Studies on classification of Korean catfish (Silurus asotus) peripheral leucocytes (한국산 메기 (Silurus asotus)의 말초혈액 백혈구의 분류)

  • Classification of peripheral leucocytes in Korean catfish (Silurus asotus) were performed. The leucocytes were classified on the basis of their morphological, cytochemieal and phagocytic characteristics. Four types of leucocytes were observed in the peripheral blood. Neutrophils were round and 12 - $15{\mu}m$ in diameter with round, oval or 2 - 3 segmented neuclei. Their cytoplam was stained whitish or grey. Lymphocytes were round and 5.5 - $8{\mu}m$ in diameter with large round neuclei which occupied most of the cells. Monocytes were round or irregular cells and 12 - $15{\mu}m$ in diameter with bean - shaped or irreular neuclei. Thrombocytes were oval or spindle - shaped cells with oval or spindle - neucle. The cells with greyish cytoplasm ranged 7.2 to $14.4{\mu}m$ in length and 5.8 to $7.2{\mu}m$ in width. Only neutrophils showed positive reaction in peroxidase and PAS. Neutrophils and monocytes showed active phagocytosis. As the results it is concluded that peripheral leucocytes of Koran catfish could be classified as lymphocytes, neutrophils, monocytes and thrombocytes.

Effects of Dietary Germanium Biotite on Growth Performance and Blood Characteristics in Broiler Chicks (육계에 있어 게르마늄 흑운모의 첨가가 성장 및 혈액성상에 미치는 영향)

  • This study was conducted to investigate the effect of germanium biotite on the growth performance, blood composition , fecal NH$_3$-N and volatile fatty acid(VFA) in broiler chicks. Two hundred forty broiler chicks were randomly allocated into five treatments with four replicates for five weeks. Dietary treatments included 1) Control, 2) GB1(basal diet + 200 mesh 0.5%), 3) GB2(basal diet + 325 mesh 0.5%), 4) GB3(basal diet + 200 mesh 1.0%) and 5) GB4(basal diet +325 nesh 1.0%) For overall period, weight gain, feed intake and feed conversion were not significantly different among treatments (P>0.05). Digestibilities of nutrients were not affected by adding GB(P>0.05). No differences occurred in red blood cell(RBC), white blood cell(WBC), hematocrit(HCT), hemoglobin(Hb), lymphocyte, monocyte and platelet of blood among the treatments (P>0.05). Fecal NH$_3$-N concentration of chicks fed GB1.0 diets(GB3 and GB4) was lower(P<0.03) than chicks fed GB0.5 diets(GB1 and GB2). Propionic acid and butyric acid in feces of chicks fed GB diets were lower(P<0.04) than in chicks fed control diet. GB diets reduced fecal acetic acid significantly compared to control (P<0.01). Also, acetic acid of feces in chicks fed GB1.0 diets(GB3 and GB4) was lower(P<0.02) than GB 0.5 diets(GB1 and GB2) treatments. In conclusion, dietary germanium biotite was an effective means of decreasing fecal NH$_3$-N and volatile fatty acid(VFA) emission.

Growth and Blood Characteristics of Red Sea Bream Pagrus major by Starvation and Stocking Density during Red Tide (적조발생시기 참돔의 절식과 사육밀도에 따른 성장과 혈액성상)

  • In order to minimize the damage on the red sea bream Pagrus major by a harmful dinoflagellate Cochlodinium polykrikoides, we investigated the effect of feeding, starvation and stocking density on the survival rate, growth, growth restoration and physiological response of P. major exposure to C. polykrikoides. The experimental groups were divided into three groups such as F-HD (feeding and high density with $6.4kg/m^3$), S-HD (starvation and high density with $6.4kg/m^3$) and S-LD (starvation and low density with $3.2kg/m^3$) according to stocking density and starvation in marine cage ($11m{\times}11m{\times}5m$). The F-HD was fed throughout the experiment for 9 weeks, whereas S-HD and S-LD were not fed for 5 weeks and then refeeding for 4 weeks. Survival rate was the lowest in F-HD (85.5%) and S-LD was the highest (97.3%). The growth rates of S-HD and S-LD were significantly lower than F-HD during starvation period for 4 weeks, but rapidly recovered after feeding. The nutritional status such as ALB, TP, TCH, TG were similar to tendency of growth data. Ht, Hb, AST, ALT and GLU levels were significantly higher in the F-HD than in the starvation groups at the same time (in 3 week) during starvation period. But starvation groups did not differ during starvation period. As a result, F-HD is more sensitive to stress than S-HD and S-LD. Thus, during C. polykrikoides bloom period, starvation and stocking density control can help survival and growth restoration of the red sea bream.

