  • Ganoderan, antitumor ${\beta}-glucan$ from Ganoderma lucidum was extracted from the mycelium of G.lucidum IY009 which was cultured in various carbon sources. The mycelium was shown to be capable of utilizing various carbon sources, e.g., soluble starch, fructose and glucose, and differs in morphology on carbon sources. In radioisotope assay, about $5.2{\sim}16%$ of glucose was to be incorporated in ganoderan of the mycelium. The monosugars of these ganoderan were mainly consisted of glucose, mannose, galactose. The galactose was not good carbon source for growing the mycelium but the best carbon source for producing the potentialized-ganoderan on the antitumor and anticomplementary activity. The tumor inhibition ratio of ganoderan-GAL, obtained from galactose medium, was 83.6% at the dose of 20 mg/kg/day. This crude polysaccaride was composed of five monosaccharide and the protein contained 16 amino acids. Also, ganoderan-GAL increased the anticomplementary activity than that obtained from any other media. This fact suggests that the structural differences of ganoderan influence the antitumor and anticomplementary activity.

  • Neutral salt soluble [0.9% NaCl (Fr. NaCl)], hot water soluble (Fr. HW) and methanol soluble (Fr. MeOH) materials were extracted from Paecilomyces sinclairii. In vitro cytotoxicity tests. Fr. HW was not cytotoxic against MH3T3, Sarcoma 180 and HepG2 cancer cell lines at the concentration of $0{\sim}2,000\;{\mu}g/ml$, while HT-29 cell line was cytotoxic at the concentration of $100\;{\mu}g/ml$. Fr. NaCl and Fr. MeOH were slightly cytotoxic to the cell lines. Intraperitoneal injection with Fr. NaCl and Fr. MeOH exhibited antitumor activity with life prolongation effect of 32.3% in mice inoculated with Sarcoma 180. Fr. NaCl improved proliferation of spleen cells and the immunopotentiation activity of B lymphocyte by increasing spleen cell numbers and the alkaline phosphatase activity by $2.4{\sim}2.6\;and\;2.7{\sim}3.9$ folds, respectively. Fr. NaCl generated $89\;{\mu}M$ of nitric oxide (NO) when cultured with RAW 264.7, a mouse macrophage cell line, at the concentration of $50\;{\mu}g/ml$, while iipopolysaccharide, a positive control, produced $79\;{\mu}M$. The Fr. NaCl showed the hightest antitumor effect and activation of B lymphocytes and macrophage. From these results, antitumor effect of P. sinclaitii was likely due to immunopotentiation activity.

  • An antitumor substance was purified from the culture filtrate of phytopathogenic fungus Alternaria brassicicola SW-3 isolated from soil of a chinese cabbage patch, and its characteristics were investigated. Antitumor activity of A. brassicicola SW-3 was measured by MTT assay. The cytotoxic activity against human cancer cell line was detected in the culture filtrate of A. brassicicola SW-3, but no activity found in mycelium. Antitumor substance was isolated from the culture broth by ethyl acetate extraction and purified by silica gel column chromatography. Structure of the purified compound was analyzed by the instrumental analysis such as $^1$H-NMR, $^{13}$ C-NMR and IR spectroscopy. The purified fungal metabolite of an A. brassicicola SW-3, consists of 11 carbon chain with two hydroxyl groups and two epoxides which is identical to depudecin. The $IC_{50}$/ values of the active compound identified as depudecin were $69\mu$g/mL and $57\mu$g/mL against mouse melanoma B16BL6 cell line, and human hepatoma SK-HEP1 cell line, respectively.

  • The purpose of this study was to investigate the effect of Kamisamchulsamja-tang (KSST) water extract on the antitumor activity. The results were summarized as follows: KSST extract exhibited a weak cytotoxicity against HT1080, A549, SK-OV-3, B16-F10 and SK-MEL-2 cells. KSST extract showed a inhibitoty effect on DNA topoisomerase I from calf thymus in a dose-dependent manner. Also, KSST extract showed antiadhesive effect on HT1080 cells but didn't showed on A549 cells to complex extracellular matrix. In pumonary colonization assay, a number of colonies in the lungs were decreased significantly in KSST treated group as compared with control group. In vitro neovascularization assays, angiogenesis was significantly inhibited in KSST treated group than control group. In CAM assay, KSST extract inhibited angiogenesis significantly at 15㎍/egg concentration as compared with control. From the above results it was concluded that KSST showed antitumor effect through the antimetastatic effect. So it is expected to be clinically helpful on the prevention of metastasis of cancer.

  • To evaluate the antioxidant and potential of Humulus japonicus, total polyphenol and flavonoid content, radical scavenging activities, and antitumor activities were measured. The total polyphenol and flavonoid contents of the methanol extracts from Humulus japonicus were $30.13{\pm}1.13$ and $13.61{\pm}0.49$ mg gallic acid equivalent/g extract, respectively. DPPH radical and hydroxyl radical scavenging activities of methanol extracts of Humulus japonicus were 60% and 35%, respectively. The Humulus japonicus higher activities of anticancer activities on liver cancer cell lines compared to other cancer cell lines.
