Growth Responses of South Korea Three Evergreen Woody Species according to Indoor Light Intensity Conditions (실내 광량조건에 따른 자생 상록 목본 3종의 생육반응)

  • 본 연구에서는 자생 상록 목본 3종의 실내도입을 위하여 몇 가지 광량조건에서의 식물생육 및 광화학 반응을 조사하였다. 식물재료는 황칠나무[Dendropanax trifidus (Thunb.) Makino ex H. Hara], 먼나무(Ilex rotunda Thunb.), 편백[Chamaecyparis obtusa (Siebold & Zucc.) Endl.]의 2년생 실생묘를 유리온실에 재배하면서 실험에 사용하였다. 실내에 조사되는 광환경을 측정하여 광량 10, 50, 100 및 200 PPFD(${\mu}mol{\cdot}m^{-2}{\cdot}s^{-1}$)를 처리구로 설정하였다. 실내환경은 광주기(12/12 h), 온도($25{\pm}1^{\circ}C$) 및 습도($55{\pm}3%$)를 유지하였으며, 1회/3일 간격으로 관수하였다. 유리온실에서 동일기간 동안 재배중인 식물을 대조구로 사용하였으며, 실험은 8주간 수행되었다. 황칠나무와 먼나무는 200 PPFD의 높은 광량에서 초장, 줄기직경, 엽장, 엽수 등의 생육수치가 우수하였다. 반면 두 종 모두 10 PPFD 광량에서는 재배기간 중 잎이 말라가는 현상이 관찰되었으며 8주차에 모든 식물체가 고사하였다. 이는 10 PPFD 광량에서 재배된 황칠나무와 먼나무의 광화학반응에서도 Fv/Fm (-0.10, -0.08) 및 Fm/Fo (0.91, 0.93)가 낮은 수치로 조사된 반응과 일치하였다. 한편 편백나무는 모든 광량조건에서 대조구에 비해 초장 및 줄기직경 등의 생육이 우수하였으나, 10 PPFD의 낮은 광량에서는 Fv/Fm (0.16) 및 Fm/Fo (1.60)가 타 처리구에 비해 낮은 수치로 조사되었다.

Effects of Dendropanax morbifera Leaf Extracts on Lipid Profiles in Mice Fed a High-Fat and High-Cholesterol Diet (황칠나무 잎 추출물이 고지방·고콜레스테롤 식이를 급여한 마우스의 지질 개선 효과에 미치는 영향)

  • The purpose of this study was to determine the effects of dried Dendropanax morbifera leaf extracts on lipid profiles of mice fed a high-fat and -cholesterol diet (HFCD). ICR mice were divided into six groups based on mice fed AIN-93G diet (Normal), HFCD (Control), HFCD+100 mg/kg/d of D. morbifera leaf aqueous extract (DA-100), HFCD+200 mg/kg/d of D. morbifera leaf aqueous extract (DA-200), HFCD+100 mg/kg/d of D. morbifera leaf ethanol extract (DE-100), or HFCD+200 mg/kg/d of D. morbifera leaf ethanol extract (DE-200) for 7 weeks. The final body weights of mice fed D. morbifera extracts were all lower than those of the control group. Mice treated with D. morbifera extracts showed significantly reduced plasma and hepatic triglyceride (TG), total cholesterol (TC), and low-density lipoprotein cholesterol levels, along with increased plasma high-density lipoprotein cholesterol level. Fecal TG level was higher in DE-100 and DE-200 groups and TC level was significantly higher in the DA-200 and DE-200 groups. Relative liver weight, spleen weight, and testicle fat weight in mice treated with D. morbifera were reduced compared to the control group. Plasma insulin, aspartate transaminase, and alanine transaminase levels of experimental groups were also lower than those of the control group. All mice treated with D. morbifera extracts had lower malondialdehyde (MDA) content and higher superoxide dismutase (SOD) activity than the control group. Particularly MDA levels of the DA-200 and DE-200 groups and SOD levels of the DE-200 group were identical to levels of the normal group. These results suggest that D. morbifera extracts have lipid improvement effects in mice fed a HFCD.

