This study was performed to investigate the effects of genistein, a kind of soy isoflavones, on fatty liver and lipid metabolism in rats fed high fat diet. Twenty four male Sprague-Dawley rats were divided into four groups by dietary fat and genistein contents then raised for six weeks. The rats(n=6/group) were fed normal fat diet(NOR), high fat diet (HF), high fat with 0.1% genistein(HF+0.1%G) or high fat with 0.2% genistein(HF+0.2%G). Hepatic total lipid, triglyceride, total cholesterol and Serum GPT, as a marker for fatty liver, were significantly increased by high fat diet. Also, serum total lipid, triglyceride, total cholesterol, glucose and insulin concentration, hepatic lipogenic enzyme (fatty acid synthase and malic enzyme) activities were significantly increased by high fat diet. However, hepatic total lipid, triglyceride, total cholesterol and Serum GPT were significantly decreased by genistein intake. Also, genistein supplementation decreased serum total lipid, triglyceride, glucose and insulin concentration, hepatic lipogenic enzyme (fatty acid synthase and malic enzyme) activities. There were no differences by genistein level except for serum insulin. These results suggest that fatty liver induced by high fat diet was caused by increased serum lipid profiles and hepatic lipogenesis, whereas, genistein may be useful in inhibiting of fatty liver by reducing serum lipid profiles and hepatic lipogenesis.
Objective: This experiment was conducted to evaluate the effects of a corn- or cassava- based diet on the production of meat-type ducks. Methods: Four hundred day-old ducks were used in this experiment. They were divided into five groups with each group replicated eight times. The ducks fed the corn-based diets served as the control group. The four other groups comprised different treatments, with each one given the cassava-based diet based on phase-feeding. Three treatments were fed the cassava-based diet from 16, 28, and 35 d; respectively up to 42 d of age and the other group was fed the cassava-based diet from 1 to 42 d of age. Results: The results indicated that ducks on either the corn- or cassava-based diets were similar in growth during 1 to 9 d of age. However, toward 35 to 42 d, the cassava-diet produced a higher weight gain (p<0.05). The cassava-based diet was better than the corn-based diet at increasing the outer and inner breast weights at 28, 35, or 42 d (p<0.05). In contrast, the corn-based diet was better at increasing abdominal fat (p<0.05). The two diets did not differ in their effects on the serum triglyceride, cholesterol, high-density lipoprotein-cholesterol, low-density lipoprotein-cholesterol, very-low-density lipoprotein-cholesterol, and liver cholesterol. The corn-based diet, however, caused a highly significantly greater level of liver triglyceride (p<0.01). Conclusion: The results of this study suggest that both the cassava- and corn- based diets are similar in their effect on meat-type ducks during the starter stage but toward the finisher stage, the cassava-based diet has a better influence on weight gain and carcass characteristics.

Total cholesterol은 인삼 4mg을 첨가한 실험군에서 가장 낮았고, 마그네슘과 칼슘이 1:2로 급여한 군에서 낮게 나타났다. 3. Free cholesterol과 cholesterol ester도total cholesterol의 경우와 같이 인삼 4mg을 첨가한 것과 마그네슘과 칼슘을 1:2로 급여한 실험군에서 가장 낮았다. 4. Total cholesterol이 가장 낮은 실험군에서 LDL이 가장 낮았으며, HDL/T-CHOL의 비는 가장 높게 나타났다. 5. Triglyceride와 phospholipid에 있어서는 인삼 첨가로 TG의 양은 점점 감소되었으나 PL의 경우는 상당히 증가되었다. 6. 전해질의 경우는 마그네슘과 칼슘을 1:2로 첨가하고 인상 4mg을 급여한 실험군의 $Na^+$이 120mEq/l, $K^+$이 6.8mEq/l를 보여 인삼이 전해질의 이동에 크게 작용하고 있는 것으로 생각된다. 이상과 같은 결과로 인삼은 토끼의 혈청 전해질의 분포상태와 콜레스테롤의 농도에 영향은 주며, 콜레스테롤 농도를 저하시키는 작용을 함과 동시에 지방질을 감소시키고 인지질은 증가시키는 작용을 하는 것으로 추정된다.
