The effects of dietary levan, high-molecular-weight $\beta$-2,6-linked fructose polymer, on adiposity, serum leptin and UCP expression in rats fed high fat diet were studied. The adipose tissue hormone, leptin has been proposed to be involved in the regulation of food intake and energy expenditure. Uncoupling protein (UCP), a mitochondrial protein that uncouples the respiratory chain from oxidative phosphorylation, generates heat instead of ATP, thereby increase energy expenditure. To determine whether the dietary levan may have the anti-obesity effect, 4 wk old Sprague Dawley male rats fed high fat diet for 6 wks to induce obesity, and subsequently fed one of three diets for further 6 wks: 1) high fat (40% of calories) diet without levan 2) with 3% (w/w) levan 3) with 5% levan. For the comparison, control group fed AIN-76A diet. Visceral and peritoneal fat masses were lower in high fat diet with levan groups compared to high fat diet group. The effect of levan was dose-dependent. Adipocyte size was significantly reduced in the levan diet groups compared to the no levan diet group. Serum cholesterol level was not affected by levan containing diet, while the serum HDL cholesterol level was higher in leven diet groups. In addition, serum triglyceride level was markedly reduced by levan containing diet, thus lower than that of control group. Serum leptin was reduced by levan containing diet and lower in 5% levan group compared to 3% levan group (p < 0.001), as a result, serum leptin and insulin levels of 5% levan group were reduced to level of control group. Futhermore, the serum leptin level reflected the adiposity. The expression of UCP 1, and UCP 2 in brown adipose tissue was up-regulated by levan containing diet. In conclusions, levan containing diet reduced adiposity and serum triglyceride but increased UCP expression in the obese rats fed high fat diet. (Korean J Nutrition 35(9) : 903~911, 2002)
Objectives : In order to investigate the effects of Gambibang-3(here in after referred to GBB3) on the obese gene and obese inhibitory, C57BL/6 mice were induced by high fat diet. Methods : C57BL/6 mice were divided into four groups(normal, high fat diet with control, high fat diet with Reductil(here in after referred to RDT), high fat diet with GBB3 extract) and fed for 8 weeks. And observed that, the weight change of the adipocytes in body and liver, total cholesterol, LDL-cholesterol, HDL-cholesterol, triglyceride, glucose, leptin change in the serum, the expression of ${\beta}3AR$ and leptin gene in 3T3 cell and primary adipocyte cell. Results & conclusions : The following results were obtained in this study. 1. GBB3 and RDT group showed that the revelation of ${\beta}3AR$ in primary adipose cell and 3T3 cell were increased considerably, and that the revelation of leptin in primary adipose cell and 3T3 cell were decreased considerably. 2. GBB3 and RDT group showed that the weight of adipocyte and liver were not different with control group significantly. 3. GBB3 and RDT group showed that the amount of HDL-Cholesterol were increased considerably, the amount of glucose, LDL-Cholesterol and Triglyceride were decreased considerably. 4. GBB3 group showed that the amount of leptin in the serum were decreased considerably.
Objectives : This research was performed to investigate the effect of acupuncture using invasive low level laser therapy (LLLT) at Yolgyol (LU7) + Yogu (LR5) on weight gain, food intake, food efficiency, lipid metabolism, atherogenic index, HTR (HDL-cholesterol to total cholesterol ratio) and morphological change of hepatic tissue in hyperlipdemia rats. Methods : Experimental groups were divided into high fat diet group (Control group), high fat diet and acupuncture therapy group at LU7 + LR5 (AT group), high fat diet and acupuncture group using 10 mW LLLT at LU7 +LR5 (LA10 group), high fat diet and acupuncture group using 20 mW LLLT at LU7+LR5(LA20 group), high fat diet and acupuncture group using 60 mW LLLT at LU7 + LR5 (LA60 group), once per 3 days during 9 weeks. Results : Body weight was decreased significantly in AT and LA20 groups compared with Control group. Food intake was increased significantly in LA60 group compared with Control group. Food efficiency was decreased significantly in LA10, LA20 and LA60 groups compared with control group. In the lipid metabolism, total cholesterol was decreased significantly in AT, LA10, LA20 and LA60 groups, triglyceride was decreased significantly in LA10, LA20 and LA60 groups, TG/HDL-cholesterol ratio was decreased significantly in LA 60 group compared with control group. In the morphological change, hepatic tissue were not showed balloning degeneration and irregular arrangement of hepatic cell in LA10 and LA20 groups with control group. Conclusions : Acupuncture using LLLT at LU7+LR5 can manage hyperlipemia by controlling body weight, food intake, food efficiency ratio and lipid metabolism.
