Effect of Antimicrobiotic of Opuntia ficus-indica on Surface Disinfection (제주도 손바닥선인장 추출물의 표면소독제로써의 항균효과)

  • Purpose: To verify the anti-microbiotic activity and to develop eco-friendly surface disinfectants using the Opuntia ficus-indica extract. Methods: The stem and fruits of the Opuntia cultivated in Jeju Island were extracted with hot water, 70% EtOH and 100% MeOH, respectively. To examine the antimicrobial activity of the extracts, gram positive bacteria (Bacillus subtilis, Staphylococcus aureus), gram negative bacteria (Escherichia coli, Salmonella typhimurium), yeast (Candida albicans), mold (Aspergillus flavus) and antibiotic resistant bacteria (E. coli, S. aureus) were measured to form a clear zone. Results: The antibacterial activity of the fruit extract was higher than that of the stem extract, and the antibacterial activity of the 100% MeOH (FM) extract and the 70% EtOH (FE) extract of the palm cactus fruit was suitable for the surface disinfectant overall, and clear-zone formed even at relatively low concentrations (250 mg/mL). The FM and FE have antibacterial activity against various bacteria, therefore they can be used as disinfectants. Conclusion: The antimicrobial activity of Opuntia ficus-indica extract from Jeju Island by stem and fruit was determined at relatively low concentrations (250 mg/mL). The Opuntia ficus-indica extract will be useful for surface disinfectants dissolving the patients' bath, oral gargling, and powdered powder in water or ethanol in appropriate concentrations.

Knowledge on the chemical disinfectants among dental workers in some Daejeon region (대전 일부 지역 치과종사자의 화학소독제에 관한 지식도 연구)

  • The usage of appropriate disinfectants is essential for infection control in dental hospitals, dental clinics. Inadequate use of disinfectants is the cause of human or environmental toxicity and is a waste of cost. This study was aimed to assess the level of knowledge on the disinfection and chemical disinfectants among dental workers in dental hospitals, dental clinics. It's ultimately intended to serve as a basis for the preparation of more effective, appropriate educational programs on the disinfection and manuals on the use of disinfectants for dental workers. This study enforced a questionnaire with 184 dental workers employed in dental hospitals, dental clinics from 15 Aug. to 15 Sep. 2007, in the area of Daejeon. The obtained result were as follows; 1. The population sociological feature of dental hospital and dental clinic showed that significant differences of dental service career(pM0.039). Dental service career of dental workers shows; below 3 years 42.9%, 4~6 years 34.7%, more than 7 years 22.4% in dental hospitals, below 3 years 30.4%, 4~6 years 26.7%, more than 7 years 43.0% in dental clinics(pM0.039). 2. The average score of dental workers knowledge in 'Critical item soaks in high-level disinfectants for 20minutes was 2.73V0.49 point, got from knowledge of dental instrument is appropriate to immerse before sterilization in the dental device disinfection(pM0.002). 3. In the general disinfection which it follows in education experience of chemical disinfectants direction for use, 'Direction for use by Spaulding process classification' responded that the correct answer was the education experience dental workers 60.0%, the education non-experience dental workers 39.5%(p=0.026). 4. In the dental device disinfection which it follows in education experience of chemical disinfectants direction for use. 'High level disinfection is not applied for the non-critical items and equipment' responded that the education experience dental workers 49.2%, the education non-experience dental workers 31.9%(pM0.045), 'Semi-critical items is applied same method in presence of the infection disease which it acts responded that the education experience dental workers 44.6% answer back, the education non-experience dental workers 24.4%(pM0.017). 5. 'A hand disinfectants of anticeptics have effect' the education experience dental workers 78.5% answer back, the education non-experience dental workers 52.9%(pM0.003). 1t uses with hand disinfectants when the instruments which be imbrued and patient contact', the education experience dental workers 78.5% answer back, the education non-experience dental workers 62.2%(pM0.026), 'Boric acid solution uses for the skin disinfectants the education experience dental workers 52.3% answer back, the education non-experience dental workers is 37.0%(pM0.016), 'Gluconate have effective difference which it follows in chemical disinfectant consistency and the solution type' education experience dental workers 72.3% answer back, education non-experience dental workers 47.9%(pM0.004). 6. The education experience dental workers were appeared higher than the education non-experience dental workers in knowledge of the disinfection and chemical disinfectants. Consequently system and the specialty education which is standardized continuously must be provided to all dental workers.

