Purpose : The purpose of this study is to evaluate epidemiological data of pathogens obtained from stool exams and compare them with the clinical course in pediatric patients with symptoms of acute gastroenteritis. Methods : Subjects were selected from patients presenting with symptoms of acute gastroenteritis who visited the outpatient clinic or who were admitted to the Dankook University Hospital from December of 2004 to December of 2005. Stool exams for 17 pathogens was performed. RT-PCR was used to detect norovirus and enzyme-linked immunoabsorbant assay (ELISA) was used to detect rotavirus, adenovirus and astrovirus in the subjects stool samples. Ten different species of bacteria(Salmonella spp., Shigella spp., Clostridium perfrigens, Campylobacter spp., Escherichia coli, Vibrio spp., Staphylococcus aureus, Bacillus cereus, Yersinia spp., and L. monocytogenes) were each selectively cultivated and enzyme immunoassays(EIA) was used to test for antigens for C. parvum, E. histolytica and G. lamblia. Retrospective chart review was performed for comparisons of clinical manifestations. Results : A total of 215 subjects was selected and of these 89 cases(41.4%) showed positive results for at least one pathogen. Male to female ratio was 1.3:1. Age distribution showed 4 cases less than one month(4.5%), 4 cases from 1~2 months(4.5%), 24 cases from 3~12 months(26.7%), 47 cases form 13~48 months(52.8%), 10 cases greater than 48 months (21.2%). Viruses showed the greatest proportion of cases with 68 subjects(77.5%), of these rotavirus being the most commonly reported in 50 cases. Bacteria was identified in 26 cases (29.2%), of these nontyphoidal salmonella was noted in 10 cases. Protozoa followed with 21 cases(23.6%), of these C. parvum was noted in 11 cases and G. lamblia was noted in 10 cases. Mixed infections with more than two pathogens were seen in 22 cases(24.7%), of these viral infection with accompanying parasitic infection was seen in 12(54.5%) cases. Conclusion : In this study we examined various pathogens known to cause acute gastroenteritis in children. Further studies for various pathogens can provide useful information for management of the acute gastroenteritis.
Throughout the years 2008 to 2010, we analyzed approximately two thousand oriental melon samples collected from Seongju, using electron microscopy and testing by RT-PCR using primers specific for eight cucurbit-infecting viruses. Data from RT-PCR indicated that Cucumber green mottle mosaic virus (CGMMV), Watermelon mosaic virus 2 (WMV2) and Zucchini yellow mosaic virus (ZYMV) were present and the other viruses were not detected. Among them, CGMMV and WMV2 were the most prevalent pathogens. CGMMV was thought to infect oriental melon from the early growing season, and reached nearly 100% in the later of growing period. Otherwise, WMV2 emerged from June, several months later compared to CGMMV. CGMMV was detected from all aerial parts of the oriental melon including seeds, but not from the roots of the grafted pumpkin rootstock. Seed of two out of five commercial varieties were shown to be CGMMV positive. Nine varieties of pumpkins used as rootstocks were not infected with CGMMV. When the seedlings of grafted oriental melon were transplanted into pots mixed with the oriental melon debris infected with CGMMV, they were not infected by CGMMV. Cutting of pruning shear and the contact of tendrils contributed 48% and 30% to the transmission of the virus, respectively.
This study was performed to observe the role of Pneumocystis carinii as an etiologic agent of interstitial pneumonia in immunocompromised hosts. Total 90 male Sprague-Dawley rats, approxi. mately 150-180 g, were used. Fifteen of them were used as control group and remaining 75 (5 groups) were as immunosuppression groups; group 1 received prednisolone (25 mg/kg twice weekly) only; group 2 Prednisolone and tetracycline (75 mk/kg/day) ; group 3 Prednisolone, tetracycline and trimethoprim-sulfamethoxasole (50~250 mg/kg/day) : group 4 prednisolone and trimethoprim-sulfamethoxasole; and group 5 prednisolone and griseofulvin (300 mg/kg/day) until death. The survival days of each group rat were calculated, and upon death their lungs were removed immediately and then stamp smears were prepared and stained by Giemsa or toluidine blue O. For histopathologic observation, lungs were fixed in 10% formalin, cut into sections and stained with Gomori's methenamine silvei, hematoxylin-rosin, and Brovkn & Brenn stain. The results obtained were as follows: 1. The mean survival time of each group rat was 19.3$\pm$5.2 days (group 1), 41.1$\pm$14.0 days (group 2), 50.5$\pm$18.4 days (group 3), 43.0$\pm$22.9 days (group 4) or 21.8$\pm$5.1 days (group 5). Significant differences were noted between group 1 and group 2(p<0.01), group 1 and group 3 (p<0.01), and group 1 and group 4 (p<0.01), which represented bacterial infections were most fatal in immunocompromised rats. Group 5 revealed no difference in the survival day from group 1, while significant differences were noted between group 2 and group 5(P<0.01), group 3 and group 5(p<0.01), and group 4 and group 5(p<0, 01), which represented little importance of fungal infection as the cause of death of the rats. 2. The first fatality due to p. carinii pneumonia occurred 17 days after the beginning of the immunosuppression. The occurrence rate of P. carinii pneumonia in the decreasing order was 92.9% (group 3), 80.0% (group 2 and group 5), 78.6% (group 4) and 33.3% (group 1). With regard to the pathological stage of P. carinii pneumonia, the stage 1 was 11.3%, the stage 2, 28.3%, and the stage 3, 60.4%. 3. Viewing from the duration of immunosuppression, bacterial pneumonia chieay appeared in 1 month, mixed infections (P. carinii and bacteria, or p. carinii and fungi) in 1~2 months, and pure P. carinii pneumonia after 2 months. The present study revealed that P. carinii pneumonia was the most important cause of death of immunocompromised rats later than 1 month after the start of immunosuppression.