Objectives: This study was performed to evaluate the effects of fermented Samjung-hwan (SJH) extracts on weight, serum lipids and blood glucose. Methods: SJH was fermented using three different probiotic bacterial strains (Lactobacillus plantarum [LP], Leuconostoc mesenteroides [LM], Bifidobacterium longum [BL]) separately. Thirty-six rats were divided into normal, control (high fat diet), SJH-UF (high fat diet+unfermented SJH 200 mg/kg), SJH-LP (high fat diet+LP fermented SJH 200 mg/kg), SJH-LM (high fat diet+LM fermented SJH 200 mg/kg) and SJH-BL (high fat diet+BL fermented SJH 200 mg/kg). For 8 weeks later, we examined body weight, total cholesterol, high-density lipoprotein (HDL)-cholesterol and blood glucose. Results: The control group showed significantly increased weight gain compared with normal group and SJH-LP and BL groups had less weight gain than control group, significantly. In the lipid serum tests, control group showed significantly increased total cholesterol levels compared with normal group and only SJH-LP represented decreased total cholesterol levels compared with control group. However there was no significant change in the HDL-cholesteol levels. In the blood glucose tests, that of control group significantly incereased more than that of normal group, SJH-BL showed significantly decreased blood glucose levels compared with control group. Conclusions: SJH-LP, SJH-BL showed weight control effect, SJH-LP decreased TC and SJH-BL reduced blood glucose.
Ethanol extract of Cinnamomi cortex inhibited potently cholesterol esterase activity in vitro. Chloroform fraction of ethanol extract showed the stronger inhibitory effect than other solvent fractions - ethylacetate fraction, butanol fraction, and aqueous fraction. The chloroform fraction of Cinnamomi cortex was studied as a candidator of plasma cholesterol-lowering material using high cholesterol-fed rats. In high cholesterol-fed rats, the diet with chloroform fraction of 150 mg/kg lowered not only plasma neutral lipids contents 25.1% but also plasma total cholesterol level 49.6% than only high cholesterol diet. Plasma HDL-cholesterol level in Cinnamomi cortex chloroform fraction-fed rats was recovered as those level of normal rats. LD$_{50}$ of Cinnamomi chloroform extract was calculated as 1,300 mg/kg.

This study was performed to investigate the effects of a diet with a red yeast sweet potato supplement on fecal amount and lipid metabolism in male Sprague-Dawley rats fed a high-fat diet for 10 weeks. Rats were fed a high-fat diet (15% fat) with additional lard (7%) and cholesterol (1%) based on AIN-93G basal diet (7% fat) for 6 weeks during the first phase. In the second phase, which lasted 4 weeks, the rats divided into four experimental groups which were composed of a high-fat diet group as a control (CON), a high-fat diet with 5% white-fleshed sweet potato supplement group (WFSP), a high-fat diet with 5% red yeast sweet potato supplement group (RYSP), and a high-fat diet with 5% purple-fleshed sweet potato supplement group (PFSP). The fecal amount of group RYSP increased significantly during the second phase compared to the other groups (p<0.05). The fecal total cholesterol (TC) and triglyceride (TG) content of group RYSP were also highest among all experimental groups. The serum TC and TG were shown to have the lowest levels in the group RYSP, and LDL-cholesterol levels were significantly decreased in groups RYSP and PFSP than in group CON (p<0.05). These results indicate that supplementation with red yeast sweet potato seemed to be effective in increasing feces and fecal lipid excretion, and also in decreasing serum lipid levels in rats fed a high-fat diet.