The Infection Control of Dental Impressions (치과용 인상체의 감염 관리)

  • The characteristics of dental treatment makes the dental staff frequently contact with patient directly. Also the daily use of high-handpieces, sharp instruments, and needles often causes bleeding on oral cavity. Therefore, the risk of cross transmission grows up. The pathogen from dental practice could spread on not only the practice itself but also the dental laboratory with contaminated impressions and prosthesis. Dental clinic staffs (dentists, dental hygienists and dental technicians) should recognize all the patients have a possibility of cross contamination, and try to prevent the transmission of infection by proper infection control. In this study, we review the articles about disinfection methods and chemical infection agents used for dental impressions, and try to figure out the suitable and effective infection control system of dental impressions.

  • The use of electric field has been studied as an alternative for biofilm control dominated by disinfectants and antibiotics. This technology would be advantageous in the environmental respect that biofilm can be controlled based on electron transfer, not using chemical disinfectants and antibiotics. Control mechanisms which were reported by earlier studies are organized as; (1) bacterial adhesion control by electrostatic repulsion at a negative current, (2) bacterial adhesion control using bacterial motion and (3) bacterial inactivation by direct oxidation at a positive current, (4) bioelectric effect leading to biofilm inactivation. In this review article, we summarized the technologies for biofilm control using electric field and provided some application examples from previous studies.

  • This study aims to investigate the effects of hydrogen peroxide and grapefruit seed extract used as a chemical and natural disinfectants on human conjunctival cells in vitro. The main component of grapefruit seed extract is a narigin. It is one of the flavonoid types in citrus fruits and f1avonoids are widely recognized as naturally occurring(삭제) antioxidants. Cytotoxicity was determined by mitochondrial activity(MTT assay) and DNA damage was analyzed by measuring Comet assay. In LDH assay, 5% of grapefruits seed extract has been observed as a material is giving recovery effect of damaged cultured conjuctival cells by hydrogen peroxide. And also, each of concentrations has been treated simultaneously with same amounts and cytotoxicity of hydrogen peroxide and grapefruit seed extract have been estimated by LDH leakage assay after 24 hours. In conclusion, H2O2-induced cytotoxicity, apoptosis were Significantly prevented by grapefruit seed extract. It is a main component of bioflavonoids that we can simply take it as food. The present results suggest that grapefruit seed extract is a useful disinfectanct having antioxidant and antiapoptopic activity as a natural product.

Susceptibility of Foodborne Pathogens Isolated from Fresh-Cut Products and Organic Vegetable to Organic Acids and Sanitizers

  • In this study, we evaluated the ability of various disinfectants to suppress the growth of microorganisms in fresh-cut products and organic vegetable. The growth of more than 50% of B. cereus isolates were suppressed by 50% ethanol, 0.1% hydrogen peroxide, 0.4% sodium hypochlorite or 1% calcium oxide. E. coli generally showed high susceptibility to concentration of 10% ethanol, 0.4% sodium hypochlorite and 1% calcium oxide. Eighty percent or more of S. aureus isolates exhibited resistance to ethanol, hydrogen peroxide and sodium hypochlorite, but the isolates were susceptible to concentrations of 1% calcium oxide. All isolates evaluated in this study were sensitive to benzalkonium chloride (BAC) and growth in the presence of $2.0{\mu}g/mL$ of BAC was completely inhibited. These pathogens showed widely different susceptibilities to different organic acids. Greater than 0.5% acetic acid and 2% and higher concentrations of malic acid and tartaric acid inhibited the growth of 60% of the isolates of B. cereus. Two percent acetic acid and tartaric acid inhibited 50% of the S. aureus isolates. Seventy percent of the E. coli isolates were resistant to malic acid and susceptible to 1% acetic acid and 10% tartaric acid. The antibacterial effects of the various sanitizers evaluated in this study were not only dependent on the type of disinfectant but also on the pathogen. Thus, it is important to select a sanitizer that is safe and effective at removing specific types of microorganisms.