The Effects of Fig Fermented Product Supplementation on Animal Performance, Serum Profile and Meat Quality in Hanwoo Bulls (무화과 발효물의 급여가 한우의 생산성, 혈액성상 및 육질에 미치는 영향)

  • We investigated the effect of fig fermented product(FFP) supplementation on growth performance, serum profile, carcass performance, meat performance and meat quality in 10 bulls of Korean cattle. Concentrates diet was supplemented with substrate fermented from fig fruit and leaves at 10% of the diet. The feed intake of FFP were slightly higher than the control, but the final weight showed no sifnificant difference between the two. Daily weight gain and feed intake were increased in FFP. The serum profile had no significant difference in the treatment. In carcass performance, the meat quantity grade of the treatment had no significant difference, but in meat quality grade the marbling score of FFP was significantly(P<0.05) increased therefore it showed a positive effect on meat quality grade. Also there was no significant(P<0.05) difference of meat cut performance in the treatment. Due to the proximate characteristics of longissimuss muscles the crude fat content of the FFP was significantly(P<0.05) increased. There was no significant difference(P<0.05) in physical characteristics ; pH level, meat color and heat loss of the treatment, but the shear force value and the cholesterol content of FFP significantly(P<0.05) decreased. Crude fat was increased(P<0.05) and cooking loss, shear force and cholesterol concentration were decreased. In fatty acids composition of the FFP, the linoleic acid from the longissimus increased significantly(P<0.05). In subcutaneous fat of longissimus of the FFP, C16:0(palmitic acid) significantly(P<0.05) decreased, but C18:1 significantly (P<0.05) increased. Therefore in FFP, the concentration of saturated fatty acid significantly decreased (P<0.05), but on the other hand the concentration of unsaturated fatty acids significantly (P<0.05) increased. In sensory evaluation of the FFP, the evaluation of odor increased slightly in a positive manner, also the appearance and the taste increased significantly(P<0.05). In conclusion when annexing additional fig fermented product to Hanwoo bulls, the carcass grade improves and the livestock production increases. Also the shear force, lower cholesterol, improved appearance and taste will open the doors to high quality meat production.

Effects of Dietary Zinc Supplements on Growth, Feed Efficiency, Organ Weight, Blood Biochemical Profiles, and Activity of Digestive Enzymes in Growing Korean Native Chicks (아연 보충급여에 따른 한국재래계의 성장, 사료이용성, 장기무게, 혈액생화학적 성상, 장기무게 및 소화효소 활성도에 미치는 영향)

  • The aim of the study was to investigate the effects of dietary supplementation of zinc (Zn) sources (zinc oxide and Zn-methionine) on performance, organ weights, blood biochemical profiles, and digestive enzymes of the pancreas and small intestine in Korean native chicks (KNC). A total of 144 KNC (n=6) were fed a basal diet (CON, 100 ppm of Zn), a basal diet supplemented with 50 ppm of Zn with ZnO (ZNO), or a basal diet supplemented with 50 ppm of Zn with Zn-methionine (ZMT) for 28 days. There was no significant difference in body weight, gain, feed intake, and feed conversion ratio among the three groups. The relative weights of the liver, spleen, and intestinal mucosa were unaffected by the dietary source of Zn, whereas pancreas weight in the ZNO group decreased (P<0.05) compared with that in the CON and ZMT groups. Blood biochemical components including aspartate aminotransferase, and alanine aminotransferase were unaffected by dietary Zn supplementation. Pancreatic trypsin activity in the ZNO and ZMT groups was significantly (P<0.05) enhanced compared with that in the CON group. However, the activities of ${\alpha}$-amylase and carboxypeptidase A were not altered by dietary Zn supplementation. The activities of maltase and sucrase were unchanged, whereas the activity of leucine aminopeptidase tended (P=0.08) to be increased by dietary Zn supplementation. In conclusion, the supplementation with 50 ppm of ZnO or Zn-methionine resulted in an activation of protein digestive enzymes in the pancreas and small intestine without affecting animal performance in KNC.