Objectives : This study was performed to investigate inhibitory effects of Gamipalmul-tang($jiaweibawu-t{\bar{a}}ng$) on the hematological and histological changes of obese mice. Methods : C57BL/6 mice were divided into four groups (normal group, high fat diet with normal saline, high fat diet with reductil, high fat diet with Gamipalmul-tang($jiaweibawu-t{\bar{a}}ng$) and fed for 8 weeks. Body weight change, final increase of body weight, ALT, AST, creatinine, total cholesterol, LDL-cholesterol, HDL-cholesterol, triglyceride, glucose, cell viability by cytotoxicity, the expression of ${\beta}3AR$ in 3T3-L1 cell, the expression of leptin, ${\beta}3AR$ and serotonin in adipocyte tissue and size of adipocyte were observed in 8 weeks. Results : 1. In 3T3-L1 cell. the expression of ${\beta}3AR$ was increased significantly. 2. The final increase of body weight in obese-mouse were decreased significantly. 3. The level of AST, ALT were decreased significantly. 4. The level of LDL-cholesterol was decreased significantly and HDL-cholesterol was increased significantly. 5. The levels of triglyceride was decreased and leptin and glucose were decreased significantly. 6. In adipocyte tissue, the expression of ${\beta}3AR$ were increased significantly. 7. In adipocyte tissue, the expression of leptin and serotonin were decreased significantly. 8. The size of Adipocyte was decreased. Conclusions : On the basis of these results, we concluded that Gamipalmul-tang($jiaweibawu-t{\bar{a}}ng$) has inhibitory effects in rat.

This study was investigated the effects of water soluble extract of Ganoderma lucidum kale juice and soidium dextrothyroxine on lipid components in serum of hypercholesterolmemic rats in vivo in order to prevent in cardiovascular disease. Total cholesterol concentrations in serum were significantly increased after feeding cholesterol diet group compared with control group and were significantly increased after feeding cholesterol diet group compared with control group and were lower in Ganoderma lucidum kale juice and sodium dextrothyrioxine diet groups than in control group. Ratio of high density lipoprotein cholesterol concentration to total cholesterol were significantly highest in sodium dextrothyroxine fed groups among diet groups and were higher in Ganodrma lucidum and kale juice fed group than in control group. Phospholipid concentrations in seurm were significantly lower in Ganoderma lucidum kale juice and sodium dextrothyroxine(1.25mg/kg diet) fed groups than in control group and triglyceride concentra-tions were lower in Ganderma lucidum sodium dextrothyroxine fed groups than in other groups. Triiodothyronine concentration in serum were lower in Ganoerma lucdum and kale juice fed groups than in the other groups while it was higher in sodium dextrothyroxine diet group than in other groups. Tetraiodothyronine concentrations is serum were remakably higher in sodium dextrothyroxine fed group than in other groups. Blood glucose concentration was lower in cholesterol diet group than in other groups but was higher in sodium dextrothyroxine diet group than in other groups.
This study was conducted to compare the effect of dietary sucrose and starch on the lipid metabolism of Rat fed by low protein diet for 3 and 6 weeks periods. Forty male weanling Spargue-Dawley rats weighing 52.7 grams each, after being adopted for 2 days with standard diet, were blocked into 8 groups and fed experimental diet as designed. Experimental diet were composed of two different levels of proteins, 6% designed as low protein diet and 15% designed as standard protein diet. In each group, the content of serum lipid, serum cholesterol and liver lipid were measured. The results of this experiment were summerized as follows. 1. liver lipid content was tended to be high in the sucrose group of low protein fed animal. 2. the content of total serum lipid was tended to be high in the sucrose group, and this tendency was showed statistical significance in the animals fed by low and standard protein after 6 weeks of experimental period. 3. The difference in the total serum cholesterol content between the sucrose and starch group was not significant

The objective of this study was to evaluate the effects of Lactobacillus acidophilus ATCC 43121 and L. fermentum MF27 on biochemical indices in the serum, cholesterol metabolism in the liver and mucin expression in the gallbladder in lithogenic diet (LD)-induced C57BL/6J mice to determine the preventive effects of lactobacilli on gallstone formation. By the end of 4 wk of the experimental period, mice fed on a LD with high-fat and high-cholesterol exhibited higher levels of total and low-density lipoprotein cholesterol in the serum compared to mice fed on control diet or LD with L. acidophilus ATCC 43121 (LD+P1; p<0.05). Cholesterol-lowering effects observed in the LD+P1 and LD with L. fermentum MF27 (LD+P2) groups were associated with reduced expression of 3-hydroxy-3-methylglutaryl coenzyme A reductase in the liver compared to the LD group (p<0.05). Furthermore, expression of the gel-forming mucin, including MUC5AB and MUC5B, was suppressed in the LD+P1 and LD+P2 groups compared to the LD group (p<0.05). Therefore, steady intake of both L. acidophilus ATCC 43121 and L. fermentum MF27 may have the ability to prevent the formation of cholesterol gallstones in LD-induced C57BL/6J mice.