  • 축산폐수, 침출수 등의 고농도 폐수를 생물학적으로 처리할 경우 최종 방류수는 강한 색도를 띠며 고분자량의 유기물질을 다량 함유한다. 이는 생물학적으로 분해하기 어려운 유기성 복합체와 생화학적 반응에 의한 중간생성물로 색도를 띠는 천연유기물질(NOM)을 포함한다. 생물학적 처리수의 색도는 심미적인 불안감, 방류수역의 수질오염 및 공중보건상의 잠재적 위해성을 갖는다. 또한, 수자원 이용측면에서 정수처리공정에서의 약품투입량 증가와 특히, 소독부산물 생성이라는 잠재적 문제점이 뒤따른다. 따라서 이러한 문제점을 해소하기 위한 생물학적 2차 처리수의 후속처리가 요구되며, 실제로 난분해성 유기물과 색도를 제거하기 위한 흡착, 막 분리, 고급산화(AOP) 및 화학적 응집 등의 물리-화학적 공정에 대한 연구가 수행되어왔다. 특히, 화학적 응집은 무기응집제 또는 고분자중합체(Polymer)를 이용하여 콜로이드성 입자와 색도를 띠는 난분해성 유기물을 전기적 불안정화를 유도함으로서 흡착 및 응집과정을 통해 제거하는 공정으로 많은 연구자들에 의해 연구되어왔다. 그러나 난분해성 유기물과 색도제거는 대상원수의 성상과 화학적 특성 등에 따라 각각의 제거효율과 최적 운전조건이 상이하게 나타난다. 화학적 응집공정은 비교적 높은 제거효율을 보이지만, 운전 및 유지관리의 기술적 어려움, 경제적 비효율성 등으로 인하여 적용에 어려움을 겪고 있는 실정이다. 본 논문에서는 생물학적 혐기-호기성 공정에서 방류되는 축산폐수의 2차 처리수를 대상으로 화학적 응집에 의한 색도 및 난분해성 유기물의 제거거동을 고찰하였다. 대상 처리수의 $TCOD_{Cr}$ 농도는 평균 410 mg/L인 반면, $BOD_5$는 7-15 mg/L 범위로 난분해성 유기물을 다량 함유하고 있음을 알 수 있었다. 이에 황산알루미늄(Aluminium sulfate; $Al_2(SO_4){\cdot}14H_2O$)과 염화철(ferric chloride)의 무기응집제를 이용하여 자 테스트(jar test)를 수행한 결과, 동일한 응집제 주입량에서 염화철의 유기물 제거 효율이 높은 것으로 나타났다. 황산알루미늄과 염화철의 경우 각각의 응집제 주입율 5.85mM에서 89%, 7.03mM에서 97.5%의 최대 유기물 제거효율을 보여주었으며, 이 때 최종 pH는 4.0-5.6 범위이었다. 한편, 대상 원수 내의 콜로이드성 입자 또는 용존성 유기물의 작용기(functional group)는 일반적으로 음으로 하전 되어 있어 응집에 의해 잘 제거되지 않는 특성을 가지고 있다. 따라서 과량의 응집제를 주입하여 다가의 양이온성 금속염을 흡착시켜 전기적으로 중화시키고, 생성된 침전성 수화물 내에 포획 또는 여과시켜 제거하게 된다. 이 때, 금속염 수화종의 전하밀도가 응집효율에 영향을 주는 것으로 알려져 있는데, 다가의 양이온은 전기적 이중층(Double layer) 압축에 의한 불안정화를 향상시킬 수 있기 때문에다. 또한, 2가 금속염은 색도유발물질과 흡착하여 humate 또는 fulvate 등의 착화합물(complex)을 형성시켜 응집효율을 향상시킬 수 있다. 따라서 본 연구에서는 생물학적 2차 처리수의 화학적 응집처리에 있어서 알루미늄염 등의 다가이온 첨가가 응집에 미치는 영향을 관찰하고, 후속되는 플록형성 및 침전공정에 의한 제거효율을 비교, 평가함으로써 2차 처리수로부터 난분해성 유기물과 색도를 보다 효과적이고 경제적으로 제거할 수 있는 최적인자를 도출하고자 하였다.