Effect of Water Extract of Green tea, Persimmon Leaf and Safflower Seed on Heme Synthesis and Erythrocyte Antioxidant Enzyme Activities in Lead-Administered Rats (납투여한 흰쥐의 헴합성과 적혈구 중의 항산화효소 활성에 미치는 녹차, 감잎, 홍화 열수추출물의 영향)

  • This study was performed to investigate the effect of water extract of green tea (GT), persimmon leaf (PL) and safflower seed (SS) on heme synthesis and erythrocyte antioxidant enzyme activities in lead (Pb)-administered rats. Male rats were divided into five groups. a normal, Pb-control (Pb-Con), Pb-GT, Pb-PL and Pb-55 groups with ten rats per group. Pb (25 mg/kg. BW) was orally administerd once a day for 4 weeks. The extract of GT, PL and 55 were administered based on 1.26 g of raw traditional tea/kg BW/day. Blood hematocrit, homoglobin level and red blood cell counts were significantly lower in rb-Con group than in normal group. However, the supplementation of GT, PL and 55 were effective to improve the hematological parameters. Plasma AST and ALT activities were significantly lower in Pb-GT, Pb-PL, Pb-SS groups than in Pb-Con group. The $\delta$ -amino-levulinic acid dehydratase (ALAD) activity of blood and liver were significantly lowered in Pb-Con group com-pared to those of the normal group. The ALAD activity in Pb administered rats was recovered to tile normal level by the water extract of GT, PL and 55 supplementation. Erythrocyte superoxide dismutase and catalse activities were significantly higher in Pb-Con group than in normal group, whereas glutathione peroxidase activity was lowered in Pb administered rats. The extract of GT, PL and SS supplement attenuated changes of these erythrocyte antioxidant enzyme activities by Pb intoxication.

Effects of Different Levels of Oxytetracycline on Physiological and Bio- chemical Responses in Olive Flounder, Paralichthys olivaceus (넙치, Paralichthys olivaceus에서의 Oxytetracyline 처리 농도가 생리ㆍ생화학적 반응에 미치는 영향)

  • Olive flounder was treated with oxytetracycline (OTC) and changes in blood physiology, antioxidant enzymes and heat shock protein (HSP) were recorded to obtain preliminary data for optimal OTC treatment. Blood parameters were measured 1 and 3 h after the OTC treatments at the concentration of 0 (control), 100, 300 and 500 ppm for I h. Hematocrit decreased with time, however the difference was not significant (P>0.05). Reduced number of red blood cell was observed with increasing OTC concentration. Serum glucose level increased as the OTC concentration increased. However, glucose level was similar to control after 3 h. Blood total protein decreased immediately after the OTC treatment but increased after 1 and 3 h. However, the increment in blood total protein was low. Activities of superoxide dismutase enzymes in 300 and 500 ppm groups increased by the OTC concentration. Catalase enzyme activity was negatively affected by the OTC concentration. However, the differences were not significant (P>0.05). High expression of HSP-70 protein was recorded for groups treated with 100 and 500 ppm compared to that of the control group. However HSP-70 mRNA showed a lower increment which was not significant (P>0.05).

Influence of Dietary δ-Aminolevulinic Acid Supplement on Growth Performance and Hematological Changes in Weaned Pigs (이유자돈에 있어 δ-아미노레불린산의 식이 내 첨가가 성장 밑 혈액학적 변화에 미치는 영향)

