Growth Inhibitory and Quinone Reductase Activity Stimulating Effects of Internal Organs of Todarodes pacificus Fractions on Human Cancer Cell Lines In vitro (오징어내장 분획물의 in vitro에서의 암세포 성장억제 및 quinone reductase유도 활성 증가 효과)

  • We investigated the growth inhibitory effect of internal organs of Todarodes pacificus (TP) on proliferation in human cancer cell lines in vitro. The internal organs of TP were extracted with methanol (TPM), which was then further fractionated into four subfractions by using a solvent partition method, resulting in hexane (TPMH), methanol (TPMM), butanol (TPMB), and aqueous (TPMA) soluble fractions. We determined the cytotoxic effect of these four fractions in three kinds of cancer cell lines - HepG2, MCF-7 and HT-29 - by MTT assay. Among the four subfractions of TPM, TPMH showed the strongest cytotoxic effects at a concentration of $300{\mu}g$/ml, displaying 91.56% on the HepG2 cell line and 85.93% on the MCF-7 cell line. Morphological changes such as membrane shirinking and blebbing of cells were also observed during TPMH treatment of HepG2 cells. In addition, we also observed quinone reductase (QR) induced effect in the methanol (TPMM) layer of HepG2 cells. TPMM showed the highest induction activity of quinone reductase on HepG2 cells among the other partition layers. The QR induced effect of TPMM was determined to be 2.7 at a level of $360{\mu}g$/ml with a control value of 1.0. Although further studies are needed, the present work suggests that internal organs of Todarodes pacificus (TP) may be a chemopreventive agent for the treatment of human cells.

  • 이 논문은 이소플라본과 골조직의 관계에 대하여 발표된 역학조사, 사람과 동물을 대상으로한 실험, 조직세포연구, 세포배양에서의 연구들을 포괄하고 있다. 가장 중요한 결론은 적절한 양을 투여했을 때 이소플라본, 특히 제니스테인과 다이드제인은 골밀도 증진에 효과가 있다는 것이다. 쥐를 대상으로 한 실험에서 적정 골밀도 향상 효과를 나타내기 위한 이소플라본의 invivo 투여량은 에스트라디올보다 보통 1000배가 높았다. 또한 적정 투여량보다 낮거나 높은 농도에서는 이소플라본은 뼈에 유익한 효과가 없었으며 그 때문에 이중적인 효과를 발휘한다. 유사 조골세포를 배양한 결과 중 in vitro실험에서 제니스테인의 효과는 관찰되었으나 다이드제인의 효과는 나타나지 않았다. 일반적으로 대두와 대두제품에 존재하는 이소플라본은 동물이나 사람이 적정한 양으로 섭취하였을 때 약한 에스트로겐 효과를 발휘하지만 고농도에서는 오히려 해로운 효과가 관찰될 수도 있다. 제니스테인에 대한 세포수준의 연구에서는 고농도 투여시에는 정상적인 세포기능의 소실을 유도하기도 한다고 제안되었으며, 반면 다이드제인은 같은 농도에서 유사 조골세포에 유익한 효과를 발휘한다고 하였다. 사람이 식이로부터 섭취하는 양은 부작용을 초래할 정도가 되기는 힘들다. 따라서 이소플라본은 적정한 양으로 섭취했을 때 사람의 골질량을 유지하거나 골질량을 증가시키는 잠재적인 능력이 있음이 분명하다.

  • This experiment was conducted to investigate the response of the fertilizer three factors on the growth of Alopeuclus aequalis Sobol. var. amurensis(Kom.) Ohwi. The growth of Alopeuclus aequalis Sobol. var. amurensis(Kom.) Ohwi. such as plant height, tiller number and dry matter production were vigorous on the plot fertilized with phosphate(P, NP, PK, NPK). The main effect of plant height elongation and dry matter production were significant for phosphate and nitrogen, and their interaction effect was recognized, too. Tiller growth and panicle number were mainly affected by phosphate. The growth of Alopeuclus aequalis Sobol. var. amurensis(Kom.) Ohwi. depended mostly on phosphate among the fertilizer three factors. The effect of nitrogen fertilizer was very different according to phosphate fertilizer treatment.

The Economic Impact Analysis on the Water Industry with Social Accounting Matrix (사회계정행렬을 이용한 수자원분야 정책 효과 분석)

  • This paper analyses the economic effects of the water industry on the Korean economy by using Social Accounting Matrix (SAM). The SAM is constructed based on the Input-Output table, National account and Family income and expenditure survey for Korea in 2009. Through the SAM multiplier analysis, I estimate the effects of water investment. As the results, this study has found the followings. i) output multiplier effects of water sector are 5.300~7.741, ii) value added multiplier effects of water sector are 0.685~1.158, iii) income multiplier effects of water sector are 0.511~0.984, iv) redistributed income multiplier effects of water sector are -0.096~0.247. The results indicate that a significant influence on the industrial production and the household income in Korea.