Purpose : This study was perfomed to analyze in detail the viral etiology of acute lower respiratory tract infections (ALRI) in Cheunan, Korea by multiplex RT-PCR, including human rhinovirus (hRV) and newly identified viruses such as human metapneumovirus (hMPV) and human coronavirus (HCoV-OC43, HCoV-229E/NL63). Method : Nasopharyngeal aspirates (NPA) were collected from 863 hospitalized children with ALRI on the first day of admission at Soonchunhyang University Cheonan Hospital and analyzed by multiplex RT-PCR from December 2005 to November 2006. Results : Viral agents were detected from 474 subjects (54.9%). The identified viral pathogens were hRV 9.2%, hMPV 6.8%, HCoV-229E/NL63 1.4%, and HCoV-OC43 2.1%. Coinfections with ${\geq}2$ viruses were observed in 108 patients (22.8%). The major period of viral ALRI was the first year of life. Clinical diagnoses of viral ALRI were pneumonia (59.5%), bronchiolitis (24.7%), tracheobronchitis (11.4%), and croup (4%). The most common causes of bronchiolitis was respiratory syncytial virus B (RSV B), whereas hMPV, hRV, HCoV-229E/NL63, and HCoV-OC43 were commonly found in patients with pneumonia. The number of hMPV infections peaked between March and May 2006. HCoV-OC43 was prevalent from November to February 2006, whereas HCoV-229E and hRV were detected throughout the year. Conclusion : Although the study was confined to one year, hMPV was not detected during winter and peaked between March and April, which was not consistent with previous studies'. This present study indicates that HCoV is a less common respiratory pathogen in cases of ALRI in Korean children
Purpose : This study was performed to investigate the epidemiologic and clinical features of 13 respiratory viruses in children with acute lower respiratory tract infections (ALRIs). Methods : Nasopharyngeal aspirates were prospectively obtained from 325 children aged 15 years or less from May 2008 to April 2009 and were tested for the presence of 13 respiratory viruses by multiplex real-time-polymerase chain reaction (RT-PCR). Results : Viruses were identified in 270 children (83.1%). Co-infections with ${\geq}2$ viruses were observed in 71 patients (26.3 %). Respiratory syncytial virus (RSV) was the most common virus detected (33.2%), followed by human rhinovirus (hRV) (19.1%), influenza virus (Flu A) (16.9%), human metapneumovirus (hMPV) (15.4%), parainfluenza viruses (PIVs) (8.3%), human bocavirus (hBoV) (8.0%), adenovirus (ADV) (5.8%), and human coronavirus (hCoV) (2.2%). Clinical diagnoses of viral ALRIs were bronchiolitis (37.5%), pneumonia (34.5%), asthma exacerbation (20.9%), and croup (7.1%). Clinical diagnoses of viral bronchiolitis and pneumonia were frequently demonstrated in patients who tested positive for RSV, hRV, hMPV, or Flu A. Flu A and hRV were most commonly identified in children older than 3 years and were the 2 leading causes of asthma exacerbation. hRV C was detected in 14 (4.3%) children, who were significantly older than those infected with hRV A ($mean{\pm}SD$, $4.1{\pm}3.5$ years vs. $1.7{\pm}2.3$ years; P =0.009). hBoV was usually detected in young children ($2.3{\pm}3.4$ years) with bronchiolitis and pneumonia. Conclusion : This study described the features of ALRI associated with 13 respiratory viruses in Korean children. Additional investigations are required to define the roles of newly identified viruses in children with ALRIs.
