Axenic Culture of Gyrodinium impudicum Strain KG03, a Marine Red-tide Microalga that Produces Exopolysaccharide

  • An exopolysaccharide-producing microalgal dinoflagellate was isolated from a red-tide bloom and des­ignated strain KG03. A bacteria-free culture of strain KG03 was achieved using a modified wash with phototaxis and antibiotic treatment. Combined treatment with neomycin and cephalosporin was the most effective for eliminating the bacteria associated with the microalgae. Strain KG03 was identified as Gyrodinium impudicum by analyzing the ITS regions of the 5.8S rDNA, 18S rDNA, morphological phenotype and fatty acid composition. The exopolysaccharide production and cell growth in a 300-ml photobioreactor were increased 2.7- and 2.4-fold, respectively, compared with that in a flask culture at the first isolation step.

  • The purpose of this study is to grasp present condition of railroad noise, and forecast noise and presented basis data that ready noise reduction countermeasure. This study has predicted the level of noise reduction according to barrier by considering the characteristics of the cause of noise. These result drew using 'Prediction Model of NIER' and 'Shall03 of Germany'. This paper found important factors in railroad noise using this results and become forward syudy direction.

  • Most radio astronomy issues at WRC-03 (World Radiocommunication Conference-03) revolved around satellite downlink allocations, particularly to NGSO (Non-Geostationary Satellite Orbit) satellite systems, in bands adjacent to or close to a radio astronomy frequency band. Out of a total of 50 agenda items, ten were of interest to radio astronomers. This paper provides some details about the important outcome of the radio astronomy related issues at the WRC-03.

  • This paper presents experimental results of the fuzzy controller for DC servo motor. The fuzzy controller consists of 9 quantized levels and 25 fuzzy rules. The fine Controller is employed in the fine control mode when the value of error is between -0.03 and +0.03, whereas the coarse controller is used in the coarse control mode when the value of error is in the outside range of -0.03 and +0.03. The experimental results show that the fuzzy controller provides a better performance (lower overshoot and error) than the PID controller regardless of the load applied.

Production of $\beta$-Glucosidase from Aspergillus niger (Aspergillus niger에 의한 $\beta$-Glucosidase 생산)

  • This study was designed to reveal the conditions for $\beta$-glucosidase production from Aspergillus niger. The maximal enzyme production was obtained when the fungus was cultured at $30^{\circ}C$ for 5~6 days in the optimal medium containing 0.8% CMC, 0.5% beef extract, 0.3% Ca(NO3)2, 0.03% K2HPO4, 0.03% FeSO4, 0.05% Li2SO4, 0.2% tween 80, trace solution 1.0ml and initial pH 4.0, and then final enzyme activity under above conditions was 8.5-9.8 unit/ml culture filtrate.

Study on Characteristics of Car Air-con Compressor Under Bench System Fuel Economy Simulation Condition (벤치 연비 모사 조건에서 차량용 에어컨 압축기의 특성에 관한 연구)

  • In this study, an experiment on an air conditioning test bench was performed to verify the possibility of fuel economy simulation for the SC03 mode, North America fuel economy certification mode with a/c on condition, one of the vehicle fuel economy evaluation modes. The air conditioning test bench used in this study had each chamber simulating the actual vehicle air conditioning system and the controlling temperature, humidity, and air flow velocity to reproduce environmental conditions. Reliable results were obtained about the compressor RPM and inlet air velocity in front of the condenser corresponding to vehicle speed and air velocity in front of the vehicle, respectively, in the simulation of the SC03 mode, previously performed in CWT, in an air conditioning test bench. It was also discovered that there was a distinct difference in the fuel economy depending on the difference in the compressor displacement in the simulation test of the SC03 mode in the air conditioning test bench under various displacement conditions of the compressor.

A Study on Nano-emulsion for Enhanced Transdermal Delivery of Hippophae rhamnoides Leaf Extract (비타민나무 잎 추출물의 피부 흡수 증진을 위한 나노에멀젼 연구)

  • In this study, nano-emulsions containing 0.01, 0.03, 0.05, and 0.10% ethyl acetate fraction of Hippophae rhamnoides (H. rhamnoides) leaf extracts were prepared. The particle size, particle size distribution and skin permeability of the nano-emulsions were evaluated for five weeks. Nano-emulsion was prepared by the sequential use of homogenizer and microfluidizer. Nano-emulsion containing the ethyl acetate fraction exhibited a monodispersed form. Nano-emulsion containing 0.03% ethyl acetate fraction was the most stable for five weeks. The in vitro skin permeation study of nano-emulsion containing 0.03% ethyl acetate fraction was carried out using Franz diffusion cell. The nano-emulsion showed a better skin permeability than that of O/W emulsion. These results indicate that the nano-emulsion containing the ethyl acetate fraction of H. rhamnoides leaf extract showed a remarkable stability and skin permeability than that of O/W emulsion.

Construction and Characterization of the Vibrio parahaemolyticus Collagenase Inactivated Mutant (Vibrio parahaemolyticus collagenase 불활성화 돌연변이체의 제조 및 특성)

  • For better understanding of the host infection mechanism of Vibrio, a Vibrio parahaemolyticus collagenase mutant was generated by insertional inactivation of a vppC gene encoding extracellular collagenase. A recombinant DNA containing vppC::nptII was cloned into a suicide plasmid pDMS197, resulted in pVCM03. The recombinant suicide plasmid pVCM03 contained in E. coli $\chi$7213 was transferred to a wild-type V. parahaemolyticus 04 through conjugation. The recombinant vppC::nptII DNA in pVCM03 was exchanged with wild-type allele by homologous recombination resulting vppC mutant, V. parahaemolyticus CM. The mutant was selected and screened on TCBS media containing 10% sucrose and kanamycin. The mutation by allele exchange was confirmed with the comparison of the size of DNAs amplified by PCR. V. parahaemolyticus CM showed at least 4-fold less collagen-degrading activity than those of wild-type, and the mutant exhibited less cytotoxicity than that of wild-type in MTT assay.
