Characterization of a Hydrogen evolving strain of Rhodopseudomonas sphaeroides (수소생성 Rhodopseudomonas sphaeroides의 특성)

  • Many photosynthetic bacteria capable of hydrogen production were isolated from samples of mud flats of paddy field collected in Kim Hae and Dae Jeo. A strain 230 was selected for the high capability of hydrogen evolution. As the results of examination in physiological, morghological and cultural characteristics, the strain 230 was identified as Rhodopseudomonas sphaeroides.

Effect of Microbes and Formic acid on Storage of Spent Mushroom (Flammulina velutipes) Substrates (미생물과 개미산 첨가가 팽이버섯 수확후배지의 저장성에 미치는 영향)

  • This study was conducted to suggest effective storage methods for the use spent mushroom (Flammulina velutipes) substrates (SMS) as animal feed. SMS was storage by deep stacking, composting and vinyl mulching after treated with 1% microbes and 0.5% formic acid. During the storage periods, all of the treatment was covered with hyphae of mushroom. The external change of SMS was shown that the storage of SMS can be improved by anaerobic condition and 1% microbial treatment. The comparatively high temperature, pH and total bacterial number was shown in the SMS treated without microbes and formic acid by composting.

  • This study investigates the effect of denitrified biofloc water on changes in the water quality parameters and the physiological characteristics of shrimps. Biofloc rearing water contains a large number of microorganisms and can rapidly stabilize the water quality and energy saving if reusable due to high water temperatures. Rearing water contain floating bacteria with both aerobic and anaerobic bacteria. Therefore, when the carbon source is added in limited air supply, the anaerobic state is activated and the denitrification process is possible. In this study, the denitrification water had the following properties: ammonia (6.9 mg L-1), nitrite (0.3 mg L-1), nitrate concentration (9.2 mg L-1), high pH (8.42) and alkalinity (590 mg L-1). The experimental group consisted of seawater (SW, control), a mixture of Seawater and denitrified biofloc water (DNW) in the ratio of 3:1, 1:1 and DNW only. All experiments were done in triplicate. As a result, the survival rate never changed even when 100% of the denitrification water was utilized. However, a body fluid analysis showed that creatine and BUN were increased due to index of stress and the tissue damage resulting from the high denitrified water content. Body fluid ions (Na+, K+, and Cl-) significantly decreased as the denitrified water content increased. It was recommended that the denitrification water be mixed with a certain ratio (less than 50%) in the future as it may affect the osmotic pressure control in shrimps.

  • The traditional ripening method of clay was analyzed. An advanced refining method of clay using enrichment cultures of iron r reducing bacteria was developed. After the traditional ripening, the whiteness of the clay was increased due to removal of | iron impurities by inhabitant dissilmaltien with iron reducing bacteria. Other characteristics of the refined clay such as v viscosity, plasticity, and strength were also improved by iron reducing bacteria. An advanced method of clay refinement with a anaerobic enrichment cultivation of iron reducing bacteria supplemented with an extra carbon source such as glucose was s suggested. When the clay was treated by the advanced method. the refinement time could be reduced to 1/6 of that r required by the traditional method. The physical properties of the refined clay by the advanced method were better than t those of the traditionally refined clay.

  • Parvimonas micra is a non-spore-forming anaerobic gram-positive coccus and a known commensal of the skin, gums, vagina, and gastrointestinal tract. It is rarely associated with severe infections, which typically follow invasive procedures such as dental treatment. We describe a case of a brain abscess caused by P. micra in an immunocompetent 11-year-old boy without periodontal disease. He presented with a 7-day history of headaches and vomiting, and complained of diplopia that began on the day of presentation. He did not have any recent dental treatment or specific past medical history. A brain abscess in the left frontoparietal lobe was noted on brain magnetic resonance imaging. P. micra was cultured from brain abscess aspirate. He was successfully treated with surgical drainage and combined antibiotic therapy with ceftriaxone and metronidazole for 6 weeks.

  • Prevotella intermedia is a Gram-negative, obligately anaerobic, nonsporeforming, and nonmotile rod. P. intermedia is associated with periodontitis, pregnancy gingivitis, acute necrotic ulcerative gingivitis, endodontic infection, and rheumatoid arthritis. P. intermedia KCOM 1107 (= ChDC KB29) was isolated from a human subgingival dental plaque of gingivitis lesion. Here, we present the draft genome sequence of P. intermedia KCOM 1107.