The study of analysis of mutagen in drinking water (음용수 중 변이원성 물질(MX)에 관한 연구)

  • Disinfection by-products(DBPs), such as volatile trihalomethanes and the nonvolatile organochlorine acids, created by chlorination have been extensively studied. However MX which contributes 20-50% of the mutagenic activity in drinking water began to people's attention since 1990. Its chemical name is 3-chloro-4-dichloromethyl-5-hydroxy-2(5H)-furanone. According to WHO guidelines its concentration should be controlled, but its value has not been set up. Due to analytical difficulties in measuring this compound at such a low concentrations and lack of information on toxicity to human. Because concentration (ng/L) of MX in drinking water is low traditional testing methods are ineffective. Therefore this study compared LLE and SPE and have chosen SPE to improve preconcentration. MX has been identified in chlorinated drinking water samples in several countries but not in korea Therefore this study analyzed concentration of MX in different water sources and in spring water. This study examined the causes of changing MX content. Chlorine dosage, seasons, water temperature and distance from the source was all discoverd to be relavant. MX was analyzed in various treatment to find optimum disinfection methods. The outcome was that the concentration of MX was minimized when using biological activated carbon-O3 and granular activated carbon.

Bacteriocidal Effect of Calcium oxide (CaO, Scallop-shell Powder) on Natural Microflora and Pathogenic Bacteria in Sesame Leaf (Calcium oxide(CaO)를 활용한 깻잎 중 존재하는 자연균총 및 주요 식중독균의 제어)

  • Bacteriocidal effect of calcium oxide (CaO, scallop shell powder) on reduction of microorganism in sesame leaf was evaluated through comparison with chemical sanitizers such as chlorine, ethanol, and hydrogen peroxide. CaO showed 0.55-1.49, 0.85-2.56, 0.16-1.08, 0.30-1.14, and $0.19-1.07\;log_{10}$ CFU/g reductions of total aerobic bacteria, total coliforms, Escherichia coli, Listeria monocytogenes, and Salmonella typhimurium respectively, indicating possible use of CaO as effective natural sanitizer.

A STUDY ON DIMENSIONAL STABILITY OF THE RUBBER IMPRESSION MATERIALS FOLLOWING IMMERSION WITH CHEMICAL DISINFECTANTS (수종의 화학소독제에 침적시킨 고무인상체의 크기안정성에 관한 연구)

  • Dental impression materials often become contaminated with patients' saliva and blood which creates the potential for cross-infection. It was the purpose of this study to investigate the effects of disinfection of three different rubber impression materials with four different disinfecting solutions. Polysulfide, vinyl polysiloxane and polyether impression materials were mixed according to the manufacturer's directions and samples were formed on a stainless steel model. On removal from the standard model, impressions were immersed in a disinfectant (acid-potentiated glutaraldehyde, phenollic compound, chlorine compound, iodophor) at room tempera tures for ten minutes. After disinfection, the distance between reference points(linear dimension) was measured using the non-contact automatic cordinate measuring projector(MZ-1, Nikon). Through statistical analyses on the data from this study,. the following conclusions were obtained. 1. Polysulfide, vinyl polysiloxane impressions were disinfected without dimensional change.(p>0.05) 2. Polyether impressions which were immersed in acid-potentiated glutaraldehyde were statistically different from control group.(p<0.05) But the amount of shrinkage(0.04%) would not be clinically significant. 3. By immersion of polysulfide, vinyl polysiloxane, polyether impressions in Banicide, Biocide, Multicide plus, sodium hypochlorite for ten minutes, clinically accurate impressions were obtained without dimensional change.
