Chemical Compositions and Antioxidative Activities of Sweet Potato Foliages Harvested by the Cultivation Period and Tips Location (재배기간 및 마디별 고구마 끝순의 성분과 항산화 활성)

  • The foliage of sweet potato [Impomoea batatas (L.) Lam] is an excellent source of food material due to the functional components of polyphenol and carotenoid. In this study, the lightness (L-value), polyphenol contents, and DPPH radical scavenging activity of the foliage of Shinwhangmi, Hayanmi and Suioh cultivars harvested after different cultivation periods (60, 90 and 120 days after planting) and according to tip locations (1st~5th, 6th~10th, 11th~15th tips) were investigated. As a result, Shinwhangmi showed a lower lightness value (60 days after planting and 1st~5th tips). The longer the cultivation periods and the closer the tips, the higher the contents of polyphenol and DPPH radical scavenging activity were in lyophilized foliage of Shinwhangmi, Hayanmi and Suioh. In the 1st~10th tip of Shinwhangmi, Hayanmi and Suioh cultivated for 90 days and dried by 2 step hot-air (1st: $70^{\circ}C$ for 8 hr, 2nd: $80^{\circ}C$ for 4 hr), the polyphenol and ${\beta}$-carotenoid contents were 5.0, 3.9 and 4.0 mg/g and 75.6, 71.6 and 63.1 mg/g, respectively. The DPPH radical scavenging activities and ABTS radical scavenging activities ($EC_{50}$) were 0.29, 0.36 and 0.33 mg and 0.12, 0.15 and 0.11 mg, respectively. These antioxidative activities were over two times stronger than spinach.

Antioxidant and Nitrite Scavenging Activities of Acanthopanax senticosus Extract Fermented with Different Mushroom Mycelia (버섯균사체를 달리한 발효가시오가피 추출물의 항산화 활성 및 아질산염 소거능)

  • This study was designed to investigate the antioxidant activities (ORAC value, DPPH and ABTS radical scavenging activity, FRAP activity, and reducing power) and nitrite scavenging activities of Acanthopanax senticosus extracts fermented with the mycelia of three different mushroom species: Ganoderma lucidum, Phellinus linteus, and Hericium erinaceus. The highest total phenol content (42.09 GAE mg/g) and ORAC value ($74,912{\mu}M$ TE/g) were observed in a hot water extract of A. senticosus fermented with G. lucidum. The highest DPPH radical and nitrite scavenging activities were observed in a 70% ethanol extract of A. senticosus fermented with G. lucidum. In addition, ABTS radical scavenging activity (8-88%), FRAP activity (0.1-0.2), and reducing power (0.3-0.7) were increased by ethanol addition in all samples in a dose-dependent manner. These results provide a basic understanding of the antioxidant and nitrite-scavenging activities of A. senticosus extracts fermented with different mushroom mycelia.

Preparation and Quality Characteristics of Makgeolli Made with Black Garlic Extract and Sulgidduk (설기떡을 이용한 흑마늘 막걸리의 제조와 품질 특성)

  • We conducted this study to develop a high value black garlic Makgeolli that was made of black garlic extract (BGE) and Sulgidduk. We investigated the quality characteristics of Makgeolli made with three different combinations of materials (control, Sulgidduk only; A, Sulgidduk combined with 15% BGE and water; B, Sulgidduk combined with 15% BGE instead of water). The pH of A and B were higher than the control, but the titratable acidity of A and B were lower. The sugar and alcohol contents of A and B increased during fermentation. A similar growth pattern was observed invisible cells, yeast, and lactic acid bacteria in all three Makgeolli. In A and B, the quantity of lactic acid bacteria was relatively higher than the yeast. The L value (lightness) was highest in the control, and the a value (redness) and b value (yellowness) were higher in A and B. The antioxidant properties of the three types of Makgeolli were evaluated using DPPH (1,1-diphenyl-2-picrylhydrazyl) and ABTS (2,2'-azino-bis[3-ethylbenzthiazoline-6-sulfonic acid]) radical scavenging activities. In these assays, B showed significantly higher radical scavenging activities than the other two Makgeolli.