  • This study was. conducted to investigate the effects of $\delta$-aminolevulinic acid supplementation on growth. performance, nutrient digestibility and hematological changes in weaned pigs. Seventy five Duroc${\times}$Yorkshire${\times}$ Landrace pigs (7.21$\pm$0.02 kg average BW) were used in a 20 d growth assay. Dietary treatments included 1) NC (negative control; basal diet), 2) PC (positive control; NC diet + 0.1% Apramycin + 0.1% Oxytetracycline), 3) ALA0.l (NC diet+0.l% $\delta$-aminolevulinic acid), 4) ALA0.2 (NC diet+0.2% $\delta$-aminolevulinic acid) and 5) ALA + AB (PC diet+0.2% $\delta$-aminolevulinic acid). In entire experimental period, ADG (Average Daily Gain) was greater in pigs fed ALA + AB diet than in pigs fed NC diet (p<0.05). However, there was no significant difference in ADG for pigs fed PC, ALA0.l or ALA0.2 diets. Also, ADFI (Average Daily Feed Intake) and G/F (Gain/Feed) were not affected by treatments. For 20 d, fecal digestibilities of DM (Dry Matter) and N (Nitrogen) were higher (p<0.05) for pigs fed ALA + AB diet than for pigs fed NC and ALA0.l diets. Total protein concentration of serum was increased in ALA + AB treatment compared to NC and PC treatments (p<0.05). In iron concentration of serum, pigs fed $\delta$-aminolevulinic acid supplementation diets were greater than for pigs fed NC and PC diets (p<0.05). TIBC (Total Iron Binding Capacity) concentration of serum was increased in ALA + AB treatment compared to NC, PC and ALA0.l treatments (p<0.05). Pigs fed ALA + AB and ALA0.2 diets were higher hemoglobin (Hb) and hematocrit (HCT) concentration of blood than pigs fed NC and PC diets (p<0.05). RBC and WEC concentrations of blood were significantly higher (p<0.05) for the pigs fed ALA0.2 and ALA + AB diets than for pigs fed NC diet. Lymphocyte concentration of blood was increased in $\delta$-aminolevulinic acid supplementation treatment compared to NC treatment (p<0.05). In conclusion, $\delta$-aminolevulinic acid supplementation for weanling pigs had improved growth performance and nutrient digestibilities and had increased total protein, iron, hemoglobin and lymphocyte concentration of blood. Also, feeding for dietary $\delta$-aminolevulinic acid and antibiotic mixture supplementation may have more higher effective growth performance in weanling pigs.

Expression of Several Biologic Markers as Prognostic Markers in Non-Small Cell Lung Cancers (폐암조직에서 생물학적 지표들의 예후인자로서의 비교검토)

  • Background: Despite modern diagnostic, staging, and therapeutic advances, esp. with molecular biologic techniques, the 5-year survival rate of all cases of lung cancer does not exceed 15%. Also, the incidence of lung cancer of both sex in Korea is increasing year by year and the lung cancer is one of the leading causes of cancer death. Therefore, it is strongly needed to develop the new combination of treatment modalities including neoadjuvant chemotherapy and to identify tumor specific characteristics with staging or prognostic markers. Here we present the clinical significance of several biologic tumor markers to use as a prognostic markers in patients with non-small cell lung cancers. Method: The survival has correlated with the expressibility of proliferative cell nuclear antigen (PCNA), epidermal growth factor receptor(EGFR), p53 and/or blood group antigen A(BGAA) using immunohistochemistry in 46 patients with non-small cell lung cancers. Results: 1) The expression rates of PCNA, EGFR, p53 and BGAA were 80.6%, 61.3%, 45.9% and 64.3%, respectively and those were not correlated to cell types or clinical stges. 2) The expression of BGAA was correlated with better survival in median survival and in 2-year survival rate and that of PCNA was correlated with worse survival in median survival and 2-year survival rate. 3) The expression of EGFR or p53 was not valuable to predict prognosis in non-small cell lung cancers. 4) With simultaneous applications of PCNA, EGFR and p53 immunostain, the patients with 2 or more negative expressions showed better prognosis than the patients with 2 or more positive expressions. Conclusion: It is suggested that the expression of blood group antigen may be a positive prognostic factor and that of PCNA may be a negative prognostic factor. Also, the combination of expressions of PCNA, EGFR and p53 may be used as a negative prognostic factor.

Effect of Gluathione on the Histopathological Changes Caused by Oxidized Diets in the Carp, Cyprinus carpio (산화(酸化)된 사료(飼料)에 의(依)한 잉어의 병리조직학적(病理組織學的) 증상(症狀)과 glutathione 첨가효과(添加效果))

  • Nutritional ceroidosis accompanied with histopathological changes such as myopathy and visceral ceroidosis, was experimentally induced by feeding the oxidized feed with the peroxide value of 98meq/kg, to common carp resulting in some motalities of carp. To see the effect of treacetment survivors of the above fish were orally administered by feeding diets containing daily doses of 3mg and 6mg of glutathione per kg fish for 10 days or 20 days. While control and 3mg dosage group retained viscerral ceroidosis and showed evidence myopathy histo pathologically, the fish treated with 6mg of glutathione for 20days showed obvious reduction of macrophages of ceroid in the viscera.
