This study was conducted to reduce the using amount of chemical fungicides for the control of red-pepper Phytophthora blight. Effect of combination application of two microbial fungicides and two chemical fungicides for the control of red-pepper Phytophthora blight was examined in vitro, in greenhouse and under field conditions. Each microbial fungicides and chemical fungicides was two-fold diluted and mixed-soil drenched. In the greenhouse pot assay, the mixed application of B. pumilus QST2808 and a mixture of dimethomorph + ethaboxam (De) among four mixed applications of two microbial fungicides (B. pumilus QST2808, P. polymyxa AC-1) and two chemical fungicides showed the highest control effect against Phytophthora blight. Also, control effect of mixed application of B. pumilus QST2808 and De was similar to that of single application of De (dimethomorph + ethaboxam) or Mo (mancozeb + oxadixyl). In the field test, when the microbial fungicides (B. pumilus QST2808, P. polymyxa AC-1) and the chemical fungicide(De) for the control of Phytophthora blight of red pepper were mixed-soil drenched four times at 7~10 day-intervals, the control values were in the range of 78.8% to 82.0%. On the other hand when each of the two chemical fungicides (De, Mo) were soil drenched four times at 7~10 day-intervals, the control value were 65.7% to 85.8%. Consequently, the mixed application of the microbial fungicides and chemical fungicides could be recommended as a control method for reducing the using amount of chemical fungicides.
In order to investigate the effect of plant growth regulators on effective in vitro micropropagation, apical meristems of Pulsatilla cernua var. koreana were cultured on Murashige and Skoog's (MS) medium with 2,4-D, NAA, TDZ and BA. Media containing 2,4-D and kinetin, 2,4-D and TDZ, NAA and TDZ were not effective on callus induction. However, embryogenic or organogenic callus was obtained on media containing NAA and BA. Especially, on MS medium with 0.5mg/L NAA and 1.0mg/L BA was optimal for a high frequency (62%) of shoot or shoot bud obtained from callus. Callus proliferation, shoot multiplication and elongation were significantly increased by adding 10% coconut water on MS media with 0.5mg/L NAA and 1.0mg/L BA. Repeated subculturing of in vitro grown shoots resulted in propagation rate of 12.9 shoots per explant every 30 days. Root formation from the adventitious shoots was not easily achieved. However, roots were only produced through callus on MS medium with 2.0mg/L NAA alone or 0.5mg/L NAA and 1.0mg/L BA. These roots were used materials for callus and shoot production repetitively.
Apical meristems of Wasabia japonica were cultured on Murashige and Skoog's medium supplemented with cytokinins alone or together with 1.0 mg/L IAA. Shoot initials could be induced from leaf primordia on apical meristems. Calli and roots were formed on the medium containing cytokinins and 1.0 mg/L IAA in combination after 30 days of culture, but there were no callus proliferation. Shoot organogenesis began after 60 days of culture and these small shoots elongated when transferred to a medium containing 1.0 mg/L BA or kinetin. Shoots were formed directly without callus induction from apical meristems all the explants on the medium containing cytokinins variously, and most of the shoots proliferated multiple shoots which could be divided to obtain plantlets. Shoot multiplication rate in response to cytokinins was best on the medium containing 1.0 mg/L BA or 2.0 mg/L zeatin. Divided plantlets rooted well on MS medium containing 0.01 mg/L IBA after 15~30 days of subculture and the rooted plantlets developed into whole plants with multiple shoots. After rooting, the regenerated plants were washed and transferred to the pots containing sterilized soil.

These experiments were conducted to improve the effect of seed disinfection on rice seed severely infected Bakanae disease by seed soaking into mixed solution of prochloraz EC and fludioxonil FS. We investigated the effects of various concentrations of two fungicides mixed solution on spore germination and mycelial growth of Fusarium fujikuroi. Mycelial growth was inhibited 100% at $10{\mu}g$/ml of prochloraz and 33.3% at $80{\mu}g$/ml of fludioxonil. Spore germination was inhibited 81.4% at $40{\mu}g$/ml of prochloraz. Interestingly, mixed solution of $5{\mu}g$/ml or $10{\mu}g$/ml of each fungicide inhibitied 100% of mycelial growoth and 99.2% of spore germination, respectively. Severely infected rice seeds soaked into mixed solution composed of $125{\mu}l$/ml of prochloraz and $50{\mu}l$/ml of fludioxonil showed 2.1% of disease symptoms compared to 20.4% of prochloraz $125{\mu}l$/ml, but higher concentrations of prochloraz decreased the seedling stand rate. When the seed soaking time was longer and temperature was higher, control effect on Bakanae disease was improved, but seedling stand was lower about 80% over $35^{\circ}C$.