Antioxidant Contents and Antioxidant Activities of Hot-Water Extracts of Aronia (Aronia melancocarpa) with Different Drying Methods (건조방법에 따른 아로니아(Aronia melancocarpa) 열수 추출물의 항산화 성분 함량 및 항산화 활성)

  • This study determined the antioxidant levels and activities of hot water aronia extracts by different drying methods such as sun drying, sun drying after steam treatment, freeze-drying, and oven drying. The total polyphenol content, calculated as gallic acid equivalent, was the highest in the freeze-dried sample (910 mg), followed by sun-dried after steam treatment (779 mg), sun-dried (769 mg), and oven-dried (757 mg) samples. Similar patterns were observed for the total flavonoid and anthocyanin contents. Freeze-dried aronia samples contained the highest polyphenol, flavonoid, and anthocyanin contents as compared to the samples dried by other methods. All antioxidant activities were found to increase in a dose-dependent manner. For the hot water-extracted freeze-dried aronia powder (200 mg/mL), the 1,1-diphenyl-2-picrylhydrazyl (DPPH) and 2,2'-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid) diammonium salt (ABTS) radicalscavenging activities were 65.5% and 61.7% and the hydroxyl and superoxide anion radical-scavenging activities were 50.5% and 52.1%, respectively. These results suggest that comparatively, freeze-drying is a better method for preserving the bioactive components and the antioxidant activities of aronia.

Antioxidative Activities and Inhibitory Effects on Tyrosinase and Elastase by Water Extracts of Allium sp. (Allium 속 식물 물 추출물의 항산화 활성과 Tyrosinase 및 Elastase 저해 효과)

  • Three different Allium plants, including wild garlic (Allium monanthum Max; AMM), onion (Allium cepa L.; ACL) and green onion (Allium fistulosum L; AFL), have been widely used as spicy food in daily life. This study was attempted to investigate the properties of antioxidant, whitening and anti-wrinkle activities in water extracts in Allium plants. The antioxidant, whitening and anti-Wrinkle activities were evaluated by performing DPPH and ABTS radical scavenging activities, inhibitory effects on tyrosinase, and elastase, respectively. The water extract (10 mg/mL) of onion, wild garlic and green onion exhibited DPPH radical scavenging activities of 28.46%, 28.45%, and 15.91%, respectively. The inhibitory effects of tyrosinase activities by wild garlic, green onion, and onion were increased by heating the extracts at $90^{\circ}C$. Additionally, the wild garlic extract showed higher elastase inhibitory activity than those of other plant extracts. These results suggest that water extracts of Allium plants may be useful as potential sources of beauty foods due to higher antioxidant, anti-melanin and anti-wrinkle activities.

Antioxidant Activity of Honeydew Honey Produced by Apis mellifera L. (양봉꿀벌이 생산한 감로꿀의 항산화 활성)

  • To evaluate biological activity of honeydew honey produced by Apis mellifera L. in Korea, we measured antioxidant activity by using DPPH (1,1-diphenyl-2-picryl hydrazyl), ABTS [2,2-azobis(3-ethylbenzothiazoline-6-sulfonate)], FRAP (Ferric reducing antioxidant power), and total polyphenol content assays. Korean honeydew honey respectively scavenged 26% and 86% of DPPH and ABTS radicals at the highest concentration of 10 mg/mL. In the result of FRAP assay, Korean honeydew honey showed activity (126 µM of FRAP value) to reduce Fe3+ to Fe2+. Total polyphenol content was 73.41 mg GAE/kg. Korean honeydew honey exhibited excellent antioxidant activity due to having high radicals scavenging ability and reducing power of ferric ion as well as the presence of phenolic compounds. These findings suggest that Korean honeydew honey has great potential as a functional food material.

Antioxidant Activity of Korea Traditional Fermented Sauces Made with Asparagus (아스파라거스를 이용한 전통장류의 항산화 효과)

  • Asparagus have anti-cancer, antioxidant, and anti-inflammatory properties. Since the high functionality of Korean traditional sauces was discovered, various studies have been conducted using asparagus. In this study, the antioxidant activity of traditional sauces containing asparagus was confirmed. The antioxidant capacity of asparagus was independent of the concentration and showed the highest activity at 70 mg/ml. For instance, concerning Meju containing asparagus, high antioxidant activity was observed in water and ethyl acetate extracts; the total polyphenol content was equivalent to 1.4 mg/g and 0.7 mg/g of tannic acid in the water and ethyl acetate fractions respectively. The DPPH and ABTS radical scavenging activities were 27.6% and 9.04%, respectively, in the water layer, and 10.7% and 52.4%, respectively, in the ethyl acetate layer. Additionally, the antioxidant activity of asparagus-containing traditional sauces was measured using 95% ethanol extracts; soy sauce showed the highest activity, retained in a wide range of concentrations.