Antioxidative Effect of Ginger Extracts (생강 추출물의 항산화 효과에 관한 연구)

  • To investigate the antioxidant effects of different concentration of ginger extracts during incubating and heating, ginger extracts were added into soybean oil, cottonseed oil at 1, 3, 5 percent level and BHT, tocophereol at 0.02% level. Also practical antioxidant effects of dried ginger slices for soybean oil were determined. In case of incubating oil, ginger extracts showed antioxidant effects at all concentration, and the antioxidant effects were similar to those of BHT but considerably higher than those of the tocopherol. In case of heating oil, the antioxidant effects of ginger extracts were almost equal to all concentration and higher than those of BHT and tocopherol. And the rate of oxidation was retarded considerably by heating with the dried ginger slices added into flying oil at 5, 10% level, but the best antioxidant effects was shown in 5% dried ginger slices added into soybean oil.

  • This study was carried out to investigate the anticarcinogenic and antioxidative activities of Hemicentrotus pulacherrimus (HP). HP was extracted with methanol (HPM), which was then further fractionated into four sub-fractions by using the solvent partition method, affording methanol (HPMM), hexane (HPMH), butanol (HPMB) and aqueous (HPMA) soluble fractions. We determined the anticarcinogenic activities of these four fractions in four kinds of cancer cell lines, such as HepG2, HT29, MCF-7 and B16-F10, by MTT assay. Among various fractions from HPM, the HPMH showed the strongest growth inhibition effect. We also determined the inductive effect on quinone reductase (QR) of HP fractions. HPMB fraction exhibited strong inductive effects in HepG2 cells at a level of 90 ${\mu}g/ml$, showing inductive indexes of 2.26 compared to the control value of 1.0. The antioxidant activities of fractions from HP were also investigated by measuring the scavenging activities of HP against reactive oxygen speicies (ROS), peroxynitrite (ONOO-) and NO. Among the various solvent fractions, HPMH fractions displayed marked antioxidative activities.

Inhibitory activities of phenolic compounds isolated from Chionanthus retusa flower on biological enzymes (이팝나무 꽃에서 분리한 페놀 화합물의 생리활성 효소 억제효과)

  • This study was designed to determine the biological activities of Chionanthus retusus flower extracts. Water and 90% ethanol extracts of C. retusus flower were prepared. The inhibitory activities of water and ethanol extracts with a phenolic content of $200{\mu}g/mL$ against xanthine oxidase were 25.60% and 15.92%, respectively. Further, the water extract did not show any inhibitory activity against ${\alpha}$-glucosidase whereas the ethanol extract showed 100.00% inhibition of ${\alpha}$-glucosidase. The inhibitory activities of the extracts against tyrosinase were 17.27% (water extract) and 36.13% (ethanol extract), which suggest that the extracts may have a whitening effect. The water extract did not inhibit elastase activity but showed a collagenase-inhibitory activity of 20.21%. On the contrary, the ethanol extract showed 96.26% and 35.93% inhibition of collagenase and elastase, respectively. These findings suggest that the extracts may have an anti-wrinkle effect. Lastly, the extracts showed a hyaluronidase inhibitory activity of 36.96% (water extract) and 88.70% (ethanol extract), suggesting that they may have an anti-inflammatory effect. The results indicate that C. retusus flower extracts containing phenolic compounds can be used as functional resources because they have anti-gout, carbohydrate degradation-inhibitory, whitening, anti-wrinkle, and anti-inflammatory effects.

저선량 $\gamma$선 조사에 의한 곡물류와 채소류의 생육촉진 효과

  • 작물과 채소류의 생육에 대한 저선량 방사선 조사효과를 보고자 온실과 포장실험을 수행한 결과, 발아율과 유묘초장을 조사한 초기생육의 경우 벼, 콩 및 들깨의 200rad, 400rad, 100rad에서 생육촉진 효과를 볼 수 있었다 포장실험에서 벼의 경우 저선량조사에 의해 수량증가 효과는 없었고 종자의 불임율이 감소하였으며 콩과 들깨의 경우 400rad에서 생육상태와 수량이 다소 양호하여 저선량에 의한 생육촉진 효과가 인정되었다. 배추와 무의 경우는 200rad에서 발아율이 증가하여 저선량조사에 의한 효과를 볼 수 있었고 800rad에서 초장 등이 다소 증가하였으나 뚜렷한 수량증가 효과는 볼 수 없었다.