This experiment was conducted to find the effects of a $GA_3$ and thidiazuron (TDZ) on seedless rate, harvest time, fruit cracking and fruit quality in 'Kyoho' grapes over two years from 2008 to 2009. In 2008, fruit clusters were dip treated with $GA_3$$25.0mg{\cdot}L^{-1}$ twice at full bloom (FB) and 14 days after full bloom (DAFB) in a combination with TDZ 0 or $2.5mg{\cdot}L^{-1}$. Berry seedless rate and berry enlargement were slightly improved only when TDZ was added to the second $GA_3$ treatment at 14 DAFB, compared to $GA_3$ + TDZ treatments at both FB and 14 DAFB. However, berry cracking rate was significantly increased by any plant growth regulator (PGR) treatments compared to non treatment. In 2009, $GA_3$ at $12.5mg{\cdot}L^{-1}$ and $25.0mg{\cdot}L^{-1}$ was dip treated twice at FB and 14 DAFB while TDZ $2.5mg{\cdot}L^{-1}$ was treated only at 14 DAFB. Berry cracking rate was depended on the concentration of $GA_3$ applied. The higher concentration at $25.0mg{\cdot}L^{-1}$ significantly increased berry cracking rate while the lower concentration at $12.5mg{\cdot}L^{-1}$ had no effect. Also, the addition of TDZ to $GA_3$$25.0mg{\cdot}L^{-1}$ at 14 DAFB, substantially decreased the cracking rate to the level of untreated control. Although all PGR treatments advanced fruit maturity, the most significant advance occurred when TDZ was added to $GA_3$$12.5mg{\cdot}L^{-1}$ only at the second dip. Considering the overall aspects related to fruit maturity and quality, we concluded that the double applications of $12.5mg{\cdot}L^{-1}$$GA_3$ at FB and 14 DAFB with addition of $2.5mg{\cdot}L^{-1}$ TDZ only at 14 DAFB was appropriate to produce about 400-500 g size of seedless 'Kyoho' grape cluster having 35-40 berries.
This study was conducted to examine the effects of explant sources and plant growth regulators on mass propagation of Cyclamen persicum. Tuber, cotyledon, and petiole tissues were cultured on MS medium supplemented with different concentrations and combinations of auxins and cytokinins. Shoots were not induced from calli on cotyledon and petiole explants cultured on MS medium containing various concentrations of 2,4-D or NAA. However, multiple shoots were formed directly from tuber explants cultured on the medium containing 0.5 and 1.0 mg/L 2,4-D or NAA. In MS medium with cytokinin alone, TDZ was more effective in shoot formation than BA or kinetin in all explants. The combinations of NAA and BA was found to be most effective in shoot formation from tuber, cotyledon and petiole explants. Especially, shoots were formed from all the tuber explants on the medium containing 0.5 mg/L of NAA and BA. Hormonal effects on root formation were examined by subculturing single shoots on MS medium containing NAA or IBA. The medium with 0.5 mg/L IBA was most effective in root induction and subsequent plantlet regeneration.
This study was carried out to investigate control efficacy of fungicides mixed with pyroligneous acid on Large patch caused by Rhizoctonia solani AG2-2. R. solani AG2-2 was not inhibited by concentration of 500 times and 1,000 times diluents of pyroligneous acid. It is thought that R. solani AG2-2 was not inhibited by Pyroligneous acid alone. Pencycuron and tebuconazol mixed with pyroligneous acid had a good effect in prevention of R. solani AG2-2 in zoysiagrass. Concurrently, it was observed that those were reduced $50\%$ of usage of fungicides. 500 times and 1,000 times diluents of pyroligneous acid treated group had a significantly effect compared to control turf in the leaf length, leaf width and dry weight. In conclusion, usage of fungicides could be reduced about $50\%$ by a fungicides mixed with pyroligneous acid for protection of R. solani AG2-2. Therefore, It is thought that fungicides mixed with pyroligneous acids can be reduced an economic load and also an environmental pollution such as a tolerance to pathogenic bacteria and a destruction of an ecological system of soil microbes according to use fungicides.