Antioxidant, Alcohol Metabolizing Enzyme, and Hepatoprotective Activities of Dendropanax morbifera Water Extract (황칠나무 물 추출물의 항산화, 알코올 대사 효소 및 간 보호 활성)

  • The leaves, stems, seeds, and roots of Dendropanax morbifera have been used since ancient times as folk medicines for the treatment of headaches, skin diseases, infectious diseases, and other ailments. This study investigated the antioxidant, alcohol metabolism, and hepatoprotective effects of D. morbifera leaf and stem water extracts. The total polyphenol content of the D. morbifera leaf and stem water extracts was 49.56 mg tannic acid equivalent (TAE)/g, and the 1,1-diphenyl-2-picrylhydrazy (DPPH) radical scavenging activity of the D. morbifera leaf and stem water extracts was 84.09% at 1,000 ㎍/ml concentration. The effects of D. morbifera leaf and stem water extracts on alcohol metabolism were determined by measuring the generation of reduced nicotinamide adenine dinucleotide (NADH) by alcohol dehydrogenase (ADH) and acetaldehyde dehydrogenase (ALDH). The ADH and ALDH activities of D. morbifera leaf and stem water extracts were increased in a dose-dependent manner at 37.68% and 41.67%, respectively, at a 1,000 ㎍/ml concentration. The D. morbifera leaf and stem water extracts showed significant protective effects against tacrine-induced cytotoxicity in HepG2 cells at 50 ㎍/ml. Based on our results, we concluded that D. morbifera leaf and stem water extracts may be used as major pharmacological agents, such as antioxidants, alcohol metabolism, and anti-hepatitis remedies.

Cutting Propagation of Dendropanax morbifera $L_{EV}$. (황칠나무 삽목번식에 관한 연구)

  • This experiment was carried out to establish cutting propagation method of dendropanax morbifera $L_{EV}\$. at Wando in Chonnam, native area. The hardwood cutting and the greenwood cutting were able to be used as propagation method, but callus formation and rooting ratio in the greenwood cutting were higher than in hardwood cutting. The optimum cutting time was February to middle of March in hardwood cutting and July to August in greenwood cutting. The earthen-ball cutting method was better than normal cutting method in callus formation and rooting ratio. The rooting in different bed soils was the best at sand-loam soil. The application of IBA 100ppm promoted rooting.

Effects of Temperature Stress and Paraquat on SOD Activity and Photochemical Efficiency of PSII in Leaves of Araliaceae Plants (두릅나무과 식물의 SOD 활성과 광계II의 광화학적 효율에 미치는 온도 스트레스와 Paraquat의 영향)

  • The effects of temperature stress and paraquat on the superoxide dismutase (SOD) activity and the photochemical efficiency of photosystem II were studied in the leaves of Araliaceae plants. The SOD activity of Acanthopanax koreanum leaf discs increased at 4$^{\circ}C$ and 28$^{\circ}C$, and increased significantly at 4$^{\circ}C$ ,28$^{\circ}C$ and 35$^{\circ}C$ in the presence of paraquat. However, the SOD activity of Dendropanax morbifera leaf discs decreased at 4$^{\circ}C$, 28$^{\circ}C$ and 35$^{\circ}C$ regardless of paraquat treatment. The photochemical efficiency of photosystem II, Fv/Fm, of leaf discs of A. koreanum and D. morbifera fell remarkably at 35$^{\circ}C$. In the presence of paraquat, the Ev/Fm values fell slightly at 4$^{\circ}C$ in A. koreanum leaf discs and at 35$^{\circ}C$, in D. morbifera leaf discs. These results indicate that A. koreanum plants are more resistant to temperature stress or oxidative stress than D. morbifera plants although their photochemical efficiency falls slightly at 4$^{\circ}C$ in the presence of paraquat.