To study the effect of prosomillet (Panicum milaceum) on lipid metabolism, male Sprague-Dawley rats weighing 190$\pm$8g were fed six experimental diets for four weeks. The six diets based on AIN-76 composition consisted of one cholesterol-free(normal) and five 1%(w/w) cholesterol diets, i.e. control, two diets containing additional 0.3 and 0.6%(w/w) methanol extracts of prosomillet and another two diets containing 15 and 30% (w/w) prosomillet powder. There was no difference in weight gains between the groups but relative liver weights increased under the cholestrol diets. Plasma levels of total cholesterol and triglyceride(TG) decreased by 23-27% and by 37-52%, respectively, in the four prosomillet diet groups compared to those of the normal and control groups. Whereas in the liver, only TG levels decreased in the prosomillet diet groups. Fecal excretions of bile acid and cholesterol significantly with methanol extracts of prosomillet. There was a significant increase in the activity of hepatic microsomal cholesterol 7$\alpha$-hydroxylase when feeding 1% cholesterol but prosomillet in the diet, either as in the form of powder or methanol extract, appeared to have only slight additional effects, namely increases in enzyme activity. The activity of liver cytosolic glucose-6-phophate dehydrogenase (G6PDH) tended to be reduced with high cholesterol diets and dropped markedly by 15% using additional prosomillet powder. Those of the liver cytoxolic malic enzyme had a similar tendency to those of G6PDH. The results indicate that certain active components in prosmillet other than fiber have the potential to exert hypolipidemic effects via regulating cholesterol excretions and lipogenesis.
Curcumin, the yellow pigment in turmeric, curry and mustard, has anti-inflammatory and anti-carcinogenic activities. In this study; we investigated the hypocholesterolemic and anti-atherogenic effect of curcumin in rabbits. Hypercholesterolemia was induced by feeding high cholesterol diet to male rabbits for 30 days, and the animals were then fed high cholesterol diet containing 0.1% (w/w) or 0.5% (w/w) curcumin for additional 30 days. Supplementation of 0.l% curcumin tended to lower serum total cholesterol and low density lipoprotein (LDL)-cholesterol levels and inhibit serum lipid peroxidation. In the 0.5% curcumin-supplemented group, serum total cholesterol was significantly lowered by 11.7%, LDL-cholesterol by 12.8% and lipid peroxidation by 47.9% compared to the control group. Hepatic cholesterol and triglyceride contents were also significantly lowered by 50.6% and 37.4%, respectively compared to the control group. Lipid staining of the arteries isolated from the curcumin-treated rabbits showed that curcumin significantly decreased formation of fatty streaks and atheromatous plaques on the intima of the arteries. These results demonstrated that curcumin lowered serum cholesterol concentration, hepatic cholesterol and triglyceride contents, and accumulation of cholesterol in the artery These cholesterol lowering effects of curcumin, together with its anti-oxidative and anti-inflammatory activities, may play some important roles in preventing atherosclerosis.
Excessive salt intake induces hypertension, but several gamma-aminobutyric acid (GABA) supplements have been shown to reduce blood pressure. GABA-salt, a fermented salt by L. brevis BJ20 containing GABA was prepared through the post-fermentation with refined salt and the fermented GABA extract. We evaluated the effect of GABA-salt on hypertension in a high salt, high cholesterol diet induced mouse model. We analyzed type 1 macrophage (M1) polarization, the expression of M1 related cytokines, GABA receptor expression, endothelial cell (EC) dysfunction, vascular smooth muscle cell (VSMC) proliferation, and medial thicknesses in mice model. GABA-salt attenuated diet-induced blood pressure increases, M1 polarization, and TNF-α and inducible nitric oxide synthase (NOS) levels in mouse aortas, and in salt treated macrophages in vitro. Furthermore, GABA-salt induced higher GABAB receptor and endothelial NOS (eNOS) and eNOS phosphorylation levels than those observed in salt treated ECs. In addition, GABA-salt attenuated EC dysfunction by decreasing the levels of adhesion molecules (E-selectin, Intercellular Adhesion Molecule-1 [ICAM-1], vascular cell adhesion molecule-1 [VCAM-1]) and of von Willebrand Factor and reduced EC death. GABA-salt also reduced diet-induced reductions in the levels of eNOS, phosphorylated eNOS, VSMC proliferation and medial thickening in mouse aortic tissues, and attenuated Endothelin-1 levels in salt treated VSMCs. In summary, GABA-salt reduced high salt, high cholesterol diet induced hypertension in our mouse model by reducing M1 polarization, EC dysfunction, and VSMC proliferation.
