Bacillus polymyxa No.26의 생전분 당화형 $\beta$-amylase를 이용한 떡의 노화억제효과

  • Bacillus polymyxa No. 26이 생산하는 생전분 당화형 $\beta$-amylase를 쌀가루 반죽시 첨가하여 45$^{\circ}C$에 5시간 둔 후 증자하여 제조한 절편을 4$^{\circ}C$ 저온실에 40시간 동안 저장하면서 기계적 검사와 관능검사에 의하여 물성과 맛을 측정하고 효소 무 첨가의 경우와 비교하여 노화억제 효과를 조사하였다. 기계적 검사시 효소 무 첨가구의 경우는 40시간까지도 굳지 않아 노화되지 않았으며, 경도, 점착성, 씹힘성에서도 큰 차이를 보였다. 관능검사시에도 경도, 촉촉함성, 단맛에 있어서 효소 첨가효과가 크게 나타났으며 떡의 제조 시 효소의 이용으로 설탕첨가를 생략할 수 있고 소화성과 맛을 좋게하므로서 품질을 향상시킬 수 있었다.

  • The effects of various antimicrobial agents on the production of organic acid and saccharifying amylase by Aspergillus usamii shirousamii $U_2$ were studied and the results are as follows. 1) The production of organic acid and saccharifying amylase was stimulated by using optimum amounts of nitrofurazone and $AF_2$ among several antimicrobial agents. 2) Wheat flour medium containing 50% volumes of 10 ppm $AF_2$ solution gave the maximum yield of organic acid. 3) Wheat bran medium containing 100% volumes of $100{\sim}500ppm$ nitrofurazone solution gave the maximum yield of saccharifying amylase.

Characterization of Cellulases from Schizophyllum commune for Hydrolysis of Cellulosic Biomass (Schizophyllum commune에 의한 Cellulase 생산 및 섬유소계 바이오매스의 당화를 위한 효소적 특성)

  • The optimum culture condition of Schizophyllum commune for the cellulase production and its enzymatic characteristics for saccharification of cellulosic biomass were analyzed. S. commune secrets ${\beta}$-1,4-xylosidase (BXL) and cellulases, including endo-${\beta}$-1,4-glucanase (EG), cellobiohydrolase (CBH), and ${\beta}$-glucosidase (BGL). The optimum reaction temperature for all cellulases was $50^{\circ}C$ and the thermostable range was $30{\sim}40^{\circ}C$C. The optimum reaction pH for all cellulases was 5.5 in a range of temperature from $0^{\circ}C$ to $55^{\circ}C$. The best nutritions for the cellulase production of S. commune among tested nutrients were 2% cellulose for the carbon source and corn steep liquor or peptone/yeast extract for the nitrogen source without vitamins. The environmental culture condition for the cellulase production was 5.5~6.0 for pH at $25{\sim}30^{\circ}C$. The enzyme activities of EG, BGL, CBH, and BXL were 3670.5, 631.9, 398.5, and 15.2 U/$m{\ell}$, respectively, after concentration forty times from the culture broth of S. commune which was grown at the optimized culture condition. Alternative filter paper unit assay showed 11 FPU/$m{\ell}$ enzyme activity. The saccharification tests using cellulase of S. commune showed the low saccharification rate on tested hardwoods but a high value of 50.5% on cellulose, respectively. The saccharification rate (50.5%) of cellulose by cellulase produced in this work is higher than 45.7% in the commercial enzyme (Celluclast 1.5L, 30 FPU/g, glucan).

  • The objective of this study was to investigate the efficient pretreatment method for bioethanol production from rice hull. Ammonia and sodium hydroxide as an alkaline solution and dilute sulfuric acid as an acidic solution were used in a batch reactor under high-temperature and high-pressure conditions. The highest enzymatic saccharification efficiency of 82.8% and ash removal rate of 94.7% were obtained in the dilute sulfuric acid treated sample after the sodium hydroxide solution treatment. The enzymatic saccharification efficiencies and ash removals of pretreated rice hull samples have very similar variation tendency. This means that the maximum obstructive factor for the enzymatic saccharification of rice hull is the ash (silicate) content in biomass. The findings suggest that the combined sodium hydroxide-dilute sulfuric acid treatment system under high-temperature and high-pressure conditions is a promising pretreatment method to enhance the enzymatic saccharification of the silica-rich biomass.