Growth Characteristics and Native Environment of Dendropanax morbifera LEV. in Wando, Korea (완도(莞島)지역 황칠나무의 자생지 환경 및 생육특성)

  • This environment of native area and growth characteristics by different age of dendropanax morbifera was grown wild widly at Wando area, particularly, distribution frequency was high at 50 to, 150 meters above the sea level, and configuration of the ground of native area descended slowly toward the southeast. The Soil characteristics of native area was $pH\;5.3{\sim}5.4,\;8.4{\sim}9.8%$ in organic matter content and $18.2{\sim}21.3%$ in soil moisture. The flowering of dendropax morbifera was began at 6 years old tree, and flowering time was at middle of July. The growth characteristics of 6 years old tree was 129cm in stem height, 34mm in stem diameter, 15.4 in number of leaf, and 12.9 cm in petiole length.

Secretion Stimulation of Golden Vernish Sap from Dendropanax morbifera Lev. (황칠(黃漆)나무의 칠액(漆液) 분필(分泌) 촉진(促進))

  • The stimulated secretion of golden varnish sap by the infection of microorganisms on the bark of Dendropanax morbifera Lev. was observed in the natural habitats. Four candidate strains of such microorganisms(i.e., Sb1, Sb2, Sb3 and Sb4) were isolated from the bark of D. morbifera which was infected in natural. Of them, Sb3 and Sb4 strains showed stimulated secretion of golden vernish sap with the greatest stimulation by Sb4 strains. The yield of golden varnish sap from the bark of D. morbifera was boosted about 3.4~7.7 times by the inoculation of Sb4 strain in compassion with that collected from lacquer tree(Rhus verniciflua Stokes) by the traditional method. The mycelium color of Sb4 strain was grayish white. The optimum growth temperatures and pH were ranged from 30 to $35^{\circ}C$ and from pH 6.5 to 7.0.

Protective Effect of Dendropanax morbifera Leaf Extract on CCl4-induced Oxidative Damage in HepG2 Cells (사염화탄소로 유발된 산화적 손상에 대한 황칠나무 잎 추출물의 간세포 보호 효과)

  • This study aimed to investigate the hepatoprotective effect of Dendropanax morbifera (D. morbifera) leaf hot-water extract on carbon tetrachloride (CCl4)-treated HepG2 cells. Treatment with D. morbifera leaf hot-water extract increased the cell viability of CCl4-treated HepG2 cells without inducing cytotoxicity. The levels of alanine transaminase (ALT) and aspartate transaminase (AST) released by CCl4-treated cells were 27.6 U/L and 52.4 U/L, respectively, and were significantly higher than those in untreated control cells (10.0 U/L and 15.2 U/L, respectively). Moreover, the level of γ-glutamyl transpeptidase (GGT) was 5.4 times higher, while that of glutathione was 44.0% lower in CCl4-treated cells than in control cells. However, treatment with D. morbifera leaf hot-water extract resulted in a dose-dependent decrease in the levels of ALT, AST, and GGT, and an increase in the level of glutathione. Moreover, the malondialdehyde (MDA) content in CCl4-treated HepG2 cells was effectively reduced after treatment with D. morbifera leaf hot-water extract. Additionally, overproduction of intracellular lipids induced by CCl4 treatment was effectively inhibited by D. morbifera leaf hot-water extract treatment. Furthermore, DCFDA staining showed that overproduction of reactive oxygen species (ROS) induced by CCl4 treatment was effectively reduced by treatment with D. morbifera leaf hot-water extract. Our results indicate that owing to its beneficial effects, D. morbifera leaf extract has considerable potential as a functional food material for liver protection.

The Effect of Mulching Materials on Agronomic Chracteristics of Dendropanox morbifera LEB. In Southern Area of Korea (남부 도서지역에서 황칠나부의 주요형질에 미치는 멀칭재료의 효과)

  • This study was carried out to obtain basic data for technical inprovement about cultivation according to micrometerological changes under various mulching materials in Dendropanox morbifera LEV. The results are summarized as follows. Transparent polyethylene film and black polyethylene film mulching materials increased the soil porosity and reduced change of the soil moisture content. The rate of missing plant during overwinter increased in the control plot. Growth of Dendropanox morbifera LEV, was accelerated by transparent polyethylene film and black polyethylene film mulched.