This study was conducted to establish the tissue culture system for Korean domestic Miscanthus sinensis, which is used in various purposes such as forage, and bio-energy resources. With the mature seed of Miscanthus, optimum concentrations of plant growth regulators were identified for an efficient callus induction and regeneration. Among the treatments of 1~10 $mg{\cdot}L^{-1}$ 2,4-D, IBA, or NAA, callus induction rate was highest (85.3%) on MS medium containing 5 $mg{\cdot}L^{-1}$ 2,4-D. Under the condition, the callus were efficiently induced and proliferated with comparably lower frequencies of callus browning. In shoot regeneration, the treatment of NAA combined with BAP seemed to contribute more efficient conditions to shoot regeneration than those of NAA with Kinetin or 2-iP. Especially, regeneration efficiency and number of regenerated plants were 83.7% and 5.5 in 3 $mg{\cdot}L^{-1}$ NAA with 5 $mg{\cdot}L^{-1}$ BAP, respectively, which were higher frequencies than those in NAA with Kinetin or 2-iP. In results, 5 $mg{\cdot}L^{-1}$ 2,4-D and 3 $mg{\cdot}L^{-1}$ NAA combined with 5 $mg{\cdot}L^{-1}$ BAP were efficient for embryogenic callus induction and regeneration of Miscanthus. This system would be useful for mass-propagation and developing new cultivars via tissue culture of Miscanthus sinensis.

벼룩이울타리(Arenaria juncea M.Bieb.)는 청열, 양혈, 허로기열, 골증, 수심작열, 도한 등의 약효가 있으며, 백색의 꽃이 취산화서로 아름답게 개화하여 조경소재로 이용가치가 높다. 본 연구는 벼룩이울타리의 육묘기술 개발을 위하여 2017년 6월 2일부터 7월 31일까지 8주간 수행하였다. 실험의 대조구는 200구 트레이에 원예상토를 충진하고 종자를 셀 당 3립씩 파종하여 무차광 조건에서 육묘하였으며, hyponex $500mg{\cdot}L^{-1}$를 2회 추비하였다. 토양 실험은 원예상토[$300-200-200mg{\cdot}L^{-1}$(N-P-K)]와 피트모스:펄라이트(3:1), 코코피트:펄라이트(3:1) 혼용토를 이용하였으며, 원예상토를 제외한 모든 토양은 0-0-0, 300-200-200, $600-400-400mg{\cdot}L^{-1}$의 비료를 첨가하였다. 종자는 셀당 1, 3, 5립을 파종하였고, 추비는 hyponex (6.5-6-19)를 0, 500, $1000mg{\cdot}L^{-1}$로 4, 6주차에 총 2회 잎이 충분히 젖을 정도로 살포하였다. 차광은 0, 55, 75%로 설정되었다. 벼룩이울타리 육묘의 결과, 토양 종류별로는 원예상토에서 초장이 가장 길었으며, 다음으로 피트모스 혼용토의 순이었다. 피트모스 혼용토에서 육묘한 유묘의 초장은 비료를 첨가하지 않은 처리구에서 길어지는 경향을 보였다. 경직경과 근장은 원예상토 단용구와 코코피트 혼용토가 피트모스 혼용토에 비해 양호하였다. 엽수는 상토(26.1 ea)에서 가장 많았고 다음으로 비료 코코피트 혼용토 $600-400-400mg{\cdot}L^{-1}$(22.9 ea)의 순이었다. 셀 당 파종립수를 달리하여 육묘한 결과, 초장은 파종립수가 적을수록 길었으며, 경직경과 엽수는 처리에 따른 유의적인 차이를 확인할 수 없었다. 추비 처리별로 전반적인 생육은 큰 차이는 보이지 않았으나, 경직경은 처리농도가 낮을수록 두꺼워지는 경향이었다. 엽수는 hyponex 처리시 무처리구에 비해 증가하는 경향을 보였으며, $500mg{\cdot}L^{-1}$ 처리구에서 가장 많았다. 차광정도에 처리에 따른 생육은 차광률이 높을수록 생육이 억제되는 경향이었다. 초장, 엽수 및 근장은 무차광에서 차광처리구에 비해 우수하였다. 경직경은 무차광구와 50% 차광구가 70% 차광구에 비해 증가하였다. 지상부, 지하부의 생 건체중도 무차광에서 가장 무거웠으며, 차광률이 증가할수록 감소하였다.
The most effective cytokinin for shoot multiplication in vitro of Prunus persica cv. Baekmijosaeng, Okubo, and Yumyeong was 2.0 mg/L BA. As the result of combinational treatment of BA and auxin sources (IAA, IBA and NAA), 2.0 mg/L BA with 1.0 mg/L IAA was the most effective for shoot multiplication of cv. Baekmijosaeng. The most effective auxin source for rooting was IAA and the concentration was 5.0 mg/L and 3.0 mg/L for cv. Baekmijosaeng and Okubo, respectively.