  • The cellulolytic activities of Aspergillus fumigatus KC-1 was investigated, which showed the most active producer of cellulase among the 256 strains of cellulose-decomposing microorganisms screened in our laboratory. All the examined cellulolytic activities (filter paper-, Avicel-, cotton-, CMC-, salicin- and xylansaccharifying activity) in a culture of A. fumigatus KC-1 grown on 1% popular sawdust pretreated with peroxide alkaline reached a maximum within 4-5 days. The optimum pH and temperature for the enzymatic activity was found to be pH 4.5 and $60^{\circ}C$ respectively. The sawdust of poplar wood delignified with 1% NaOH and 20% peracetic acid succesively recorded the highest hydrolysis rate in the tests of enzymatic saccharification. The major end product of hydrolysis of poplar wood with the cellulolytic enzymes obtained from A. fumigatus KC-1 was glucose with small amount of cellobiose and xylose. It can be concluded from these results that A. fumigatus KC-1 is an advantagous source of a cellulase that is capable of hydrolyzing cellulose to glucose rapidly. The influence of degree of delignification, substrate size and its concentration on the rate of hydrolysis of poplar wood was also discussed.

  • 쌀을 주식으로 하는 우리나라의 여건상 연간 추정치로 싸라기 약 12만톤, 미강 약 49만톤, 왕겨 약 79만톤의 벼 도정 부산물이 발생하고 있다. 본 연구에서는 벼 도정 부산물 중 비식량 자원인 왕겨를 대상으로 고효율 효소 당화를 위한 바이오매스 전처리 방법을 탐색하였다. 왕겨 원시료의 초기 조성은 셀룰로스 34.5%, 헤미셀룰로스 20.5%, 리그닌 25.3%, 회분 14.6%로 나타났는데, 억새 등 초본계 바이오매스와 비교하여 특이하게 높은 성분은 회분으로 이는 벼에 대한 규산질 비료의 시용에서 기인한 것이다. 바이오매스 전처리에 많이 사용되는 암모니아, 희황산 용매와 규산염에 침식성을 가지는 가성소다 용매를 이용하여 각 용매별 단독 및 알칼리-산 복합 처리 하였을 때 효소 가수분해 효율, 고상시료 성분변화 등을 상호 비교하였다. 예비실험을 통하여 암모니아 처리조건은 15%(w/w) $150^{\circ}C$ 20분, 가성소다 처리조건은 1.5%(w/w) $150^{\circ}C$ 20분, 희황산 처리조건은 1.0%(w/w) $150^{\circ}C$ 10분으로 설정하였다. 암모니아 단독, 희황산 단독, 암모니아-희황산 복합 처리 시료의 효소 가수분해 효율은 각각 37.8%, 39.1%, 42.8%로 약 40%선에서 큰 차이가 없었다. 반면 가성소다 단독, 가성소다-희황산 복합 처리시료의 효소 가수분해 효율은 각각 62.7%, 82.8%로 나타나 앞선 3가지 처리방법 대비 50%, 100%에 가까운 효소 가수분해 효율 향상을 보였다. 이 때 전처리 고상시료의 성분 변화를 살펴보면 회분 함량에서 큰 차이를 보였는데 암모니아 단독, 가성소다 단독, 희황산 단독, 암모니아-희황산 복합, 가성소다-희황산 복합 처리에서 각각 47.8%, 77.1%, 43.5%, 55.8%, 94.7%의 회분 성분 기각률(rejection rate)을 나타냈다. 이는 왕겨 효소 가수분해 효율의 최대 저해요인이 회분임을 추정할 수 있다. 왕겨 전처리 알칼리 용매는 암모니아보다 가성소다가 더 효과적이었고 희황산 복합 처리시 그 효과가 크게 상승하였다. 따라서 규산염(회분) 함량이 높은 바이오매스는 고온 고압 조건에서 가성소다 용액으로 처리한 후 그 고상분을 희황산 용액으로 복합 처리하는 시스템이 효소 당화 증진에 매우 유리함을 확인하였다.

