, using 2-thiobarbituric acid(TBA) and by evaluation the radical scavenging activity on 1,l-diphenyl-2 picrylhydrazyl (DPPH) radical. Bioassay guided fractionation of MeOH extract isolated six flavonoid compounds as active components from EtOAc fraction. Adenosine and two saponins were isolated from the weak active BuOH fraction. The antioxidant effect by DPPH radical scavenging activity showed that quercetin was the most active among these compounds. Hyperoside and kaempferol were also active, while 6"-O-acetyl astragalin, astragalin, trifolin, adenosine, oleanolic acid 28-O-glucosyl ester and salsoloside C methyl ester were almost inactive. All the compounds were identified by spectroscopic methods.