Change of Chemical and Microbial Properties during Fermentation of Cotton Waste for Oyster Mushroom Cultivation (느타리 재배용 폐면 발효 중의 화학성 및 미생물 상의 변화)

  • The changes of microflora and chemical characteristics during fermentation process of cotton waste for oyster mushroom cultivation were investigated with 5 l bench-scale reactors placed in an incubator at different temperatures ($40,\;50\;and\;60^{\circ}C$). Cotton waste was wetted to 70% moisture, and air flow rates to the substrate were 50, 100 and 300 cc/min. In processing of composting, the mesophilic bacterial population decreased sharply but thermophilic bacterial population increased. In case of fungi, both mesophilic and thermophilic population decreased. The daily $CO_2$ evolution showed little difference in all treatments, while $NH_3$ dropped sharply after 3 days. The desirable composting temperature and air flow based on the mycelial growth of oyster mushroom were $50^{\circ}C$ and 100 cc/min, respectively.

  • This study investigated the diversity of intestinal microbial communities isolated from the intestine of topshell (Turbo cornutus) from the coast of Jeju Island (Beobhwan, Seogwipo city). Pure cultivation using the standard marine agar (MA) medium showed the most significant number of clusters. Aerobic and anaerobic culture allowed isolation of strains of 1.8×105 CFU·g-1 and 0.4×10 CFU·g-1 on average, respectively. The microbial population in the topshell intestine was classified into 4 phyla, 12 families, 26 genera, and 67 species. The microbes in the topshell intestine were detected by homology with 93~100% with standard strains. The microbes in the topshell intestine consisted of Proteobacteria 39%, Firmicutes 34%, Actinobacteria 21%, and Bacteroidets 6%. The identified families were Alteromonadaceae (1), Shewanellaceae (4), Vibrionaceae (12), Phyllobacteriaeceae (1), Rhodobacteraceae (8), Bacillaceae (21), Paenibacillaceae (2), Cellulomonadaceae (1), Mycobacteriaceae (6), Nocardiaceae (4), Streptomycetaceae (3) and Flavobacteriaceae (4). Bacillus sp. and Vibrio sp. accounted for the greatest portion of the separated strains. Among the isolated microorganisms, some strains had probiotic functions.

Antagonistic Action of Lactobacilli Toward Pathogenic Bacteria in Associative Cultures (Lactobacillus spp.에 의한 병원성 세균의 생육저해)

  • Three species of lactobacilli (L. casei, L. acidophilus, L. bulgaricus) were tested for their antibacterial activity. They all were antagonistic to growth of enteropathogenic Escherichia coli and Salmonella enteritidis in associative cultures in YS-medium (0.1 % yeast extract + skimmilk). Sal. enteritidis was more sensitive to the inhibition than was E. coli. Control cultures of E. coli and Sal. enteritidis were pH 5.08 and 5.70 in 72 hrs of incubation and the associative cultures were pH 3.35-4.48. The increases in pH resulting from growth of the lactobacilli in the associative cultures appeared to be sufficient and mainly responsible for the antagonistic actions exerted on the pathogens.

Isolation and characterization of an Enterococcus faecalis bacteriophage (Enterococcus faecalis 특이적 박테리오파지의 분리와 특성규명)

  • Enterococcus faecalis is a Gram-positive and facultative anaerobic bacterium that causes many hospital-acquired infections. Novel E. faecalis specific bacteriophage (phage) ECP3 that had been isolated from thirty-four environmental samples and characterized phenotypically and genotypically. ECP3 phage belongs to the family Myoviridae with contractile tail and lysed E. faecalis specifically but other bacteria including Enterococcus faecium. The genome was double-stranded linear DNA and its size was 145,518 bp comprising of 220 open reading frames. The GC content was 35.9%. The genome sequence showed 97% identity in 90% coverage region with Myoviridae phage PhiEF24C. ECP3 is the first E. faecalis-specific Myoviridae phage isolated in Korea which can be a promising antimicrobial agent against E. faecalis infections.