Preparation and characteristics of yogurt added with enzymatically saccharified Korean rice wine lees powder (효소로 당화시킨 주박 분해물을 첨가한 요구르트의 발효 특성)

  • This study was carried out to determine the effect of enzymatically saccharified Korean rice wine lees powder (eKRWLP) on the quality characteristics and storage stability of curd yogurt. Yogurt with different contents [0.5-2.0% (w/w)] of eKRWLP was incubated with commercially available mixed lactic acid bacteria (Lactobacillus acidophilus, Streptococcus thermophilus, Bifidobacterium longum) at $40^{\circ}C$ for 18 h. The production of acid measured at pH and titratable acidity of yogurts increased with increasing eKRWLP content. After 12 h fermentation, titratable acidity of eKRWLP yogurt was 0.77-0.90% and was higher than that (0.72%) of yogurt made without eKRWLP. The viable cell counts of lactic acid bacteria in eKRWLP yogurts were increased in proportion to the addition of eKRWLP, and increased up to 8.01-8.13 log CFU/g after 12 h incubation. The repressive effect of whey separation in eKRWLP yogurt curd significantly decreased than that in Korean rice wine lees powder (KRWLP) yogurt. With sensory evaluation, yogurt with 0.5% eKRWLP obtained the highest scores among all eKRWLP yogurts. When eKRWLP yogurts and the control preparations fermented for 12 h were incubated at $4^{\circ}C$, their pHs and titratable acidities were slightly changed and the number of viable lactic acid bacteria were well maintained above $10^7CFU/g$ for 16 days.

Enzyme Deactivation During Enzyme Recycling with Ultrafiltration Hollow Fibers (한외여과막을 이용한 효소재순환 시스템에서의 효소역가감소)

  • The enzyme deactivation in a membrane recycling system for the simultaneous saccharification and fermentation(SSF) was studied under various temperature and pressure. The optimum molecular weight cut off(MWCO) of the ultrafiltration membrane for recycling cellulase and ${\beta}$-glucosidase was 50,000. When the cellulase was recycled continuously through the membrane system, it was not deactivated. But the activity of ${\beta}$-glucosidase was decreased with an increase in operating temperature and transmembrane pressure. After 720 minutes at $42^{\circ}C$ and 24.8 psig , the activity of ${\beta}$-glucosidase was reduced by 35% of the initial activity. Such tendencies could be well explained by the results of highly induced shear at the fiber surface of membrane when temperature and transmembrane pressure became higher.

Usefulness of Freshwater Alga Water-net (Hydrodictyon reticulatum) as Resources for Production of Fermentable Sugars (발효 당용액 생산자원으로서 담수조류 그물말의 유용성)

  • To investigate the usefulness of freshwater alga Water-net (Hydrodictyon reticulatum, HR) as resources for production of fermentable sugars, the easiness of enzymatic saccharification was evaluated at first. When 6 plant materials (HR, Spirulina, Chlorella, Scenedesmus, Cladophora, Corn stover) were enzymatically hydrolyzed with 2% solid loading at the same condition, HR showed the highest ratio of saccharification based on glucose production. No milled HR was also completely saccharified at the amounts of optimal enzyme mixture. Glucose yield was not changed though the citrate buffer strength for saccharification was decreased from 0.1 M to 0.1 mM. Only about 10% yield reduction was observed compared to that of $120^{\circ}C$ treatment when HR was enzymatically hydrolyzed at room temperature. The saccharification was normally occurred at $37^{\circ}C$ and pH 6.5 which is general growth condition of fermentable microrganisms, suggesting that HR have a biomass characteristics applicable for the simultaneous saccharification and fermentation. The saccharification was occurred by more than 70~80% of one of the best condition although the supplied enzyme amounts was reduced to 1/10 volume. And the glucose yield by enzymatic hydrolysis was not decreased by 10% HR solid loading and began to decrease at more than 15% solid contents. Above these results show that HR is an interesting algal biomass which is relatively easy to be saccharified by hydrolyzing enzymes. In addition, HR is a flilamentous alga and very easy to be collected. Therefore, HR seems to be an useful and valuable resources in the economical production of fermentable sugars for manufacture of bio-chemical products.