Inhibitory Effect of Red Bean (Phaseolus angularis) Hot Water Extracts on Oxidative DNA and Cell Damage (팥(Phaseolus angularis) 열수 추출물의 산화적 DNA와 세포 손상 억제 효과)

  • In this study, we evaluated the protective effects of the hot water extract from red bean (Phaseolus angularis) against oxidative DNA and cell damage induced by hydroxyl radical. The antioxidant activities were evaluated by hydroxyl radical and hydrogen peroxide scavenging assay, and $Fe^{2+}$-chelating assay. Although the extract with hot water didn't scavenge the hydroxyl radical, it removed and chelated hydrogen peroxide and ferrous iron necessary for the induction of hydroxyl radical by 71% and 64% at 200 ${\mu}g/ml$, respectively. Its protective effect on oxidative DNA damage was carried using ${\Psi}$X-174 RF I plasmid DNA comparing the conversion level of supercoiled form of the plasmid DNA into open-circular form and linear form and the expression level of phospho-H2AX in NIH 3T3 cells. In ${\Psi}$X-174 RF I plasmid DNA cleavage assay, it inhibited oxidative DNA damage by 96% at 200 ${\mu}g/ml$. Also, it decreased the expression of phospho-H2AX by 50.1% at 200 ${\mu}g/ml$. Its protective effect against oxidative cell damage was measured by MTT assay and the expression level of p21 protein in NIH 3T3 cells. In MTT assay for the protective effect against the oxidative cell damage, it inhibited the oxidative cell death and the abnormal cell growth induced by hydroxyl radical. Also, it inhibited p21 protein expression by 98% at 200 ${\mu}g/ml$. In conclusion, the results of the present studies indicate that hot water extract from red bean exhibits antioxidant properties and inhibit oxidative DNA damage and the cell death caused by hydroxyl radical.

Antioxidative Activity of Mushroom Water Extracts Fermented by Lactic Acid Bacteria (유산균 발효에 의한 버섯 추출물의 항산화 활성)

  • This study was focused on the development of fermented mushroom water extracts with antioxidative activities. Mushroom water extracts were fermented with Bifidobacterium bifidum, Lactobacillus plantarum, Lactobacillus acidophilus, Leuconostoc lactis, Streptococcus thermophilus and Lactobacillus sakei subsp. LI033 was isolated from kimchi. Fermented mushroom water extracts increased DPPH and ABTS radical scavenging activities in a dose-dependent manner. However, radical scavenging activity of fermented Phellinus linteus and Ganoderma lucidum water extracts was decreased compared to non-fermented mushroom water extracts. Antioxidative activity of fermented mushroom water extracts was also confirmed by xanthin oxidase (XO) inhibition and superoxide dismutase (SOD) activities at the same concentration. As the fermentation progressed, fermented mushroom water extracts increased XO inhibition activity and SOD activity. In conclusion, fermented mushroom water extracts were tentatively identified to enhance enzyme activity.

Antioxidant activities from the Different Parts of Artemisia argyi H. using an in vitro System (In vitro 시스템에서 남해 약쑥(Artemisia argyi H.)의 부위별 항산화 활성)

  • The antioxidant activities of 60% ethanol extracts from different parts (leaf, stem, and root) of Artemisia argyi using an in vitro system were examined to find a possibility as natural antioxidant substances. The highest total phenolics (23.08 mg/g) was obtained by 60% ethanol extract of leaf. The scavenging activity of 60% ethanol extract from different parts of A. argyi on the DPPH and ABTS radical was raised with increasing amount of extracts, and 60% ethanol extract from leaf showed the highest radical scavenging activities. The leaf extract presented the highest reducing power and strong inhibitory effect on autooxidation of linoleic acid. In MDA formation using mouse brain homogenates, the 60% ethanol extracts of A. argyi also exhibited antioxidant activity as inhibition of MDA (71.38% at $200{\mu}g/mL$). Therefore, these results suggest that the 60% ethanol extract of A. argyi leaf possess excellent antioxidant activities and thus it has great potential as a source for natural antioxidant.