Oak mushroom (Lentinus edodes) is cultivated by using oak logs and sawdust medium. Green mold (Trichoderma) infection on these media severely suppresses the growth of oak mushroom. Usages of antibiotics and chemicals are not generally allowed to control of green mold since the mushroom is a fresh food. Tolaasin and its analogues, peptide toxins secreted by Pseudomonas tolaasii, have the antifungal activity and they have been successful to control the green mold disease. When the green mold, Trichoderma harzianum H1, was cultured in the presence of these toxins, the growth of fungus was effectively suppressed. In sawdust media, when the bacterial culture supernatants were sprayed on the aerial hyphae of green molds, the fungal growth was completely suppressed. Particularly, the antifungal activity was greatly increased by the combined culture extracts of P. tolaasii 6264 and HK11 strains. Therefore, these bacterial strains and their peptide toxins were able to suppress the growth of green molds and these can be good candidates to prevent from Trichoderma disease in oak mushroom cultivation.
Curing the studies on the mechanism of the nodule formation and the infection of rhizobia using the continously cultured plant cell tissues, it was found that some calluses possess high nitrogen fixation activity. This experiment was conducted to know the differences among the calluses and the Rhizobia. The results obtained were as follows; 1) In a single inoculation the nitrogenase activity of soybean cell cultures-rhizobium was moderately higher than non-leguminous cell cultures, however, in the mixing inoculation that was reversely found. 2) Host factor, which was characteristics for the nodule formation and the nitrogenase activity, was not appeared generally in the plant cell cultures except for Kuamusume and Toyozuzu in soybean, and Datura in non-leguminous cell cultures. 3) In the 012 rhizoblium cultured on soybean cell cultures and in the 010, 023 and 024 rhizobia cultured on non-leguminous cell cultures the nitrogenase activity higher than the others.
The present study was undertaken to estimate the antibacterial effect of a combination of C. rhizoma, L. Flos, and P japonica (1:1:1) extracts (CLP1000) and a combination of the herbal extract mixture and dioctahedral smectite (CLPS1000) against murine salmonellosis. At the concentration of CLP1000 and CLPS1000 0.5 mg/ml, the antibacterial effect was not showed on Salmonella typhimurium (S. typhimurium). On the other hand, the antibacterial effect against S. typhimurium was observed at the concentration of CLP1000 and CLPS1000 1.0 mg/ml. Oral administration of Smectite, CLPl000, and CLPS1000 at the dose of 10 mg/ml showed a therapeutic effect for S. typhimurium infected BALB/c mice. The mortality of Smectite, CLP1000 and CLPS1000-treated mice was 90%, 90%, and 70% at 12 days, respectively, while that of untreated mice was 100% at 9 days after a lethal dose of S. typhimurium infection. The results of our study strongly indicate that CLPS1000 has potential as an effective of salmonellosis.
Polymethyl methacrylate (PMMA) is concerned with promoting oral infection due to its low antibacterial activity. To overcome this, the nanoparticles of Ag-MSN, nGO, and CNP were mixed with MMA liquid in a weight ratio of 0, 0.25, 0.5, 1.0, 2.0% compared to resin powder using Orthocryl from Dentarum, a calibration resin, and then instructed by the manufacturer. Accordingly, a specimen for calibration was prepared by mixing PMMA: MMA (1.2: 1) ratio, and physical properties of the calibration resin, antifungal experiments, and statistical analysis were performed. As a result of antibacterial experiments, the antibacterial properties of Ag-MSN increased. In nGO, the antibacterial adhesive effect increased hydrophilicity, not a change in surface roughness. The higher the CNP concentration, the higher the antibacterial activity. This suggests its potential usefulness as an antibacterial dental material for orthodontic devices and temporary restorations.
Porcine respiratory disease complex (PRDC) is a common respiratory disease in nursery and grow-finishing pigs. A complex of viral and bacterial agents is known to be involved in the etiology of PRDC. The purpose of this study was to investigate common etiologic agents associated with PRDC in the field and compare detection methods for identifying these agents. To understand the mechanism of polymicrobial infection in PRDC, changes in the expression of cytokines were investigated. In 461 pig samples examined, most of the affected pigs ranged from 3 to 10 weeks old (73.4%), and 348 (75.4%) samples were confirmed as polymicrobial infection. Of the polymicrobial-infected cases, two (50.3%), three (32.2%), four (13.8%), five (3.2%), and six (0.5%) agents were detected. Two- or three-agent infections were the most common, with PRRSV/PCV-2 (44.6%) the most common two-agent infection. PRRSV/PCV-2/H. parasuis (11.0%) was the most common three-agent infection. Comparison of two detection methods (PCR and IHC) in the polymicrobial cases showed that 78.4% were PCV-2 positive with the PCR method, and 26.2% were PCV-2 positive with IHC. SIV was 7.8% by the PCR method and 3.7% positive by the IHC. This result indicates that the PCR method is more useful than IHC for detecting causative agents in PRDC. In the analysis of cytokines in the two- and three-agent infected samples, interleukin (IL)-$1{\alpha}$, IL-2, IL-4, IL-6, IL-10, and INF-${\alpha}$ showed the same expression pattern. All cytokines were suppressed, except IL-6. These findings indicate that changes in cytokine expression could be used to understand the mechanism of polymicrobial infection in PRDC.