Objective: Ramulus mori (RM) has been known to be effective for the treatment of obesity. To show the effectiveness of RM in a more scientific way, RM extract was prepared and evaluated in high fat diet rats by measuring the changes of body weight and lipid metabolism as described briefly below. Methods: 200 g of crushed RM was extracted withmethyl alcohol. The extract was evaporated under reduced pressure to give 33.4 g. For 10 weeks, control group rats were fed a high fat diet, while the test group rats were fed with the same diet plus RM extract. The normal group was fed with a normal diet. 150 mg of RM extract per 1 kg of body weight was added to the diet in the test group rats. Results: The control group rats on the high fat diet gained weight significantly, whereas the test group rats on the high fat diet plus RM extract gamed less weight. The significant increase of liver weight caused by the high fat diet was also inhibited by the RM extract treatment. Total lipid, triglyceride and total cholesterol levels of serum in the high fat diet rats were remarkably increased, whereastheir levels on the high fat diet plus RM extract were less increased. While serum HDL-cholesterol levels were remarkably decreased in the high fat diet, its level was less decreased in the high fat diet plus RM extract. Furthermore, we observed that the activities of hepatic acetyl-CoA carboxylase and fatty acid synthetase increased under the high fat diet, while their activities under the high fat diet plus RM extract were getting back nearly to the normal levels of the normal diet rats. Conclusions: These result show that the obesity caused by a high fat diet was effectively inhibited by an RM extract. Our results also showed that the abnormal lipid metabolism caused by a high fat diet was effectively cured by adding RM extract.
Objectives : This study was aimed to investigate the effect of ACLL extract (Artemisia capillaris Thunb, Curcuma longa L, Lycium chinense, Ligustrum lucidum Aiton complex extract) in a dyslipidemia animal model induced by a high-fat diet. Methods : ACLL extracts was administered at 200 mg/kg, and 400 mg/kg to Apoe mice, where arthritis was induced by high cholesterol diet. After mice were treated with GO for 4 weeks, we measured AST, ALT, BUN and lipid levels (total cholesterol, HDL cholesterol, LDL cholesterol, triglyceride), atherogenic index(AI) and cardiac risk factor(CRF). Results : Serum AST, ALT, creatinine, BUN levels were not changed by ACLL extract do not show any toxic effects. Also, ACLL extract groups were found to have atherogenic Index and cardiac risk factor as well as lipid metabolism improvement (total cholesterol, LDL cholesterol and triglyceride decrease). Conclusions : We suggest that ACLL extract may have the control effects of Dyslipidemia by improving lipid metabolism.
Objectives : The present study was conducted to examine whether Cynanchi wilfordii radix (CWR) with or without fermentation has an ameliorative effect on hyperlipidemia in rats.Methods : We analyzed the contents of Conduritol F on Cynanchi wilfordii radix. The experimental animals were divided into six groups; normal diet fed group (N), high cholesterol fed control group (Con), Lovastatin 20 mg/kg (L), CWR-W 300 mg/kg (CWR-W), and CWR-F 300 mg/kg on hyperlipidemia model induced by feeding 1.25% cholesterol. Rats were administrated orally every day for 8 weeks. And lipid profile of serum and weight change were observed.Results : The vehicle displayed a markedly increased body weight and significantly increased liver and epididymal fat weight, however, the administration of CWR improved the body, liver, and epididymal fat weights. All drug treatment reduced significantly the serum level of total cholesterol and LDL-cholesterol elevated by intake of high cholesterol diet. TG displayed a reducing tendency all drug treatment, however, CWR-W decreased significantly. Atherogenic index and cardiac risk factor increased high cholesterol diet fed control group, while the administration of CWR-W and CWR-F decreased significantly. The major index of liver injury such as AST and ALT improved in all drug treatment.Conclusions : These results suggest that CWR extended the effect of lipid enhanced. Therefore CWR with or without fermentation may be useful for therapeutic treatment of clinical conditions associated with hyperlipidemia. Finally, these require more investigations about the action mechanism of CWR in the future.
The dietary effects of soy fiber beni-koji(SBCA) containing chitosan-ascorbate on the body weight and serum lipids in obese rats induced by a high fat diet were investigated. The experimental plots(6 rats per each group) were divided into a normal group(NC), a high fat diet group(HF), a 1% SBCA diet group(SBCA1) and a 2% SBCA diet group(SBCA2), and fed for 6 weeks. Compared with the HF group, the mean body weight and the $R\'{o}hrer$ index of the SBCA1 and SBCA2 groups decreased by $3.4{\sim}7.4%$ and $8.1{\sim}13.9%$, respectively. In particular, the body lipids in the SBCA2 group decreased by 42.3%. The serum triglyceride content decreased by approximately $25.54{\sim}27.34%$ in the experimental diet groups. There was no significant difference in the serum total lipids between the HF and SBCA1 groups. On the other hand, the total lipids in SBCA2 group decreased by 19.12% compared with the HF group. In the SBCA2 group, the total serum cholesterol and LDL-cholesterol content decreased by 6.57% and 41.20% compared with the HF group, respectively, while the HDL-cholesterol increased by 10.0%. The risk factor index(RFI) in the SBCA2 group was decreased remarkably by 58.57%.
